Antioxidant and skin whitening effects of Inonotus obliquus methanol extract (차가버섯 메탄올 추출물의 항산화 및 미백효과)

  • This study was initiated to investigate the skin whitening activities of methanol extracts from fruiting bodies of I. obliquus. The total polyphenols and flavonoids contents of I. obliquus methanol extracts were 31.85 mg/g and 28.33 mg/g, respectively. The methanol extract of the mushroom treated on B16/F10 melanoma and NIH3T3 cell lines did not show cytotoxic activity. 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical scavenging activity and chelating activity on ferrous ions of I. obliquus methanol extract were lower than those of positive control, tocopherol and BHT. The tyrosinase and L-DOPA inhibitory activities of the extract were lower than those of positive control, kojic acid and ascorbic acid. The tyrosinase and melanin synthesis inhibitory activities of the melanoma cells treated with the extract were comparable with positive control, arbutin. The experimental results suggested that methanol extract of I. obliquus contained inhibitory activities of tyrosinase and melanin synthesis in the B16/F10 melanoma cells by dose dependent manner. High ultra-violet absorption spectra in the range of 280-350 nm showed that I. obliquus extract could protect skin from UV radiation damage. Therefore, fruiting bodies of I. obliquus can be used for developing skin whitening, anti-UV and skin care agents.

  • Capsaicin (8-methyl-N-vanillyl-6-nonenamide), a pungent ingredient of hot chili peppers, has been reported to possess substantial anticarcinogenic and antimutagenic activities. In the present study. we investigated the effects of capsaicin on pulmonary metastasis of B 16-F10 murine melanoma cells. as well as its mechanism of action. Capsaicin (i.p.. 2.5mg/kg) suppressed the number of lung colonies (58%) in experimental lung metastasis assay. (omitted)

  • In the present study, we investigated the antitumor effects of Ginsenoside Rh2 and $\beta$-glucan using an experimental metastatic mouse model intravenously injected with B 16 melanoma F10 cells. Oral administration to various concentration of $\beta$-glucan (50mg/kg, l00mg/kg and 200mg/kg) dose-dependently reduced the lung-metastatic potential of metastatic BI6 melanoma F10 cells in syngenic mice. At same dose, Ginsenoside Rh2(50mg/kg) has more antitumor effect than $\beta$-glucan(50mg/kg). (omitted)

  • Ligularia stenocephala has been used as a traditional medicine for the treatment of asthma, arthritis, jaundice, and hyperpiesia. In this study, we investigated the anti-metastatic and hypnotic effects of the methanolic extract of L. stenocephala (MLS). Gelatin zymographic analysis revealed that MLS suppresses matrix metalloproteinase-2 (MMP-2) and MMP-9 activities in B16F10 cells. The gene expressions of MMPs were also down-regulated by MLS treatment in a dose-dependent manner. In addition, cancer cell invasion and migration were attenuated by MLS via suppression of NF-${\kappa}B$ activation. The in vivo lung metastasis of B16F10 melanoma cells was also inhibited by the treatment of MLS. These findings show that MLS has anti-metastatic properties, and, therefore, it might be applicable as a valuable anti-metastatic agent.

Nootkatol prevents ultraviolet radiation-induced photoaging via ORAI1 and TRPV1 inhibition in melanocytes and keratinocytes

  • Skin photoaging occurs due to chronic exposure to solar ultraviolet radiation (UV), the main factor contributing to extrinsic skin aging. Clinical signs of photoaging include the formation of deep, coarse skin wrinkles and hyperpigmentation. Although melanogenesis and skin wrinkling occur in different skin cells and have different underlying mechanisms, their initiation involves intracellular calcium signaling via calcium ion channels. The ORAI1 channel initiates melanogenesis in melanocytes, and the TRPV1 channel initiates MMP-1 production in keratinocytes in response to UV stimulation. We aimed to develop a drug that may simultaneously inhibit ORAI1 and TRPV1 activity to help prevent photoaging. We synthesized nootkatol, a chemical derivative of valencene. TRPV1 and ORAI1 activities were measured using the whole-cell patch-clamp technique. Intracellular calcium concentration [Ca2+]i was measured using calcium-sensitive fluorescent dye (Fura-2 AM). UV-induced melanin formation and MMP-1 production were quantified in B16F10 melanoma cells and HaCaT cells, respectively. Our results indicate that nootkatol (90 μM) reduced TRPV1 current by 94% ± 2% at -60 mV and ORAI1 current by 97% ± 1% at -120 mV. Intracellular calcium signaling was significantly inhibited by nootkatol in response to ORAI1 activation in human primary melanocytes (51.6% ± 0.98% at 100 μM). Additionally, UV-induced melanin synthesis was reduced by 76.38% ± 5.90% in B16F10 melanoma cells, and UV-induced MMP-1 production was reduced by 59.33% ± 1.49% in HaCaT cells. In conclusion, nootkatol inhibits both TRPV1 and ORAI1 to prevent photoaging, and targeting ion channels may be a promising strategy for preventing photoaging.

Antioxidative and antimelanogenic effects of ethyl acetate fractions of Korean domestic honeys from different floral sources (화종에 따른 국내산 벌꿀 에틸 아세테이트 분획물의 산화방지능 및 멜라닌 생성 억제 효과)

  • Honey is known to have a variety of health-promoting effects. Ethyl acetate fractions (EAFs) of four Korean domestic honeys from basswood, Korean raisin, chestnut, and acacia as well as a New Zealand manuka honey were analyzed comparatively to evaluate their antioxidative and antimelanogenic effects. The EAFs of basswood, Korean raisin, and chestnut honey had higher antioxidant capacities and tyrosinase inhibition activities than those of manuka honey. Pretreatment of B16F1 melanoma cells with EAFs at $100{\mu}g/mL$ resulted in relative retention of melanin contents as follows: acacia honey (141.0%)>basswood honey (134.3%)>manuka honey (131.5%)>Korean raisin honey (107.3%)>chestnut honey (88.0%). Intracellular tyrosinase activities of B16F1 melanoma cells were significantly (p<0.05) lowered by EAFs of Korean raisin and chestnut honey than by EAF of manuka honey. Consequently, these results suggest that Korean domestic honeys from different floral sources serve as good sources of antioxidative and antimelanogenic agents.

Effects of Banha Extract on the Melanin Biosynthesis and Tyrosinase mRNA Level in Bl6 Mouse Melanoma Cells (반하 추출물이 B-16 마우스 흑색종 세포의 멜라닌 생성과 타이로시네이즈 mRNA 양에 미치는 영향)

  • Melanin pigmentation in human skin is a major defense mechanism against ultraviolet light of the sun. Tyrosinase(EC 1.14.18.1) plays a key role in the biosynthesis of ultraviolet of melanin. This is why much researches have been focused on its regulation in controlling the epidermal melanization. We have found that the water-extract of Banha(Pinelliae ternate B.), an oriental medicinal plant, has no tyrosinase inhibitory activity, but does inhibit the melanin biolsynthesis in B16 mouse melanin cells. We also found that Banha extract lowers the tyrosinase activity in cultured cells. To elucidate the action mechanism of Banha extract we have investigated its effect on the tyrosinase mRNA level using reverse transcription-polymerase chain reaction technique. It was revealed that Banha extract reduced the tyrosinase mRNA level in dose dependent manner; when B16 mouse melanoma cells were cultured with 2mg/ml and 5mg/ml of Banha extract, there were 20% and 44% decrease in tyrosinase mRNA level, respectively. These data suggest that the Banha extract exerts its melanogenic inhibitory effect through the transcriptional regulation of tyrosinase mRNA.

  • The diterpene acid 1 was isolated from the roots of Anisotome lyallii(Apiaceae/Umbelliferae). The structure of the compound was elucidated as anisotomenoic acid 1 on the basis of spectroscopic method. This compound was evaluated against p388 murine leukaemia and B16/F10 melanoma cells.

Effect of Lindera obtusiloba extract on cancer metastasis (생강나무 추출물의 암전이 억제효과)

  • Objectives : In the present study, anti-metastatic properties of the methanol extract of L. obtusiloba (MLO) were evaluated. Methods : To determine the effect of MLO on cancer metastasis, we checked matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) activities and expressions in B16F10 melanoma cells. In addition, we performed cell migration assay as well as invasion assay using Matrigel. Finally, we used an in vivo lung metastasis model to confirm the anti-metastatic activity of MLO. Results : 1. MLO showed potent inhibitory effects on MMP-2 and MMP-9 activities and expressions via down-regulation of activation of NF-${\kappa}B$ in B16F10 melanoma cells. 2. Melanoma cell migration and invasion were down-regulated by MLO treatment. 3. Not only in vitro model, but MLO also significantly suppressed lung metastasis in vivo. Conclusions : The present results indicate that MLO has strong inhibitory effect on cancer metastasis. Therefore, L. obtusiloba could be a valuable anti-metastatic agent.

Marked Expansion of CD11c+CD8+ T-Cells in Melanoma-bearing Mice Induced by Anti-4-1BB Monoclonal Antibody

  • 4-1BB (CD137), a member of the tumor necrosis factor receptor superfamily, is expressed on activated T-cells, and 4-1BB signaling due to interaction with 4-1BB ligand or ligation with anti-4-1BB monoclonal antibody (mAb) costimulates T cells. It has been shown that administration of anti-4-1BB mAb induces anti-tumor immunity in mice, but the nature of the cellular subsets responsible for this immunity is uncertain. In this study we found that anti-4-1BB mAb administration to B16F10 melanoma-bearing mice induced marked expansion of $CD11c^+CD8^+$ T-cells in parallel with suppression of pulmonary tumors. The mAb-treated mice produced higher levels of $IFN-{\gamma}$ in their tumor tissues, spleen and lymph nodes than mice exposed to control antibody. When the $CD11c^+CD8^+$ T-cells were purified and re-stimulated in vitro, they produced high levels of the Th1 cytokines, $IFN-{\gamma}$ and IL-2, but low levels of the Th2 cytokines, IL-4 and IL-10. Furthermore, they expressed high levels of 4-1BB and CD107a, a marker of activated cytotoxic T-lymphocytes. Our results suggest that $CD11c^+CD8^+$ T-cells play a role in the anti-tumor immunity induced by anti-4-1BB mAb.
