The study investigated the effect of thermo-alkaline pretreatment on the solubilization of polyhydroxybutyrate (PHB) and its potential to enhance of thermophilic anaerobic digestion, focusing on biochemical methane potential (BMP) and methane production rate, using two different particle sizes of PHB (1500 ㎛ and 400 ㎛). Thermo-alkaline pretreatment tests were conducted at 90 ℃ for 24 hours with varying NaOH dosages from 0-80% (w/w). BMP tests with untreated PHB exhibited methane production ranging from 150.4~225.4 mL CH4/g COD and 21.5~24.2 mL CH4/g VSS/d, indicating higher methane production for smaller particle sizes of PHB, 400 ㎛. Thermo-alkaline pretreatment tests achieved a 95.3% PHB solubilization efficiency when 400 ㎛ PHB particles were treated with 80% NaOH dosage at 90 ℃ for 24 hours. BMP tests with pretreated PHB showed substantial improvement in thermophilic anaerobic digestion, with an increase of up to 112% in BMP and up to 168% in methane production rate. The results suggest that a combined pretreatment process, including physical (400 ㎛ PHB particles) and thermo-alkaline (90 ℃, 40-80% NaOH dosage, and 24 hours reaction time), is required for high-rate thermophilic anaerobic digestion of PHB with enhanced methane production.
Luteolin is a flavonoid that exists in a glycosylated form in celery and green pepper. Flavonoids possess antioxidant and anti-inflammatory properties and can reduce the expression of key inflammatory molecules in macrophages and monocytes. It has been reported also that some flavonoids have effects on bone metabolism. The effects of luteolin on the function of osteoblasts were investigated by measuring cell viability, alkaline phosphatase activity, type I collagen production, osteoprotegerin secretion, Wnt promoter activity, BMP-2 and Runx2 expression and calcified nodule formation. Luteolin has no effects upon osteoblast viability but induced an increase in alkaline phosphatase activity, type I collagen production and a decrease in osteoprotegerin secretion in these cells. Luteolin treatment also upregulated BMP-2 mRNA expression. These results suggest that luteolin may be a regulatory molecule that facilitates the differentiation of osteoblasts.
1년령의 중성화된 수컷 요크셔테리어와 7년령의 암컷 푸들이 골절의 지연유합으로 내원하였다. 요크셔테리어의 경우 오른쪽 요골과 척골의 원위부가 골절되었고, 푸들의 경우 왼쪽 경골과 비골의 원위부가 골절되었다. 두 환자에서 신체검사 및 방사선검사를 통해 골절의 유합이 지연되고 있음을 확인하였다. 골절부는 금속판 및 나사못으로 고정시키고 자가해면질골을 골절 틈새에 이식하였다. 또한 빠른 골유합을 위해 $20{\mu}g$의 rh-BMP 2 가 포함된 matrigel을 골절부에 주입하였다. 수술 후 방사선검사를 통해 골절부의 치유 상태를 확인하였다. 장기간의 방사선검사에서 두 마리 모두 빠른 골유합을 보였다. 첫 증례에서는 방사선사진상에서 2주째에 요골과 척골 골절 틈새에 골밀도가 증가하였다. 20주째에는 골절선이 더 이상 관찰되지 않았으며, 정상 외관을 회복하였다. 두 번째 증례에서는 방사선사진상에서 11주째에 골절 치유가 진행되고 있음이 확인되었다. 이 두 증례 모두 예상한 것보다 골절의 치유가 빠르게 일어났다. 따라서 rh-BMP 2와 matrigel은 지연유합된 골절의 치유를 촉진하는데 효과가 있다고 생각된다.
The ultimate goal of periodontal therapy is the regeneration of periodontal tissue and repair of function. For more than a decade there have been many efforts to develop materials and bioactive molecule(such as growth factor and differentiation factors) to promote periodontal wound healing. Among the bioactive molecules, bone morphogenetic protein(BMP) was studied for periodontal wound healing. Since Urist demonstrated that demineralized bone matrix could induce the formation of cartilage and bone in ectopic site, many studies on BMP have been reported. Among those BMPs, it was reported that rhBMP-2 enhanced the healing of bone defects in animal studies and clinical studies. However, its efficacy in periodontal regeneration, especially 1-wall intrabony defects is still unknown. The purpose of this study was to examine the effect of rhBMP-2/ACS on the epithelial migration, gingival connective tissue adhesion, cementum formation, alveolar bone regeneration in intrabony defects of dogs. Four millimeter deep and four millimeter wide 1-wall defects were surgically created in the mesial aspects of the 3rd incisors. The test group received rhBMP-2/ACS with a flap procedure and the control underwent buffer/ACS with a flap procedure. Histologic analysis after 8 weeks of healing revealed the following results: 1. The length of epithelial growth(the distance from alveolar crest to the apical end of JE) was $0.9{\pm}1.5mm$ in the control group and $1.2{\pm}1.4mm$ in the test group. There was no statistically significant difference between the two groups. 2. The length of connective tissue adhesion was $2.4{\pm}1.3mm$ in the control group and $1.2{\pm}1.1mm$ in the test group. The control group showed significantly enhanced adhesion(P<0.05). 3. The length of new cementum was $0.9{\pm}1.0mm$ in the control group and $1.7{\pm}0.8mm$ in the test group. The test group showed significantly enhanced cementum regeneration(P<0.05). 4. The length of new bone height was $1.9{\pm}0.6mm$ in the control group and $2.4{\pm}0.9mm$ in the test group. There was no statistically significant difference between the two groups. 5. The new bone area was $4.7{\pm}1.7mm^2$ in the control group and $8.0{\pm}2.0mm^2$ in the test group. The test group showed significantly enhanced bone formed area(P<0.05). 6. The new bone density was $73.0{\pm}8.6%$ in the control group and $66.6{\pm}15.3%$ in the test group. There was no statistically significant difference between the two groups. These results suggest that the use of rhBMP-2 in 1-wall intrabony defects has significant effect on new cementum and new bone formation area, but doesn't have any significant effect on the prevention of junctional epithelium migration and new bone formation height.
본 연구는 부산시에 위치한 하천퇴적물에 대한 현재의 오염상태와 혐기성 상태에서의 잠재메탄발생량을 평가하기 위해 실시하였다. 먼저 부산시 하천중 총 5곳을 선정하였고 대상하천의 퇴적물을 채집하였다. 그리고 퇴적물특성을 알아보기 위해 퇴적물의 화학적산소요구량, 강열감량, 총 유기탄소에 대해서 분석을 실시하였고 그 결과 각각 15.20~75.07mg $15.20{\sim}75.07mg\;g^{-1}$, 2.34~11.54%, 1.28~34.21%로 나타났다. 또한 실험실규모의 BMP Test를 실시하였고 그 결과 pH가 약간 증가하다가 약 7.11~7.35에서 평형을 이루었으며, C/N, 최종메탄 및 이산화탄소수율 그리고 생분해도는 각각 1.05~10.27, 10.1~179.4, 10.3~34.4, 4.0~30.1을 나타내었다. 그리고 최종메탄수율과 C/N비, 최종이산화탄소수율, COD, 강열감량, 총유기탄소와의 상관관계를 알아보기 위해 선형모델에 최소자승법을 적용해 평가하였고 그 결과, COD($r^2=0.7586$)와 강열감량($r^2=0.7876$)을 제외한 나머지 모든 항목에서 최종메탄수율과 높은 상관관계를 띔을 알 수 있었다($r^2=0.9795{\sim}0.9858$). 따라서 C/N 또는 TOC 분석결과를 통해 메탄발생수율을 예측 가능할 것으로 판단된다.

The present study aimed to investigate the osteogenic potentials of differentiated osteoblast-like cells (DOCs) induced from bone marrow-derived mesenchymal stem cells (MSCs) on ${\beta}-tricalcium$ phosphate (${\beta}-TCP$) with recombinant human bone morphogenetic protein (rhBMP-2) in vitro. Osteoblast differentiation was induced in confluent cultures by adding 100 nM dexamethasone, 10 mM ${\beta}$-glycerophosphate, 50 mM L-ascorbic acid. The Alizarin red S staining and reverse transcriptase-polymerase chain reaction (RT-PCR) were perfomed to examine the mRNA expression of alkaline phosphatase (ALP), bone sialoprotein (BSP), osteocalcin (OCN), receptor activator for nuclear factor ${\kappa}B$ ligand (RANKL), runt-related transcription factor 2 (RUNX2), collagen-Ⅰ (COL-Ⅰ). There were no significant differences in the osteogenic potentials of DOCs induced from MSCs on ${\beta}-TCP(+/-)$. According to the incubation period, there were significant increasing of Alizadin red S staining in the induction 3 weeks. The mRNA expression of ALP, RUNX2, and RANKL were higher in DOCs/${\beta}-TCP(-)$ than DOCs/${\beta}-TCP(+)$. According to rhBMP-2 concentrations, the mRNA expression of BSP was significantly increased in DOCs/${\beta}-TCP(+)$ compared to that of DOCs/${\beta}-TCP(-)$ on rhBMP 10 ng/ml. Our study presented the ${\beta}-TCP$ will have the possibility that calcium phosphate directly affect the osteoblastic differentiation of the bone marrowderived MSCs.
PURPOSE. This randomized clinical trial was conducted to assess the safety and effectiveness of the ErhBMP-2 in alveolar bone regeneration as well as preservation of the ${\beta}$-TCP bone graft material that contains ErhBMP-2. MATERIALS AND METHODS. This study involved 72 patients at the 3 study centers. The patients, who were divided into 2 groups: the experiment group who had ErhBMP-2 coated TCP/HA and the control group who had TCP/HA graft material alone transplanted immediately after tooth extraction. CT was taken before and 3 months after the transplantation and healing status was compared between the two groups. The efficacy endpoints that were used to measure the degree of bone induction included alveolar bone height and 3 measurements of bone width. The paired t test was used to determine the significance of the changes (P<.05). RESULTS. Changes in alveolar bone height were $-1.087{\pm}1.413$ mm in the control group and $-.059{\pm}0.960$ mm in the experimental group (P<.01). At 25% extraction socket length [ESL], the changes were $0.006{\pm}1.149$ mm in the control group and $1.279{\pm}1.387$ mm in the experimental group. At 50% ESL, the changes were $0.542{\pm}1.157$ mm and $1.239{\pm}1.249$ mm, respectively (P<.01 for 25% ESL, and P<.05 for 50% ESL). During the experiment, no adverse reactions to the graft material were observed. CONCLUSION. ErhBMP-2 coated ${\beta}$-TCP/HA were found to be more effective in preserving alveolar bone than conventional ${\beta}$-TCP/HA alloplastic bone graft materials.
Nowadays chronic degenerative diseases such as arthritis are increasing rapidly, even though acute infectious diseases are decreasing due to the advance of modern medicine. Although many of remedies are developed for arthritis healing, there is no precise medical prescription, and pathogenesis is not examined exactly. In this study, we confirmed gene expression of BMP 1A, BMP 2B, osteonectin and MGP which are genes related with bone formation in osteoblast by using ultrasonic stimulation. Through this study, we also evaluated the fact that ultrasound could be applied to arthritis healing by making activated osteoblast induce remodeling of cartilage. As a result of this research, BMP 2B and MGP have higher rates of expression in specific ultrasound mode. In conclusion, it is expected that ultrasound could be used efficiently for healing arthritis if we use ultrasounds according to symptoms of arthritis on reasonable terms.
Mixed electrolytes formed by the combination of 1-butyl-1-methylpyrrolidinium bis(trifluoromethanesulfonyl) imide (BMP-TFSI) ionic liquid and standard liquid electrolyte are prepared and characterized. Linear sweep voltammetry measurements demonstrate that these mixed systems exhibit a wide electrochemical stability window, allowing them to be suitable electrolyte for carbonaceous anode-based lithium-ion batteries. Lithium-ion cells composed of graphite anode and $LiCoO_2$ cathode are assembled using the mixed electrolytes, and their cycling performances are evaluated. The cell containing proper content of BMP-TFSI shows good cycling performance comparable to that of a cell assembled with organic electrolyte. The presence of BMP-TFSI in the mixed electrolyte contributes to the reduction of the flammability of electrolyte solution and the improvement of the thermal stability of charged $Li_{1-x}CoO_2$ in the electrolyte solution.
Locus coeruleus (LC)에는 전체 노르아드레날린성 뉴런의 절반 가량이 모여 있는데, 여기서 노르아드레날린성 뉴런이 뇌의 거의 모든 부위로 신경자극을 보내게 된다. LC는 알츠하이머병, 파킨슨병, 헌팅턴병 같은 여러 가지 신경퇴행성 질환에서 공통적으로 타격을 받는 주요 부위이다. 뇌 유래 신경영양인자, BDNF가 LC 노르아드레날린성 뉴런을 포함한 중추신경계 뉴런들의 분화와 신경세포 생존에 중요한 조절자로 작용한다. 본 연구에서는 LC 노르아드레날린성 신경세포에서 여러 가지 작은 분자들과 성장인자들이 BDNF 생산을 촉진할 수 있는지를 조사하였다. 실험에 사용한 분자들로는 neuropeptides, cytokines, 성장인자, 신경전달물질들과 세포내 신호전달물질들이 포함되었다. 여러 가지 작은 분자들과 성장인자들 중에서 FGF8b, BMP-4, forskolin 그리고 dibutyrl cGMP가 LC 노르아드레날린성 뉴런에서 BDNF 분비를 뚜렷하게 증대시킨 것으로 판명되었다. 특히, BMP-4는 BDNF 생산을 2.5배 이상 증가시켰다. LC 노르아드레날린성 뉴런에서 BDNF를 증가시킨 물질들은 여러 가지 신경퇴행성 질환에서 신경세포가 손실되는 것을 막거나 지연시킬 수 있을 것이므로, 치료제나 증상완화제로서의 가능성이 높다.
