Virus Free Stock Production by In vitro Stem Cutting of Shoot Tip Cultures of Grapes (포도 경정배양에서 얻은 유묘의 기내삽목에 의한 무병묘 생산)

  • The experiment was conducted to know the effect of plant growth regulators on axillary bud elongation from in vitro stem cutting and the possibility of virus-free stock production. Axillary buds were well elongated in 3/4 strength MS medium supplemented with 0.1 or 0.5 mg/1 BA and 0.05 mg/1 NAA. Transferred plantlets could be established well in vermiculite and peat moss mixture (3:1, v/v) compare to other mixtures. In virus indexing, all the varieties of mother plants were infected by GLRV Ⅲ. Infected percentages of the three varieties were ranged from 30% to 75%. But negative response was revealed against the other species of virus, GLRV Ⅰ, GFLV and ArMV. Plantlet of 'Schuyler' and 'Muscat of Alexandria', which were cultured in vitro, showed positive response against GLRV Ⅲ and infected percentage of the former was 37.5% but the latter, 12.5%. On the other hand, that of 'Campbell Early' negativiely responded against all the species of virus indexed.

In Vitro Propagation of Wild Cynanchum wilfordii Through Axillary Bud Culture (액아배양을 통한 야생 백하수오(Cynanchum wilfordii)의 기내증식)

  • This study was conducted to establish the optimal condition for in vitro propagation of wild Cynanchum wilfordii. The highest in vitro seed germination rate of 91.6% was obtained from the seed treated with gibberellic acid ($GA_{3}$) (100 ppm) for 24 hours followed by cultured on Woody Plant Medium (WPM) media supplemented with 1.0 mg/L 6-benzyl adenine (BA). The best shoot height obtained (5.2 cm) in medium with 1.0 mg/L $GA_{3}$, but the plants was grown abnormally and eventually died. The highest number (2.4) was shown when shoot were in cultured on the media including 0.1 mg/L BA after 4 weeks. Root induction from shoot obtained in vitro culture was effective on ventilation and without plant growth regulators (PGRs) and root length was highly developed (2 cm) at 0.1 mg/L naphthalene acetic acid (NAA). The highest survival rate (70%) was when plantlet grew pre-ventilation of in vitro condition.

  • Different kinds of cytokinins and auxins were tested for both shoot induction and rooting in a rare species, Abeliophyllum distichum Nakai. BA in WPM medium was the most effective in shoot induction, whereas zeatin seemed to be the most suitable for shoot elongation. Kinetin, at the concentration of 2.0~5.0 mg/L showed an effect in shoot induction, but the effect was inferior to BA and zeatin. Rapid shoot elongation could be achieved when the cultures were maintained on the diffuse light condition (below 500 lux) regardless of cytokinin treatments. For in vitro rooting, IBA was investigated as the best type of auxin tested when half strength GD medium was incorporated. The frequency of rooting using the plant growth regulator and medium just mentioned above was revealed as approximately 90%. In addition, the survival rate of rooted plantlets was almost 100% in an artificial soil mixture.

Micropropagation of Juvenile and Mature Tree of Corylopsis coreana by Axillary Bud Culture (액아배양에 의한 유묘 및 성숙 히어리나무의 기내번식)

  • We have developed an in vitro micropropagation system via shoot formation from axillary buds using nodal segments of Corylopsis coreana. Explants from both juvenile tree (one-year-old greenhouse stock seedlings) and mature tree (ten-years-old tree in nursery) were compared with regard to propagation efficiency. Combined treatment of both BA and zeatin were effective on shoot proliferation since the best result was obtained on MS medium supplemented with 0.5∼3.0 mg/L zeatin and 0.2 mg/L BA. Generally, juvenile explants were better in both shoot proliferation and growth than mature explants. However, as the duration of in vitro culture was proceed to 6 months, explants from mature tree also produced three shoots per explant. Distinctive differences in rooting and adaptability to soil of shoots obtained from mother trees. Whereas shoots originated from juvenile explants rooted as high as 97%, those from adult explants showed 62% rooting. Similar result was also observed in soil acclimatization. The plantlets derived from juvenile plants survived 67%, while only 48% of those from adult trees survived. The results showed a possibility of the micropropagation of Corylopsis coreana through shoot formation from axillary buds. In addition, the advance of the research still remain to enhance the frequency of acclimatization of plantlets from mature trees for practical application.

  • Axillary bud explants from 3-year-old seedlings of Forsythia saxatilis N., rare and endangered species in Korea, were cultured on Murashige and Skoog's medium. The effect of various cytokinins(BAP, kinetin, and zeatin) at the different concentration(0.2, 0.5 and 1.0mg/L) was tested. Although an apparent shoot proliferation was not observed, zeatin showed slight promotional effect on normal shoot and leaf development. Both shoots and adventive roots could be induced simultaneously when the explants were cultured on the medium with kinetin, but adventive rooting was gradually reduced according as BAP and zeatin concentrations increased. Axillary shoot growth was promoted by the etiolation treatment. Shoot proliferation has been maintained more than three years with consecutive subculture. Rooted plantlets were successfully acclimatized in the artificial soil mixture and showed normal growth after transplantation into field.

An Effective Selection of PAT Gene Transformed Populus alba $\{times}$ Populus glandulosa No.3 using Herbicide Basta Treatment (제초제 Basta를 이용한 Phosphinothricin Acetyltransferase 유전자로 형질전환된 현사시 3호의 효율적인 선발)

  • This study was conducted to simple transformants selection by herbicide Basta treatment after transformation with Agrobacteium tumefaciens MP90/PAT in hybrid poplar(Populus alba ${\times}$ P. glandulosa No. 3). In preliminary study, we determined that the lethal concentration of herbicide Basta was 1.0mg/L in callus culture, adventitious bud formation and axillary bud elongation experiment. By the treatment of 1.0mg/L Basta, we could be selected the transformed shoots effectively from the various cultures. In addition, the treatment was useful on selection of transformants which are growing in soil pot. Finally, we also confirmed the transformation by PAT assay, Above results show that the herbicide Basta treatment and PAT assay can be a very simple and effective method for the identification of PAT gene transformed hybrid poplar.

High-frequency regeneration by stem disc culture in selected clones of Populus euramericana

  • An efficient regeneration protocol for stem disc culture of Populus euramericana, which is important species for bioenergy resource in agroforestry, was established. The number of explants that were obtained and the number of explants that regenerated varied with the genotypes. However, in all the genotypes, stem disc culture produced more regenerated shoots than did in axillary bud culture. A comparison of the effects of cytokinin type and concentration on shoot regeneration in different explants (i.e., petiole, leaf, and root segments of P. euramericana) revealed that a concentration of $0.002mg\;l^{-1}$ thidiazuron (TDZ) used on petiole segments resulted in the greatest shoot regeneration (95.83%). The hormonal requirements for the greatest shoot regeneration in the three explant types varied. Different concentrations of $AgNO_3$ and $CoCl_2$ were added separately to the medium to stop the yellowing and subsequent necrosis of the regenerated shoots. Lower concentrations (3 and $5mg\;l^{-1}$) of these compounds improved shoot regeneration and elongation, compared with the control. The in vitro-regenerated shoots were transferred to rooting medium and subsequently acclimatized. The highly efficient regeneration system of P. euramericana reported here can be used for mass propagation of this recalcitrant for regeneration, economically important tree species.

Morphogenetic Responses according to Media and Growth Regulators in vitro Culture of Eight Populus spp. (배지(培地) 및 생장(生長)호르몬에 따른 기내배양(器內培養)된 포푸라속(屬) 8개(個) 수종(樹種)의 기관형성반응(器官形成反應))

  • To examine the morphogenetic response, stem segments of 8 Populus spp. and 3 different explants of P. nigra var. italica were cultured on MS (Murashige and Skoog 1962) and WPM (Woody Plant Medium) medium containing various phytohormones. The results obtained were as follows: 1. Shoot regeneration and development from stem segment of 8 Populus spp. showed a quite difference according to the section and the species. All of the species of Leuce and Tacamahaca section did not form adventitious buds, while most of explants showed axillary or dormant bud elongation after 4 weeks. But P. nigra var. italica of Aigeiros section showed a successful adventitious bud formation (mean 5.4 buds per explant). 2. Leaf, petiole, and internode segment of P. nigra var. italica showed a quite differences according to media and ex plants upon the morphogenetic response. Adventitious bud formation from leaf was more abundant and readily initiated on the abaxial side than on the adaxial side. Mean number of 103 adventitious buds per explant was obtained from abaxial side of leaf segment cultured on WPM medium containing $0.2mg/{\ell}$ BAP for 5 weeks. 3. 2,4-D (2,4-dichlorophenoxy acetic acid) supplemented to media appeared to be negative upon the adventitious bud formation of P. nigra var. italica, while it promoted callus formation from all explants. Especially, NAA (${\alpha}$-naphtalene acetic acid) or NAA combination with BAP (6-benzylaminopurine) promoted root regeneration from the all explant of P. nigra var. italica in this study.

A Study on the Landscape Characteristics and Propagation Methods of Korean native Lindera obtusiloba Blume (자생 생강나무의 조경적 특성 및 번식방법에 관한 연구)

  • This study was carried out to investigate the growth characteristics and propagation methods of L. obtusiloba as a woody landscape plant. The results are obtained as follows: L. obtusiloba was dioecious shrub and shaped with bush type. Leaves were ovate, obtuse, cordate, 7.05 cm long, 7.20 cm wide, and petiole 2.0 cm long. Flowers of L. obtusiloba were diclinous. Soil acidity ranged from pH 4.06 to 5.53 with the lowest at the Mt. Soo-Ri. Mt. Soo-Ri located near factory district, which was considered to damaged by environmental deteoration. While soil organic matter was highest at Mt. Soo-Ri, inorganic nutrients were low. L. obtusiloba grows in the area with low soil acidity and low content of inorganic. Therefore it seemed to be tolerant to air pollution. L. Obtusiloba was high seed germination rate in the plug box and its shoots were longer than seeding box and softwood cutting of L. obtusiloba showed the rooting rate of 50% at 5,000ppm on June 23. To develop a mass propagation method of Korean native L. obtusiloba through an axillary bud culture as a woody landscape plant, about 2∼3 cm shoots induced from explant were subcultured to new media contained different growth regulators. Shoots multiplied most effectively on a WPM containing 1.0 mg/l BA, producing 5.5 shoots with a shoot length of 2.5 cm per shoot explant.

Survival and Early Growth of Populus alba × P. grandidentata In Vitro Culture Plantlets in Soil (Populus alba × P. grandidentata 조직배양묘(組織培養苗)의 토양(土壤)에서의 활착(活着)과 생장(生長))

  • This study was undertaken to find out the effects of three kinds of potting media and two sources of explants on the survival and early growth of new plantlets of Poputus alba ${\times}$ P. grandidentata in the greenhouse. The results obtained can be summarized as follows; 1) Among three potting media, Terralite was best for early growth and survival of plantlets. 2) Like humidifier, an intermittent misting system can be effective in keeping relative humidity high for the plantlets. 3) Survival rates over 80% could be obtained if humidity was kept high during the hardening period. 4) During hardening period, the plantlets showed the juvenile characteristics such as smaller leaves, thinner stems, and shorter internodes. 5) There were no differences on morphological characteristics between the plantlets originating from axillary buds and the plantlets originating from multiple shoots while they were growing at the greenhouse. 6) The plantlets originating from bud culture grew normally comparing to regular cuttings.
