Developing a Virus-Binding Bacterium Expressing Mx Protein on the Bacterial Surface to Prevent Grouper Nervous Necrosis Virus Infection

  • Grouper nervous necrosis virus (GNNV) infection causes mass grouper mortality, leading to substantial economic loss in Taiwan. Traditional methods of controlling GNNV infections involve the challenge of controlling disinfectant doses; low doses are ineffective, whereas high doses may cause environmental damage. Identifying potential methods to safely control GNNV infection to prevent viral outbreaks is essential. We engineered a virus-binding bacterium expressing a myxovirus resistance (Mx) protein on its surface for GNNV removal from phosphate-buffered saline (PBS), thus increasing the survival of grouper fin (GF-1) cells. We fused the grouper Mx protein (which recognizes and binds to the coat protein of GNNV) to the C-terminus of outer membrane lipoprotein A (lpp-Mx) and to the N-terminus of a bacterial autotransporter adhesin (Mx-AIDA); these constructs were expressed on the surfaces of Escherichia coli BL21 (BL21/lpp-Mx and BL21/Mx-AIDA). We examined bacterial surface expression capacity and GNNV binding activity through enzyme-linked immunosorbent assay; we also evaluated the GNNV removal efficacy of the bacteria and viral cytotoxicity after bacterial adsorption treatment. Although both constructs were successfully expressed, only BL21/lpp-Mx exhibited GNNV binding activity; BL21/lpp-Mx cells removed GNNV and protected GF-1 cells from GNNV infection more efficiently. Moreover, salinity affected the GNNV removal efficacy of BL21/lpp-Mx. Thus, our GNNV-binding bacterium is an efficient microparticle for removing GNNV from 10‰ brackish water and for preventing GNNV infection in groupers.

The Change of Serum Gastrin Concentration with Oculo-acupuncture in Dogs (개에 있어서 안침(眼針)에 따른 혈청 가스트린 농도의 변화)

  • This study was performed to clarify the effect of oculo-acupuncture (OA) on change of serum gastrin concentration. Twenty-three clinically healthy mongrel dogs were used in this study. This study was comprised of two experiments. In experiment 1, the effect of OA (stomach/spleen region) with comparison of non-a cup oint was examined. On the other hand, the effect of OA (stomach/spleen region) with comparison by body acupuncture (BL-21) was also examined in experiment 2. Serum gastrin levels were measured on before, 0, 30, 60 and 120 minutes after treatment in both non-acupoint and stomach/spleen region groups, and BL-21 The results showed that significant decreases in serum gastrin level was detected on 30 mins(p<0.05) after OA when compared with that of non-acupoint group. Serum gastrin concentrations with OA at stomach/spleen region group were significantly decreased on 60 mins (p<0.05) and 120 mins (p<0.01) compared with those of the AP at BL21 group. In conclusion, it was demonstrated that OA at stomach/spleen region had significantly decreased serum gastrin levels in dogs and OA at stomach/spleen region decreased serum gastrin concentration more than that body acupuncture at BL-21.

A Clinical Study on Adolescent Idiopathic Scoliosis using DITI (DITI를 활용한 청소년기 특발성 척추측만증에 대한 임상적 고찰)

  • Objectives : This study was performed to investigate the correlation between Cobb's angle and digital infrared thermographic imaging(DITI) on adolescent idiopathic scoliosis. Methods : A total of 56 patients with adolescent idiopathic scoliosis were classified into four groups according to their Cobb's angle. Four groups were made for each increment of $5^{\circ}$: group $5-10^{\circ}$, group $11-15^{\circ}$, group $16-20^{\circ}$, and group $21-25^{\circ}$. Next we checked thermal differences of 6 acupoints(GB21, BL13, BL15, BL20, BL23, BL26) on DITI and analyzed correlation by SPSS ver. 20.0.0 for windows. Results : 1. The average patient age was approximately 13 years old. 2. The majority of patients complained of pain on both sides. 3. Significant correlation was found between group $11-15^{\circ}$ and group $16-20^{\circ}$, group $16-20^{\circ}$ and group $21-25^{\circ}$ on Simsu(BL15) by Duncan test. Conclusion : Using DITI, Simsu(BL15) is expected to be a valid indicator for the diagnosis and treatment of adolescent idiopathic scoliosis.

Influence of Gigaltang on Alcohol Intake Amount in C57BL/6 mice (지갈탕(枳葛湯)이 C57BL/6형 생쥐의 알코올섭취량에 미치는 영향)

  • Objective : This study was designed to test the anti-craving effect of Gigaltang(extracts of Oriental medicinal herbs ; Hovenia dulcis and Puerariae flos) in C57BL/6 mice. Anti-craving effect was substituted by alcohol intake amount of C57BL/6 mice. Method : 60 C57BL/6 mice was randomized into vehicle(water) group and Gigaltang group. Only 39(vehicle group(21) and Gigaltang group(18)) was eligible for this study, and 21 C57BL/6 mice was dropped out. The procedure was composed of 5 days of adjustment period and 32 days of alcohol exposure and withdrawal period with limited access paradigm and 10 days of treatment period. Alcohol was offered only for 2 hours a day from 2 to 4PM and water was offered for 22 hours left. Food was offered for 24 hours a day. 1140mg/Kg/day of Gigaltang was administrated to 18 mice of Gigaltang group for 10 days whereas water to 21 mice of vehicle group. The amount of alcohol intake, water intake, food intake, body weight were measured every other day. Result : There weren't significant differences in 2 hours of alcohol intake, 22 hours of water intake, 24 hours of food intake and body weight for ten days between Gigaltang and vehicle group. Conclusion : Anti-craving effect of Gigaltang on C57BL/6 mice wasn't verified. To verify the anti-craving effect, further study with advanced animal model, various dose of Gigaltang and diverse period of drug administration should be taken.

  • The Present study was examined the response of mice of 5 strains to superovulation treatment. Female mice of CF#1, C57BL/6, ICR, dd and CHN strains, aged 60 days were used. Superovulation was induced by the subcutaneous injection of 5 IU PMS and followed 48 hours later by the subcutaneous injection of 5 IU HeG. All females were killed 24 hours after RCG and then ova in the oviduct were counted under a microscope. The average number of ova ovulated in CF#1, C57BL/6, ICR, dd and CHN were 21.3, 17.9, 20.7, 21.0 and 20.5, respectively.

Effect of Traditional Acupuncture on Canine Gastric Motility

  • The effect of traditional acupuncture on gastric motility was investigated in dogs. Six healthy dogs, of mixed breed, were used in this study. The gastric motility was evaluated by electrogastromyograms (EGMG) of the smooth muscle of the pyloric region. The acupoints investigated were ST-36, ST-40, ST-41, ST-42, ST-45, BL-21 and CV-12. The gastric motility was increased by traditional acupuncture at ST-36 and BL-21 but decreased by traditional acupuncture at CV-12 However, there were no significant changes in the gastric motility after acupuncture at ST-40, ST-41, ST-42 and ST-45.

A Retrospective Study on Needling Depth of Hwangmun (BL51) and Jisil (BL52) with MRI (MRI를 통한 황문혈(BL51)과 지실혈(BL52)의 자침 깊이에 대한 연구)

  • Objectives : The aim of this study is to investigate the needling depth of Hwangmun (BL51) and Jisil (BL52) retrospectively using an L-spine MRI. Methods : We measured the shortest distance from the skin to transversalis fascia at both sides of Hwangmun and Jisil, and analyzed the difference between male and female using a student t-test, and between the left and right sides with a paired t-test. Results : In the case of males, the average depth of left Hwangmun was $48.24{\pm}10.16mm$, and that of right was $47.23{\pm}9.59mm$ ; left Jisil was $56.91{\pm}9.00mm$ and right was $55.74{\pm}8.75mm$. In the case of females, the average depth of left Hwangmun was $42.26{\pm}9.29mm$, and that of right was $41.63{\pm}9.32mm$ ; left Jisil was $49.21{\pm}10.77mm$ and right was $48.41{\pm}11.38mm$. The depth of male insertion was deeper than that of female in Jisil, but there was no significant difference according to gender in Hwangmun. Conclusion : For males, the needling depth of left Hwangmun is 37.21 ~ 68.23 mm and right is 36.91 ~ 70.50 mm, while left Jisil is 42.97 ~ 70.84 mm and right is 43.75 ~ 72.00 mm. For females, the needling depth of left Hwangmun is 26.80 ~ 68.28 mm and right is 25.70 ~ 65.59 mm, while left Jisil is 30.94 ~ 79.06 mm and right is 28.13 ~ 77.27 mm.

Effect of Morus alba extract for hair growth promotion in C57BL/6 mouse (C57BL/6 mouse에서 뽕나무 추출물의 모발성장효과)

  • Morus alba has been well known for its tonic effect or preventing of hypertension, aging, or diabetes mellitus in Asian countries. To evaluate the its effect on hair growth promotion, we performed the hair regrowth experiment with leaf, branch, root and fruit extract of Morus alba on animal model of C57BL/6 mouse. The hair regrowth effect was remarkable in the mouse treated branch and root extract (with what %), however, little effect on hair regrowth appeared on the mouse treated with leaf extract. Morus alba extract displayed antibacterial effects on Bacillus subtilis, Staphylococcus aureus, Pseudomonas and Candida albicans. These results indicated that branch and root extract of Morus alba could be applicable for hair regrowth or prevention of hair loss in human.

Immunomodulating Activity of BL18 (Ganshu) Acupuncture on the Experimental Liver Metastasis Model of Mice

  • We investigated that the immunomodulating activity of BL18 (Ganshu) acupuncture on the experimental liver metastasis model of mice. NA (non-acupoint)- and BL18-treatment enhanced the mitogenic activity of BALB/c whole splenocytes induced by various mitogenic stimuli. Acupuncture treatment tended to increase splenocytes differentiation even though did not show significance. Acupuncture treatment caused a marked increase of production of Th1 cytokine (IFN-${\gamma}$) and Th2 cytokine (IL-4) by splenocytes and IL-12 and IFN-Y by macrophages. The increase of cytokine production on BL18-treated group was more pronounced compared to NA-treated group. The liver weight of NA- and BL18-treated group decreased compared to tumor group, but did not showed significant differences.
