Enhanced Natural Purification of Crude Oil Contaminated Tidal Flat (원유로 오염된 갯벌 지역의 자연정화 기능 향상 기술의 개발)

  • Tidal flats which are ecologically sensitive, are hard to remediate once they are contaminated by oil spill accidents. Traditional oil remediation measures focus on removal efficiency, and their improper implementation can adversely affect crude oil contaminated coastal areas and greatly disrupt the structure and functions of crude oil contaminated tidal flats. In this study, the oil degradation due to the implementation of remediation measures naturally enhanced using air and natural oil sorbents was evaluated in the lower strata of tidal flats. The effects of air and natural oil sorbents on oil degradation for two concentration levels (< 500 ppm and > 500 ppm) were tested at artificially contaminated tidal flats. Fifty days after these treatments, the natural oil sorbent treatment showed the lowest total petroleum hydrocarbon (TPH) concentration ($4.46{\pm}1.47%$) at the low concentration level, whereas both air and natural oil sorbent treatments showed high degradation efficiencies at the high concentration level ($29.30{\pm}4.39%$). Although the phosphatase activity decreased for all treatments, there was no significant difference between the decreases for the different treatments; on the other hand, B-glucosidase activities were high for both air and natural oil sorbent treatments. Although degradation efficiencies decreased as the concentration increased, the air provision and natural oil sorbent treatment could be an effective ecological restoration measure for oil contaminated tidal flats while minimizing the environmental impact of the remediation efforts.

Purification of ginseng rare sapogenins 25-OH-PPT and its hypoglycemic, antiinflammatory and lipid-lowering mechanisms

  • Background: Panax ginseng Meyer has been used as a nourishing edible herb in East Asia for thousands of years. 25-OH-PPT was first discovered as a natural rare triterpenoid saponin in ginseng stems and leaves by our group. Research found that it showed strong inhibitory effects on α-glucosidase and protein tyrosine phosphatase 1B, and protected cardiocytes (H9c2) through PI3K/Akt pathway. Methods: In the research, in order to optimize the 25-OH-PPT enrichment process, optimal macroporous resins and optimal purification conditions were studied. Meanwhile, the hypoglycemic effect and mechanism of 25-OH-PPT were evaluated by using STZ to establish insulin-dependent diabetic mice and the spontaneous type 2 diabetes DB/DB mice. Results and Conclusion: Research found that 25-OH-PPT can reduce blood glucose and enhance glucose tolerance in STZ model mice. It increases insulin sensitivity by upregulating GLUT4 and AMPK in skeletal muscle, and activating insulin signaling pathways. In DB/DB mice, 25-OH-PPT achieves hypoglycemic effects mainly by activating the insulin signaling pathway. Meanwhile, through the influence of liver inflammatory factors and lipids in serum, it can be seen that 25-OH-PPT has obvious anti-inflammatory and lipid-lowering effects. These results provide new insights into the study of ginseng as a functional food.

Evaluation of Immunomodulatory and Biological Effects of Aquilaria crassna Extracts (침향 추출물의 면역조절 및 생리활성 분석)

  • Objectives : Aquilaria crassna is a traditional herbal medicine, which is used to treat allergies, diabetes, neurological diseases. Recently, Aquilaria crassna extracts have been reported in anti-bacterial and anti-inflammatory activities. In this study, various solvents fraction of Aquilaria crassna were investigated on various physiological activities. Methods : According to the polarity, the solvents fraction of Aquilaria crassna were confirmed through TLC, and the activities of the extracts were confirmed in anti-diabetes, anti-obesity, whitening, anti-gout, and anti-inflammation. Results : TLC results showed that ACM and ACM/E have similar patterns and most of the components were transferred to ACM/E. Treatment with ACM and ACM/E fraction were significantly decreased the generation of NO in lipopolysaccharide (LPS)-stimulated macrophage cells. Analysis of biological activities such as α-glucosidase, protein tyrosine phosphatase (PTP1B), tyrosinase, xanthine oxidase (XO) and pancreatic lipase inhibition, showed that ACM and ACM/E have more inhibitory effects than other fractions. Conclusions : Therefore, the results of the present study clearly demonstrate that Aquilaria crassna and its constituents might be beneficial in the prevention or treatment of immune-regulating effects.

A Studies on the Chemical Composition and in vitro Biological Activities of a Hot Water Extracts of Gastrodia elata (천마추출물의 성분분석 및 in vitro 생물활성에 관한 연구)

  • A hot water extract was prepared from the artificially grown Gastrodia elata to investigate its chemical composition and various in vitro biological activities as an effort to develop G. elata as health/functional food materials. The contents of crude protein, ash, fat, fiber, moisture and total sugar were 5.4, 2.6, 3.6, 3.3, 8.1 and 77% (w/w), respectively. The extract of G. elata had greater amount of potassium (1,150 mg/100 g) than phosphorus (300 mg/100 g). Dose-dependence against human carcinoma (Hep3B, MCF-7, A549 and AGS) were observed from 0.2 mg/ml to 1.0 mg/ml. Especially, the treatment of 1.0 mg/ml extracts showed the highest cytotoxicity with 83% against gastric carcinoma (AGS). The extracts showed weak antimicrobial activities against Bacillus subtilis and Pseudomonas aeruginosa, but practically no antimicrobial activity against the other microorganisms tested. The effect of ${\alpha}$-glucosidase inhibition was 64% at the concentration of 1.0 mg/ml. The inhibitory effect of angiotensin converting enzyme (ACE) of the extract in the range of $0.2{\sim}1.0mg/ml$ showed $63{\sim}89%$, and the highest ACE inhibition was 89% at the concentration of 1.0 mg/ml of extracts. The highest activity of glutathion S-transferase (GST) was 221 % at the concentration of 1.0 mg/ml of the G. elata extracts. These results suggest that G. elata may be used as health/functional food materials.

Anti-Diabetic, Alcohol-Metabolizing, and Hepatoprotective Activities of Moringa (Moringa oleifera Lam.) Leaf Extracts (모링가 잎 추출물의 항당뇨, 알코올 대사 및 간 보호 활성)

  • This study was carried out to investigate anti-inflammatory, anti-diabetic, alcohol metabolizing, and hepatoprotective effects of hot water (MOW) and 80% ethanol (MOE) extracts from moringa (Moringa oleifera Lam.) leaf. The total phenol content of MOW and MOE were 45.49 and 63.06 mg tannic acid equivalents/g, respectively. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activities of MOW and MOE were remarkably elevated in a dose-dependent manner, and about 60.8% and 71.3% at 1 mg/mL, respectively (P<0.01). Superoxide dismutase-like activities of MOW and MOE were 2.8% and 7.4% at 5 mg/mL, respectively (P<0.05). ${\alpha}-Glucosidase$ inhibitory activity also increased in a dose-dependent manner in both extracts, and MOE was higher about two times than MOW at 5 mg/mL (P<0.001). The effects of MOW and MOE on alcohol metabolizing activity were determined by measuring generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). ADH and ALDH activities significantly increased upon addition of MOW and MOE (P<0.05). Anti-inflammatory activity was examined in lipopolysaccharide-stimulated RAW 264.7 cells. Nitric oxide production was reduced to 32.1% and 81.2% by addition of MOW and MOE at 1 mg/mL, respectively (P<0.05). MOW and MOE showed significant protective effects against tacrine-induced cytotoxicity in Hep3B cells at $100{\mu}g/mL$. These results suggest that moringa leaf extracts have great potential as natural health products.

Physicochemical properties, phytochemicals, and biological activities of heat-treated Elaeagnus multiflora juice and vinegar (열처리 보리수 과즙과 식초의 이화학적 특성, phytochemicals 및 생리활성)

  • In this study, vineger was produced after heat treatment of Elaeagnus multiflora juice and its fermentative characteristics were investigated. The heat-treated juice and vinegar of E. multiflora were similar in fruit color, with b values (redness) of 39.48 (juice) and 37.56 (vinegar). After 10 days of fermentation of E. multiflora fruit, the acetic acid bacteria viable cell number, pH, acidity, reducing sugar content, and alcohol content were 4.59-4.62 log CFU/mL, 3.14-3.45, 0.2-2.12%, 0.69-35.24 mg/mL, and 0.2%, respectively. The heat-treated juice and vinegar showed significantly higher radical scavenging and digestive enzyme inhibitory activities than untreated samples, and the levels of soluble phenolics, soluble flavonoids, flavan-3-ol derivatives, and phenolic and derivatives were increased. Additioinally, the heat-treated vinegar contained major organic acids, such as acetic acid (21.82 mg/mL), and major flavan-3-ols and phenolic acids, such as catechin ($72.24{\mu}g/mL$), catechin gallate ($273.36{\mu}g/mL$), epigallocatechin gallate ($68.35{\mu}g/mL$), protocatechuic acid ($12.84{\mu}g/mL$), and salicylic acid ($42.29{\mu}g/mL$). At $25{\mu}L/mL$ treatment, DPPH and ABTS radical scavenging activities and ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities were 79.66%, 93.99%, 90.12%, and 64.85%, respectively. This result suggested that it is possible to produce new types of vinegar and beverages, using heat-treated E. multiflora juice.

Physiological Activity and Physicochemical Properties of Condensed Prunus mume Juice Prepared with Pectinase (Pectinase처리를 한 매실 농축액의 이화학적 특성 및 생리활성)

  • Prunus mume Siebold & Zucc., a member of the Rosaceae family (called Maesil in Korea), has been widely distributed in East Asia, e.g. Korea, Japan and China, and its fruit has been used as a traditional drug and health food. In this study, we evaluated physicochemical properties and physiological activities of condensed Prunus mume juice treated with pectinase (PJ). The values of total acidity, pH, sugar contents, turbidity moisture content of the PJ were 35.81%, 2.73, $54.36^{\circ}Brix$, 2.75 and 51.32%, respectively. The PJ had effective DPPH radical scavenging activity, reducing power effect, $H_2O_2$ scavenging activity and ${\beta}$-carotene bleaching effect. DPPH radical scavenging activities of PJ was 46.31%; their reducing power ($OD_{700}$) was 1.80; $H_2O_2$ scavenging activity of PJ was 91.62%; and ${\beta}$-carotene bleaching effect of PJ was 73.02%. Also, PJ showed effective levels of ${\alpha}$-glucosidase inhibition activity. The cell viability was measured by SRB assay. The PJ significantly decreased the cell viability of mouse melanoma cells (B16) and human melanoma cells (SK-MEL-2 and SK-MEL-28) in a dose-dependent manner, however, there was no effect on human keratinocyte HaCaT. In morphological study, PJ-treated SK-MEL-2 cells showed distorted and shrunken cell masses. Total polyphenol contents and total flavonoid contents of PJ were 588.31 mg% (gallic acid equivalent) and 860.45 mg% (rutin equivalent). The antiproliferative effect of PJ seems to be associated with the antioxidant activity of its flavonoid and polyphenol contents. In conclusion, PJ may be beneficial in development of a functional food material.

Bioactive compounds of Cheonggukjang prepared by different soybean cultivars with Bacillus subtilis HJ18-9 (Bacillus subtilis HJ18-9 이용하여 제조한 품종별 청국장의 품질특성과 isoflavone 함량의 변화)

  • This study was conducted to investigate the changes of isoflavone composition (glycoside and bio-active aglycone) in Cheonggukjang and to evaluate its quality characteristics depending on different soybean cultivars (Daewon, Daepoong, Wooram, Hwangkeumol and Saedanback). The bioactivity of Cheonggukjang was enhanced during fermentation at $37^{\circ}C$ for 48 h. Activities of ${\alpha}$-amylase, protease, and cellulase increased significantly after 48 h fermentation (p<0.05). In addition, amino-type nitrogen and reducing sugar contents in Cheonggukjang fermented with B. subtilis increased significantly after 48 h fermentation (p<0.05). Among the isoflavones, the content of $\beta$-glucosides and acetyl-glucosides decreased, while aglycone content increased during fermentation. Especially, Cheonggukjang fermented with Daepoong cultivars showed the greatest increase in daidzein, genistein and glycitein contents. After 48 h fermentation, the contents of daidzein, genistein and glycitein in the Cheonggukjang fermented with Daepoong cultivars increased significantly up to $503.65{\pm}2.76$, $111.40{\pm}0.42$, and $633.95{\pm}6.01{\mu}g/g$ (p<0.05), respectively. Total aerobic and anaerobic cell counts increased with increase in fermentation time. Therefore, it would be beneficial for the food industry if components of Cheonggukjang could be separated and used to develop functional products.

Nutritional Components and Biological Activities of Barnyard Millets(Echinochloa spp.) (식용피의 영양성분과 생리활성)

  • The edible barnyard millets(Echinochloa spp.) which have essentially vanished in the farmhouses environment and in agricultural germplasm were evaluated with the aim of restoration as a crop. The proximate components and mineral elements of milled millet were nutritionally similar or better than brown rice, and the vitamin contents of $B_1$ and $B_2$ exceeded those of rice by 1.3 times and 2.3 times, respectively. ${\beta}$-Carotene which is absent from brown rice was detected at levels ranging from 15~31 ${\mu}g$ in millet samples. Nine essential amino acids, including histidine and arginine and eight non-essential amino acids, such as aspartic acid were detected. The sum of all amino acids was determined to be IEC518>525>510 in the range of 69~106 mg/g. Analysis of physiological active substances via their electron donating ability(EDA) revealed values ranging from 3.4~8.2%, with the total polyphenol component being 51.1~69.4 mg/g and ${\alpha}$-glucosidase inhibition ability determined as 8.3~10.9%. In terms of agronomical characteristics and yields of barnyard millet, three millet varieties(IEC510, 518, and 525) were suitable as edible crops. IEC525 was selected as optimum variety for cultivation on the basis of nutritional ingredients, physiological active substances, and yield.

Antimelanogenic Effect and Whitening of Anthocyanin Rich Fraction from Seeds of Liriope platyphylla (맥문동 종실 안토시아닌 분획물의 멜라닌 생성 억제 및 미백 효과)

  • This study was performed to determine the antimelanogenic effect and tyrosinase inhibitory activities of anthocyanin rich fraction (AN-SLP) from Liriope platyphylla Wang et Tang seeds. Anthocyanins isolated from L. platyphylla seeds revealed the presence of four major anthocyanin components, which were tentatively identified as delphinidin-3-Oglucoside, delphinidin-3-O-rutinoside, petunidin-3-O-rutinoside, and malvidin-3-O-rutinoside using semipreparative HPLC, $^1H$-NMR, $^{13}C$ NMR, FAB-MS and LC/ES-MS. The inhibitory effect of AN-SLP on tyrosinase activity was studied using in vitro (against mushroom tyrosinase) and ex vivo (against B16 melanoma cell tyrosinase) models. Cellular tyrosinase activity was decreased by AN-SLP treatment in B 16 melanoma cells through dose dependent manner, but AN-SLP did not inhibit mushroom tyrosinase and L-DOPA oxidation directly. AN-SLP showed melanin inhibition by 53.2% at 50 ${\mu}g/m{\ell}$ which was 0.7 times more efficient than the antimelanogenic effect of commercial arbutin and kojic acid (36.5%) also did not show cell toxicity. Additionally, AN-SLP inhibited the activity of ${\alpha}$-glucosidase and the glycosylation of tyrosinase in melanoma cell. The resulting unsaturated glycosylation of tyrosinase makes it unstable and disturb correct transportation. From theses results, we conclude that AN-SLP could be used as anti-melanogenic agent for skin whitening.
