Saudi Women's Interest in Breast Cancer Gene Testing: Possible Influence of Awareness, Perceived Risk and Socio-demographic Factors

  • Background: Development of effective educational strategies should accompany increases in public awareness and the availability of genetic testing for breast cancer (BC). These educational strategies should be designed to fulfill the knowledge gap while considering factors that influence women's interest in order to facilitate decision making. Objective: To determine the possible correlates of Saudi women's interest in BC genes testing including socio-demographics, the level of awareness towards BC genes, the family history of BC and the perceived personal risk among adult Saudi women in Al Hassa, Saudi Arabia. Subjects and methods: This cross-sectional study was carried out during the second BC community-based campaign in Al Hassa, Saudi Arabia. All Saudi women aged ${\geq}18$ years (n=781) attending the educational components of the campaign were invited to a personal interview. Data collection included gathering information about sociodemographics, family history of BC, the perceived personal risk for BC, awareness and attitude towards BC genes and the women's interest in BC genes testing. Results: Of the included women (n=599), 19.5% perceived higher risk for BC development, significantly more among < 40 years of age, and with positive family history of BC before 50 years of age. The participants demonstrated a poor level of awareness regarding the inheritance, risk, and availability of BC genetic testing. The median summated knowledge score was 1.0 (out of 7 points) with a knowledge deficit of 87.8%. The level of knowledge showed significant decline with age (> 40 years). Of the included women 54.7% expressed an interest in BC genetic testing for assessing their BC risk. Multivariate regression model showed that being middle aged (Odds Ratio 'OR'=1.88, confidence intervals 'C.I'=1.14-3.11), with higher knowledge level (OR=1.67, C.I=1.08-2.57) and perceiving higher risk for BC (OR=2.11, C.I=1.61-2.76) were the significant positive correlates for Saudi women interest in BC genetic testing. Conclusion: Saudi women express high interest in genetic testing for BC risk despite their poor awareness. This great interest may reflect the presence of inappropriate information regarding BC genetic testing and its role in risk analysis.

Heavy Metal Removal of Acrylic Acid-grafted Bacterial Cellulose in Aqueous Solution (아크릴산으로 그라프트된 미생물셀룰로오스의 수용액 내 중금속 흡착거동)

  • Electron beam-induced grafting polymerization was employed to prepare Acrylic acid-grafted bacterial cellulose (BC-g-AAc). BC-g-AAc as an adsorbent was applied to remove heavy metals (e.g., As, Pb, and Cd). This study examined followings; morphological change of surface, adsorptive behavior of BC-g-AAc, and interpretation of adsorptive kinetics. Specific surface areas of BC and BC-g-AAc were $0.9527m^2g^{-1}$ for BC and $0.2272m^2g^{-1}$ for BC-g-AAc, respectively as measured by BET nitrogen adsorption, revealing the morphological change of the surface of BC-g-AAc. Batch adsorption test was performed to investigate adsorptive behavior of BC-g-AAc in aqueous solution. The amounts of Pb and Cd adsorbed on BC-g-AAc were $69mg\;g^{-1}$ and $56mg\;g^{-1}$, respectively. However, As was not adsorbed on BC-g-AAc due to its neutral nature. Both the Benaissa model and the Kurniawan model were applied in the study to interpret adsorptive kinetics. From the value of correction coefficient ($R^2$), adsorptive kinetics of Pb and Cd were subjected to Kurniawan model referred to pseudo-second-order. Taken together, the results of this study show that BC-g-AAc has potential as a heavy metal (eg., Pb, Cd)-adsorbent made of an environmentally friendly material.

Increased Expression of the ${\alpha}_2$ Isoform of (Na,K)ATPase in the Differentiated Murine Muscle Cell Line BC3H-1 (BC3H-1 분화세포에서의 (Na,K)ATPase ${\alpha}_2$ isoform의 표현증대)

  • The development of the alpha2 isoform of (Na,K)ATPase which is high affinity ouabain receptors was studied in the differentiating nonfusing muscle cell line BC3H-1. T he differentiation process of BC3H-1 cell line was confirmed by 2-dexy-D-[$^3$H] glucose uptake experiment and the quantity of the expression of ${\alpha}_2$ isoform was measured using a whole cell [$^3$H] ouabain-binding assay. Undifferentiated growing BC3H-1 cells, myoblasts, exhibited low levels of insulin-stimulated glucose uptake and [$^3$H] ouabain-binding sites. In contrast, differentiated BC3H-1 cells, myocytes, had a 5.6-fold increase in insulin-stimulated glucose uptake and 5-fold increase in [$^3$H] ouabain-binding sites. Scatchard analysis showed that myocytes developed more [$^3$H] ouabain-binding sites than myoblasts vath a dissociation constant (kd) of 6${\times}10^{-8}$M and capacity of 6.l${\times}10^{-5}$ sites/cell. Therefore. it seems that myoblasts express low levels of ${\alpha}_2$ subunit and probably the majority of ${\alpha}_1$ subunit, whereas myocytes express high levels of ${\alpha}_2$ isoform. The results indicate that the expression of ${\alpha}_2$ isoform is developmentally regulated during differentiation and that BC3H-1 culture system provides an excellent model for the study of differentiation and mechanism of (Na,K)ATPase action in muscle which requires electrical excitability.

  • This study presents the Mid-Lateglacial vegetation history of southwestern Korea. From the result of AMS dating and pollen analysis, four local pollen assemblage zones(BS) were recognized. The four local pollen assemblage zones are BS 1. 12,222cal BC$\sim$9,160cal BC: cool-temperate northern/altimontane mixed coniferous & deciduous broad-leaved forest, BS 2. 9,160cal BC$\sim$ 4,210cal BC: cool-temperate central/montane deciduous broad-leaved forest, BS 3. 4,210cal BC$\sim$125cal BC: cool-Temperate southern/submontane mixed coniferous & deciduous broad-leaved forest. BS 4. BC125cal ${\sim}$present: warm-temperate/cool-temperate southern/submontane coniferous forest. Pine forest expanded since 4,210cal BC and full-scale rice cultivation might started since 2,120cal BC. A radical expansion of Pinus densiflora forest after postglacial stage might be caused by human impacts including full-scale agriculture.

Micro-computed tomographic evaluation of single-cone obturation with three sealers

  • Objectives: This study used micro-computed tomography (µCT) to compare voids and interfaces in single-cone obturation among AH Plus, EndoSequence BC, and prototype surface pre-reacted glass ionomer (S-PRG) sealers and to determine the percentage of sealer contact at the dentin and gutta-percha (GP) interfaces. Materials and Methods: Fifteen single-rooted human teeth were shaped using ProTaper NEXT size X5 rotary files using 2.5% NaOCl irrigation. Roots were obturated with a single-cone ProTaper NEXT GP point X5 with AH Plus, EndoSequence BC, or prototype S-PRG sealer (n = 5/group). Results: The volumes of GP, sealer, and voids were measured in the region of 0-2, 2-4, 4-6, and 6-8 mm from the apex, using image analysis of sagittal µCT scans. GP volume percentages were: AH Plus (75.5%), EndoSequence BC (87.3%), and prototype S-PRG (94.4%). Sealer volume percentages were less: AH Plus (14.3%), EndoSequence BC (6.8%), and prototype S-PRG (4.6%). Void percentages were AH Plus (10.1%), EndoSequence BC (5.9%), and prototype S-PRG (1.0%). Dentin-sealer contact ratios of AH Plus, EndoSequence BC, and prototype S-PRG groups were 82.4% ± 6.8%, 71.6% ± 25.3%, and 70.2% ± 9.4%, respectively. GP-sealer contact ratios of AH Plus, EndoSequence BC, and prototype S-PRG groups were 65.6% ± 29.1%, 80.7% ± 25.8%, and 87.0% ± 8.6%, respectively. Conclusions: Prototype S-PRG sealer created a low-void obturation, similar to EndoSequence BC sealer with similar dentin-sealer contact (> 70%) and GP-sealer contact (> 80%). Prototype S-PRG sealer presented comparable filling quality to EndoSequence BC sealer.

Production and Structural Analysis of Cellulose by Acetobacter sp. V6 Using Static Culture (정치배양을 이용하여 Acetobacter sp. V6의 셀룰로오스 생산 최적화 및 구조 분석)

  • The optimal medium compositions for the production of bacterial cellulose (BC) by a Acetobacter sp. V6, which was isolated from the traditionally fermented vinegar in Korea, were investigated in static cultures. The optimum medium compositions for BC production were 3% glucose, 3% soytone, 0.8% $K_2HPO_4$, and 0.4% ethanol, respectively. Adding $NaH_2PO_4$ or $KH_2PO_4$ had not shown the increase in BC production. Under the optimum medium compositions, the highest BC production was 44.67 g/$m^2$ in 8 days and the thickness of BC pellicle was about 1 cm. Structural properties of BC produced in the optimal medium were studied using Fourier-transform infrared spectroscopy and X-ray diffractometer. BC from the optimal medium was found to be of cellulose type I, the same as typical native cellulose. No difference in the compositions between bacterial and plant celluloses, but BC showed unique micro-network structure and high crystallinity (82%).

Nucleotide Sequence of Cellulolytic Xylanase Gene (bglBC2) from Bacillus circulans (Bacillus circulans 유래 cellulolytic xylanase 유전자(bglBC2)의 염기서열 결정 및 분석)

  • The nucleotide sequence of the cloned cellulolytic xylanase gene (bglBC2) from B. circulans ATCC21367 was determined. bglBC2 consists of an 1,224 bp open reading frame (ORF) coding for a polypeptide of 407 amino acids with a deduced molecular weight of 45 kDa. The Shine-Dalgarno (SD) sequence (5'-AAAGGAG-3') was found 9 bp upstream of the initiation codon, ATG. A promoter region corresponding closely to the B. subtilis consensus sequence (-35: TTGACA,-10: TATAAT) was detected, the putative -35 and -10 sequences of which were TTTACA and TATACT, respectively. The deduced amino acid sequence of the cellulolytic xylanase showed 97% homology with that of the alkaline $endo-\beta-1,4-glucanase$ from B. circulans KSM-N257, 75% homology with that of the $endo-\beta-1,3-1,4-glucanase$ from B. circulans WL-12, and 45% homology with that of the $endo-\beta-1,4-glucanase$ (cellulase) from Bacillus sp. KSM-330. The bglBC2 sequence was deposited in Gen-Bank under the accession number AY269256.

Effects of Phellinus linteus-Barley corn Water Extract in Triton WR-1339(TX)-Induced Hiperlipidemic Rats (상황버섯보리의 기능성에 관한 연구)

  • This study was designed to observe the effects of the Phellinus linteus-Barley corn water extract (PLBC) on the improvement of the blood biochemical aspects; serum AST, ALT, LDH, ALP, Phospholipid, Trigryceride, Total Cholesterol, HDL, LDL, of Triton WR-1339(TX)-induced hiperlipidemic rats. Experimental groups were designed to normal control group(Normal), hiperlipidemic control group(Control), hiperlipidemic control group treated with Barley corn water extract(1000mg/1kg ; BC), hiperlipidemic control group treated with Phellinus linteus-Barley corn water extract (500, 1000, 1500mg/1kg; PLBC-1, PLBC-2, and PLBC-3, respectively). Serum AST levels of BC and all PLBC treated groups were significantly decreased (p<0.005) compared to Control group. Serum ALT levels of BC and PLBC-2 groups were decreased (p<0.05), PLBC-1 and PLBC-3 groups were significantly decreased (p<0.005) compared to Control group. Serum LDH levels of BC group was decreased but no significant, PLBC-1 and PLBC-2 groups were decreased (p<0.05), PLBC-3 group was significantly decreased (p<0.025) compared to Control group. Serum ALP levels of BC and PLBC-1 groups were decreased (p<0.05), PLBC-2 group was decreased but no significant, PLBC-3 group was significantly decreased (p<0.005) compared to Control group. Serum Phospholipid levels of BC group was decreased but no significant, PLBC-1 group was decreased (p<0.025), PLBC-2 and PLBC-3 groups were significantly decreased (p<0.005) compared to Control group. Serum Triglyceride levels of BC group was increased but no significant, PLBC-1 group was decreased but no significant, PLBC-2 and PLBC-3 groups were significantly decreased (p<0.005) compared to Control group. Serum Total cholesterol levels of BC group was decreased (p<0.05), PLBC-1, PLBC-2 and PLBC-3 groups were significantly decreased (p<0.005) compared to Control group. Serum HDL levels of BC group was increased (p<0.05), PLBC-1 and PLBC-2 groups were significantly increased (p<0.005), PLBC-3 group was increased (p<0.025) compared to Control group. Serum LDL levels of BC, PLBC-1 and PLBC-2 groups were decreased but no significant, PLBC-3 group was significantly decreased (p<0.005) compared to Control group. These results suggested that Phellinus linteus-Barley corn water extract improve Triton WR-1339(TX)-induced hiperlipidemic symptom in rats.
