Acute toxicity of pectenotoxin 2 (PTX2) was examined in mice. Treatment of mice with a toxic dose of PTX2 resulted in clinical signs such as ataxia, cyanosis and an abrupt decrease in body temperature. Histopathological studies revealed that the liver is the major target organ for PTX2. Activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and sorbitol dehydrogenase (SDH) were significantly elevated by PTX2 administration. Glucose-6-phosphatase activities were not changed by the treatment. The PTX2 treatment decreased relative liver weight without changing the body weight. The effect of PTX2 on hepatic drug metabolizing enzyme system was determined. An ip dose of PTX2 (200 $\mu$g/kg) induced a significant decrease in the hepatic microsomal protein content. Cytochrome P-450 content, cytochrome b$_5$ content, NADPH cytochrome c reductase, aminopyrine N-demethylase activities, or hepatic glutathione content were not altered by PTX2 treatment.
A series of new derivatives of 1-(5-oxohexyl)-3,7-dimethylxanthine has been prepared for the treatment of the vascular dementia. To investigate hepatoprotective effect of these derivatives, the serum biochemical activity and the histological change of liver tissue were evaluated in carbon tetrachloride ($CCl_4$) treated rats. The activities of aspartate aminotransferase and alanine aminotransferase and the activities of total cholesterol, triglyceride and total bilirubin in 5-oxohexyl-3,7-dimethylxanthine derivatives pretreated rats were significantly decreased compared to the $CCl_4$ only treated rats but the content of glutathione S-transferase was increased compared to the $CCl_4$ only treated rats. These results indicated that 5-oxohexyl-3,7-dimethylxanthine derivatives have hepatoprotective effect in $CCl_4$ intoxicated rats.
Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine(CRE), total cholesterol(TCHO), triglyceride(TG), low density lipoprotein cholesterol(LDL-CHO), high density lipoprotein cholesterol(HDL-CHO), total protein(TP), albumin(ALB) and total bilirubin(BIL) in serum. Hepatic parameters monitored were levels of cholesterol(CHO), triglyceride(TG), malondialdehyde(MDA), content of cytochrome P450(CYP), level of glutathione(GSH), and activities of NADPH-CYP reductase, superoxide dismutase(SOD), catalase(CAT), glutathione S-transferase(GST), glutathione reductase(GR), glutathione peroxidase(GPx). The histopathological examination showed that the treatment of Sachungwhan relieved the ballooning degeneration of hepatocytes which had been generated by $CCl_4$. The results suggested that hepatoprotective effects of Sachungwhan possibly are due to their promising antioxidative activity.

The purpose of this study was to investigate the pharmacological effects of low-esterified pectin on carbon tetrachloride $(CCL_4)-induced$ hepatotoxicity in rats. The study included two experiments. In the first experiment the animals were given daily $CCL_4$ through gavage for 7 days and then 10, 50, or 250 mg/kg b.w. of pectin for 21 days. At the end of experiment rats were killed within 24 hours. The increased bilirubin level, enhanced alanine aminotransferase and aspartate aminotransferase activity in plasma induced by $CCL_4$ were partly normalized by pectin administration in a dose-dependent manner. The pectin treatment also resulted in significant recovery of $CCL_4-induced$ decrease of the liver glycogen content. In addition, pectin significantly improved $CCL_4-induced$ alterations of pro-oxidant and antioxidant biochemical parameters in liver and plasma compared to those of rats administered $CCL_4$. In the second experiment the animals were given daily 10, 50 or 250 mg/ kg b.w. of pectin for 21 days before a 7-day administration of $CCL_4$. Rats were killed 24 hours after the end of experiment. Pretreatment with pectin before $CCL_4$ administration resulted in significantly inhibited increase of the blood enzymatic activities of alanine and aspartate aminotransferases and bilirubin level in a dose-dependent manner. Also, preliminary administration of pectin prevented elevation of malondialdehyde and conjugated diene levels in liver and plasma as well as a reduction of glutathione content in liver of rats given $CCL_4$. These results suggest that low-esterified pectin exert healing and preventive effects on $CCL_4-induced$ hepatotoxicity in rats.

This study was investigated the biochemical mechanism on reducing blood lipids in second-generation rats fed diet with different omega 6 to omega 3 fatty acid ratio. The experiment treatment groups were classified into the groups with the omega 6 to omega 3 ratios of 0 (control group), 1:1, 8:1, and 19:1, respectively. The levels of the blood triglyceride, total cholesterol, low density lipoprotein-cholesterol (LDL-C), aspartate aminotransferase (AST), alanine aminotransferase (ALT) and glucose were lowest in the group with the omega 6 to omega 3 ration of 1:1. The levels of high density lipoprotein-cholesterol and phospholipid were highest in the group with the omega 6 to omega 3 ration of 1:1. The HMG-CoA reductase activity was suppressed in the groups with the omega 6 to omega 3 ratio of 1:1 and 8:1 compared with that in the control group, but the excretion of sterol through feces was promoted. The blood omega 6 to omega 3 ratio decreased in a concentration-dependent manner depending on the increase in the omega 6 to omega 3 ratio within the ingested diet. The results of this study demonstrated a new finding that when the parent generation and second-generation rats ingested the diet with the omega 6 to omega 3 ratio of below 8:1, harmful lipids in the blood were reduced, the liver functions were maintained, and the growth was promoted due to the nutrient metabolism activation mechanism.
Objectives : Effects of Fructus Foeniculi extract on liver function were investigated in carbon tetrachloride(CCl4) intoxicated rats. Methods : Thirty two male Sprague-Dawley rats with mean weight of $227.28{\pm}7.92g$ were used in these experiments and housed with food and water ad libitum. Fructus Foeniculi extract was administerd at dose 100mg/kg/day and 200mg/kg/day p.o. for 2 weeks after that CCl4 was treated 3 times at dose of 2.5ml/kg, p.o. in alternate day basis. Then serum AFP(${\alpha}$-Fetoprotein), Total protein, Albumin, Triglyceride, Total cholesterol concentrations and ALP (Alkaline phosphatase), AST(Aspartate Aminotransferase), ALT(Alanine Aminotransferase), ${\gamma}$-GT( ${\gamma}$-Glutamyl transferase), LDH(Lactate Dehydrogenase) activities were determined with commercial kit by autoanalyzer. Results : Plasma ${\alpha}$-fetoprotein and total protein concentration showed a tendency to decrease in Fructus Foeniculi extract-treated groups. However, plasma albumin concentration showed no significant differences in all treatment groups. Activity of plasma aspartate aminotransferase and alanine aminotransferase in Fructus Foeniculi extract-treated groups showed a lower value than that of control group. Alkaline phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Fructus Foeniculi treated groups. However, ${\gamma}$-glutamyl transferase activity showed no significant difference in all treated groups. Concentration of plasma triglyceride and total cholesterol showed a high level in CCl4 intoxicated rats but not in Fructus Foeniculi treated groups. Conclusion : Reviewing these experimental results, it appears that Fructus Foeniculi extract have recovering effect against liver injury.
This study was conducted to investigate the effects of cadmium (Cd) exposure on biochemical factors in the hemolymph and hepatopancreas of the abalone, Haliotis discus hannai. The abalone were exposed to 0, 5, 10, 20 and 40 ${\mu}g/L$ Cd for 4 weeks. The phenoloxidase (PO) activity was decreased in hemolymph of abalone exposed to 40 Cd ${\mu}g/L$ for 4 weeks compared to the control (P < 0.05). The hemolymph enzymes, alkaline phosphatase (ALP), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were markedly elevated in 40 Cd ${\mu}g/L$ after 4 weeks. The hemolymph calcium concentrations were significantly decreased in 20 and 40 Cd ${\mu}g/L$ for 4 weeks. Hepatopancreas superoxide dismutase (SOD) and catalase (CAT) activities were significantly increased by Cd. SOD was increased in both 20 and 40 Cd ${\mu}g/L$ and CAT, in 40 Cd ${\mu}g/L$ after 2 weeks (P < 0.05). These results suggested that the abalone SOD and CAT including PO may serve as a protective mechanism against oxidative stress by Cd. We conclude that a Cd concentration, 40 ${\mu}g/L$ in water may curtail hemolymph homeostasis and anti-oxidative reactions in abalone hepatopancreas. From these results, these biochemical factors may represent a convenient method of monitoring heavy metal pollution in coastal areas.
This study was proposed to investigate the effects of water extract of aged black garlic on ethanol induced hangovers in rats. Male Sprague-Dawley rats weighing $180{\pm}10\;g$ were divided into the following three groups; control, 130 mg/kg, and 260 mg/kg of aged black garlic extract. Aged black garlic was administered orally 30 min before and 30 min after ingestion of 40% ethanol (5 g/kg, B.W.). The rats were killed 24 hr after ethanol treatment, and blood was taken from the caudal artery at 1, 3, and 5 hr to test for ethanol or acetaldehyde in the serum. Groups that were administered aged black garlic extract pre- and post-alcohol consumption showed a significant decrease in ethanol levels in the blood at 1, 3, and 5 hr. The acetaldehyde concentrations decreased in both 130 mg/kg and 260 mg/kg groups that were administered aged black garlic extract pre- and post-alcohol consumption. The activities of alcohol dehydrogenase seemed to be unaffected, although the aged black garlic showed slightly higher activities of acetaldehyde dehydrogenase in pre- or post-alcohol consumption. The aspartate aminotransferase (AST) activity in the serum, elevated by ethanol, was decreased by administering a high dosage of aged black garlic extract, but resulted in no significant change in serum alanine aminotransferase (ALT) activity. These results concluded that aged black garlic extract can reduce hangover syndrome through the elevation of ALDH.
This study was investigated the effects of the lower power He-Ne IR laser treatment on the changes of blood biochemical components in the rat liver damaged by the carbon tetrachloride (CCl₄). The twenty one Sprague-Dawley adult male rats weights (260±18.6 g) were designed to the three groups: one control group and two experimental groups, the experimental groups were divided into the CCl₄-treated groups and the laser therapy group (CCl₄+ Laser). The experimental groups were injected twice with CCl₄(1.0 ml/kg body weight) intraperitoneal for two days. Each group was sacrificed after two weeks irradiated with the lower power He-Ne IR laser for ten minutes per every day. The activity of alanine aminotransferase (ALT), lactic dehydrogenase (LDH), alkaline phosphatase (ALP) and the concentration of serum glucose treated with He-Ne IR laser groups was significantly decreased to the conrtol (treated with carbon CCl₄) group. The activity of aspartate aminotransferase (AST) was decreased in the laser group but not significantly, the concentration of the serum cholesterol in the laser group was significantly increased comparing with the control and case control groups. In conclusion, the effect of the lower power He-Ne IR laser treatment is believed to be a possible protective effects for CCl₄ induced acute hepatotoxicity in rats.
Rubus coreanus, commonly known as 'red raspberry' is used as a traditional oriental medicine in Korea for the management of diseases such as impotence, spermatorrhea and athsma, and for allergies, in combination with other herbal preparations, in many centuries. We undertook a comparison of the hepatoprotective effect of ethanol extracts of the unripe (UREx) and ripe (RREx) R. coreanus extract against acetaminophen (AAP) induced hepatotoxicity in rats. UREx reduced the elevated alanine aminotransferase (ALP), aspartate aminotransferase (AST), total bilirubin (TB), alkaline phosphatase (AP), lipid peroxide and nitric oxide content which had been increased by AAP administration. UREx also increased the cellular glutathione (GSH) content and induced the glutathione-S-transferase (GST), glutathione peroxidase (GSH-Px) content which had been decreased by AAP. RREx did not exhibit strong hepatoprotective effect or antioxidant activity under the same conditions. The experimental results show that the degree of the ripening of R. coreanus affects the hepatoprotective activity in the AAP-intoxicated rats. These findings of a protective mechanism are supportive evidence for the utility of unripened R. coreanus in traditional medicine for liver ailments.
