Effect of respiratory syncytial virus on the growth of hepatocellular carcinoma cell-lines

  • In several reports, the respiratory syncytial virus (RSV) was identified as an oncolytic virus in cancer cells (e.g., lung and prostate cancer). However, the effects of RSV in hepatocellular carcinoma (HCC) cells have not yet been investigated. Here, we observed the inhibitory effects of RSV infection in HCC cell-lines. Cell growth was significantly decreased by RSV infection in BNL-HCC, Hep3B, Huh-7 and SNU-739 cells. After RSV infection, plaque formation and syncytial formation were observed in affected Hep3B and Huh-7 cells. RSV protein-expression was also detected in Hep3B and Huh-7 cells; however, only Huh-7 cells showed apoptosis after RSV infection. Furthermore, inhibition of cell migration by RSV infection was observed in BNL-HCC, Hep3B, Huh-7 and SNU-739 cells. Therefore, further investigation is required to clarify the molecular mechanism of RSV-mediated inhibition of HCC cell growth, and to develop potential RSV oncolytic viro-therapeutics.

  • SK-302B, an antibiotic purified from soil Streptomyces sp. 302, was structurally identified as echinomycin (C/sub 50/H/sub 66/N/sub 11/S/sub 2/). In the present experiment, the possibility of SK-302B as an anticolon cancer agent was investigated by using chemosensitivity system (MTT assay, clonogenic assay). Treatment of SK-302B on various colon cancer cell lines resulted in a significant cytotoxicity and tumor colony formation inhibition. These studies showed that SK-302B had a potent inhibition on colon cancer cells.

Emerging Co-signaling Networks in T Cell Immune Regulation

  • Co-signaling molecules are surface glycoproteins that positively or negatively regulate the T cell response to antigen. Co-signaling ligands and receptors crosstalk between the surfaces of antigen-presenting cells (APCs) and T cells, and modulate the ultimate magnitude and quality of T cell receptor (TCR) signaling. In the past 10 years, the field of co-signaling research has been advanced by the understanding of underlying mechanisms of the immune modulation led by newly identified co-signaling molecules and the successful preclinical and clinical trials targeting co-inhibitory molecules called immune checkpoints in the treatment of autoimmune diseases and cancers. In this review, we briefly describe the characteristics of well-known B7 co-signaling family members regarding the expression, functions and therapeutic implications and to introduce newly identified B7 members such as B7-H5, B7-H6, and B7-H7.

Expression of Chimeric Chicken-Yeast-Chicken H2B Histone Gene

  • A chicken H2B histone gene was cloned and expressed in Rat 3 cell line. Its messenger RNA level was about 10 times higher during S phase than during $G_1$ phase. A chimeric chicken-yeast-chicken H2B histone gene was made to change some of wobble sequences of chicken H2B gene. When the chimeric H2B gene was transfected into the Rat 3 cell line, it showed a pattern of expression similar to that of the original chicken H2B gene. At least in this gene, it was concluded that the wobble sequences were not required for the cell-cycle regulated pattern of expression.

Activation-induced Cytidine Deaminase in B Cell Immunity and Cancers

  • Activation-induced cytidine deaminase (AID) is an enzyme that is predominantly expressed in germinal center B cells and plays a pivotal role in immunoglobulin class switch recombination and somatic hypermutation for antibody (Ab) maturation. These two genetic processes endow Abs with protective functions against a multitude of antigens (pathogens) during humoral immune responses. In B cells, AID expression is regulated at the level of either transcriptional activation on AID gene loci or post-transcriptional suppression of AID mRNA. Furthermore, AID stabilization and targeting are determined by post-translational modifications and interactions with other cellular/nuclear factors. On the other hand, aberrant expression of AID causes B cell leukemias and lymphomas, including Burkitt's lymphoma caused by c-myc/IgH translocation. AID is also ectopically expressed in T cells and non-immune cells, and triggers point mutations in relevant DNA loci, resulting in tumorigenesis. Here, I review the recent literatures on the function of AID, regulation of AID expression, stability and targeting in B cells, and AID-related tumor formation.

Brazilin Inhibits of TPA-induced MMP-9 Expression Via the Suppression of NF-${\kappa}B$ Activation in MCF-7 Human Breast Carcinoma Cells

  • Metastasis is the primary cause of from breast cancer mortality. Cell migration and invasion play important roles in neoplastic metastasis. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix (ECM), plays an important role in cancer cell invasion. NF-${\kappa}B$ is transcription factor important in the regulation of MMP-9, as the promoter of MMP-9 gene contains binding sites for NF-${\kappa}B$. Brazilin, an active component of sappan wood (Caesalpinia sappan), decreases TPA-induced MMP-9 expression and invasion in MCF-7 cells. Also, brazilin suppressed NF-${\kappa}B$ activation in TPA-treated MCF-7 cells. Taken together, we demonstrated that the inhibition of TPA-induced MMP-9 expression and cell invasion by brazilin is mediated by the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells. This result suggest brazilin provide a potential therapeutic app roach for the treatment of breast cancer.

인간 진피섬유아세포에서 Bifidobacterium bifidum 배양액 및 추출액의 자외선B에 대한 보호 효능 (Protective Effects of Bifidobacterium bifidum Culture Supernatants and Intracellular Cell-Free Extracts on Human Dermal Fibroblasts against UV-B Irradiation)

  • 본 연구는 장내세균 Bifidobacterium bifidum 배양액과 추출액을 사용하여 자외선B를 조사한 인간 진피섬유아세포에서 세포생존율과 세포 노화 및 세포주기의 변화를 살펴봄으로써 자외선B에 의한 세포 보호 효능을 평가하였다. 우선 자외선B를 조사한 결과 광량에 비례하여 HDFs의 생존율이 감소하였으며 $100mJ/cm^2$ 조사 시 67.3%로 떨어졌으나 B. bifidum 배양액과 추출액 처리 후 세포생존율을 증가시켜 자외선B에 대한 보호 효능이 있었다. 그리고 $SA-{\beta}-gal$ activity 변화를 측정한 결과에서 세포 노화율을 감소시켰음을 확인하였다. 또한, propidium iodide staining을 통하여 세포주기상 Sub-G1기 세포 수가 감소하였으므로 apoptosis를 억제하였고 이는 세포주기를 정상화하는 데 긍정적인 영향을 미쳤다. B. bifidum 배양액과 추출액 처리한 결과 세포 내 활성산소종이 감소함에 따라 p53과 p21의 발현을 감소시켰으며, 따라서 이에 본 연구에서 B. bifidum 배양액과 추출액은 UV-B로 인한 손상을 보호하는 효과가 있음을 규명하였다.

Dural Marginal Zone Lymphoma Confused with Meningioma en Plaque

  • We report a case of dural marginal zone lymphoma which showed the usual radiological findings resembling meningioma. A 59-year-old woman presented with headache. Initial computed tomography and magnetic resonance images showed a frontal convexity meningioma. The patient underwent a craniotomy and subtotal (simpson grade II) resection of tumor was done. Pathological examination confirmed an extranodal marginal zone B-cell lymphoma of Mucosa-Associated Lymphoid Tissue (MALT). The lesion was composed of a lymphoid mass with irregularly shaped follicles surrounded by many monomorphic small lymphocytes and a stained marginal zone for B-cell markers CD20 and CD29a. The natural history of primary CNS lymphoma and MALT type lymphoma are different. B-cell MALT lymphoma can mimic meningioma in its radiological features. Accordingly, MALT lymphoma of the CNS must be considered in the differential diagnosis of meningioma.

A case of nodal marginal zone B-cell lymphoma of the lower eyelid

  • A marginal zone B-cell lymphoma is a type of B-cell lymphoma which is normally located at the margins of the secondary lymph node follicles. According to 2008 World Health Organization (WHO) classification, there are three main types classified according to the location of invasion: nodal lymphoma in the lymph nodes, splenic lymphoma in the spleen, and extranodal lymphoma in other locations. Recently, we have experienced a rare case of primary nodal marginal zone lymphoma that arose in the lower eyelid. Therefore, we report this case with a review of literature.

카드뮴이온에 의한 Hansenula anomala B-7의 형태 변이 (Morphological Changes of Hansenula anomala B-7 by Cadmium Ion)

  • Yeast-form cells of cadmium ion-tolerant Hansenula anomala B-7 were changed to mycelial cells in medium containing more than $400\mu$g/ml of cadmium. Moreover, the mycelial cells were exchanged into clumped cells in a medium containing more than $1,000\mu$g/ml of cadmium. Optimal conditions of mycelial cell formation were achieved in the presence of .$1,000\mu$g/ml of cadmium with shaking cultivation for 7 days. Glucan and mannan contents of the yeast cell wall frown with $1,000\mu$g/ml of cadmium decreased by 10% compared with those grown without cadmium. However, protein and lipid contents increased about 20% respectively. By cadmium, no significant findings in specific amino acid contents were discovered.
