Effect of GA3 Treatment on Bud Formation, Fruit Set, and Enlargement in Ardisia pusilla (GA3에 의한 산호수의 화아형성과 착과 및 비대 증진효과)

  • The objective of this study was carried out to investigate the proper plant growth regulator for increasing the number of flower, fruit set, and to enlarge the size of the berries in Ardisia pusilla. Flower bud formation was used rooted cutting, and fruit set, enlargement, and coloration of fruit were used with two years-old. $GA_3$ concentrations were treated with 0, 100, 200, or $400mg{\cdot}L^{-1}$. Flower bud formation was effective in $400mg{\cdot}L^{-1}$ $GA_3$ and it was 1.8 times greater than control. Plant growth regulators were applied by foliar spray at full bloom stage to increase the fruit set. As a result, $GA_3$ was the most effective for increasing fruit set. Also, auxins of 4-CPA (Tomatotone, Donbu hitech Co., Korea) and dichloprop triethanol amine (Antifall, Bayer Crop Science Co., Ltd., Korea) were effective. When $GA_3$ concentrations of 0.5 and $1.0mg{\cdot}L^{-1}$ were used, fruit set (%) reached to 70% and 77%, respectively. Effectiveness of $GA_3$ was 1.8 times greater than control. Also, auxins, dichloprop triethanol amine increased to about 7-12% during fruit setting, but cytokinin and anti-gibberellin were ineffective. To investigate the fruit enlargement and coloration, $GA_3$ was treated with 0.3, 0.6, and $1.2mg{\cdot}L^{-1}$. Fruit enlargement was achieved to about 15% by $GA_3$ $0.6mg{\cdot}L^{-1}$ when $GA_3$ was treated 3 times at the interval of 1 month per treatment when fruit size was about 2-3mm (after full-blooming two months). But anthocyanin contents for coloration of fruit skin were not significant according to $GA_3$ concentration. The results showed that $GA_3$ enhanced bud formation, fruit set and enlargement of fruit size in Ardisia pusilla.

The Promotive Effect of NAA, IBA and Ethychlozate on Rooting Cuttings of Certain Ornamental Plants and Some Physiological Studies. (관상식물 삽목발근에 있어서 NAA, IBA 및 Ethychlozate의 발근촉진효과와 그 생리학적연구)

  • The present studies were undertaken to elucidate the influence of auxins, auxin-like substance-ethychlozate ("Figaron"),and pH and sort of rooting media on rooted propagation of certainornamental woody plant cuttings, and to see possible changes in internal compositions characterizing after root-promoting treatment as the cutting stage proceeded. The experimental check-up srevealed and summarized as seen in the following;I. Effect of three different auxin treatments on rooting cuttings: 1) Promotive influence of auxin varied according to different concentration levels, hours of dipping treatment of the auxins, and kind of plants. The greatest effect was obtained for Forsythia ksreana with NAA and IBA, for Ligustrurn obtusifolium var. variegatum with NAA and ethychlozate, for Hydrangea macrophylla, Magnolia kobus, and Magnolia liliflora with NAA, lBA and ethychlozate also. The most effective level of the promotive agents was found 200mg/l for NAA, 1000mg/l for IBA, and 200mg/l for ethychlozate. For Weigela florida and Gardenia jasminoides, range of the most effective level was shown relatively wide spread. 2) NAA was more effective at its optimal level of the rooting agent than ethychiozate for Weigela florida, Viburnum awabuki, Forsythia koreana, Acer palmatum 'Nomura', Bouga invillea glabra, Elaeagnus umbellata, Prunus tomentosa, Ligustrum obtusifolium, Pyracantha coccinea, Cestrum noctu rnum, Hydrangea macrophylla, Codiaeum variegatum, Rhododen dron lateritium, and Ilex crenata var. macrophylla, and yet ethychlozate was found either as equally as effective or more so than NAA for Zebrina pendula, Hibiscus syriacus, Fatshedera lizei, Schefflera arboricola, Campsis grandiflo ra, Ixora chinensis, Euonymus japonica, and Magnolia liliflora. On the contrary, no the auxin effect was noted with Lagerstroemia indica, Trachelospermum asiaticum, and Syringa vulgaris. This probably indicates that these species are genetically different for the auxin response.II. Effect of different pH and sorts of cutting media on rooting cuttings: 1) Bougainvillea showed best in rooting for the number and dry weight at pH 6.5, more with ethychlozate than NAA, while Ligustrum did at pH 5.0 more with NAA than ethychlozate. pH 4.0 medium resulted in the best rooting for Rhododendron with NAA, more than ethychlozate. 2) Use of cutting medium with peat: perlite: vermiculite = 1:1:1 showed to give the greatest rooting percent and dry weight, apart from considering the number of roots. This apparently meant the fact that cutting medium has more to do with root growth than root differentiation. Rhododendron yet showed results with cutting media that use of peat: perlite = 2:1 mixed is more effective on rooting than using peat alone.III. Effect of auxinic treatments on rooting cuttings and change in some cutting compositions: 1) Under the climatic conditions of July having temperature $26.3\pm$$2.4^{\circ}C$for cutting bed, new roots of Magnolia started to show up generally 20 days after the cutting was made, whereas Cestrum did much earlier than that, namely 14 days after. 2) Although total carbohydrate content of Magnolia cuttings showed no marked change without auxin treatment, it did so with the treatment, especially 30 days after the start of cutting. Cestrum cuttings demonstrated a gradual in crease in total carbohydrate content as rooting took place, and the content became reduced more with auxin than with out, just about when rooting proceeded to 14 days after the start of cutting. 3) Magnolia generally showed an increase in total nitrogen content as rooting proceeded more, and Cestrum showed a decrease in total nitrogen of cuttings. The auxin treatment exhibited no pertinent relation with change in plant nitro gen when rooting is promoted with auxin treatment. 4) An abrupt drop of total sugar and reducing sugar was noticed as Magnolia rooting started, and this reduction was parti cularly outstanding with auxin treatment. Starch content also was decreased in the later stage of cutting with auxin treatment, and was rather increased without auxin. Although sugar content soon increased as cutting started with auxin treatment in the case of Cestrum, it became reduced after rooting took place. 5) Total phenol content increased with rooting, and this was especially true when rooting started. This increase was reversed somehow regardless of auxin treatment. A decrease in phenol of Magnolia was found more striking with auxin than without in the later stage of the cutting period. 6)Avena coleoptile test for auxin-like substances presented the physiologically active factor is more in easy-to-root Magnolia liliflora than hard-to-root Magnolia kobus, and the activity of auxin-like substances was much increased with auxin treatment. The increase in the growth promoting substances was markedly pronounced when rooting just started. The active growth substances decreased in the later stage of cutting, and certain inhibitory substances started appearing. Cestrum also showed physiologically similar growth promoting substances accompanying auxin-like active substances if auxin is treated, and some strong inhibitory substances seemed to appear in the later stage of cutting. 7) Mung-bean-rooting test indicated biologically that endogenous growth substances in Magnolia all promoted mung-bean rooting, and activity of the growth substances apparently stimulated mung-bean rooting with auxin more than without. Here auxin treatment seemed to give a rise to an increased activity of endogenous growth substances in cuttings. This activity was found much greater with either NAA or IBA than ethychlozate, and showed its peak of the activity when rooting first started taking place. Certain inhibitory substances for Avena coleoptile growth strongly promoted mung-bean rooting, and it was also much like in the case of Cestrum.

Studies on the Anther Culture of Some Woody Species (목본식물(木本植物)의 약배양(葯培養)에 관(關)한 연구(硏究))

  • Recently successful induction of haploid plant by means of anther culture method has become a big topic among geneticists and plant breeders. The haploid plant can be used as a precious material for such basic researches as mutation or genetics. Once the haploid is obtained, production of homozygous plant is not a difficult problem. The method of producing homozygous plant can, also, be applied to the practical breeding works. When applied to the hybridization of self-fertilizing breeding period would be greatly shortened and in cross-fertilizing vegetables production of uniform hybrid seed would be very easily obtained. Last few years many scientists attempted anther cultures using various plant species, but it was successful only in several species. Unlike the other tissue cultures which use somatic organs or tissues as explants, anther culture seems to be very difficult because the plants or calli have to be induced from the haploid microspores or pollen grains. In the present experiment anther culture of fruit trees and ornamental shrubs of four genera and seven species was attemped. Anthers of Various stages ranging from tetrad and late microspore were cultured on the modified Murashige and Skoog's medium supplemented with various concentrations of auxins and kinetin as growth regulators. Handling of materials, sterilization, and other operations of culture were done by routine methods. The results were summarized as follows: 1. Calli were induced in the anthers of Forsythia Koreana Nak., Rhododendron mucronuratum Turcz., R. yedoense Max. var. Poukhanense Nak., and Prunus armeniaca L. var. ansu Max. No signs of callus were observed in Prunus persica Sieb. et Zucc. var. vurgaris Max., Pyrus ussuriensis var. macrostipes (Nak.), and Prunus salcina Lindley. 2. Calli were easily formed in any of the media with differing concentrations of auxins and kinetin. 3. In F. Koreana calli developed from anther surface and connective. Callus emerging out of anther locule was not observed. 4. Somatic calli arose from filament, connective, and inside of anther wall in R. mucronulatum. Many of the microspores accumulated starch grains. 5. The anther lobes located opposite the filament of R. yedoense turned easily to calli. This phenomenon was not observed in R. mucronulatum. Microspore embedded for a period in the medium became starch pollen. No callus was observed arising from microspore. 6. In P. armeniaca calli were not induced from somatic anther tissues. Instead, callus emerged out of anther locule rupturing the anther slit. Starch was not formed in the microspore. 7. In P. persica, Pyrus ussuriensis, and P. salcina, calli were not observed in the anthers examined more than 60 days after culture. Microspores of these species, however, were free of starch grains even after long period of subculture. 8. It was learned that somatic calli of the species examined arose usually from endothelium of anther wall, septum of two neighboring anther locules, parenchyma tissues of connectives, or anther lobes. 9. In the anther locule of P. armeniaca cultured long in medium, swollen microspores, polynucleate microspores, multicellular pollen grains, or callus mass were frequently observed, this indicating that the callus of this species was microspore-origin. 10. It was clarified that in P. armeniaca production of haploid plant by anther culture might be possible.

  • aA series of experiments were conducted to investigate the effect of plant growth regulator treatments on the growth and lateral root formation in soybean sprouts in order to establish the effective method of producing root-less or short-rooted soybean sprouts with larger diameter in the hypocotyl. Major results can be summarized as follows. 1. Soybean sprouts showed fairly uniform elongation rate from 3 to g days after imbibition with daily increase of 3.8cm. The speed of elongation of hypocotyl was reduced whereas that of root accelerated 7 days after imbibition. Lateral roots began to emerge fairly evenly from 5 to 9 days after imbibition with a daily increase of 4.4. 2. Auxins(IAA, IBA, NAA, 2,4-D) inhibited hypocotyl elongation and formation of lateral roots and increased hypocotyl diameter without influencing root length and hook diameter at higher concentrations. The dry weight of cotyledon was increased significantly as compared to that of hypocotyl and root. Among the tested auxins, 2, 4-D was the most effective. 3. BA and 4PU-30 significantly reduced elongation of hypocotyl and root and resulted in the biggest diameter of hypocotyl when treated at higher concentrations. The lowest effective concentration of BA to prevent the formation of larval gal roots was 12.5ppm. The formation of lateral roots could be completely prevented by BA and 4PU-30 treatment but kinetin, zeatin, zeatin riboside resulted in many lateral roots and increased thickness of soybean sprouts with little influence. Cotyledon deformation was found in soybean sprouts treated by 4PU-30. 4. 2, 4-D was the most effective for increasing the hypocotyl diameter while 4PU-30 was the most effective for reducing no. of lateral roots. 5. It can be concluded that among the plant growth regulators tested, BA was effective in reducing root length and increasing hypocotyl diameter. BA 12.5 ppm or 15 ppm may thus be the more practical for production of soybean sprouts. 6. ABA showed no significant effect of growth parameter, however ABA 25 ppm inhibited only no of lateral roots with little influence on the growth of seedling. 7. Ethephon inhibited the elongation of hypocotyl and root and increased hypocotyl diameter at higher concentrations. 8. The combined effect of cytokinins and ethephon was very similar to result of BA treatment alone. As the ethephon concentration increased, hypocotyl diameter and dry weight of cotyledon tended to increase.

Effect of tissue proliferation and somatic embryo induction in Larix kaempferi following treatment with organic nitrogen sources and plant growth regulators (일본잎갈나무(Larix kaempferi) 유기질소원 및 식물생장조절물질 처리에 따른 조직증식 및 체세포배 유도 효과)

  • This study was conducted to evaluate the effects of different types and concentrations of organic nitrogen sources (${\small{L}}$-Glutamine and casein hydrolysate, CH) and plant growth regulators (auxins and cytokinins) on embryogenic tissue proliferation and somatic embryo production in L. kaempferi. Overall, the highest tissue fresh weight was obtained at either 2 or 4 weeks in culture when 1,000 mg/L ${\small{L}}$-Glutamine was added to the culture medium, which showed similar results with other treatments. In experiments with different types and concentrations of plant growth regulators on somatic embryo production, the highest production (426.3/90 mg tissue) was found when 0.2 mg/L IBA was added; however, no somatic embryos were induced following treatment with 0.2 mg/L BA or Kinetin. The effect of various concentrations of IBA on somatic embryo production was also tested. The best result (303/90 mg tissue) was obtained when plants were treated with 0.2 mg/L IBA; 1.0 mg/L IBA was also effective (281/90 mg tissue). The lowest result (109.3/90 mg tissue) was obtained with 5.0 mg/L IBA.

The Combination Effect of Kinetin and Auxin on the Growth Root Development and Vitamin C Content of Soybean sprouts (콩나물의 성장과 Vitamin C 생성에 미치는 Kinetin과 Auxin의 혼합효과)

  • This study was conducted to investigate the growth of soybean sprouts and the effective hormonal refutation of vitamin C content by the concentration and combination of kinetin and auxins such as IAA, 2.4-D and NAA. The results were as follows; 1. The growth of soybean sprouts were fine under the respective conditions of kinetin $10^{-8}M+IAA\;10^{-6}M$, kinetin $10^{-7}M+IAA\;10^{-5}M$, kinetin $10^{-6}M+IAA\;10^{-6}M$, kinetin $10^{-5}M+IAA\;10^{-7}M$, kinetin $10^{-8}M+2,4-D\;10^{-8}M$ and kinetin $10^{-7}M+NAA\;10^{-5}M$. 2. The root development of soybean sprouts were almost in accord with the growth of soybean sprouts by the concentration of kinetin and auxin. 3. The content of vitamin C were more increased under in kinetin $10^{-8}M+IAA\;10^{-8}M$, kinetin $10^{-6}M+2,4-D\;10^{-5}M$ and kinetin $10^{-8}M+NAA\;10^{-8}M$ and both growth and vitamin C content were more activated under kinetin $10^{-8}M+IAA\;10^{-6}M$ and $10^{-8}M+NAA\;10^{-8}M$. 4. The growth, root development and vitamin C content were increased in the low kinetin and high IAA, NAA concentration. Root development and vitamin C content of soybean hypocotyl were decreased in the low kinetin and high 2,4-D concentration.

Induction and growth of adventitious roots and bioreactor culture in Codonopsis lanceolata (더덕 (Codonopsis lanceolata)의 부정근 유도 및 생장에 미치는 배양조건과 생물반응기 배양)

  • This paper reported the establishment of mass production system of adventitious roots of Codonopsis lanceolata through shake flask and bioreactor culture. Induction of adventitious roots was started from the explants of leaf, stem and root on 1/2 strength Murashing and Skoog (MS) solid medium. Stem segments were the best explants for induction of adventitious roots compared to leaf and root segments. Among the different auxins tested (NAA, IBA and IAA), number of adventitious root per explant was highest on solid medium with 1.0 mg/L IBA, and produced $9.9{\pm}1.2$ roots per explant. However, growth of adventitious roots was fast in the presence of IBA at low concentration (0.1 mg/L). In shake flask culture, maximum production of adventitious roots (fresh weight) was obtained in half-strength MS medium compared to full-strength and one-third MS medium. When the adventitious roots produced in shake-flask culture were transferred to 5 L air-lift bioreactor, 16 times of fresh weight increase was gained after one month of culture. These results indicate that this protocol for the production of C. lanceolata adventitious roots can be applied to large scale culture for practical application.

Auxin and Cytokinin Affect Biomass and Bioactive Compound Production from Adventitious Roots of Eleutherococcus koreanum (섬오갈피 부정근 배양 시 오옥신과 사이토키닌이 생장과 생리활성물질 생산에 미치는 영향)

  • In an attempt to improve biomass and bioactive compound production, we cultured adventitious roots of $Eleutherococcus$ $koreanum$ in 250 mL Erlenmeyer flasks using Murashige and Skoog (MS) medium with different concentrations of auxins (IBA, NAA, IAA) and cytokinins (BA, kinetin, TDZ). Root biomass (fresh and dry weight) was enhanced at $5mg{\cdot}L^{-1}$ indole-3-butyric acid (IBA) after 5 weeks of culture. The content of total phenolics and flavonoids was also increased with $5mg{\cdot}L^{-1}$ IBA compared to ${\alpha}$-naphtalene acetic acid (NAA) or indole-3-acetic acid (IAA) treatments. The combination of $5mg{\cdot}L^{-1}$ IBA with $0.1mg{\cdot}L^{-1}$ thidiazuron (TDZ; N-phenyl-N'-1,2,3,-thidiazol-5-ylurea) enhanced root fresh and dry weight (1.4- and 1.6-fold, respectively) as well as the content of total phenolics and flavonoids compared to the relative control (without cytokinin). On the contrary, $N_6$-benzyladenine (BA) and 6-furfurylaminopurine (kinetin) did not significantly affect root biomass and bioactive compound production in adventitious roots of $E.$ $koreanum$. These results suggested that $5mg{\cdot}L^{-1}$ IBA combination with $0.1mg{\cdot}L^{-1}$ TDZ supplementation was most suitable for both biomass and bioactive compound production from adventitious roots of $E.$ $koreanum$.

Induced Systemic Resistance in plants by Bacillus sp. Isolated from Dok-do Islands (독도 자생식물 번행초로부터 분리한 바실러스 속 식물생장촉진근권 세균에 의한 식물병 저항성 유도)

  • In September 2017, the rhizospheric soil of Tetragonia tetragonoides (Pall.) Kuntze was further sampled. One hundred and thirty eight species of microorganisms were isolated from the soil. Indole-3-acetic acid (IAA) production, siderophore production, and phosphate degradation were examined in order to confirm bacterial growth from isolated microorganisms. As a result, most strains were able to produce auxins or siderophores and to solubilize phosphate. In addition, 138 isolated strains were treated with tobacco extract and conferred pathogen resistance to host plants upon treatment. As a result, 35 strains that were able to reduce pathophysiology by more the 60% were selected. Among them, 6 strains with high induced systemic resistance (ISR) activity were found. All of these strains belong to the genus Bacillus according to the 16S rDNA sequence analysis. Bacillus aryabhattai KUDC6619 showed outstanding effects with reduced infection in tobacco and pepper plants. Probably, these Bacillus species play a beneficial role by association with T. tetragonoides for its survival in the harsh conditions found on the island of Dokdo.

Micropropagation of Cucurbita foetidissima and Asclepias syriaca through Shoot Tip Culture of Seeding (Cucurbita foetidissima 와 Asclepias syriaca의 정단배양을 통한 기내 대량증식)

  • In order to establish a micropropagation system for buffalo gourd (Cucurbita foetidissima ) and common milkweed (Asclepias syriaca), the effects of several plant growth regulators and culture temperature on shoot multiplication and rooting were investigated. In buffalo gourd, the greatest number of shoot from shoot tip culture and well growth of formed shoot were obtained on the MIS medium supplemented with 1.0 mg/L BA and 0.3 or 0.6 mg/L IAA. Whereas kinetin and 2iP were not effective for shoot multiplication in vitro. It was found that 22$^{\circ}C$ and $25^{\circ}C$ were suitable for shoot multiplication. Roots were easily formed by the addition of auxins, especially 1.0 or 2.0 mg/L IBA and 2.0 mg/L IAA. Over 90% of plants survived successfully after being transferred into the field. In common milkweed, BA was more effective than kinetin or 2iP for its micropropagation in vitro. The increased shoot weight and number of nodes per shoot were obtained on the medium containing 3.0 mg/L BA and 0.3 or 0.6 mg/L IAA. But 2iP promoted the shoot elongation. In addition. common milkweed was sensitive to culture temperature in vitro. Temperature around 22$^{\circ}C$ was favorable for shoot multiplication and growth, whereas temperature higher than $25^{\circ}C$ usually reduced the rate of shoot survival rate.
