Metabolic Engineering of Escherichia coli for the Biological Synthesis of 7-O-Xylosyl Naringenin

  • Flavonoids are a group of polyphenolic compounds that have been recognized as important due to their physiological and pharmacological roles and their health benefits. Glycosylation of flavonoids has a wide range of effects on flavonoid solubility, stability, and bioavailability. We previously generated the E. coli BL21 (DE3) ${\Delta}pgi$ host by deleting the glucose-phosphate isomerase (Pgi) gene in E. coli BL21 (DE3). This host was further engineered for whole-cell biotransformation by integration of galU from E. coli K12, and expression of calS8 (UDP-glucose dehydrogenase) and calS9 (UDP-glucuronic acid decarboxylase) from Micromonospora echinospora spp. calichensis and arGt-4 (7-O-glycosyltransferase) from Arabidopsis thaliana to form E. coli (US89Gt-4), which is expected to produce glycosylated flavonoids. To test the designed system, the engineered host was fed with naringenin as a substrate, and naringenin 7-O-xyloside, a glycosylated naringenin product, was detected. Product was verified by HPLC-LC/MS and ESI-MS/MS analyses. The reconstructed host can be applied for the production of various classes of glycosylated flavonoids.

대장균에서 진핵세포 펩타이드 호르몬 전구물질의 대량생산과 특성규명 (Overexpression and Characterization of Eukaryotic Peptide Hormone Precursors in E. Coli.)

  • 대장균에서 진핵세포의 펩타이드 호르몬 전구체를 대량으로 생산할 목적으로 T7 overexpression system을 이용하여 angler 어류의 prepro-SRIF I 유전자와 T7 phage coat 단백질 S10 유전자를 결합시켜 일련의 융합유전자를 합성하였다. 이 융합유전자를 가지고 있는 숙주 대장균, BL21 DE3는 3가지 종류의 서로 다른 SRIF 관련 폴리펩타이드를 대량 합성하였다. 본 연구에서는 대량합성된 폴리펩타이드의 특성을 규명하였으며, 펩타이드 호르몬 전구체를 얻기 어려운 이종이에서의 대량발현의 중요성과 표적 펩타이드 호르몬인 SRIF의 적용성을 논의하였다.

  • Salmonella typhimurium LT2에서 deoxy-thymidine diphosphate-D-gluxose-4,6-dehydratase의 유전자를 재조합한 Escherichia coli BL21 clone으로부터 dTDP-D-glucose dehydratase를 분리 정제한 후, 이 효소의 단결정을 상온에서 sitting drop 기체확산법으로 성장시켰다. 결정은 효소에 기질이 포함되어 있는 것과 포함되어 있지 않는 것 모두가 얻어지며, 이 때 침전제는 1.6-2.0 M Na, K 인산 완충용액(pH 8.0)을 사용하였다. 이 단결정은 최소 2.5Å의 분해능으로 회절하였으며, 공간군은 hexagonal한 P61이고, 격자의 크기는 a=b=168.54Å, c=81.08Å이었다. Asymmetric unit에는 이량체 한분자를 포함하고 있으며 단백질 질량당 결정의 부피는 VM=2.4Å3/Da, 용매의 함유율은 부피를 기준으로 64%였다.

Identification of hrcC, hrpF, and maA Genes of Xanthomonas campestris pv. glycines 8ra: Roles in Pathogenicity and Inducing Hypersensitive Response on Nonhost Plants

  • Nonpathogenic mutants of Xanthomonas campestris pv. glycines were generated with Omegon-Kim to isolate genes essential for pathogenicity and inducing hypersensitive response (HR). Three nonpathogenic multants and two mutants showing slow symptom development were isolated among 1,000 colonies tested. From two nonpathogenic mutants, 8-13 and 26-13, genes homologous to hrcC and hrpF of X. campestris pv. vesicatoria were identified. The nonpathogenic mutant 8-13 had a mutation in a gene homologous to hrpF of X. campestris pv. vesicatoria and failed to cause HR on pepper plants but still induced HR on tomato leaves. The nonpathogenic mutant 26-13 had an insertional mutation in a gene homologous to hrcC of X. campestris pv. vesicatoria and lost the ability to induce HR on pepper leaves but still caused HR on tomato plants. Unlike other phytopathogenic bacteria, the parent strain and these two mutants of X. campestris pv. glycines did not cause HR on tobacco plants. a cosmid clone, pBL1, that complemented the phenotypes of 8-13 was isolated. From the analysis of restriction enzyme mapping and deletion analyses of pBL1, a 9.0-kb Eco RI fragment restored the phenotypes of 8-13. pBL1 failed to complement the phenotypes of 26-13, indicating that the hrcC gene resides outside of the insert DNA of pBL1. One nonpathogenic mutant, 13-33, had a mutation in a gene homologous to a miaA gene encoding tRNA delta (2)-isopentenylpyrophosphate transferase of Escherichia coli. This indicated that tRNA modifications in X. campestris pv. glycines may be required for expression of genes necessary for pathogenicity. The mutant 13-33 multiplied as well as the parent strain did in the culture medium and in planta, indicating that loss of pathogenicity is not due to the inability of multiplication in vivo.

Assay of ${\beta}$-Glucosidase Activity of Bifidobacteria and the Hydrolysis of Isoflavone Glycosides by Bifidobacterium sp. Int-57 in Soymilk Fermentation

  • The isoflavone glycosides are hydrolyzed by ${\beta}$-glucosidase from gut microbes to the bioactive aglycones. However, the specific bacteria from the human intestinal tract that are involved in the metabolism of these compounds are not known. This study was undertaken to develop a fermented soymilk which converts isoflavones to the more bioactive aglycones form using a Bifidobacterium strain. The ${\beta}$-glucosidase activity of 15 Bifidobacterium strains were measured during cell growth. Among them, Bifidobacterium sp. Int-57 was selected for this study, because it has the highest ${\beta}$-glucosidase activity. Growth, acid development, ${\beta}$-glucosidase activity, and the hydrolysis of daidzin and genistin were investigated in four soymilks inoculated with Bifidobacterium sp. Int-57. After 12 h of fermentation, the counts of viable Bifidobacterium sp. Int-57 in all the soymilks reached a level of more than $10^8$ cfu/ml, which was then maintained. The pH of soymilks started to decrease rapidly after 6 h of fermentation and leveled off after 18 h. The titratable acidity of BL# 1 soymilk, BL#2 soymilk, and JP#l soymilk increased from 0.18 to 1.21, 1.15, and $1.08\%$ over the fermentation period, respectively. After 24 h of fermentation, the $\beta$-glucosidase activity in BL#1 soymilk, BL#2 soymilk, JP#l soymilk, and JP#2 soymilk increased to 59.528, 40.643, 70.844, and 56.962 mU/ml, respectively. The isoflavone glycosides, daidzin and genistin, in soymilks were hydrolyzed completely in the relatively short fermentation time of 18 h. These results show that Bifidobacterium sp. Int-57 can be used as a potential starter culture for developing fermented soymilk which has completely hydrolyzed isoflavone glycosides.

자기를 이용한 경락 자극요법 시행 후 요부근육통 환자의 체열변화 관찰: 무작위배정, 환자-평가자 눈가림, 위기기 대조군, 예비 임상 연구 (Observation on Change of Body Temperature for Patients with Low Back Muscular Pain after Electromagnetic Theraphy: a Randomized, Patient-assessor Blind, Placebo-controlled, Pilot Trial Study)

  • Objectives: This study was resigned to observe change of body temperature for patients with low back muscular pain after NUGA MRT-II(pulsed electromagnetic therapy) treatment. Methods: This study was a randomized, patient-assessor blind, placebo-controlled, pilot trial study. After the approval of institutional review board(IRB), we have recruited 38 patients suffering from low back muscular pain and divided them into two groups randomly: the treatment group and control group. To the treatment group, NUGA MRT-II was practiced and to the control group sham device was practiced and their low back muscles and acupuncture points three times a week for 2 weeks from February 2011 to May 2011. After 1 week of last treatment, We compared body temperature of two groups. Results: 1. There was significant decrease of body temperature on both Shinsu(BL23), Chishil(BL52), Kwanwonsu(BL26) acupunture points for the treatment group. 2. There was no difference in the decrease of body temperature between treatment group and control group except. Left Kwanwonsu(BL26). Conclusions: We found out that treatment of NUGA MRT-II on low back muscular pain can reduce the temperature of low back.

Shotgun Phage Display of Lactobacillus casei BL23 Against Collagen and Fibronectin

  • Lactobacilli are normal constituents of the intestinal microbiota, and some strains show the capacity to bind to extracellular matrix proteins and components of the mucosal layer, which represents an adaptation to persist in this niche. A shotgun phage-display library of Lactobacillus casei BL23 was constructed and screened for peptides able to bind to fibronectin and collagen. Clones showing binding to these proteins were isolated, which encoded overlapping fragments of a putative transcriptional regulator (LCABL_29260), a hypothetical protein exclusively found in the L. casei/rhamnosus group (LCABL_01820), and a putative phage-related endolysin (LCABL_13470). The construction of different glutathione S-transferase (GST) fusions confirmed the binding activity and demonstrated that the three identified proteins could interact with fibronectin, fibrinogen, and collagen. The results illustrate the utility of phage display for the isolation of putative adhesins in lactobacilli. However, it remains to be determined whether the primary function of these proteins actually is adhesion to mucosal surfaces.

Isolation of Cysteine Protease Actinidin Gene from Chinese Wild Kiwifruit and its Expression in Escherichia coli

  • The actinidin (EC 3.4.22. 14) found in kiwifruit is a cysteine protease. In order to obtain the actinidin gene from the Chinese wild kiwifruit, primers were designed on the basis of the actinidin gene of Actinidia deliciosa, the New Zealand kiwifruit. The 1.2 kb DNA fragment was acquired from the total RNAs of Chinese wild kiwifruit via reverse transcription polymerase chain reaction (RT-PCR), and its DNA sequence was analyzed. Its sequence was determined to share 98.4% homology with the actinidin gene of A. deliciosa. In order to verity the actinidin gene isolated from the Chinese wild kiwifruit in Escherichia coli, the mature gene was amplified via PCR and expressed in E. coli under the control of the T7lac promoter. The actinidin was expressed in E. coli as inclusion bodies, which were solubilized with urea and refolded. The protease activity of the refolded protein was approximately twice as high as that of E. coli BL2l (DE3).

신경작용제 분해효소의 Soman 및 Sarin Gas 분해 능력 측정 및 제독응용 가능성 (Test of Degradation of Soman and Sarin Gas by Organophosphorus Acid Anhydrolase and Applicability of the Enzyme to the Development of Nerve Agent Decontaminant)

  • A gene expressing organophosphorus acid (OPA) anhydrolases have been cloned from Alteromonas haloplanktis strain and expressed in bacterial strain BL21. Crude extract was prepared from the transformed bacterial strain BL2l and used in testing its degrading capability of real nerve gas, soman and sarin. Within 1 minute after the start of the reaction, nearly 65% of the soman added to the reactant(3mM) was degraded by adding 1 mg of the crude extract enzyme(20.0 Unit $mg^-{1}$ crude protein). In 6 minutes, the reaction reached at its steady state, which indicates that soman was completely degraded by that time. In the case of sarin, the degradation efficiency was observed to be about 0.7 times of that of soman. If the specific activity of OPAA is enhanced by both increased expression efficiency and purification, OPAA seems to be applied for the development of decontaminant of skin, especially of eye.

Xanthomonas oryzae pv. oryzae로 부터 aspartate aminotransferase 유전자의 분리 및 생화학 특성 (Cloning and Biochemical Characterization of Aspartate Aminotransferase from Xanthomonas oryzae pv. oryzae)

  • Xoo로 부터 aspartate aminotransferase로 추정되는 유전자를 분리한 다음 발현시켜 생화학 특성을 조사하였다. 분리된 유전자는 His6 pET-21(a) 운반체에 삽입시켰으며 E. coli BL21(DE3)에서 발현시켰다. 재조합된 Asp-AT는 affinity chromatography를 이용하여 분리하였으며 SDS-PAGE분석에서 43kDa의 단일 밴드를 나타내었다. 분리된 효소는 amino donor 로서 L-aspartate에 대하여 효소활성도가 가장 높았고, L-leucine 및 L-cysteine에 대하여서도 상당한 활성도를 나타내었다. 효소의 최적 pH는 약 7.5 근처에서 나타났고 효소의 안정성은 산성조건 보다는 알칼리 조건에서 훨씬 높았다. 최적 온도는 약 $35-40^{\circ}C$로 나타났고 $55^{\circ}C$에서 20분간 열처리한 이후의 잔여 활성도는 약 78%로 나타났다. 여러 중금속 중에서 망간 이온에 의해 효소활성이 촉진되었다.
