Anti-allergic Activity of Butanol Fraction of Asterias amurensis (불가사리 부타놀 분획의 항알러지 활성)

  • We investigated the effects of butanol fraction of Asterias amurensis (BFA) on the anti-allergic activity. BFA inhibited both cyclooxygenase-2 dependent prostaglandin $D_2$ and 5-lipoxygenase dependent leukotriene $C_4$ generation in a concentration-dependent manner with $IC_{50}$ values of 174.6 and 22.2 ${\mu}g$/ml, respectively. In addition, BFA also inhibited the degranulation in a dose dependent manner. Furthermore, oral administration of BFA inhibited IgE-mediated passive cutaneous anaphylaxis in mice. These results suggested that BFA may be useful in regulating mast cell-mediated allergic diseases.

Distribution Pattern and Feeding Preference of Asterias amurensis (Echinodermata: Asteriidae) in Tongyeong, Korea (통영해역에 서식하는 아므르불가사리의 분포 특성과 서식처에 따른 먹이 선호도 비교)

  • This study examined distributional patterns of Asterias amurensis in Tongyeong, the central South Sea of Korea. The density of the sea star was estimated at 10 chosen sites in the inner and the outer parts of the Tongyeong coast from December 2000. The mean density of the species in this area was $2.4ind./m^{2}$. The seasonal surveys conducted at 3 arbitrary chosen sites (i.e., sea cage, reef and soft sediment) also showed that the abundance of the species at the sea cage site $(density:\;3.6\;ind./m^{2};\;biomass:\;250.7\;gwwt/m^{2})$ was significantly higher than at the reef site $(density:\;1.7\;ind./m^{2};\;biomass:\;63.5\;gwwt/m^{2})$ and the soft sediment site $(density:\;0.4\;ind./m^{2};\;biomass:\;18.9\;gwwt/m^{2})$. Densities were higher at sea cages areas than at reefs and soft bottom sites. At sea cage site, A. amurensis population exhibited a strong aggregated distributional pattern. In contrast, at reef and soft bottom sites, A. amurensis population showed a random distributional pattern. The spatial difference in prey species and its abundance was the primary factor determining the spatial heterogeneity of the sea star in its behavior characteristics. Experiments on the feeding preference indicated that A. amurensis had a strong selectivity on its prey, but this selectivity varied between populations living in different sites. In particular, A. amurensis populations at the reef site showed a strong selectivity on various sessile and mobile animals living in reef areas, suggesting that these animal groups may play a role as "windows for the survival of A. amurensis". These results suggest that the distribution of A. amurensis in Tongyeong is closely associated with abundance of prey species and the bottom composition.

Predation of Asterias amurensis and Asterina pectinifera on Valuable Bivalves at Different Water Temperature (유용 양식패류에 대한 아무르불가사리(Asterias amurensis)와 벨불가사리 (Asterina pectinifera)의 수온별 포식 특성)

  • Effect of predation of starfish Asterias amurensis and Asterina pectinifera under different water temperature in the recirculating seawater vessel was conducted at the invertebrate culture laboratory of Yosu National University from February 1 to February 25, 2000. The highest activity of Asterias amurensis and Asterina pectinifera according to water temperature was obtained at 14$\^{C}$ and 22$\^{C}$. The average individual numbers of Anadara broughtonii predated by Asterias amurensis and Asterina pectinifera were 2.20 and 1.60 at 6$\^{C}$ , 3.60 and 2.00 at 10$\^{C}$, 7.20 and 2.50 at 14$\^{C}$, 3.40 and 6.80 at 22$\^{C}$, 0.80 and 3.90 at 26$\^{C}$, respectively.

Optimal Conditions for the Bioassay Using Sperms of a Starfish, Asterias amurensis (아무르불가사리(Asterias amurensis) 정자를 이용한 생물검정의 최적조건)

  • 성게의 정자를 이용한 생물검정은 해양환경의 독성평가에 널리 사용되고 있다. 그러나, 국내산 성게류의 경우 산란기가 3~11월로 겨울에는 생물검정 실칩이 불가능한 단점이 있다. 이를 보완하기 위하여 겨울에 산란이 가능한 아무르불가사리(Arterias amurensis)를 생물검정용으로 개발하였다. 불가사리류의 생식과 발생과정은 성게류와 유사하므로 성게류를 이용한 실험 방법을 아무르불가사리에 적용할 수 있다. 그러나, 생물검정 실험 결과의 유효성은 실험조건에 의하여 달라질 수 있으므로, 본 연구에서는 실험 결과에 큰 영향을 미칠 수 있는 조건들 즉. 수정막의 발달 시간, 정자와 알의 비율, 정자의 활력이 유지되는 시간, 정자의 노출 시간, 그리고 시료의 염분 범위 등을 최적화 하고자 하였다. 수정막의 발달이 완전하게 이루어지는 최소 시간은 60분이었고, 시료에 주입할 정자와 알의 비율은 3,000이상일 때 수정률이 높게 유지되었다(70%이상). 정자의 활력은 해수에 희석한 후 3시간 이상이 경과하면 급격히 감소하였다. 정자의 노출시간은 20~60분의 범위에서 구리에 대한 $EC_{50}$이 크게 달라지지 않았다(변동계수: 17%).실험에 적절한 시료의 염분 범위는 26~38 psu이었다. 아무르불가사리 정자의 민감도는, 일반적으로 해양환경 평가에 흔히 사용되는 생물들의 민감도 범위의 중간 정도에 해당하였다. 따라서 아무르불가사리의 정자는 해양환경 평가를 위한 생물검정용으로 적합한 것으로 여겨진다.

Starfish smooth muscle relaxing activity of SALMFamide isotype peptide and its analog derived from starfish, Asterias rubens

  • An organism's physiological processes and behaviors are regulated by neuropeptides and hormone peptides. The first neuropeptide identified from echinoderms is SALMFamide. The two most well-studied SALMFamide neuropeptides are S1 and S2, which possess myoactivity on apical muscle, tube feet, and the cardiac stomach of starfishes. However, neuropeptide candidates identified from SALMFamide's precursor protein sequence have not been investigated. This study aims to compare the bioactivity of SALMFamide neuropeptides from the starfish Asterias rubens using various starfish muscle preparations. In this study, the bioactivity of the L-type SALMFamide neuropeptides from the starfish A. rubens, AYHTGLPFamide (SALMFa-A) and the derivative AYHSALMFamide (SALMFa-B) was investigated. The neuropeptides were applied on Asterias amurensis apical muscle, tube feet, which revealed that the neuropeptides exhibit relaxing activity on apical muscle but no activity on tube feet. The native SALMFa-A peptide had lower relaxing activity on the apical muscle compared to the derivative peptide SALMFa-B. The relaxing activity of two neuropeptides also was compared with those on the apical muscle of Patiria pectinifera, which revealed relaxing activity as well as SALMFamide-S1 and S2 neuropeptides. Moreover, the investigation of SALMFa-A and SALMFa-B peptides' bioactivity on P. pectinifera cardiac stomach muscle also showed slight relaxing activity.

The Diversity of Heterotrophic Bacteria Isolated from Intestine of Starfish(Asterias amurensis) by Analysis of 16S rDNA Sequence (16S rDNA염기서열에 의한 불가사리(Asterias amurensis) 장내에서 분리된 종속영양세균 군집의 다양성)

  • To study the diversity of heterotrophic bacteria isolated from intestine of starfish, Asterias amurensis, we collected starfishes from the coastal area near Jangheung-Gun, Jeollanam-Do, Korea during July, 2000. Population density and bacterial diversity in the intestine of starfish were measured. The results were as follows; The population densities of heterotrophic bacteria in the intestine of starfish were 8.65${\pm}$0.65${\times}10^3\;dfu\;g^{-1}$. Gram positive bacteria occupied 59% among 29 isolates. The community structure of dominant heterotrophic bacteria in the intestine of starfish consisted of Bacillaceae in the low G+C gram positive bacteria subphylum, Microbacteriaceae in the high G+C gram positive bacteria subphylum, and Alteromonadaceae in ${\gamma}$-Proteobacteria subphylum. Among eight strains of Bacillus spp., three strains showed more than 97% identity, but five strains showed about 90% identity with type strain on the basis of partial 16S rDNA sequence.

  • Gonadal development and reproductive cycle of the North Pacific seastar, Asterias amurensis captured from the Gosung, Gyeongsangnamdo, between November 2003 and February 2005, was investigated monthly changes of gonadosomatic index(GSI), gonadal development and oocyte size-frequency distribution. Monthly changes of GSI values showed similar trends in female and male. GSI values were reached the maximum in March($3.88{\pm}3.04$ in female, $0.87{\pm}0.57$ in male), and then gradually decreased. Based on the monthly changes of GSI, histological observation of gonadal development, reproductive cycle was divided into following successive stage: growing stage(October to January), mature stage(February to March), spent stage(March to April), degeneration and resorption stage(April to May), recovery stage(July to September). based on these result, this species seemed to have a synchronous oocyte development and one spawning season a year.

Biological Activity of Extracts of the Starfish Asterias amurensis (아무르불가사리 추출물의 생리활성)

  • Nowadays, starfish is one of problems about interruption of marine ecosystem, so many researchers are focusing on application of the starfish (Asterias amurensis). In this study, we investigated the contractile and relaxant activity of the smooth muscles and the antimicrobial and antioxidant activity of six different tissues (muscle, gut, liver, tube feet, gonads, and body) of A. amurensis. Frozen samples were extracted with distilled water containing 1% acetic acid. Extracts from all the tissues, except the body tissue, showed potent antimicrobial activity against Escherichia coli D31. The dorsal retractor muscles (DRM) of muscle and gut extracts showed strong contraction responses. On the other hand, contractile activity on esophagus of squid Todarodes pacificus could be detected in all tissues tested. The contractile activity of the liver extract was higher than that of the other tissues. The body, tube feet, and liver extracts showed the contractile activity on the intestine of the panther puffer fish (Takifugu pardalis). Relaxation response on the DRM of starfish (A. pectinifera) was observed in all tissues tested. Increased antioxidant activity was observed in the gut, liver, and body extracts. The results suggest that the starfish (A. amurensis) is a potential source of novel bioactive compounds.

Immune Enhancement Effect of Asterias amurensis Fatty Acids through NF-κB and MAPK Pathways on RAW 264.7 Cells

  • Asterias amurensis is a marine organism that causes damage to the fishing industry worldwide; however, it has been considered a promising source of functional components. The present study aimed to investigate the immune-enhancing effects of fatty acids from three organs of A. amurensis on murine macrophages (RAW 264.7 cells). A. amurensis fatty acids boosted production of immune-associated factors such as nitric oxide (NO) and prostaglandin E2 in RAW 264.7 cells. A. amurensis fatty acids also enhanced the expression of critical immune-associated genes, including iNOS, $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6, as well as COX-2. Western blotting showed that A. amurensis fatty acids stimulated the $NF-{\kappa}B$ and MAPK pathways by phosphorylation of $NF-{\kappa}B$ p-65, p38, ERK1/2, and JNK. A. amurensis fatty acids from different tissues resulted in different levels of $NF-{\kappa}B$ and MAPK phosphorylation in RAW 264.7 cells. The results increase our understanding of how A. amurensis fatty acids boost immunity in a physiological system, as a potential functional material.
