Traditional Chinese herbal drugs, namely, Atractylodes Rhizome and Atractylodes Rhizome White were identified by means of chemical analysis such as TLC and GC/MS. In Korea, the old-grown and the newly-grown rhizomes of Atractylodes japonica Koidzumi (Compositae) are used as the Atractylodes Rhizome and the Atractylodes Rhizome White, respectively. Both rhizomes contain the atractylon asa major spot on TLC. The content of atractylon in a newly-grown rhizome of A. japonica (Atractylodes Rhizome White called in Korea) appears to contain much larger quantity in comparison with that of an old-growon one (Atractylodes Rhizome). The TLC spot pattern analysis showed that rhizomes of A. japonica and A. macrocephala contain the atractylon as a major indicator spot, but no atractylodin (extreamly small amount). However, rhizomes of A. lancea De Candolle and A. koreana Kitamura contain the atractylodin as a major indicator spot.
Traditional herbal drugs of Korea, namely, Atractylodes Rhizome(蒼朮) and Atractylodes Rhizome White(白朮) which are derived from and old-grown rhizome and a newly-grown rhizome of Atractylodes japonica Koidz.(Compositae), respectively, were investigated their potential liver-protective activities against hepatotoxicity induced by $CCl_4$ intoxication in mice. Each herbal drug was extracted separately with chloroform, methanol and butanol. No extract of Atractylodes Rhizome showed a significant reduction of the duration of hypnosis produced by hexobarital after $CCl_4$ intoxication in mice. In addition, the the treatments of Atractylodes Rhizome extracts produced no decrease of aspartate aminotransferase(EC 2.6.1.1) activity in serum. However, the methanol extract of Atractylodes Rhizome White exhibited a marked protection from hepatotoxicity induced by $CCl_4$ intoxication in mice. It produced significant reductions of the duration of hypnosis and serum enzyme activity, but no other extract showed liver-protective activity against $CCl_4-induced$ hepatotoxicity.
Objectives : Atractylodes rhizomes, which have been widely used to treat gastrointestinal disorders, consist of numerous chemical compounds. Polyacetylenes are the parts of characteristic compounds of importance required to understand the therapeutic properties of Atractylodes rhizomes. It is necessary to understand the physicochemical and pharmacological properties of polyacetylenes in the Atractylodes rhizomes.Methods : The literatures from 1970 to January 2016 were searched using Korean and international electronic databases. The chemical structures of polyacetylenes were drawn by structure-drawing software.Results : The reported polyacetylenes were classified by their chemical skeletons and original resources, and their physicochemical and pharmacological features were discussed. Polyacetylenes with skeletal moieties were reported, such as diene-diyne types (two double and two triple carbon-bonds), triene-diyne types (three double carbon bonds and two triple carbon bonds), and monoene-diyne types (one double carbon bonds and two double carbon bonds), with various functional groups. Atractylodin was most frequently reported from many Atractylodes species. Atractylodin-related polyacetylenes showed chemical instability in both high and freezing temperatures. Processing of the Atractylodes rhizomes by stir-frying with bran could affect the contents of polyacetylenes and their bioavailability in vivo. Several polyacetylenes showed structure-related anti-inflammatory activities and gastrointestinal activities.Conclusion : Polyacetylene compounds in Atractylodes rhizomes were based on three chemical backbones and showed diverse physicochemical and pharmacological features. The present study provides structural, physicochemical, and pharmacological information of polyacetylene from Atractylodes rhizomes. This information provides fundamental data for further research.
Objectives : When herbal medicines are extracted together, they may interact with each other, leading to change of chemical characteristics. This study aimed to evaluate the influence of Atractylodes rhizomes (Atractylodes japonica, A. macrocephala, and A. chinensis) on the chemical features of the roots and rhizomes of Glycyrrhiza uralensis, which is are commonly combined with herbal medicines in many herbal formulae, when they are co-decocted. Methods : Liquiritin apioside, liquiritin, ononin, and glycyrrhizin levels of G. uralensis in hot-water extracts prepared by the combination of Atractylodes rhizomes with various weight ratios (G. uralensis : Atractylodes rhizomes = 10:0, 10:5, 10:10, and 10:20) and extraction times (60, 90, and 120 min) were quantified using a HPLC-diode array detector and compared by statistical analysis. Results : The concentrations of liquiritin apioside, liquiritin, ononin, and glycyrrhizin from G. uralensis roots and rhizomes mostly reduced when co-extracted with Atractylodes rhizomes, and the addition of A. chinensis most reduced their contents between Atractylodes combination groups. A. japonica and A. macrocephala rhizomes also showed differences of liquiritin and glycyrrhizin levels at 10 g and 20 g groups of Atractylodes rhizomes. Extraction times also affected the concentrations of liquiritin, ononin, and glycyrrhizin mostly during 60 and 90 min. Conclusions : Atractylodes rhizomes might alter the chemical characteristics of G. uralensis when these herbs are co-decocted. This study provides the understanding of the chemical interactions of herbal medicines during the extraction in hot water.
This study was aimed at investigating hepatic toxicity and immunomodulating effects of defatted methanol extracts of two kinds of medicinal plants, Rubus coreanus Miq. and Atractylodes japonica Koidz. in mice. Defatted methanol extracts of fruits of Rubus coreanus Miq. and rhizome of Atractylodes japonica Koidz. were added at the level of 0.5% or 5%(w/w) to cholesterol-supplemented AIN-76 diet. Each diet was fed to 8 ICR male mice for 30 days. Weight gain and food efficiency ratio of the mice fed 5.0% extract of Rubus coreanus Miq. were significantly lower than those of the mice fed 0.5% extract Relative liver weight and activity of plasma alanine aminotransfernse were significantly increased only in the mice fed 5% extract of Atractylodes japonica Koidz. compared with the others. Splenocyte proliferation was not significantly different between the groups fed 0.5% or 5.0% extract of Rubus coreanus Miq. However, splenocyte proliferation was significantly decreased in the mice fed 5.0% extract of Atractylodes japonica Koidz. compared with that in the mice fed 0.5% Production of interleukin-2 by splenocytes from the mice fed 0.5% extract of Atractylodes japonica Miq. was significantly higher than the control value and it became lower with 5.0% dietary level. Secretion of $interferon-\gamma$ was not significantly different among groups. In conclusion, the defatted methanol extract of Atractylodes japonica Koidz. was likely to exert immunomodulating effect at the level of 0.5% but it may exert adverse effects on immune and liver functions at the level of 5.0%.
본 연구는 방사선 육종 차조기와 백출 복합물의 조골세포 분화 활성 및 파골세포 형성 억제를 조사하였다. 차조기와 백출 복합물은 MG-63 세포에서 ALP 활성 및 arlizarin red 염색을 확인하였고 조골세포 형성의 영향은 RAW 264.7 세포에서 TRAP 활성과 TRAP 염색을 진행하였다. 세포 독성시험에서 차조기와 백출 복합물은 50 ㎍/㎖ 농도 이하에서 안전한 것으로 확인되었다. ALP 활성 및 골석회화 형성 능력은 대조군보다 활성이 낮았으나, 파골세포에서 TRAP 활성을 유의적으로 감소시켰으며, 효과적으로 TRAP(+) 다핵세포를 억제하였다. 따라서 차조기와 백출 복합물은 골 흡수 억제 활성을 향상시켜 뼈 관련 질환의 예방 및 치료에 효과적인 것으로 보여진다.
This study was carried out to compare the major volatile components in essential oil from different origin of Atractylodes spp. which is being traded as a crude herbal drug in Korean herbal markets. From the two Atractylodes of major volatile components were similarly detected such as the ${\beta}-selinene,\;{\beta}-sesquiphellandrene$, germacrene B, 2,7-dimethoxy-2-methylnaphthalene and 9-methoxy-2,3-dihydrofuro{3,2-q}coumarin. Among the volatile components, the major components were 2,7-dimethoxy-2-methylnaphthalene (40.98%), 9-methoxy-2,3-dihydrofuro {3,2-q} coumarin (15.74%), and ${\beta}-sesquiphellandrene$ (1.98%) in both Atractylodes. As a results, It was found that the two Atractylodes were the same species which was being traded in the Korean herbal markets as the A. japonica. not to different species of A. japonica and A. macrocephalla, respectively.

Atractylodes japonica and Atractylodes macrocephala belongs to Atractylodes genus and theirs roots are used as 'Baek-chul'(White atractylodes rhizome) in Korea Pharmacopoeia. 'Baek-chul' widely used in traditional herbal remedies in Asia. Concurrently, the 'Baek-chul' demand enormously increased in the market due to the growing interest in favoring health benefits but it is depend on import from China. Thus we conducted breeding research to solve the problems associated with domestically cultivated 'Baek-chul' such as low productivity and low disease tolence. We crossed A. japonica (female parent) and A. macrocephala (male parent) in the greenhouse in September 2000. the next season 119 crossed line seeds were collected and germinated in the green house. Among them we selected the good lines in the same season. then conducted replicated yield trials (RYT) in 2014 to local adaptability test (LAT) in 2015-2016. The results showed that the 'AJM16' line has beneficial charactersitcs like, increased plant height, large rhizome diameter, narrow leaf width compared with a 'Sang-chul' (check cultivar) and varied flower color from A. japonica (female parent). The line 'AJM16' was also exhibited decreased anthracnose symptoms(Colletotrichum gloeosporides Penz.). Simultaneously the root yield of AJM16 was 2,409 kg per are, which was increased 130% higher yield compared to 'Sang-chul' (check cultivar) at three various regions from 2015 to 2016. Therefore, we named AJM16 as new cultivar 'Man-chul'. According the results, we have developed a new Atractylodes spp. hybrid cultivar 'Macn-chul' with high yielding and disease tolerance by National Institute of Horticulture and Herbal Science, RDA, in 2016.
Objective : In this study, the effect of Atractylodes japonica against LPS induced inflammation in mouse microglia BV2 cells was investigated. Method : Microglia BV2 Cells viability was determined using the MTT assay. We used water, ethanol extract from Atractylodes japonica and studied on the anti-inflammatory effect of lipopolysaccharide-induced inflammation using reverse transcription polymerase chain reaction (RT-PCR), western blot, and nitric oxide detection on mouse microglia BV2 cells. Result : The MTT assay revealed that it's extract has no significant cytotoxicity in the microglia BV2 cell. Various solvent extract from Atractylodes japonica inhibited nitrite production, iNOS protein and mRNA expression levels. And also it's extracts significantly reduced lipopolysaccharide-induced COX-2 activation in RT-PCR and western blot in lipopolysaccharide-induced microglia BV2 cells Conclusion : In this study, it's extracts was shown to suppress NO production by inhibiting iNOS expression and COX-2 activity. With this effects of anti-inflammation, we suggests that, it's extracts may be a useful candidate for the development of a drug on the related inflammatory diseases in brain.
Scopolia rhizome is mistaken as an atractylodes rhizome because of their similarities in shape. That is why atractylodes rhizome imported from China sometimes contain scopolia rhizome, which is very toxic. 8 persons were intoxicated atractylodes after taking imported atractylodes rhizome which is tainted. In kampo medicine prepared with such imported atractylodes rhizome, the level of tropane alkaloids ranged from 1.12∼4.34 mg/dose. In this study, we tried to investigate the tissue distribution of scopolia rhizome in rats. The extracts of scopolia was administered orally to rats (a single dose of 10mg/kg, 20mg/kg and 7 days repeated dose of 10mg/kg). Their blood was collected at 0.5, 1, 2, 4, 6 hrs, and liver, kidney, lung and spleen were collected after 6 hrs. The tissue homogenate was applied to solid phase extraction column for the determination of tropane alkaloids. After the oral administration of 20mg/kg scopolia extracts, l-hyoscyamine was detected in rat blood to 2 hrs after dosing. The concentration of tropane alkaloids was the highest in liver followed by lung, kidney and spleen. However, lung, kidney and spleen were similar in amount.
