  • A monoclonal antibody(MAb), KCT-23ER, with specificity for E-rosetted T cells of Korean native cattle was prepared by cell hybridization of myeloma P3/NS-1/1-Ag-4-1 and spleen cells from BALB/c mice hyperimmunized with E-rosetted lymphocytes. The isotype of KCT-23ER to T lymphocytes was mouse $IgG_{2b}$. KCT-23ER was reacted with 53.6% to peripheral blood lymphocytes and with 67.8% to nylon wool nonadherent blood lymphocytes. And it was reacted with 72.2%, 59.2% and 35.3% to thymocytes, prescapular lymph node cells and splenocytes, respectively. Immunocytological reactive rates to E-rosetted and non-E-rosetted cells were 72.5% and 22.4%, respectively. These results indicated that KCT-23ER reacted to E-rosetted cells was one of the MAb for investigate of $CD_2$ receptor positive cell subset in the Korean native cattle.

The Effects of Scutellaria baicalensis GEORGI on Allergic contact dermatitis induced by DNCB in Mice (황금(黃芩) 추출물(抽出物)이 DNCB로 유발(誘發)된 생쥐의 Allergy성(性) 접촉피부염(接觸皮膚炎)에 미치는 영향(影響))

  • This study was performed to investigated the effects of an immunosuppression and a mitigation of inflammation of Scutellaria baicalensis GEORGI on Allergic contact dermatitis. In order to study, after oral administration of Scutellaria baicalensis GEORGI extract, contact hypersensitivity assay, general morphologic change of skin, sulfated acid mucosubstance, mast cell, IL-2R, ICAM-1 and CD11b were observed in BALB/C mouse induced allergic contact dermatitis by the contact sensitizing DNCB. Ear swelling in contact hypersensitivity assay was decreased in the Scutellaria baicalensis GEORGI extracts administered(HEGT) group, compared with DNCB painted group. In the general morphologic change of skin, hyperplasia of keratinocytes, increase of lymphocytes and inflammatory cell, increase of vasculogenesis and epidermal lymphocytes infiltration, increase of cell damage in epidermal basal layer and prickle layer were decreased in the HEGT group, compared with DNCB painted group. Increase of sulfated acid mucosubstance, mast cell, IL-2R positive cell, ICAM-1 positive cell, CD11b positive cell were decreased in the HEGT group, compared with DNCB painted group.

Cultivation and Electron Microscopy of Bovine Leukemia Virus from Peripheral Blood Lymphocytes of Holstein-Friesian Dairy Cattle (젖소 말초혈액 림프구로부터 소백혈병 바이러스 배양 및 전자현미경적 관찰)

  • Many studies have been performed on the bovine leukemia virus (BLV) since bovine leukosis had been reported in 1968 in Korea. However, there was no report on the ultrastructural examination of BLV. An attempt to detect C-type viral particles in the cultured peripheral blood lymphocytes of Holstein-Friesian dairy cattle, was made to determine whether in vitro viral expression might be used as a reliable method to identify the cow which is likely to transmit BLV. In transmission electron microscopic (TEM) examination, the virus particles were found predominantly outside of the lymphocytes even though a few particles were also observed within the membrane bound cytoplasmic vacuoles. All of them were C-type particles consisting of a central, electron-dense core separated by a clear area from a limiting envelope with a unit membrane structure. Virus particles were easily detected in the lymphocyte which was cultured with medium supplemented with either T-lymphocyte mitogen (conconavalin A) or B-lymphocyte mitogen (lipopolysaccharide). Identical viral particles, although fewer, were also consistently present in the lymphocytes cultured with medium which was containing foetal bovine serum (FBS) only and which was containing neither FBS or mitogen. By contrast, no virus particle was detected in extensive examination of lymphocytes before culture. In conclusion, the BLV cultivation and detection methods established in this study could be used as a tool to identify and eliminate the cattle which can transmit the BLV.

Immunomodulating Effect of Edible Mushrooms in Mice (식용버섯의 면역조절에 미치는 영향)

  • This study was carried out to evaluate the immunomodulatory capacity of edible mushrooms, including Lepista nuda, Corprinis comatus, Letinus edodes, and Pleurotus eryngii, in mice. BALB/c mice were administered 1, 50, and 500 mg/kg body weight of various mushrooms five times a week over 4 weeks through gastric intubation. The control mice were administered distilled water. No significant changes in body weight were observed. IL-4 and IFN${\gamma}$ production was evaluated with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hr. The mice group administered L. edodes showed significantly higher ratio of IFN${\gamma}$ versus IL-4 than the other groups. In addition, the ratio of plasma IgG2a versus IgG1 was also significantly elevated in mice treated with L. edodes. However, no significant change was observed in ratio of IgG2a versus IgG1 in splenic B lymphocytes stimulated in vitro with lipopolysaccharides for 7 days. These results indicate that L. edodes can enhance type-1 helper T cell-mediated cellular immunity.

Comparison of sheep erythrocytes and Korean native goat erythrocytes-rosette forming rate of pig peripheral blood mononuclear cells (돼지 말초혈액 단핵세포의 면양 및 재래산양 적혈구 rosette 형성능 비교)

  • To develope the methods for isolation and enumeration of lymphocyte subpopulations in pigs, we carried out the rosette-assay using sheep erythrocytes(SRBC) and Korean native goat erythrocytes(GRBC) as a target cells. To enumerate T lymphocytes, E-rosette methods were applied with RBC treated with various concentration of polymers such as Aet and Dex, singly or in combination. And to enumerate B lymphocytes, EAand EAC-rosette assay was adopted. The results were as follows; 1. E-RFR with polymer-untreated SRBC and GRBC was $32.9{\pm}7.9%$ and $31.3{\pm}9.4%$, respectively. On the other hand, RFR with 0.1M Aet plus 8% Dex treated SRBC and GRBC was increased about two-fold($67.8{\pm}7.4%$ and $69.8{\pm}8.5%$), respectively. 2. EA-RFR with SRBC and GRBC were $ 39.1{\pm}10.2%$ and $32.6{\pm}6.1%$, respectively. 3. EAC-RFR with SRBC and GRBC were $27.6{\pm}7.0%$ arld $21.0{\pm}3.2%$, respectively. These results showed that both SRBC and GRBC could be recommanded as a binding cells for rosetteassay to isolate of lymphocyte-subpopulations in pigs.

Fine Needle Aspiration Cytology of Extranodal Marginal Zone B cell Lymphoma with Abundant Plasma Cells and Eosinophilic Histiocytes in Parotid Gland (귀밑샘의 형질세포와 호산성 조직구를 다량 함유한 림프절외 변연부 B-세포 림프종의 세침흡인 세포검사 소견 -1예 보고-)

  • The authors present the fine needle aspiration cytology (FNAC) cytologic findings of a case of extranodal marginal zone B cell lymphoma (MZBCL), which featured abundant plasma cells and eosinophilic histiocytes arising in both parotid glands. A 49-year-old female presented with palpable masses in both parotid glands. She had been suffering from systemic lupus erythematosus and rheumatoid arthritis. The lesions were evaluated by FNAC and smears showed a small number of clusters of oncocytic cells with abundant eosinophilic granular cytoplasm and small nuclei, intermixed with small to medium-sized lymphoid cells containing round to lobulated nuclei, which suggested Warthin's tumor. Some of lymphoid cells had a plasmacytoid appearance, and some scattered large cells contained a large amount of eosinophilic cytoplasm. Bilateral superficial parotidectomy was performed and a histopathologic study indicated MZBCL with abundant plasma cells, intermixed with eosinophilic histiocytes. The presence of oncocytic cells and a mixture of lymphoid and plasma cells indicates Warthin's tumor, but the cytologic features of a relatively monotonous small to medium-sized lymphoid infiltrate suggest the possibility of MZBCL in the clinical setting of an FNAC study performed on a patient suffering from a connective tissue disease.

Radioprotective Effects of Aronia on Radiation Irradiated Rats (방사선에 조사된 쥐에서 아로니아의 방사선 방호효과)

  • The present study was intended to orally administer aronia to rats, irradiate radiation once to the whole bodies of the rats, and conduct blood tests to observe, compare, and analyze changes in blood cells, such as leukocytes, erythrocytes, and platelets, in order to examine the radioprotective effects of aronia. As experimental animals, 15 male Sprague-Dawley (SD) rats aged six weeks weighing 200~250 g were taken and divided into the normal group (A) of five rats, the 5 Gy control group (B) of five rats, and the 5 Gy experimental group (C) of five rats. The normal group (A) was not~irradiated at all, the control group (B) was administered with general diets and irradiated, and the experimental group(C) was orally administered with 50 mg/kg/day of aronia two times per day to achieve a distilled water oral dose of 100 mg/kg/day and irradiated thereafter (5 Gy at 500 cGy/min) for 14 days. After the experiment, differences in leukocytes, erythrocytes, and platelets among the normal group (A), the control group (B), and the experimental group (C) were examined by comparing the counts of the blood cells and the results showed no statistically significant differences. However, on a detailed review, the normal group (A) showed statistically higher mean values for all of lymphocytes, hemoglobin, and mean corpuscular hemoglobin as compared to the control group (B) and the experimental group (C). Statistically significant differences in the counts of lymphocytes were shown between the normal group (A) and the control group (B), and between the normal group (A) and the experimental group (C); furthermore, statistically significant differences in mean corpuscular hemoglobin were shown between the normal group (A) and the experimental group (C). Given the results of the present study, in irradiated rats, aronia was generally considered as having no radioprotective effect on leukocyte, erythrocyte, and platelet while having statistically significant radioprotective effects on lymphocytes, hemoglobin, and mean corpuscular hemoglobin. Based on the present experiment, diverse studies should be conducted hereafter.

Effect of Quaternary Ammonium Compounds(QACs) on the Immune Response of eel(Anguilla japonica) In vitro (In vitro에서 Quaternary Ammonium Compounds(QACs)이 뱀장어의 면역반응에 미치는 영향)

  • The immunostimulatory effects of quaternary ammonium compounds(QACs) were investigated in leucocytes of eel(Anguilla japonica) in vitro. Proliferation of peripheral blood lymphocytes(PBLs) was no significantly affected by QACs, regardless of mitogen(PHA, ConA and LPS) and the concentration of QACs added. QACs heightened the leucocytes function such as respiratory burst activity, phagocytosis and pinocytosis, resulting in significantly increased the bactericidal activity of macrophages. These results suggested that QAC might modulate the immune responses by activation of leucocytes function but not by increment of immunocompetent cell numbers.
