On the Breeding of “CSR18${\times}$CSR19”- A Robust Bivoltine Hybrid of Silkworm, Bombyx mori L. for the Tropics

  • Earlier breeding experiments undertaken at Central Sericultural Research and Training Institute, Mysore, India since a decade had resulted in the development of many productive and qualitatively superior bivoltine hybrids. However, the hot climatic conditions of tropics prevailing particularly in summer are not conducive to rear these high yielding bivoltine hybrids. This has necessitated breeding of compatible bivoltine hybrids for year-round rearing. Accordingly, the Japanese hybrid, B2Ol ${\times}$ BCS12 which was found to be tolerant to high temperature was used as breeding resource material. Following hybridization and selection rearing of silkworms was taken up in SERICATRON (Environmental chamber with precise and automatic control facilities for uniform maintenance of temperature and humidity) at high temperature of $36{\times}1^{\circ}C$ and 85${\times}$5% RH in fifth instar and the control batches at $25{\times}1^{\circ}C$ and 65{\times}$5% RH. Directional selection was resorted to the batches reared at 36$\pm$1$^{\circ}C$ till F$_{5}$ keeping pupation rate as important selection criteria. From $F_{2}% onwards the experiment was modified in such a way as to conduct normal rearing every alternate generation to regain the lost vitality due to continuous exposure to high temperature and high humidity stress. At $F_{2}$ , Oval and dumb-bell cocoons were separated out and designated as CSR18 and CSR19, respectively. By utilizing these lines at $F_{12}$, the hybrid CSR18$\times$CSR19 was prepared and studied for the thermotolerance by subjecting to stress condition at high temperature of 36$\pm$1$^{\circ}C$ and 85$\pm$5% RH in fifth instar and the control batches at $25{\times}1^{\circ}C$ and 65${\times}$5% RH. The better performance of CSR18${\times}$CSR19 (survival > 88%) at $36{\times}1^{\circ}C$ clearly indicates the general superiority of CSR18${\times}$CSR19 with regard to high temperature tolerance over the productive hybrids and CSR18$\times$CSR19 can perform well in varied agro-climatic conditions of the tropics with optimum qualitative and quantitative characteristics.s.

Characterization and application of the proteins isolated from edible insects (식용곤충 분리단백의 특성 및 활용연구)

  • Proteins were isolated from four species of edible insects, Bombyx mori, B; Tenebrio molitor, T; Protaetia brevitarsis, P; and Gryllus bimaculatus, G, and their properties were analyzed and compared with those of whey proteins. The yield of the protein isolated from raw materials was 65.0-75.0%. Among three branched chain amino acids (BCAA), the amounts of valine in all the insects were similar to that of the whey protein. The BCAA content was 16.8 and 16.4% in G and T, respectively. The total polyphenolic content and the antioxidant activity of the insect proteins were significantly higher than those of the whey proteins. Based on the sensory test of the protein powder, the whey protein could be replaced by 25% of the isolated protein from the insects, but the taste and flavor needs to be improved. Although the BCAA content of insect proteins was lower than that of the whey protein (by 73-76%), their application could be useful because of high polyphenolic content and the anti-oxidative activity.

Characterization of Microbial Pathogen Bacillus thuringiensis Isolates from Soil Against Mosquito and Silkworm Larvae (II) (토양에서 분리한 살충성 Bacillus thuringiensis의 모기와 누에 유충에 대한 독성효과 (II))

  • Eight strains of Bacillus thuringiensis were isolated from soil in Korea and characterized. The isolates were named HL-24, HL-25, HL-33, HL-34, HL-35, HL-38, HL-39, HL-40. Strains HL-24 and HL-25 produced irregular parasporal crystals, HL-33 and HL-35 produced bipyramidal crystals, and others were round form in their cells. The biochemical characteristics of the eight isolates were only minor different in specific characteristics to the known serotypes of Bacillus thuringiensis. The HL-25, HL-33 and HL-34 strains showed resistances to cephalothin, colistin and penicillin G, and HL-39 and HL-40 strains were resistant to penicilin G. The strains of HL-24, HL-25, HL-33 and HL-34 were toxic to Bombyx mori lavae and HL-24, HL-25, HL-38, HL-39 and HL-40 strains killed Culex pipiens 3rd instar larvae. The HL-24 and H25 strains showed lethal activity against two kinds of the larvae, however lethality against mosquito larvae was low.

Effect of Silkworm Hemolymph Collected Large-scale Bleeding Method in Insect Cell Culture (절피원심법으로 대규모 채취한 누에체액의 곤충세포 증식효과)

  • This study was carried out to investigate the utilization of hemolymph of silkworm, Bombyx mori, as a substitute for fetal bovine serum(FBS) in the insect cell culture. Hemolymph is collected on a small scale by clipping the abdominal leg; however, this method is not appropriate for large scale collection. The hemolymph was collected from 5 th instar larva by centrifugation after cutting of the abdominal legs was more appropriate procedure for large scale collection. The cell growth in the medium supplemented with hemolymph(Baekokjam) collected in large scale was almost same as that in the medium hemolymph supplemented with hemolymph collected in small scale. However, the mutant($wE^b$) hemolymph collected in large scale was still less effective in the cell growth, as compared to the Baekokjam hemolymph collected in large scale. The optimum centrifugation condition for large-scale bleeding was 500 rpm and 15 min.

A Study on the Development of an Immune Related Genes from Midgut of Silkworm (누에 중장유래 생체방어 관련 유전자 개발 연구)

  • This study was aimed for identification of a useful genetic resources from the entomopathogenic bacteria infected-midgut of the silkworm, Bombyx mori L. We analyzed the appropriately midgut-immunizing condition of $4^{th}$ instar larvae by a feeding infection using several entomopathogenic bacteria. Xenorhabdus nematophila was selected as a suitable bacteria for midgut immunization of Jam 123, B. mori. We constructed a subtraction cDNA library from the mRNA of the immunized midgut, respectively. A total of 1,000 clones were randomly selected from the subtracted cDNA library, and then performed a differential display hybridization analysis with forward and reverse probes. In conclusion, nine clones were identified as differential expressed genes, which presumed that these genes were involved in gut immunity of silkworm. The total number of clones analyzed in this work is not enough to have a brief overview of a understanding on the midgut immunity factors of silkworm. Therefore, further defined studies on these molecules biological roles will give us well-fined information about the innate immune mechanism of silkworm.

Characteristics of Tenebrio molitor larvae and Bombyx mori pupae sequentially fermented with Aspergillus oryzae and Bacillus subtilis (Aspergillus oryzae와 Bacillus subtilis를 순차접종 발효한 갈색거저리유충과 누에번데기 발효물의 품질특성)

  • Defatted soybean, larvae of brown mealworm (Tenebrio molitor), and powdered pupae of silkworm (Bombyx mori) were fermented in solid and liquid forms using Aspergillus oryzae and Bacillus subtilis. The protein degradation rate (NDR) through solid fermentation was the highest in the fermented soybean control sample (54.69±6.54%), followed by silkworm pupae (34.82±5.99%) and brown mealworm larvae (30.54±3.80%). When these edible insects were fermented in liquid form, solid extraction yield was 37.73-46.88%, and protein yield was 47.47-63.02%. NDR of fermented liquid form products increased to 58.90, 52.62, and 50.13% for soybean, brown mealworm larvae, and silkworm pupae, respectively. SDS-PAGE of the liquid fermented products confirmed that microbial fermentation decomposed higher-molecular-weight proteins into small polypeptides. In vitro digestibility of liquid forms of edible insects increased by 1.26 to 1.53 times after fermentation. The protein solubility, foaming ability, and foam stability of liquid-fermented edible insects all tended to increase through fermentation.

Selection of Mutant Silkworm with Oxidation-deficient Haemolymph for Insect Cell Culture (곤충세포 배지 개발을 위한 체액산화지연 돌연변이 누에계통 선발)

  • Insect cell culture system has been demonstrated the effective means of producing medical and agricultural products. Furthermore, Fetal bovine serum (FBS) is in wide use in insect cell culture. Silkworm hemolymph was tested to develop as a substitute for FBS and was effective in insect cell growth. Hemolymph is oxidized and darkens visibly during the collection from silkworms due to the activity of tyrosinase in it. Toxic quinones are produced by the oxidation and consequently inhibit the cell growth. Heat treatment can be used to prevent the oxidation; however, the oxidation may occur during the collection of hemolymph before it is heat-treated. Hemolymphs collected from 257 different strains of silkworms were examined to select the slowly oxidized hemolymphs. Hemolymphs collected from mutant strains such as $Y_4$, TBO and $wE^b$ showed relatively slow color changes. Oxidation rates of the hemolymphs were measured by the absorbance change using a spectrophotometer. The absorbance of mutant hemolymph reached the saturation value at $20^{\circ}C$ in each 330 min ($Y_4$), 360 min (TBO) and 450 min ($wE^b$) min, whereas the total oxidation time of the wild-type (Baekokjam) hemolymph at the same temperature was 120 min. The cell growth in the medium supplemented with mutant species hemolmph was more effective that in the medium supplemented with Baekokjam species hemolymph.

Electrophoretic Patterns of Hemolymph Proteins of Varieties with Long and Short Life Span in the Silkworm Bombyx mori L. (${\cdot}$단명 누에 품종의 체액단백질의 전기영동상)

  • Hemolymph protein patterns of silkworms in terms of short and long life span were analyzed by native- and SDS-polyacrylamide gel electrophoresis (PAGE) with the developmental stages. From the native-PAGE patterns of silkworm major hemolymph proteins there were varietal differences on the first day of the pupal stage and were classified into there groups MHP-a, b and c SA 10, JAM109 and J037 were grouped into MHP-Ⅰ, Hangang, Chungmun and Daizo(sdi) into MHP-Ⅱ and NTZN, Sulak, Qoichuk and PR varieties into MHP-Ⅲ group. It was found that the MHP in each group revealed similar patterns and changes with development of pupal stage. In the first day of adult, MHP-c was clearly detected in both female and male of the Daizo(sdi), but not in the J037, indicating that there was significantly varietal differences in electrophoretical protein pattern. In addition, the results of protein pattern of hemolymph by SDS-PAGE showed also varietal differences in the concentration of hemolymph protein.

Effects of a Natural Medicinal Multi-plant Extract on Blood Glucose, Insulin Levels, and Serum Malondialdehyde Concentrations in Streptozotocin-induced Diabetic Rats (천연 생약 복합 추출물이 Streutozotocin 유발 당뇨성 흰쥐의 혈당 및 인슐린, 혈청 과산화지질 농도에 미치는 효과)

  • This study was designed to investigate a natural medicinal multi-plants extract (BG515), which consisted of multi extracts of Mori folium, Rehmannia glutinosa Liboschitz, Dioscorea japonica, Lycii fructus, and Astragalus radix, on blood glucose, insulin levels, and serum malondialdehyde (MDA) concentrations in streptozotocin-induced diabetic rats. Streptozotocin (STZ) induces a type 1 diabetes mellitus in rats. Type 1 is usually characterized by the presence of islet cell autoantibodies (ICA), autoantibodies to insulin (IAA), and autoantiboides to glutamic acid decarboxylase (GAD), which identify the autoimmune process that leads to $\beta-cell$ destruction. Thirty-five male Sprague-Dawley (SD) rats weighing $150\sim170g$ each (6 weeks old) were randomly divided into one control (Group A) and 4 STZ-induced diabetic groups, and were subjected to one of the following treatment for 12 weeks. Groups A and B were fed basal diets and Group C, D, and E received the same diets as groups A and B, but with supplements of 150 mg/kg, 300 mg/kg, and 600 mg/kg of BG515 orally for 12 weeks, respectively. Diabetes was induced in Groups B, C, D, and E by intravenous injection of 45 mg/kg of STZ per body weight in sodium citrate buffer (pH 4.5) via the tail vein. In the BG515 groups, we found increases in serum insulin levels, compared to the STZ-control group, but these data were not significant. In contrast, blood glucose and serum MDA concentrations decreased in the BG515 groups compare to the STZ-control group. At the 5th week, in all the BG515 administered groups, there were decreases in serum blood glucose levels compared to the STZ- control group, and this activity was very strong in the BG515-1 group at the 12th week. These results suggest that natural bio-complex compounds (BG515) may slightly suppress STZ-induced changes in serum MDA concentration via the maintenance of serum insulin levels, due to the prevention of $\beta-cell$ and glucagon destruction by STZ.

AKARI MID- TO FAR-INFRARED OBSERVATIONS OF DIFFUSE GALACTIC EMISSION

  • We have collected dozens of mid-infrared spectra showing UIR bands from diffuse Galactic emitting regions with the AKARI's Infrared Camera (IRC) onboard AKARI, as part of the ISMGN Mission Program. The datasets cover various directions in the inner Galactic Plane ($|l|$ < 70 deg), in the outer Galactic Plane ($|l|$ > 70 deg), and in the off-Plane ($|b|$ > 2 deg). The variations in the UIR band ratios are examined in terms of the radiation environments judged from the far-infrared ($50-170{\mu}m$) spectral energy distribution (SED) made with AKARI/FIS All Sky Survey data at each slit position where mid-IR spectra were obtained. We have found that the band ratios of $6.2{\mu}m/11.2{\mu}m$ and $7.7{\mu}m/11.2{\mu}m$ toward the inner Galaxy are systematically higher than those toward the outer Galaxy and off the Galactic plane. Likely causes of the variations in properties of UIR bands in diffuse emission on a Galactic scale are discussed in this paper.
