Inhibitory Effect of Agaricus Mixed Prescription on Metastasis and Tumor Formation

  • Antimetastatic effects of Agaricus mixed prescription (AMP) were studied in the respect of blood-borne metastasis. For this aim, cytotoxicity against various cancer cells and normal cells, Chicken Chorioallantoic membrane (CAM) assay, cancer cell adhesion assay, platelet aggregation assay, pulmonary colonization, life span of S-180 implanted mice, and cytokine release assay were evaluated, respectively. The results were summarized as follows; AMP did not exert any cytotoxicity against all cell lines with IC50 of 25mg/ml on B16BL6. AMP disrupted formation of CAM at 1mg/ml. AMP was suppressive in adhesion assay of B16BL6. AMP also inhibited tumor induced platelet aggregation. In pulmonary colonization assay by B16BL6, the number of colonies in the lungs was significantly decreased in sample group than in control group. In animal study with S-180, the life span of AMP treated group was extended than that of control group. IL-12 was effectively increased in AMP treated group in cytokine release assay. Taken together, AMP can be possibly applied to cancer or metastasis.

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata THUNB extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 $\mu$g/$m\ell$ concentration of methanol extract (63.81 %) of Houttuynia cordata THUNB was shown significantly antitoxic activity on A549 cell lines. (omitted)

Different Immunology Mechanisms of Phellinus igniarius in Inhibiting Growth of Liver Cancer and Melanoma Cells

  • To assess inhibition mechanisms of a Phellinus igniarius (PI) extract on cancer, C57BL/6 mice were orally treated with PI extractive after or before implanting H22 (hepatocellular carcinoma ) or B16 (melanoma) cells. Mice were orally gavaged with different doses of PI for 36 days 24h after introduction of H22 or B16 cells. Mice in another group were orally treated as above daily for 42 days and implanted with H22 cells on day 7. Then the T lymphocyte, antibody, cytokine, LAK, NK cell activity in spleen, tumor cell apoptosis status and tumor inhibition in related organs, as well as the expression of iNOS and PCNA in tumor tissue were examined. The PI extract could improve animal immunity as well as inhibit cancer cell growth and metastasis with a dose-response relationship. Notably, PI's regulation with the two kinds of tumor appeared to occur in different ways, since the antibody profile and tumor metastasis demonstrated variation between animals implanted with hepatocellular carcinoma and melanoma cells.

Cannabidiol Induces Cytotoxicity and Cell Death via Apoptotic Pathway in Cancer Cell Lines

  • In view of obtaining potential anticancer compounds, we studied the inhibitory activity and the cytotoxic effects of a candidate compound in cancer cells. The cytotoxic effects of cannabidiol (CBD) in vitro were evaluated in NIH3T3 fibroblasts, B16 melanoma cells, A549 lung cancer cells, MDA-MB-231 breast cancer cells, Lenca kidney cells and SNU-C4 colon cancer cells. The cells were cultured in various concentrations of CBD for 48 h and 25 ${\mu}$M of CBD for 6-36 h. The cells were observed to exhibit inhibitory effects of the cell viability in their growth, and then cytotoxicity was estimated. The inhibitory activity of CBD was increased in all cancer cells and showed especially strong increment in breast cancer cells. The cytotoxicity of CBD increased in a dose- and time-dependent manner with growth inhibition in all cancer cell lines. Also, to assess the membrane toxicity induced by CBD, we investigated lactate dehydrogenase (LDH) release. After treatment with various concentrations of CBD, LDH release rate of cancer cells was accelerated. On the other hand, in the induction of cell death, caspase-3, -8 and -9 activations were detected in cancer cells after treatment with various concentrations of CBD, and CBD effectively induced activity of caspase-3, -8 and -9 in A549 lung cancer cells, MDAMB-231 breast cancer cells and Renca kidney cells. Therefore these results suggest that CBD has a possibility of anticancer agents and anticancer effects against cancer cells by modulation of apoptotic pathway in the range of 5-80 ${\mu}$M concentration.

금은화(金銀花) 열수 추출물이 피부 미백(美白)에 미치는 영향 (Effects of Hot Water Extracts from Lonicera Japonica Flos Extracts on whitening using B16F10 cell lines)

  • Objectives : This study was designed to investigate effects of Lonicera Flos Extracts(LFE) on whitening using B16F10 cell lines. Methods : In this experiment, we observed effects of LFE on cell viability, inhibitory effects of melanin synthesis, inhibitory effect on tyrosinase, tyrosinase activity, superoxide dismutase(SOD)-like activity and mRNA expression. Results : In results, more than $2000{\mu}g/ml$ of LFE treated group showed lowered cell viability rates significantly compared to albutin treated group. More than $2000{\mu}g/ml$ of LFE treated groups were lower levels of melanin synthesis. Inhibitory effects of melanin production showed in $1000{\mu}g/ml$ of LFE treated group. $1,000{\mu}g/ml$ of LFE treated group significantly suppressed tyrosinase activities in vitro. LFE and albutin treated group significantly decreased tyrosinase activity compared to non treated group. SOD-like activity of LFE treated group was lower than vitamin C treated group but increased depending on concentration. $500{\mu}g/ml$ of LFE treated group and $1,000{\mu}g/ml$ of LFE treated group was significantly increased. Tyrosinase mRNA expression of ${\alpha}$-MSH and LFE $250{\mu}g/ml$ treated group significantly decreased compared to ${\alpha}$-MSH treated group. Conclusions : These results suggest that LFE can inhibit melanin synthesis through inhibitory action on tyrosinase activity. And LFE suppressed tyrosinase activities B16F10 cells significantly. So I suggest LFE can apply to whitening.

세포배양에 의한 고려인삼 성분의 생산연구 (The Production of Ginseng Saponins with the Cell Culture of Korean Ginseng Plant)

  • Panax ginseng root has been widely used as an important drug for thousands years in China, Korea and Japan. The main effective components of ginseng have been believed to be saponins. However, ginseng cultivation is very difficult and needs many years for growth. It has already been shown that Panax ginseng callus produces a considerable amount of the same kinds of saponins as in intact plants. Various culture conditions were examined for increased production of ginseng saponins by cell culture. The saponin contents and the growth rates in two cell lines of ginseng callus were compared in static and suspension cultures, rotary and reciprocal shaking cultures. It was shown that the growth rate in rotary shaking cultures of D5-B2K-B2K callus was the highest and ginseng saponin production was most effective in reciprocal cultures of D5-B2K-B2K callus. The saponin content per fresh weight of the culture was 1.03 times higher than that of the fresh ginseng root.

생맥산가미방 추출물이 멜라닌 생합성 저해 효과와 SOD 활성에 미치는 연구 (Study of ShengmaisanJiaweifang Extracts on the Inhibitory Effects of Melanin Synthesis and Superoxide Dismutase Activity)

  • This study aims to evaluate the effects of Shengmaisan (SMS) and three types of ShengmaisanJiaweifang on the inhibitory effect of melanin synthesis in B16F10 cells, the mechanism of action through tyrosinase, and the antioxidant effect through superoxide dismutase (SOD) activity. In this study, we used ShengmaisanJiaweifangs (SMS, SMSRR, SMSAD, SMSAR) to research the whitening effects in B16F10 cell lines. Shengmaisan (SMS) was a herbal medicine composed of Ginseng Radix, Liriopis Tuber, and Schisandrae Fructus. ShengmaisanJiaweifangs included SMSRR (SMS added with Rehmanniae Radix), SMSAD (SMS added with Asparagi Radix) and SMSAR (SMS added with Astragali Radix). We measured the cell viability, the inhibition rate of the melanin biosynthesis, and the activity of tyrosinase and SOD in malignant melanoma, B16F10 cells, to survey the whitening effect and the mechanism of the impact on the sample. As a result, SMSRR significantly suppressed the cell viability of B16F10 at more than $500{\mu}g/m{\ell}$ and significantly inhibited the generation of melanin induced by ${\alpha}$-MSH at more than $250{\mu}g/m{\ell}$. SMSRR ($500{\mu}g/m{\ell}$) decreased the activity of tyrosinase while increased the activity of SOD. Therefore, we considered that the SMSRR would be able to produce high value-added products more SMS if used as a commercial.

Baicalein Inhibits the Migration and Invasion of B16F10 Mouse Melanoma Cells through Inactivation of the PI3K/Akt Signaling Pathway

  • Baicalein, a natural flavonoid obtained from the rhizome of Scutellaria baicalensis Georgi, has been reported to have anticancer activities in several human cancer cell lines. However, its antimetastatic effects and associated mechanisms in melanoma cells have not been extensively studied. The current study examined the effects of baicalein on cell motility and anti-invasive activity using mouse melanoma B16F10 cells. Within the noncytotoxic concentration range, baicalein significantly inhibited the cell motility and invasiveness of B16F10 cells in a concentration-dependent manner. Baicalein also reduced the activity and expression of matrix metalloproteinase (MMP)-2 and -9; however, the levels of tissue inhibitor of metalloproteinase-1 and -2 were concomitantly increased. The inhibitory effects of baicalein on cell motility and invasiveness were found to be associated with its tightening of tight junction (TJ), which was demonstrated by an increase in transepithelial electrical resistance and downregulation of the claudin family of proteins. Additionally, treatment with baicalein markedly reduced the expression levels of lipopolysaccharide-induced phosphorylated Akt and the invasive activity in B16F10 cells. Taken together, these results suggest that baicalein inhibits B16F10 melanoma cell migration and invasion by reducing the expression of MMPs and tightening TJ through the suppression of claudin expression, possibly in association with a suppression of the phosphoinositide 3-kinase/Akt signaling pathway.

ginsenoside Rg3에 의한 B16F10 흑색종 세포의 세포사멸 유도 (Ginsenoside Rg3 Induces Apoptosis in B16F10 Melanoma Cells)

  • Ginsenoside Rg3는 홍삼으로부터 추출한 활성 성분들 중 하나로 한방 의학에선 원기를 회복시키는 약제로 잘 알려져 있는 인체에 유효한 화학 성분이다. Rg3는 지금까지 많은 연구들에 의하여 다양한 암세포로부터 강력한 항암효과를 가진다고 알려져 있다. 그러나 Rg3가 악성 흑색종 세포에서 어떻게 세포사멸을 유도하는지에 대한 작용 기작은 명백하게 밝혀지지 않았다. 따라서, 본 연구에서는 ginsenoside Rg3가 B16F10 흑색종 세포에서 세포 사멸 유도 활성 및 기전에 관한 영향을 조사하였다. 세포 생존력을 MTT assay 법으로 수행한 결과, B16F10 세포에선 농도 의존적으로 세포증식 저해 효과가 나타났고 정상세포인 EA.hy.926 과 NIH3T3 에서는 나타나지 않았다. B1610 세포에 Rg3를 농도 별로 처리 후, TUNEL 염색을 한 결과 세포사멸이 농도 의존적으로 증가 하는 것을 확인 할 수 있었다. Western blot 분석을 실시한 결과, Rg3를 처리한 B16F10 세포에서 p-FAK, Bcl-2, pro-caspase3 단백질들의 발현이 감소 되었고 이와 반대로 Bax, p-p38의 발현은 증가되었다. 따라서, 본 연구에서는 Rg3가 B16F10 흑색종 세포에서 항암제의 agent로써 사용 될 수 있다는 것을 입증하였다.

Effects of N-acetylphytosphingosine on melanogenesis of B16F10 murine melanoma cells.

  • The effects of N-acetylphytospingosine(NAPS), one of the phytospingosine derivatives, on melanogenesis of B 16F 1 0 mouse melanoma cell lines were investigated. We assessed the effect of NAPS on the depigmentation of B16F10 cells. The melanin content of cells was significantly reduced by NAPS. We examined the inhibitory effect of NAPS on tyrosinase activity using L-dopa as a substrate and the results showed that tyrosinase activity was inhibited in a does-dependent manner. The mRNA level of tyrosinase as well as that of tyrosinase related protein-l (TRP-l) and tyrosinase related protein-2 (TRP-2) genes were not affected by NAPS based on a reverse transcription-polymerase chain reaction (RT-PCR) assay. We also performed a Western blotting analysis using anti-tyrosinase antibody. It showed that there is no change in tyrosinase protein level after treatment of NAPS. These results suggest that the depigmenting mechanism of NAPS in B16F10 melanoma cells involves inhibition of melanosomal tyrosinase activity, rather than the mRNA expression or protein level of tyrosinase.
