Effect of 2,4-Dichlorophenoxyacetic Acid (2,4-D) and Naphthoxyacetic Acid (NOA) on the Biochemical Changes in the Fat Body and Haemolymph of the Silkworm, Bombix mori L.

  • The effect of topical application with 200,400 and 600 $\mu$g/ml 2,4-Dichlorophenoxyacetic Acid (2,4-D) and Naphthoxyacetic Acid (NOA) to the fifth stadium larvae of the silkworm, B. mori on fat body glycogen, protein, total lipids, phospholipids, neutral lipids and haemolymph trehalose and protein has been studied. The fat body glycogen significantly increased in all the groups treated with 2,4-D whereas there is no significantly change in fat body glycogen in all the groups treated with NDA. However, the haemolymph trehalose was significantly increased in all 274-D and NOA treated groups except in the groups treated with 400 and 600 $\mu$g/ml 2,4-D where the increase was not significant.The fat body protein did not show any significant change in all groups treated with 2,4-D and NOA except in the groups treated with 200 $\mu$g/ml 2,4-D where the fat body protein was significantly increased. The total lipids, phospholipids and neutral lipids of the fat body increased significantly in all the groups treated with 2,4-D and NOA when compared with those of the corresponding parameters of the carrier control.

  • The effect of topical application with 5,10 and 15 $\mu$g/ ml androstenedione and methyltesterone to the fifth stadium larvae of the si1kworm,B. mori on fat body glycogen, protein, tota1 lipids, phospholipids, neutral lipids and haemolymph trehalose and protein has been studied. The fat body glycogen increased significantly in all the treated groups except in the 15$\mu$g/ml androstenedione treated group. However the fat body glyco- gen increased significantly in 10 ${\mu}g$/ml methyltesterone treated group. The androstenedione treated groups showed increased haemolymph trehalose but the increase was not significant. However the haemolymph trehalose increased significantly in all the methyltestosterone treated groups. The fat body protein decreased in all the androstenedione and methyltesterone treated groups except 15 ${\mu}g$/ml methyltesterone where the decrease in fat body protein was significant. Whereas that of haemolymph protein significantly decreased in all the groups except in the group treated with 5 ${\mu}g$/ml androstenedione where the decrease was not significant. The total lipids, phospholipids and neutral lipids of the fat body decreased significantly in all the groups treated with androstenedione and methyltestosterone when compared with those of carrier control.

Development of a Simple Cell Lysis Method for Recombinant DNA Using Bacteriophage Lambda Lysis Genes

  • In this study, we describe the development of a simple and efficient method for cell lysis via the insertion of a bacteriophage lambda lysis gene cluster into the pET22b expression vector in the following order; the T7 promoter, a gene for a target protein intended for production, Sam7 and R. This insertion of R and Sam7 into pET22b exerted no detrimental effects on cellular growth or the production of a target protein. The induction of the T7 promoter did not in itself result in the autolysis of cells in culture but the harvested cells were readily broken by freezing and thawing. We compared the efficiency of the cell lysis technique by freezing and thawing to that observed with sonication, and determined that both methods completely disintegrated the cells and released proteins into the solution. With our modification of pET22b, the lysis of cells became quite simple, efficient, and reliable. This strategy may prove useful for a broad variety of applications, particularly in experiments requiring extensive cell breakage, including library screening and culture condition exploration, in addition to protein purification.

  • One of the essential functions of virus surface proteins is the recognition of specific receptors on target cell membranes, and cellular receptors play an important role in viral pathogenesis. But the earliest steps of hepatitis B virus (HBV) infection, such as hepatocyte receptor interaction with the virus, are poorly understood. Previous work has suggested an important role of the preS1 region of HBV envelope protein in mediating viral binding to hepatocytes. Although hepatitis B virus (HBV) infection appears to be initiated by specific binding of virions to cell membrane structures via one or potentially several viral surface proteins, data showing the identification or isolation of the HBV receptor (s) are not yet available. The receptor-like proteins on the plasma membrane surface of HepG2 cells that bind to PreS1 were separated and identified using affinity chromatography, and the amino-terminal amino acid sequences of the receptor-like proteins were determined.

Ruminal Protein Degradation Characteristics of Cell Mass from Lysine Production

  • Chemical analysis and in vitro studies were conducted to investigate the nutritive value for ruminants of cell mass from lysine production (CMLP) which is a by-product of the lysine manufacturing process. Proximate analysis, protein fractionation, and in vitro protein degradation using protease from Streptomyces griseus and strained ruminal fluid were carried out to estimate ruminal protein degradability of CMLP with two reference feedstuffs-soybean meal (SBM) and fish meal (FM). Amino acid composition and pepsin-HCl degradability were also determined to evaluate postruminal availability. CMLP contained 67.8% crude protein with a major portion being soluble form (45.4% CP) which was composed of mainly ammonium nitrogen (81.8% soluble CP). The amount of nucleic acids was low (1.15% DM). The total amount of amino acids contained in CMLP was 40.60% DM, which was lower than SBM (47.69% DM) or FM (54.08% DM). CMLP was composed of mainly fraction A and fraction B2, while the protein fraction in SBM was mostly B2 and FM contained high proportions of B2 and B3 fractions. The proportion of B3 fraction, slowly degradable protein, in CP was the highest in fish meal (23.34%), followed by CMLP (7.68%) and SBM (1.46%). CMLP was degraded up to 51.40% at 18 h of incubation with Streptomyces protease, which was low compared to FM (55.23%) and SBM (83.01%). This may be due to the insoluble portion of CMLP protein being hardly degradable by the protease. The in vitro fermentation by strained ruminal fluid showed that the amount of soluble fraction was larger in CMLP (40.6%) than in SBM (17.8%). However, because the degradation rate constant of the potentially degradable fraction of CMLP (2.0%/h) was lower than that of SBM (5.8%/h), the effective ruminal protein degradability of CMLP (46.95%) was slightly lower than SBM (53.77%). Unavailable fraction in the rumen was higher in CMLP (34.0%) compared to SBM (8.8%). In vitro CP degradability of CMLP by pepsin was 80.37%, which was lower than SBM (94.42%) and FM (89.04%). The evaluation of protein degradability using different approaches indicated that soluble protein in CMLP may supply a large amount of ammonia in the rumen while insoluble protein can be by-passed from microbial attacks due to its low degradability. The results from this study suggest that CMLP can be used as a protein supplement to ruminants for supplying both non-protein nitrogen to rumen microbes and rumen undegradable protein to the host animal.

  • The tumor suppressor and transcription factor p53 is a key modulator of cellular stress responses, and can trigger apoptosis in many cell types including neurons. In this study, we have shown that Microtubule-associated protein 1B(MAP1B) light chain interacts with tumor suppressor p53. MAP1B is one of the major cytoskeletal proteins in the developing nervous system and essential in forming axons during elongation. We also demonstrate that both p53 and MAP1B-LC1 interact in the nucleus in HEK 293 cells. Indeed, we show that the MAP1B-LC1 negatively regulates p53-dependent transcriptional activity of a reporter containing the p21 promoter. Consequently, MAP1B light chain binds with p53 and their interaction leads to the inhibition of doxorubicin-induced apoptosis in HEK 293 cells. Furthermore, these examinations might be taken into consideration when knock-down of MAP1B-LC1 is used as a cancer therapeutic strategy to enhance p53's apoptotic activity in chemotherapy.

Evaluation of Squid Liver Powder as a Dietary Protein Source Replacing Fish Meal in Juvenile Flounder, Paralichthys olivaceus (넙치 사료내 어분대체 단백질원으로써 오징어간분말의 평가)

  • This experiment was conducted to determine the each level of a newly developed squid liver powder (SLP) as a dietary protein source replacing fish meal (FM) in juvenile flounder. A diet excluding SLP (FM diet) and a commercial diet were two control diets. Six experimental diets were formulated to replace FM with three types of SLP at 5 and 10% level, respectively (SLP-A5, SLP-A10, SLP-B5, SLP-B10, SLP-C5, SLP-C10). Prior to the start of a feeding trial, fish were fed the commercial diet for 1 week to adjust to the experimental conditions. Fish with average weight of $22.8{\pm}0.40g$ ($mean{\pm}SD$), were randomly distributed to each aquarium as groups of 20 fish and fed the experimental diets in triplicate at satiation for 8 weeks. After 8 weeks, weight gain and specific growth rate of fish fed SLP-C10 was lower than those of fish fed FM diet and SLP-B5. Meanwhile, no significant difference was observed in WG and SGR among fish fed diets excluding SLP-C10. Feed efficiency (FE) and protein efficiency ratio (PER) of fish fed each level of three types of SLP were not different from those of fish fed FM diet. However, fish fed SLP-C5 and SLP-C10 has lower FE and PER than those of fish fed a commercial diet, SLP-A5 and SLP-B5. Therefore, these results indicate clearly that dietary SLP-B could be a good protein source to develop inexpensive commercial diet in juvenile flounder.

TUMOR-ASSOCIATED PROTEINS IN RAT SUBMANDIBULAR GLAND INDUCED BY DMBA AND IRRADIATION (DMBA 매식과 방사선 조사로 유발된 백서 악하선 암에 존재하는 단백질에 관한 연구)

  • This study was performed in order to identify changes of the plasma membrane proteins in rat submandibular gland tumors induced by 7,12-dimethylbenz[a]anthracene [DMBA] and X-irradiation. Two kinds of tumor associated membrane proteins (protein A and B) were isolated with 3 M KCl extraction from rat submandibular gland tumors induced by DMBA and X-irradiation. To identify their antigenicities, immunoelectrophoresis and double immunodiffusion was carried out with various proteins extracted from liver, heart, skin and pancreas of adult rats and from embryonic liver, heart and skin. The rabbit antisera against the protein A did not cross-react with any of the proteins extracted from the above mentioned tissues, suggesting that protein A might be tumor specific antigen. However, the rabbit antisera against protein B was precipitated with proteins extracted from the liver of adult and embryonic rats. Polyacrylamide gel electrophoresis of these two proteins (A and B) showed that protein A was a dimer with molecular weights of 69,000 and 35,000 dalton, whereas protein B was a monomer with molecular weight of 50,000 dalton.

Bacterial Expression of Cytochrome $b_5$ Type III Pseudogene

  • Cytochrome $b_5$ is involved in the reduction of methemoglobin back to hemoglobin, thereby maintaining normal function of the blood to carry oxygen around. Congenital abnormal condition of this enzyme causes a rare disease called methemoglobinemia. At least 4 different retropseudogenes are reported so far for cytochrome $b_5$. However, type III pseudogene has attracted most attention because it contains open reading frame in its structure. Although there is no evidence yet if this pseudogene is actually expressed in the cell or the blood the possibility of its expression needs to be elucidated. We have isolated type III pseudogene by polymerase chain reaction and cloned into pGEX-4T-1 expression vector followed by SDS-PAGE. Protein was expressed and the size of the expressed protein was 28 kDa as expected in its genetic code. This result also shows that the protein is not harmful for the viability of the microorganism. This study may contribute to the genetic diagnosis of cardiac diseases, possibly caused by cytochrome $b_5$.

The Study on The Nutritional Status of The Packed Lunch Boxes for Junior High School Students in Seoul City (서울시내(市內) 중학생(中學生)의 도시락 영양(營養) 실태조사(實態調査))

  • This study is concerned with the nutritional status of the packed lunch boxes which are brought by the junior high school students in Capital City of Seoul. Four hundred and eights students of the 8 school districts in Seoul had been randomly selected as subjects. The contained nutrients in the packed lunch were analysed by the Food Composition Table. To observe the influence of home economical status and mother's educational level on the nutrient concents of packed lunches, the chosen subjects were classified into three large groupings, which are upper, middle and low classes respectively. In addition, comparison between the Recommended Daily Allowanced for Korean people-13 to 15 age group-and the contained nutrients in the lunch boxed had been conducted. T-test was applied to clarify the significance of the differences between each group both economical and educational level. 1. The averaged rations between the Recommended Daily Allowances and the contained nutrients in the lunches stand: Calorie 59.7% (566 Cal.) protein 53.1% (18g), animal protein 48.6% (5g), fat 39.8% (5g), calcium 371.% (0.1g), ferret 66.4% (2.9g) Vitamin A 642 1.U. (31.3%), Vitamin $B_1$ 70.2% (0.3mg), Vitamin $B_2$ 41.4% (0.2g), Niacin 77.0%(4mg), Vitamin C 51.9% (13mg). All of the nctrients fall far behind the Recommended Daily, Allowances for 13 to 15 age group. 2. Home economical status brings influence on the kinds of foods which could been. Protein, animal protein, fat, Vitamin $B_2$, Vitamin C showed significance of diffierences between the upper and middle classes. Between the middle and low classes, Protein, animal protein, fat, calcium and Vitamin C showed significance of difference. And finally, between the upper and low classes, protein, fat, calcium, ferret, Vitamin $B_2$ and Vitamin C showed a great significance. 3. Regardless to the living conditions of the subject students, all the nutrients of the lunches packed by the mothers in the entire educational levels did not reach the Recommended Daily Allowances. Protein, animal protein, fat, ferret, Vitamin A, and Niacin showed the significance of the differences between the upper and middle classes. On the other hand, calorie, animal protein, fat, Vitamin A and Vitamin C showed the significance between the middle and low classes. Between the upper and the low classes, protein, animal protein, fat, and ferret showed significance. 4. The mairdish-ice of the lunch boxes supplied calorie, protein, Vitamin $B_1$, Vitamin $B_2$ and Niacin which stand at 83.8%, 56.1%, 52.5%, and 54.8% respectivly when compared with the whole nutritional contents. 5. The side-dishes of the packed lunch lack in variety of cooking methods. One interesting fact is that entire subject students are very favorable to the food cooked with every kind of grains.
