재조합 대장균으로부터 항고혈압 Angiotensin I-Converting Enzyme 저해제의 특성연구 (Characterization of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor from Recombinant E. coli)

  • 안지오텐신(ACE) 저해제는 항고혈 효과를 갖고 있으므로 오랫동안 고혈압의 예방이나 치료에 이용되어 왔다. 본 연구는 재조합 대장균으로부터 새로운 ACE 저해제를 생산하고 정제하며 나아가 이들이 구조-기능 관P를 규명하기 위해 수행되었다. Saccharomyces cerevisiae의 ACE 저해 펩타이드 유전자를 함유하고 있는 재조합 pGEX-4T-3을 대장균 BL21(DE3)로 형질전환 시켰다. 재조합 pGEX-4T-3을 갖고 있는 대장균 BL21(DE3)로부터 생산된 Glutathione-s 전이효소(GST) 융합 단백질을 얻어서 그중 ACE저해 펩타이드를 Sephadex G-25 컬럼 크로마토그래피로 정제하였다. 정제된 ACE 저해 펩타이드는 타이로신-아스파틱엑시드-그리신-글리신-발린-페닐알라린-아르기닌-발린-타이로신-트레오닌의 서열을 가진 새로운 decapeptide이었고 ACE에 대하여 경쟁적으로 저해하였다.

Bacillus cereus ASK-202에서 cloning 된 agarase의 물리 ${\cdot}$ 화학적 특성

  • 본 연구에서는 기능성 식품 첨가물 소재 또는 의약품으로 사용 가능한 한천올리고당의 대량생산을 위하여 agarase 생산균주인 Bacillus cereus ASK202에 대해 유전자 cloning 방법을 이용하여 한천분해효소 고생산성 균주로의 개발을 시도하였다. 원균주의 chromosomal DNA를 무작위적으로 절단하여 agarase를 생산하는 gene 부위를 선별한 결과, 83,300 Da의 agarase(Eba1)를 생산하는 재조합 균주 E. coli BL21(DE3)/pEBA1를 얻을 수 있었다. E. coli BL21(DE3)/pEBAl에서 유도물질로 IPTG를 첨가한 후 induction 5시간 후에 agarase가 원균주에 비해 8배 증가한 양이 고발현되었다. 발현된 agarase는 Asx(Aspartic acid, Asparagine), Glycine와 같은 산성 아미노산의 함량과 Alanine과 같은 중성 아미노산의 함량이 높았으며, pH 5.6, $40^{\circ}C$에서 최적의 활성을 나타내었다. 최적 기질 agar에 대한 $K_m$$V_{max}$값은 0.068 mg/$m{\ell}$과 0.094mg/$m{\ell}{\cdot}min$으로 한천의 ${\beta}$-결합을 자르는 ${\beta}$-agarase로 판명되었다.

Antibody 제작을 위한 human serine palmitoyltransferase 유전자의 발현 (Expression of Human Serine Palmitoyltransferase Genes for Antibody Development)

  • 사람의 serine palmitoyltransferase(SPT, EC 2.3.1.50)에 대한 항체를 제작하기 위하여 E. coli발현 vector인 pRset vector에 SPTLC1 및 SPTLC2 유전자를 subcloning하고 BL21 (DE3)pLys cell에 발현시켰다. 포유동물의 SPT는 원핵세포의 SPT homodimer와는 달리 SPTLC1 및 SPTLC2 2개의 sub-unit로 된 heterodimer이다. Human embryo kidney cell인 HEK293 cell의 total RNA로부터 RT-PCR을 행하여 cDNA library를 얻은 다음 SPTLC1 및 SPTLC2의 특이적인 primer 들을 이용하여 PCR을 행하였다. SPTLC1 및 SPTLC2 DNA를 hexahistidine fusion 단백질을 발현시킬 수 있는 pRset vector에 cloning하여 pRsetB/SPTLC1 및 pRsetA/SPTLC2를 얻고 염기서열을 확인하였다. 재조합 plasmid를 발현세포인 BL21 cell에 형질전환시킨 다음 ampicillin 및 chroramphenicol 배지에서 선별하여 재조합세포를 얻었다. 1 mM IPTG로서 발현을 유도하였으며 세포 단백질을 SDS-PAGE로 분리한 다음 His-tag antibody로 western blotting을 행하여 SPTLC 및 SPTLC2가 발현되었음을 확인하였다.

합곡(合谷)·태충(太衝), 간유(肝兪)·대장유(大腸兪) 동시(同時) 취혈(取穴)이 정상(正常) 백서(白鼠) 위기능(胃機能)에 미치는 영향(影響) (Effects of Treatment with Acupuncture, Moxibustion and Electro-Acupuncture at Large Intestine Meridian and Liver Meridian on the Serum Gastrin Level in Rats)

  • Objective : This study was designed to investigate the effects of acupuncture, moxibustion and electro-acupuncture at LI4 and LR3, BL18 and BL25 on serum gastin level in rats. Methods : LI4 and LR3 which belong to Large Intestine Meridian and liver Meridian are acupuncture points frequently used for healing gastrointestinal disease in oriental medicine. BL18 and BL25 which belong to Bladder Meridian are acupuncture points used for healing hepatic disease and intestinal disease in oriental medicine. Serum gastrin level by radioimmnunoassay was measured at 3 days after acupuncture, moxibustion and electro-acupuncture of those acupoints. Results & Conclusion : Moxibustion applied to the BL18 acupoint, BL25 acupoint decreased gastrin level of serum, but moxibustion applied to the BL18+BL25 acupoints did not produced significant effect. Acupuncture applied to the LI4 acupoint, LR4 acupoint did not produced significant effect, but acupuncture applied to the LI4+LR4 acupoints increased gastrin level of serum, Electro-acupuncture applied to the LI4 acupoint, LR4 acupoint increased gastrin level of serum, and electro-acupuncture applied to the LI4+LR4 acupoints also increased gastrin level of serum.

Inhibition of Experimental Systemic Inflammation (Septic Inflammation) and Chronic Bronchitis by New Phytoformula BL Containing Broussonetia papyrifera and Lonicera japonica

  • Broussonetia papyrifera and Lonicera japonica have long been used in the treatment of inflammatory disorders in Chinese medicine, especially respiratory inflammation. Previously, a new phytoformula (BL) containing B. papyrifera and L. japonica was found to exert strong anti-inflammatory activity against several animal models of inflammation, especially against an animal model of acute bronchitis. In the present investigation, the effects of BL on animal models of septic inflammation and chronic bronchitis are examined. Against lipopolysaccharide (LPS)-induced septic inflammation in mice, BL (200-400 mg/kg) reduced the induction of some important proinflammatory cytokines. At 1 h after LPS treatment, BL was found to considerably inhibit TNF-${\alpha}$ production when measured by cytokine array. At 3 h after LPS treatment, BL inhibited the induction of several proinflammatory cytokines, including IFN-${\gamma}$ and IL-$1{\beta}$, although dexamethasone, which was used as a reference, showed a higher inhibitory action on these biomarkers. Against chronic bronchitis induced by LPS/elastase instillation in rats for 4 weeks, BL (200-400 mg/kg/day) significantly inhibited cell recruitment in bronchoalveolar lavage fluid. Furthermore, BL considerably reduced lung injury, as revealed by histological observation. Taken together, these results indicate that BL may have a potential to treat systemic septic inflammation as well as chronic bronchitis.

배유혈(背兪穴)과 복유혈(腹募穴)의 주치증(主治症)에 관한 문헌적(文獻的) 비교(比較) 연구(硏究) (Documentary comparative study on the chief virtues of the Back-Su(兪) points and the Front-Mo(募) points)

  • From documentary comparative study on the chief virtues of the Back-Su(兪) points and the Front-Mo(募) points, the following results are obtained : 1. Contrast the Back-Su(兪) points with the Front-Mo(募) points, the needle-steadying depth of the Front-Mo(募) points is deeper than that of the Back-Su(兪) points. and Moxibustion dosage of the Front-Mo(募) points is more than that of the Back-Su(兪) points. 2. In the case of Kimun(LR14) and Kansu(BL18), Kergwol(CV12) and Shimsu(BL15), Chungwan(CV12) and Wisu(BL21), Kyungmun(GB25) and Shinsu(BL23), the Back-Su(兪) points can be chiefly used for the treatment of acute disease, external disease, exess disease and Jang(臟)-disease. contrary the Front-Mo(募) points can be chiefly used for the treatment of chronic disease, internal disease, deficiency disease and Bu(腑)-disease. 3. In the case of Kwangwon(CV4) and Sojangsu(BL27), Serkmoon(CV5) and Samchosu(BL22), Changmun(LR13) and Bisu(BL20), Chungbu(LV1) and Pesu(BL20), Chungguk(CV3) and Banggwangsu(BL28), the Front-Mo(募) points can be used for the treatment of unbalanced visceral funtion. contrary the Back-Su(兪) points can be used for not only the treatment of unbalanced visceral funtion but also the treatment of attached organs. 4. The Back-Su(兪) points can be used for the treatment of acute disease, external disease, exess disease, Jang(臟)-disease and attached organs-disease. contrary the Front-Mo(募) points can be chiefly used for the treatment of chronic disease, internal disease, deficiency disease, and Bu(腑)-disease.

대장균에서 증식속도 조절에 의한 수용성 재조합 인간 과립구 콜로니 촉진인자의 생산 (Production of Soluble Recombinant Human Granulocyte Colony Stimulating Factor in E. coli by Control of Growth Rate.)

  • 인간 과립구 성장인자(hG-CSF)는 골수에서 생산되는 단백질로 호중구의 분화 및 생성을 촉진시키는 역할을 한다. 현재 재조합 hG-CSF는 암화학요법에 의한 호중구감소증, 골수이식시 호중구 감소증, 재생불량성 빈혈에 수반되는 호중구 감소증 등으로 적응증이 확대되고 있다. 본 연구에서는 OmpA signal sequence를 삽입하여 인간 과립구 성장인자(hG-CSF)가 분비발현되도록 고안된 T7 promoter 에 의하여 발현되는 pYRCl 발현백터를 제조하였다. E. coli BL2l (pYRCl) 발현시 $37^{\circ}C$에서 배양하는 경우 많은 양의 봉입체(aggregates)를 형성한다. 이에 비하여 $10\mu$M ucose를 포함하는 변형된 MBL배지에서 10 g/$\ell$IPTG를 유도물질로 7시간동안 $25^{\circ}C$에서 배양하였을 때 전체 periplasm단백질의 15%가 soluble rhG-CSF이었다. 또한, 유가식 배양방법을 사용하여 E. coli BL2l(pYRCl)에서 soluble rhG-CSF의 생산조건을 조사하였다. 유가식 배양에서 rhG-CSF의 발현량이 비증식속도를 $0.43 h^{-1}$ 에서 0.14 $h^{-1}$ 으로, 유도 배양시간을 최적화함으로써 rhG-CSF의 발현량이 4.4mg/$\ell$에서 24mg/$\ell$ 로 증가하였다.

대장균에서 환경적 요인이 한탄바이러스 뉴클레오캡시드 유전자의 발현에 미치는 영향 (Effects of Environmental Conditions on the Expression of Hantaan Viral Nucleocapsid Gene in Escherichia coli)

  • Viruses belonging to the Hantavirus genus cause two acute severe illness in humans, i.e., Haemorrhagic Fever with Renal Syndrome (HFRS) and Hantavirus Pulmonary Syndrome(HPS). Among them, Hantaan virus is one of the most important viruses causing HFRS. Recombinant expression vectors, pKK-NP and pET-NP, with Hantaan viral nucleocapsid gene were constructed, and used to transform Eschericia coli BL21(DE3). Stability of the vectors in the host strain, and effects of some environmental conditions on the expression of nucleocapsid gene were studied. Expression vector, pKK-NP, was very unstable, and the expression level of nucleocapsid gene was very low compared to that of pET-NP. BL21(pET-NP) produced about 100 mg of N protein per liter of culture broth. Induction time did not show any significant difference on the expression level of nucleocapsid gen and cell growth. BL21(pET-NP) culture at 35$^{\circ}C$ showed a little higher expression level than at 30$^{\circ}C$ during growth phase, but reached to the same level at stationary phase. Total expression level was proportional to supplemented glucose concentration of media up to 0.5% along with cell growth, but expression level per unit cell mass was inversely proportional to glucose concentration and maximal when glucose was not supplemented at all.

침술자극이 개의 위 배출시간에 미치는 영향에 관한 X선 및 초음파학적인 평가 (Radiographic and Ultrasonographic Evaluation of Gastric Time of Dogs after Acupunctural Stimulation)

  • This study was performed to evaluate the effect of acupuncture on gastric motility in dogs with radiography and ultrasonography. All experimental dogs were fed with the soup with barium (3% W/V), and thereafter gastric emptying time(GET) was measured with radiography and ultrasonography with Bolondi's method. The acupoints were CV-12, PC-6, ST-36 and BL-21 in this experiment. On these acupoints, electroacupunctral stimulation(15 MHz, continous wave) was applied for 20 minutes. The results were as follows: In the control group, normal GET was 70-85 minutes with ultrasonography, and 110-130 minutes with radiography. In acupuncture stimulation groups, there was no effect of gastric motility in CV-12, PC-6, and 57-36, whereas there was significant stimulating effect of gastric movement in BL-21 with ultrasonography (P < 0.05) and radiography (P < 0.01). Above these results, it is concluded that acupunctural stimulation of BL-21 was more effective in gastric motility compared to those of CV-l2, PC-6 and ST-36.

Interaction of brassinosteroids and cytokinin in modulating light mediated signaling in Arabidopsis

  • Brassinosteroids (BRs) are a special class of plant steroid hormones that are essential for normal growth and development. Part of confusion is whether BRs are unique to plants, because they have overlapping physiological roles with other better-studied hormones and with physiological responses caused by light. In systems designed to assay for cytokinins, the effects of BRs vary. We measured hypocotyl length for testing the ability of brassinolide (BL) to rescue double mutant between det2 and the photoreceptor null mutant phytochrome B (phyB). PHYB involved in controlling hypocotyl elongation in increased concentration of BL whereas phyBdet2 double mutant just partially rescue to phyB in white and red light indicated the involvement of BRs in PHYB regulated cell elongation. BRs regulated hypocotyl growth was delayed by BAP, a cytokinin treatment but inhibitory effects of BAPs on hypocotyl growth was slightly recovered by BL. The result indicated that the mode of action of BR and cytokinin is independent or sequential in the downstream light-regulated response control on hypocotyl elongation and also light modulated the action of BR and cytokinin in some extent.
