Auranofin is a US Food and Drug Administration (FDA)-approved anti-arthritis medication that functions as a thioredoxin reductase inhibitor. Spermidine, a polyamine present in marine algae, can exert various physiological functions. Herein, we examined the synergistic anticancer activity of auranofin and spermidine in hepatocellular carcinoma (HCC). Combined treatment with auranofin and spermidine suppressed cell viability more efficiently than either treatment alone in HCC Hep3B cells. The isobologram plotted by calculating the half maximal inhibitory concentration (IC50) values of each drug indicated that the two drugs exhibited a synergistic effect. Based on the analysis of annexin V and cell cycle distribution, auranofin and spermidine markedly induced apoptosis in Hep3B cells. Moreover, auranofin and spermidine increased mitochondria-mediated apoptosis by promoting mitochondrial membrane potential (Δψm) loss. Auranofin and spermidine significantly increased reactive oxygen species (ROS) production in Hep3B cells, and the blocking ROS suppressed apoptosis induced by spermidine and auranofin. In addition, auranofin and spermidine reduced the expression of phosphorylated phosphatidylinositol-3 kinase (PI3K) and protein kinase B (Akt), and PI3K inhibitor accelerated auranofin- and spermidine-induced apoptosis. Using ROS scavenger and PI3K inhibitor, we revealed that ROS acts upstream of auranofin- and spermidine-induced apoptosis. Collectively, our study suggests that combination treatment with auranofin and spermidine could afford synergistic anticancer activity via ROS overproduction and reduced PI3K/Akt signaling pathway.

Signal transducer and activator of transcription 3 (STAT3) and telomerase are considered attractive targets for anticancer therapy. The in vitro anticancer activity of the gold(I) compound auranofin was investigated using MDA-MB 231 human breast cancer cells, in which STAT3 is constitutively active. In cell culture, auranofin inhibited growth in a dose-dependent manner, and N-acetyl-L-cysteine (NAC), a scavenger of reactive oxygen species (ROS), markedly blocked the effect of auranofin. Incorporation of 5-bromo-2'-deoxyuridine into DNA and anchorage-independent cell growth on soft agar were decreased by auranofin treatment. STAT3 phosphorylation and telomerase activity were also attenuated in cells exposed to auranofin, but NAC pretreatment restored STAT3 phosphorylation and telomerase activity in these cells. These findings indicate that auranofin exerts in vitro antitumor effects in MDA-MB 231 cells and its activity involves inhibition of STAT3 and telomerase. Thus, auranofin shows potential as a novel anticancer drug that targets STAT3 and telomerase.
Auranofin has been developed as antirheumatic drugs, which is currently under clinical development for the treatment of chronic lymphocytic leukemia. Previous report showed that auranofin induced apoptosis by enhancement of annexin A5 expression in PC-3 cells. To understand the role of annexin A5 in auranofin-mediated apoptosis, we performed microarray data analysis to study annexin A5-controlled gene expression in annexin A5 knockdown PC-3 cells. Of differentially expressed genes, plasminogen activator inhibitor (PAI)-2 was increased by annexin A5 siRNA confirmed by qRT-PCR and western blot. Treatment with auranofin decreased PAI-2 and increased annexin A5 expression as well as promoting apoptosis. Furthermore, auranofin-induced apoptosis was recovered by annexin A5 siRNA but it was promoted by PAI-2 siRNA. Interestingly, knockdown of annexin A5 rescued PAI-2 expression suppressed by auranofin. Taken together, our study suggests that induction of annexin A5 by auranofin may enhance apoptosis through suppression of PAI-2 expression in PC-3 cells.
Axl receptor tyrosine kinase has been implicated in cancer progression, invasion, and metastasis in various cancer types. Axl overexpression has been observed in many cancers, and selective inhibitors of Axl, including R428, may be promising therapeutic agents for several human cancers, such as breast, lung, and pancreatic cancers. Here, we examined the cell growth inhibition mediated by R428 and auranofin individually as well as in combination in the human breast cancer cell lines MCF-7 and MDA-MB-231 to identify new advanced combination treatments for human breast cancer. Our data showed that combination therapy with R428 and auranofin markedly inhibited cancer cell proliferation. Isobologram analyses of these cells indicated a clear synergism between R428 and auranofin with a combination index value of 0.73. The combination treatment promoted apoptosis as indicated by caspase 3 activation and poly (ADP-ribose) polymerase cleavage. Cancer cell migration was also significantly inhibited by this combination treatment. Moreover, we found that combination therapy significantly increased the expression level of Bax, a mitochondrial proapoptotic factor, but decreased that of the X-linked inhibitor of apoptosis protein. Furthermore, the suppression of cell viability and induction of Bax expression by the combination treatment were recovered by treatment with N-acetylcysteine. In conclusion, our data demonstrated that combined treatment with R428 and auranofin synergistically induced apoptosis in human breast cancer cells and may thus serve as a novel and valuable approach for cancer therapy.

The thioredoxin (Trx) system plays critical roles in regulating intracellular redox levels and defending organisms against oxidative stress. Recent studies indicated that Trx reductase (TrxR) was overexpressed in various types of human cancer cells indicating that the Trx-TrxR system may be a potential target for anti-cancer drug development. This study investigated the synergistic effect of auranofin, a TrxR-specific inhibitor, on sulforaphane-mediated apoptotic cell death using Hep3B cells. The results showed that sulforaphane significantly enhanced auranofin-induced apoptosis by inhibiting TrxR activity and cell proliferation compared to either single treatment. The synergistic effect of sulforaphane and auranofin on apoptosis was evidenced by an increased annexin-V-positive cells and Sub-G1 cells. The induction of apoptosis by the combined treatment caused the loss of mitochondrial membrane potential (ΔΨm) and upregulation of Bax. In addition, the proteolytic activities of caspases (-3, -8, and -9) and the degradation of poly (ADP-ribose) polymerase, a substrate protein of activated caspase-3, were also higher in the combined treatment. Moreover, combined treatment induced excessive generation of reactive oxygen species (ROS). However, treatment with N-acetyl-L-cysteine, a ROS scavenger, reduced combined treatment-induced ROS production and apoptosis. Thereby, these results deduce that ROS played a pivotal role in apoptosis induced by auranofin and sulforaphane. Furthermore, apoptosis induced by auranofin and sulforaphane was significantly increased through inhibition of the phosphoinositide 3-kinase (PI3K)/Akt pathway. Taken together, the present study demonstrated that down-regulation of TrxR activity contributed to the synergistic effect of auranofin and sulforaphane on apoptosis through ROS production and inhibition of PI3K/Akt signaling pathway.

Activation of the NLRP3 inflammasome is critical for host defense as well as the progression of inflammatory diseases through the production of the proinflammatory cytokine IL-1β, which is cleaved by active caspase-1. It has been reported that overactivation of the NLRP3 inflammasome contributes to the development and pathology of acne vulgaris. Therefore, inhibiting activation of the NLRP3 inflammasome may provide a new therapeutic strategy for acne vulgaris. In this study, we investigated whether auranofin, an anti-rheumatoid arthritis agent, inhibited NLRP3 inflammasome activation, thereby effectively treating acne vulgaris. Auranofin suppressed NLRP3 inflammasome activation induced by Propionibacterium acnes, reducing the production of IL-1β in primary mouse macrophages and human sebocytes. In a P. acnes-induced acne mouse model, injection of P. acnes into the ears of mice induced acne symptoms such as redness, swelling, and neutrophil infiltration. Topical application of auranofin (0.5 or 1%) to mouse ears significantly reduced the inflammatory symptoms of acne vulgaris induced by P. acnes injection. Topical application of auranofin led to the downregulation of the NLRP3 inflammasome activated by P. acnes in mouse ear skin. These results show that auranofin inhibits the NLRP3 inflammasome, the activation of which is associated with acne symptoms. The results further suggest that topical application of auranofin could be a new therapeutic strategy for treating acne vulgaris by targeting the NLRP3 inflammasome.
내재면역반응의 중요한 조절자인 Nrf2와 NF-${\kappa}B$는 염증시에 교차 작용을 통하여 서로의 전사활성을 조절할 수 있다고 보고된 바 있으나 상반된 결과도 제시되고 있어서 아직까지 확실하게 규명되어 있지 않다. 저자들은 선행연구에서 NF-${\kappa}B$ 저해제인 금(I)-화합물 오라노핀이 인간 관절활막세포와 단핵구성 세포에서 Nrf2를 활성화시킴을 확인한 바 있기 때문에, 본 연구에서는 Nrf2를 knockdown 시킨 류마티스성 활막세포를 사용하여 오라노핀에 의해 저해되는 NF-${\kappa}B$ 신호전달 과정에 Nrf2가 관여하는지를 조사하였다. 세포를 Nrf2 siRNA로 transfection시켰을 때 Nrf2 발현은 대부분 차단됨을 확인하였다. 하지만 Nrf2 knockdown은 TNF-$\alpha$에 의해 유도되는 $I{\kappa}B-{\alpha}$ 분해를 막는 오라노핀의 작용에는 영향을 주지 않았다. Nrf2 target 단백질로서 항염 작용에 관여하는 HO-1을 knockdown 시켰을 경우에도 $I{\kappa}B-{\alpha}$ 분해를 저해하는 오라노핀의 작용에 영향을 미치지 않았다. 또한, Nrf2 knockdown은 오라노핀에 의해 저해된 ICAM-1 발현을 다시 복원시키지 못했다. 이러한 결과들은 염증성 싸이토킨에 의해 유도되는 NF-${\kappa}B$ 활성화를 오라노핀이 저해하는 기전에 Nrf2 및 HO-1이 관련되어 있지 않음을 시사한다. 따라서 류마티스성 관절활막세포에서 오라노핀의 항염작용 기전으로 알려진 Nrf2/HO-1 활성유도와 NF-${\kappa}B$ 활성저해는 교차작용 없이 각각 독립적인 기전을 통해 나타나는 것으로 생각된다.
산화적 스트레스에 의한 망막 색소상피 세포의 손상 및 퇴화는 시력 소실을 포함한 다양한 망막질환의 원인으로 알려져 있다. 본 연구에서는 천연물 유래 14종 단일 성분과 합성 화합물 2종을 대상으로 망막질환 개선 가능성을 평가하기 위해, 사람 유래 망막 색소상피 세포를 대상으로 산화적 스트레스에 의한 세포 손상에 대한 보호 효능을 평가하였다. 그 결과, 16종의 후보물질 중 H2O2에 의해 유발된 세포독성을 개선시키는 효능을 보인 5종(auranofin, FK-509, hemistepsin A, honokiol 및 spermidine)을 선별하였다. 이를 기반으로 H2O2에 의한 미토콘드리아 기능 손상 및 활성 저하 억제능을 평가하였으며, auranofin을 제외한 4종에서 유의한 세포 보호 효능이 관찰되었다. 더불어 H2O2에 의해 유도된 DNA 손상에 미치는 영향을 조사한 결과, FK-506, honokiol과 spermidine이 DNA 손상을 개선시켰음을 확인하였다. 그러나 5종의 후보물질이 활성 산소종의 생성에는 뚜렷한 억제 효능을 나타내지 않았으며, 이는 5종의 후보 물질이 ROS-비의존적 기전을 통해 세포 보호효과를 나타낼 것으로 사료된다. 이상의 결과를 바탕으로, 조사 대상 후보물질 중에서 spermidine이 산화적 스트레스로부터 망막 색소상피 세포의 보호 효능이 가장 우수한 천연물 유래 단일 물질임을 규명하였으며, 동시에 합성물질로는 FK-506이 망막 세포 보호 효능이 우수한 것으로 나타났다. 비록 작용 기전에 대한 추가 연구가 필요하겠지만, 본 연구 결과는 spermidine과 FK-506이 산화적 스트레스에 의한 망막질환의 위험을 억제할 가능성이 있음을 시사한다.
식품 원료로도 널리 애용되는 검은콩은 풍부한 천연 페놀 화합물을 함유하고 있기 때문에 기능성 소재로서의 개발에도 매우 유용한 자원이다. 본 연구에서는 3가지 검은콩 품종[소청자(SCJ), 소청2호(SC2) 및 청자2호(CJ2)]을 대상으로 TPCs과 항산화 능을 조사하였다. 그 중에서도 TPCs는 CJ2 $H_2O_2$ 처리에 의한 HaCaT 세포의 생존력 감소를 현저히 억제하여 산화적 스트레스에 대한 보호 효과가 있음을 알 수 있었다. SCJ와 SC2 전처리는 또한 HaCaT 세포에서 mitochondrial dysfunction의 차단과 pro-apoptotic Bax의 발현 변화의 정상화를 통해 $H_2O_2$에 의하여 유도된 apoptosis를 효과적으로 억제하였으며, DNA 손상에 대한 보호 효과와 연관성이 있었다. 또한 SCJ와 SC2는 Nrf2와 연관된 TrxR1의 발현을 효과적으로 유도하였으나, 산화적 스트레스에 대한 SCJ와 SC2의 보호 효과는 TrxR 억제제에 의하여 상쇄되었다. 이러한 결과는 SCJ와 SC2가 Nrf2 신호전달 경로 활성을 통하여 산화적 스트레스와 관련된 세포 손상을 차단함으로써 세포 보호 활성을 갖는다는 것을 의미한다. 결론적으로, SCJ와 SC2는 산화스트레스로 인한 피부 질환의 치료와 예방을 위한 응용 가능성이 높음을 보여주었다.
