Effect of skin and seed of Grape and on Dimethylnitrosamine-Induced Liver Damage in Rats

  • Polyphenolic compounds have been reported to exhibit a wide range of pharmacological properties. In this study. we investigated the hepatoprotective effect of skin and seed of grape which contain abundant polyphenol compounds on dimethylnitrosamine(DMN)-induced liver damage in rats. Ingestion of skin and seed of grape (10% diet, daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels.   • Proanthocyanidins, one of the major natural polyphenolic compounds of grape has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the hepatoprotective effect of proanthocyanidins on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of proanthocyanidins (20, 50mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels. Association between extract from fruit of Opuntia ficus-indica and streptozotocin-induced diabetic rats

  • Opuntia ficus-indica has traditionally been used in prevention and treatment of various diseases such as diabetes mellitus. The current study was performed to determine whether Opuntia ficus-indica is associated with diabetes. Diabetic rat models were induced with streptozotocin (STZ). This study divided rats into 1 day (short-term) and 4 consecutive weeks (long-terms) of daily administration. These groups were subdivided into four groups each other for assessment of blood glucose level as follows: Group 1, untreated rats given distilled water; Group 2, untreated rats given Opuntia ficus-indica; Group 3, STZ-induced diabetic rats given distilled water; Group 4, STZ-induced diabetic rats given Opuntia ficus-indica. Blood glucose level was measured for one day and four weeks. In addition, serum markers of alanine aminotransferase (ALT), aspartate transaminase (AST), cholesterol, and creatinine were determined, and total protein triglycerides were measured at four weeks. Blood glucose level was highest in both groups (Group 3 and Group 4) at 30 minutes and two weeks and gradually decreased in a time-dependent manner. The difference in blood glucose among the four groups was significant (p < 0.05). Additionally, the levels of ALT, AST and triglycerides were significantly decreased by Opuntia ficus-indica.

수점산(手拈散)이 심장(心臟)과 소화기(消化器)에 미치는 영향(影響) (Effects of Soo Jeom San on the Functions of Heart and Digestive Organs)

  • The Present experiment was designed to investigate the effects of Soo Jeom San on the function of heart and digestive organs. And thus it was analyzed the total acidity, recovery effect, and the other various enzyme activities such as ATPase, Creatine kinase, Aspartate transaminase, and Lactate dehydrogenase. The results were obtained as follows : 1. The Total acidity decreased after Soo JeomSan administration for 6 days, however the total acidity inoreased after the drug administration for 9 days, these phenomena demonstrate that Soo Jeom San acts as a dual factor. The mechanism of decreasing the total acidity was considered to the inhibition of ATPase activity used for HCI active transport from parietal cells. 2. Soo Jeom San recovered the islets of Langerhans which was disrupted by streptozotocin. The recovery mechanism was suggested that Soo Jeom San stimulates the ${\beta}-cell$ proliferation. 3. Soo Jeom San inhibited the enzyme activities such as Creatine kinase and Aspartate transaminase, however the drug activated Lactate dehydrogenase. According to the obtained results, Soo Jeom San may be used for curing gastric ulcer and myocardiac infarction.

도꼬마리 열매 복용으로 발생한 중독증상(中毒症狀)의 한방치료 1례 (A Case Study of Poisoning Symptoms Caused by Misusing Xanthii Fructus, Treated with Korean Medicine)

  • This is a clinical report of one patient with mental disorientation, abnormal behavior, spasm, and other symptoms caused by poisoning with Xanthii Fructus, who recovered completely with treatment. The patient was treated with Korean medical treatments, such as herbal medicine and acupuncture. After treatment, various symptoms caused by the Xanthii Fructus poisoning disappeared completely and laboratory findings, such as liver function tests (aspartate transaminase and alanine transaminase), were also close to the normal ranges. This case shows that Korean medicine therapy can be applicable in treating various symptoms caused by Xanthii Fructus poisoning. It is considered that further research is needed to confirm the safe use of Xanthii Fructus.

  • Morin, one of the major natural flavonoids has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the hepatoprotective effect of morin on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of morin (10, 20mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phospatase, and bilirubin levels. Morin also increased serum protein level and reduced the hepatic level of malondialdehyde in DMN-treated rats. (omitted)

  • Quercetin, one of the most abundant flavonoids in human diet has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the protective effect of quercetin on hepatic fibrosis induced by dimethylnitrosamine (DMN) in rats. Treatment with DMN caused a significant decrease in body and liver weight. Oral administration of quercetin (10 mg/kg daily for 4 weeks) remarkably prevented this DMN-induced loss in body and liver weight and inhibited the elevation of serum alanine transaminase. aspartate transaminase, and bilirubin levels. Identification, Expression and Preliminary Characterization of a Recombinant Bifunctional Enzyme of Photobacterium damselae subsp. piscicida with Glutamate Decarboxylase/Transaminase Activity

  • Glutamate decarboxylase catalyzes the conversion of glutamate to gamma-aminobutyric acid (GABA), contributing to pH homeostasis through proton consumption. The reaction is the first step toward the GABA shunt. To date, the enzymes involved in the glutamate metabolism of Photobacterium damselae subsp. piscicida have not been elucidated. In this study, an open reading frame of P. damselae subsp. piscicida, showing homology to the glutamate decarboxylase or putative pyridoxal-dependent aspartate 1-decarboxylase genes, was isolated and cloned into an expression vector to produce the recombinant enzyme. Preliminary gas chromatography-mass spectrometry characterization of the purified recombinant enzyme revealed that it catalyzed not only the decarboxylation of glutamate but also the transamination of GABA. This enzyme of P. damselae subsp. piscicida could be bifunctional, combining decarboxylase and transaminase activities in a single polypeptide chain.

흰쥐에 있어서 Symphytum officinale L. 추출액의 독성에 관한 연구 (Toxicological Study on the Water Extract of Symphytum officinale L. in Rats)

  • This study was performed to investigate the toxic effect of pyrrolizidine alkaloids from symphytum officinale i n rat. For this experiment, 120 male and female rats of Sprague-Dawley strain were used. The experimental groups were divided into five: Group CM and CF served as normal control with its gender. Group EM1 and EF1 were fed a 1% Symphytum officinal extract diet for 8 weeks. Group EM2 and EF2 fed a diet containing 2% extract diet. 4% extract diet into group EM3 and EF3 and 8% extract diet into group EM4 and EF4 were given. The results were as follows: 1. The major alkaloids of Symphytum officinale extract were symphytine, echmidine, and lasiocarpine. The amounts of total alkaloid were 168 $\mu\textrm{g}$ PAs/$m\ell$ extract. And contents of Pas in leaves were 0.05% wt.. 2. Total serum bilirubin concentrations increased significantly in group EM2, EM3, and EM4. Group EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 3. Aspartate transaminase activities were increased significantly in group EM3 and EM4 (p<0.05). Aspartate transaminase activities of EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 4. Alanine transaminase activities increased significantly in group EM3, EM4 (p<0.05). Alanine transaminase activities of EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 5. Alkaline phosphatase activities increased significantly in group EM2, EM3, and EM4 (p<0.05). Alkaline phosphatase activities of EF1, FE@, EF3, and EF4 showed statistical sigmificance for the group CF (p<0.05). 6. istopathological analysis of liver specimens from group EM3 and EM4 showed focal necrosis, periportal necrosis and apoptpsis. Hepatocytes obtained from group EM2 showed fatty change and hydropic degeneration in group EM3 and EM4. Chromatolysis and chromatin margination was shown in group EF2 and EF3. With the above results, it was demonstrated that the Symphytum officinal extract could induce functional change of liver, and histopathological change of liver in rats fed a diet containing extract. In conclusion, because of the risk of intoxication or adverse effect, the composition, dosage and mode of administration of herbal products should be monitored strictly. And this study serves as a reminder that herbal as well as orthodox medications may have serious side effects.

Dimethylnitrosamine에 의해 유도된 흰쥐 간독성에 대한 배풍등 추출물의 보호효과 (Effect of Solanum lyratum Extract on Dimethylniatrosamine-Induced Liver Damage in Rats)

  • Solanum lyratum(Solanaceae) has been used as a traditional analgesic, antipyretic and hepatoprotective agents in Korea. In this study, we investigated the hepatoprotective effect of ethylacetate extract of Solanum lyratum (SL) on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of SL (150, 300 mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase levels. SL also increased serum protein level and reduced the hepatic level of malondialdehyde in DMN-treated rats. Furthermore, DMN-induced elevation of hydroxyproline content was reduced by the treatment of SL. In conclusion, these results demonstrated that SL exhibited in vivo hepatoprotective effect against DMN-induced liver injury, and suggest that SL may be useful in the prevention of liver damage.
