Renoprotective Effect of Hominis Placenta Aqua-acupuncture in Streptozotocin Induced Diabetic Rats (자하차약침(紫河車藥鍼)이 당뇨유발(糖尿誘發) 흰쥐의 신장보호기능(腎臟保護機能)에 미치는 영향(影響))

  • Objective : This study was undertaken to determine whether Hominis Placenta(HP) aqua-acupuncture exerts renoprotective effect on diabetic nephropathy induced by streptozotocin(STZ) in rats. Methods : In order to study the renoprotective effect of HP aqua-acupuncture, experimental animals were divided into 5 groups and treated for 2 weeks as follows: control group was injected subcutaneously by saline aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, 5% HP aqua-acupuncture group was injected subcutaneously by 5% HP aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, 10% HP aqua-acupuncture group was injected subcutaneously by 10% HP aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, normal group was injected subcutaneously by saline aqua-acupuncture into Sinsu(BL23) in normal rats, and Captopril group was administrated with captopril at a dose of 50mg/kg in STZ induced diabetic rats. Results : While HP aqua-acupuncture did not reduce any body weight, index of kidney hypertrophy, the plasma glucose concentration and BUN respectively, HP aqua-acupuncture showed lowering urinary albumin excretion rate and serum creatinine as compared with the control group. Gene and protein expressions of $TGF-{\beta}1$ and fibronectin in kidney, one of the extracellular matrix proteins were investigated. There were significant differences in expression levels in HP aqua-acupuncture group as compared with the control group, and $100{\mu}g/m{\ell}$ and $500{\mu}g/m{\ell}$ of HP depressed apoptosis, showing in a dose dependent manner. In the HE staining, HP aqua-acupuncture inhibited the injury of glomerulus and proximal convoluted tubule. Conclusions : HP aqua-acupuncture showed the renoprotective effect possibly through suppressions of $TGF-{\beta}1$ and fibronectin expressions in kidney.

Clinical Practice Guideline for acupuncture in Post-stroke urinary retention (뇌졸중후 요저류에 대한 침치료 임상진료지침)

  • Objectives: Objectives : This study is aimed to develop a Clinical Practice Guideline (CPG) on acupuncture treatment for the patients with Post-stroke Urinary Retention; PSUR. Methods: Experts committee, consisting of stroke or methodology specialists, searched Medline, EMBASE, Cochrane Library, China National Knowledge Infrastructure, and 19 Korean medicine journals. The search terms were selected to screen the randomized controlled trials (RCTs) or systematic reviews for the effectiveness of acupuncture on PSUR compared with placebo or conventional group. Levels of evidence and grades of recommendations were appraised based on Recommendations for Development of Clinical Practice Guideline in Korean Medicine. Results & Conclusions: 4 RCT were included to build the CPG. There was a strong evidence to support the effectiveness of acupuncture treatment for PSUR. The moderate evidence was presented that over 3 times a week of the acupuncture should be performed over 4 weeks on the acupoints, such as SP6, CV3, CV6, CV4, SP9, BL28, BL23, BL22, KI3 or BL67, for 15-30 minutes. 20-140Hz frequency and 10-20 minutes of treatment is suggested if electro-acupuncture treatment is performed with. It was also suggested that the procedure should begin at the acute stage just after the vital signs of the patients are stabilized. Finally, there was a moderate evidence to support safety of acupuncture treatment for PSUR.

The Effect of Scutellariae Radix Combined with Metformin on Obesity-Relating Biomarker in High Fat Fed C57BL/6 Mice (고지방식이로 비만이 유도된 C57BL/6 마우스에서 Metformin과 황금추출물의 병용 투여 시 비만관련 지표에 미치는 영향)

  • Objectives: The purpose of this study was to investigate anti-obesity effect of Scutellariae Radix extract combined with metformin. Methods: Male C57BL/6 mice, 4 weeks of age, were used to set high-fat diet induced obesity model. They were grouped NOR (normal control), HFD (high fat diet control), MET (metformin, 100 mg/kg/day), MH2 (metformin 50 mg/kg/day+Scutellariae Radix 200 mg/kg/day), and HG4 (Scutellariae Radix 400 mg/kg/day). MET, MH2, and HG4 were orally administered for 10 weeks. Body weight was measured every week. Fasting blood glucose, triglyceride, total cholesterol (TC), high density lipoprotein, glutamic oxaloacetic transaminase (GOT), and glutamic-pyruvic transaminase (GPT) were measured before sacrifice. Oral glucose tolerance test (OGTT) was also performed. Organ weight and internal fat weight were measured after sacrifice. Results: MH2 group showed a reduction of body weight when compared with HFD group. MH2 group showed stable blood level control which was calculated areas under the curves by OGTT. TC, GOT, GPT level, internal fat, and organ weight in MH2 group reduced. Conclusions: The combined treatment of Scutellariae Radix and Metformin has impact on treating obesity. Further studies are needed to evaluate the effect of Metformin and herbal medicine combination therapy.

Expression of Human Serine Palmitoyltransferase Genes for Antibody Development (Antibody 제작을 위한 human serine palmitoyltransferase 유전자의 발현)

  • For antibody development of human serine palmitoyltransferase (SPT, EC 2.3.1.50), SPTLC1 and SPTLC2 genes were subcloned in pRset vector and expressed in E. coli BL21 (DE3)pLys cells. Eucaryotic SPT is a membrane-bound heterodimer enzyme, while all other members are soluble homodimer enzymes. cDNA library were obtained from total RNA from human embryo kidney cell line, HEK293, using RT-PCR and PCR with specific primers was carried out for preparing SPTLC1 and SPTLC2 genes. pRset vector which can express hexahistidine-tag fusion protein was used and the DNA sequences of pRsetB/SPTLC1 and pRsetA/SPTLC2 were confirmed. Recombinant BL21 cells with SPTLC subunits were selected with LB plate containing ampicillin and chroramphenicol. SPTLC1 and SPTLC2 proteins were induced with 1 mM IPTG and seperated on 10% SDS-PAGE gel. Expressed proteins were confirmed by western blotting with His-tag antibody.

  • To evaluate the antitumor activity and antimetastatic effects of HDBT, studies were done experimentally. The results were obtained as follows: 1. HDBT extracts didn't show cytotoxicity against BALB/C mouse lung fibroblast cell. 2. In cytotoxicity against A549, SK-OV-3, B16-BL6 and HT1080 concen- tration inhibiting cell growth up to below 30% of control was recognized at $10^{-3}g/ml$ of HDBT. 3. The concentration inhibiting adhesion of A549 and B16-BL6 to complex extracellular matrix up to below 30% of control was recognized at $10^{-3}g/ml$ of HDBT. 4. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $200-300{\mu}g/m{\ell}$ of HDBT. 5. The T/C% was 137.9% in HDBT-treated group in S-180 bearing ICR mice. 6. In CAM assay, HDBT extracts inhibited angiogenesis significantly at $15{\mu}g/egg$ concentration as compared with control. 7. In pumonary colonization assay, a number of colonies in the lungs were decreased but insignificantly in HDBT-treated group as compared with control group. 8. In hematological changes in B16-BL6 injected C57BL/6, numbers of WBC were decreased significantly in HDBT-treated group but numbers PLT were increased insignificantly as compared with control. From above results it was concluded that HDBT could be usefully applied for the prevention and treatment of cancer.

Haeae-tang including Artemisia argyi Folium promotes hair growth in hair-removed C57BL/6 Mice (애엽을 포함하는 해애탕의 에탄올 추출물이 제모된 C57BL/6 마우스의 발모에 미치는 영향)

  • Objectives : The experiment was performed to investigate promotive effects of haeae-tang (HET) extract, a traditional Korean medicinal recipe, on hair growth, protein and gene expression in hair-removed C57BL/6. Methods : In experiment, animals were divided into 3 groups including normal (vehicle), HET ethanol extract and 5% minoxidil-treated groups (Minoxidil, positive control). The vehicle or testing samples were daily treated with 0.2ml per on hair-shaved dorsal skin of C57BL/6mice for 15 days. Effects of testing samples on hair growth was monitored through phototrichogram analysis by folliscope on the initial, $5^{th}$, $10^{th}$, $15^{th}$ day, respectively. Also, gene and protein expressions of vascular endothelial growth factor (VEGF) and Insulin like growth factor-1 (IGF-1), relevant to hair growth, were examined. Results : Hair density and hair thickness of Minoxidil treated-group was significantly increased compared to that of vehicle application on the $15^{th}$day, respectively. Dorsal hair density of HET treated-group was significantly increased compared to that of vehicle application on the $15^{th}$day. In addition, the Minoxidil group significantly increased the expression of cutaneous IGF-1 protein and mRNA compared to that of the vehicle-applied group on the $15^{th}$ day. And HET-treated group significantly increased the expression of dorsal VEGF protein compared to that of the vehicle-applied group on the $15^{th}$ day. Conclusions : These results suggest that this Korean medicinal recipe, HET has promoting activity on hair growth in an Alopecia animal model thus it can be used as a material of agent or products for improvement or prevention of alopecia.

  • The effects of dietary mannan oligosaccharides, Lactobacillus plantarum, Bacillus subtilis, and Bacillus licheniformis supplementation on hybrid grouper Epinephelus akaara ♀×Epinephelus lanceolatus ♂ were evaluated. The fish were fed a basal diet and five other diets consisting of 0.6% mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis and mixture of each 0.15% prebiotic and all the probiotics (designated as MOS, LP, BS, BL, and SYN) for 56 days. Growth performance and feed utilization showed no significant differences among all experimental groups. Lipid level of whole-body was significantly high in MOS and BL groups. Plasma aspartate aminotransferase was significantly low in BL and SYN groups. Nitro-blue tetrazolium, lysozyme and anti-protease, and glutathione peroxidase in BS, SYN, and all probiotic groups, respectively, were significantly high. Intestinal Vibrio bacteria was significantly low in all probiotic and SYN groups. Gene expression of interleukin-1β and interleukin-10 in SYN group; transforming growth factor β2 in MOS and BS groups, toll-like receptor 2-2 in BS and BL groups; and C-type lectin in MOS, LP and SYN groups were significantly upregulated. Our findings indicate that mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis could improve innate immunity, antioxidant capacity, anti-inflammation, and intestinal microbiota of hybrid grouper.

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffuse herba infusion solution(ELP-HAS), we injected ELP-HAS into Chok-samni(St36) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. We have reached the following conclusion through the effect on the number of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1+/CD3e^+$ cells in mouse PBMCs, the effect on the pulmonary colony number, and the effect on MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma The results were obtained as follows: 1. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the spleen cell proliferation in Balb/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$ cells in C57Bl/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the pulmonary colony number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Eclipta prostrata)Herbal-Acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group.

  • The generating functions of the divisor function σs(n) = Σ0<d|n ds are quasimodular forms. In this paper, we find the basis of the space of quasimodular forms of weight 6 on Γ0(10) consisting of Eisenstein series and η-quotients. Then we evaluate the convolution sum Σak+bl+cm=n σ(k)σ(l)σ(m) with lcm(a, b, c) = 10 and Σal+bm=n lσ(l)σ(m) and Σal+bm=n σ3(l)σ(m) with lcm(a, b) = 10.

Effect of High-fat Diet on Peritoneal Macrophage Immunocompetence in C57/BL6 Mice (고지방 식이가 C57/BL6생쥐의 복강대식세포 면역능력에 미치는 영향)

  • This study was to investigate the effect of high-fat diet on macrophage immunocompetence in C57BL/6 mice. C57BL/6 male mice (4 weeks aged, n=16) were divided into two groups. HD groups fed high-fat diet (45% of fat) and ND groups fed chow diet (10% of fat). Peritoneal macrophages were obtained from each mouse intra-peritoneal by sterile lavage method. Macrophage were stimulated with $1{\mu}g/ml$ of lipopolysaccharide (LPS) for 24 hr. Body weight was significantly increased by high-fat diet. Macrophage phagocytosis of HD was significantly lower than that of ND. After 24 hr of LPS stimulation, NO, IL-$1{\beta}$ and IFN-${\gamma}$ production of HD were significantly lower than those of ND. There were no significant differences in the production of TNF-${\alpha}$ and IL-12 between HD and ND. These findings suggest that high fat diet-induced obesity is associated with decreased Immunocompetence and antigen-stimulated sensitivity of peritoneal macrophage, and lower production of NO, IL-$1{\beta}$ and IFN-${\gamma}$ may contribute to these changes.
