Advanced Weed Control and Soil Environmental Survey in Lawn (잔디밭의 효과적인 잡초 방제와 토양 환경 조사)

  • The effects of salt are studied on control of weeds and growth of Korean turfgrass(Zoysia japon-ica Steud.). And the mobility of salt were determined in a lawn field. At post-emergence stage, the growth of the 13 weeds such as Trifolium repens, Erigeron canadensis, Artemisia prieceps, Equi-setum arvense, etc were controlled by treatment of salt and salt water, Pea-annua and Equisetum arvense were tolerant to salt compared with the other 11 weeds. At pre-emergence stage, the salt controlled the germination of the weeds in soil. 300~500 kg /10a salt and 20~30 % salt waterwere enough for the control of weeds, However, salt water injuried the lawngrass except the dormancy stage. The mobility of salt in soil was increased by rainfall. After 160mm of rainfall, the salinity in the soil treated with salt 500kg /l0a was below 0.3 ms/cm in surface soil and about 0.1 ms /cm in soil 30 cm depth. Repellent Activity of Five Different Plant Extracts Against Aedes albopictus (흰줄숲모기에 대한 5종 식물추출물의 기피효능 평가)

  • Five different methanol extracts of plants were evaluated as a mosquito repellent against Aedes albopictus in nude mice model. The volatile components of plants were analyzed with GC-MS. The effective dose was the highest in Nepeta cataria with 72.9, 83.7, 86.4, and 97.3% efficiencies at 10, 50, 100, and 200 ㎍/mL, respectively. Nepeta cataria, Actinidia polygama, and Artemisia annua extracts were the most effective in duration time test. And there was no difference compared to negative control in the low mosquito repellent efficiency from 30 minutes. As a result of analyzing the volatile components of each plant, a total of 28 components in Mentha suaveolens, 19 components in Actinidia polygama, 27 components in Artemisia annua, 26 components in Nepeta cataria, and 19 components in Taraxacum platycarpum were detected. Especially, nepetalactone known as an effective ingredient of a mosquito repellent was identified (27.95 mg/Kg) in one of volatile components of Nepeta cataria. Overall, our results provide the possibility to develop mosquito repellents material using natural plants which contain various volatile components.

Cell Cycle Arrest of Extract from Artemisia annua Linné. Via Akt-mTOR Signaling Pathway in HCT116 Colon Cancer Cells (HCT116 대장암세포에서 Akt-mTOR 신호경로를 통한 개똥쑥 추출물 (AAE)의 세포주기 억제 효과)

  • In this study, extract from Artemisia annua in L. (AAE) is known as a medicinal herb that is effective against cancer. The cell cycle is regulated by the activation of cyclin-dependent kinase (CDK)/cyclin complex. We will focus on regulation of CDK2 by cyclin E. cyclin E is associated with CDK2 to regulate progression from G1 into S phase. Akt is known to play an important role in cell proliferation and cell survival. Activation of Akt increases mTOR activity that promotes cell proliferation and cancer growth. In this study, we investigated that AAE-induced cell cycle arrest at G1/S phase in HCT116 colon cancer. Treatment of AAE shows that reduced activation of Akt decreases mTOR/Mdm2 activity and then leads to increase the activation of p53. The active p53 promotes activation of p21. p21 induces inactivation of CDK2/cyclin E complex and occurs cell cycle arrest at G1/S phase. We treated LY294002 (Akt inhibitor) and Rapamycin (mTOR inhibitor) to know the relationship between the signal transduction of proteins associated with cell cycle arrest. These results suggest that AAE induces cell cycle arrest at G1/S phase by Akt/mTOR pathway in HCT116 colon cancer cell.

Screening for Herbicidal Medicinal Plants against Digitaria Sanguinalis and Taraxacum Platycarpum in Turf (잔디밭 잡초 바랭이와 민들레 방제를 위한 살초활성 약용식물의 탐색)

  • Crabgrass (Digitaria sanguinalis L.), Dandelion (Taraxacum platycarpum H. DAHLST.), Annual bluegrass (Poa annua L.), Gree kyllinga (Kyllinga brevifolia var.), Red clover (Trifolium pratense L.), Annual lespedeza (Kummerowia striata (Thunb.)Schindl.), Mugwort (Artemisia capillaris Thunb.), Horseweed (Erigeron canadensis L.), Field horsetail (Equisetum arvense L.) are major weeds in turf. In this study, 23 medicinal plant extracts were tested for herbicidal activities against crabgrass and dandelion in the course of major weed in turf. In pot experiment, we implemented pre-emergence application used medicinal plant extracts showed herbicidal activity in petridish experiment. In effect, inhibition rates of germination were 61.0% at Curcuma longa extract and 62.5% at Cnidium officinale extract in crabgrass pot experiment, and 77.8% at Ailanthus altissima extract in dandelion pot experiment.

Antibacterial effect of medicinal plants against Helicobacter pylori (헬리코박터 파이로리균에 대한 약용식물의 항균 효과)

  • The study was designed to investigate the antibacterial effect of Helicobacter pylori against 32 medicinal plants commonly used as health foods. The medicinal plants used in this study were 32 kinds of medicinal plant extracts using the disk diffusion method for H. pylori activity, which can be eaten every day by everyone. Amoxicillin sodium (150 mg / ml, Ildong Pharmaceutical) and Metronidazole 50 mg / ml) was used as a control group. We measured the area of the transparent area and evaluated that the larger the area, the more effective it is for H. pylori. As a result of this study, the clear zone of inhibition was highest at $372.90mm^2$, second was $358.30mm^2$, and Chungho was $348.32mm^2$. The positive control group, Metronidazole (50 mg / ml CJ), was $503,29mm^2$. In the future, the development of antimicrobial materials of various medicinal plants is expected to be effective for the inhibition of H. pylori.

Environmental Fate and Effect of ZnO Nanoparticles (산화아연 나노입자의 환경 거동 및 영향 연구)

  • ZnO nanoparticles (ZnO NPs) are mainly used in semiconductors, solar cells, biosensors, and cosmetics (sunscreen). In this study, we investigated the behavior of ZnO NPs in aquatic and soil environments and their effects on plants (Artemisia annua L.) in hydroponic cultivation. It was confirmed that the ZnO NPs size increased and their dissolution decreased with increasing in pH. Leaching distance of ZnO NPs was less than 2.5 cm, indicating that ZnO NPs had a little potential to leach into deeper soil layers. When ZnO NPs were exposed to plant, the total weights of plants decreased. The effects on the length of root and shoot were not observed. In addition large amount of ZnO NPs were adsorbed on the surface of plant root and didn't translocate into shoot. These results suggest that ZnO NPs block the pores of the root cell wall and decrease the bioavailability of plant nutrients. Therefore it can be speculated that the particles increase in size and settle down in the water environment and may adversely affect the plant growth by firmly adhering to the root surface when the ZnO NPs are exposed to the environment.

The Extract from Artemisia annua Linné. Induces p53-independent Apoptosis through Mitochondrial Signaling Pathway in A549 Lung Cancer Cells (A549 폐암세포에서 미토콘드리아 경로를 통한 개똥쑥 추출물의 apoptosis 유도 효과)

  • The extract from Artemisia annuain L.(AAE) is known as a medicinal herb that is effective against cancer. Apoptosis is the process of programmed cell death, and mitochondria are known to play a central role in cell death control. In this study, we evaluated the p53-independent apoptosis of extract of AAE through downregulation of Bcl-2 and the mitochondrial pathway in A549 (lung cancer cells). AAE may exert cancer cell apoptosis through regulating p-Akt, Cox-2, p53 and mitochondria-mediated apoptotic proteins. p-Akt/cox-2 is known to play an important role in cell proliferation and cell survival. The Bcl-2 pro-apoptotic proteins (such as Bax, Bak and Bim) mediate the permeabilization of the mitochondrial outer membrane. Treatment of AAE reduces p-Akt, p-Mdm2, cox-2 and anti-apoptotic proteins (such as Bcl-2), while tumor suppressor p53 and pro-apoptotic proteins. Activation of Bax/Bak releases cytochrome c from mitochondria to the cytosol to activate a caspase. Caspase-3 is the major effector caspase associated with apoptotic pathways. Caspase-3 generally exists in cytoplasm in the form of a pro-enzyme. In the initiation stage of apoptosis, caspase-3 is activated by proteolytic cleavage and activated caspase-3 cleaves poly (ADP-ribose) polymerase (PARP). We treated Pifithrin-α (p53 inhibitor) and Celecoxib (Cox-2 inhibitor) to learn the relationship between the signal transduction of proteins associated with apoptosis. These results suggest that AAE induces apoptosis through a p53-independent pathway in A549.

Effect of Black Garlic and Gaeddongssuk (Artemisia annua L.) Extracts on the Lipid Profile and Hepatic Antioxidant Enzyme Activities of Exercised Rats (흑마늘 및 개똥쑥 추출물의 급이가 강제운동 시 흰쥐의 체내 지질 성분 및 간조직의 항산화효소 활성에 미치는 영향)

  • To develop functional products based on black garlic, a black garlic extract (BG) of 7 brix, a gaeddongssuk extract (GS) of 0.7 brix and two types of mixtures (MBS-I, black garlic 7 brix : gaeddongssuk 0.7 brix; MBS-II, black garlic 14 brix : gaeddongssuk 1.4 brix, 93:7, v/v) were supplemented to rats training on a treadmill for 4 weeks. Body weight from the training did not decrease during the experimental period. Serum albumin content significantly increased in the groups fed an experimental diet compared to the control. The BUN content significantly decreased in BG and MBS-II groups compared to the control. AST and ALP activities significantly decreased in the groups fed an experimental diet compared to the control. Serum triglyceride and total cholesterol levels in MBS-I and MBS-II groups significantly decreased compared to the control. Lipid levels in the serum and liver tissue were not significantly different between the MBS-I and MBS-II groups. The lipid peroxide content in the serum and liver tissue was significantly reduced in the groups fed all extracts compared to the control; the serum and liver lipid contents was lowest in the MBS-I and MBS-II groups, respectively. Hepatic catalase activity in the GS and MBS groups increased by 1.8~2.3 times compared to the control. SOD and GSH-px activities significantly increased from treatment with the extracts by 1.3~1.5 times and 1.2~1.7 times, respectively. These results indicate that a mixture of BG and GS extracts has higher biological activity than a single supplementation of BG or GS extract. Therefore, the addition of gaeddongssuk to black garlic (MBS-I and MBS-II) is effective as a defense material against oxidative stress. MBS-I may be especially effective for its biological activities.
