Antiradical activity of Azadirachta indica extracts and fractions

  • In present study, total phenolic content of crude extract and antiradical activity of crude extract and different solvent fractions of Azadirachta indica were evaluated. Crude extract and most of the polar fractions showed higher radical scavenging activity. Among the crude extract and nine different fractions, the aqueous/methanol (3:1) fraction showed the highest activity.

Anti-Inflammatory Effect of Aqueous Extract of Scolopendrae Corpus in RAW 264.7 Cells (마우스 대식세포주인 RAW 264.7 세포에서 오공(蜈蚣)의 항염증 효과)

  • Objective : The purpose of this study was to investigate the anti-inflammatory effects of aqueous extract from Scolopendrae Corpus (SC) on lipopolysaccharide (LPS)-induced inflammatory response. Methods : To evaluate the anti-inflammatory effects of SC, we examined the inflammatory mediators such as nitric oxide (NO) and pro-inflammatory cytokines (TNF-a, inteleukin (IL)-$1{\beta}$ and IL-6) on RAW 264.7 cells. We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and inhibitory kappa B a ($I{\kappa}$-Ba) using western blot. Furthermore, we also investigated the effect of SC on LPS-induced endotoxin shock. Results : Extract from SC itself had not any cytotoxic effect in RAW 264.7 cells. Aqueous extract from SC inhibited LPS-induced NO production and iNOS expression. SC pre-treatment also inhibited IL-$1{\beta}$, IL-6 production in RAW 264.7 cells. To investigate inhibitory effects of SC on inflammatory mediators, activation of MAPKs was examined. SC inhibited the phosphorylation of p38 kinases (p38), c-Jun $NH_2$-terminal kinase (JNK) and also the degradation of $I{\kappa}$-$B{\alpha}$ in RAW 264.7 cells stimulated with LPS. Furthermore, SC administration reduced LPS-induced endotoxin shock. Conclusion : SC down-regulated LPS-induced production of inflammatory mediators through inhibition of activation of p38, JNK and degradation of $I{\kappa}$-$B{\alpha}$. Taken together, our results suggest that SC may be a beneficial drug against inflammatory diseases such as sepsis.

The Prophylactic and Therapeutic Effects of Saffron Extract and Crocin on Ethanol Withdrawal Syndrome in Mice

  • Objectives: Ethanol withdrawal following its chronic use is a serious outcome and challenging to treatment. The chronic use of ethanol induces a progressive neuroplasticity in different reigns of brain. In this study we evaluated the effects of aqueous extract of Crocus sativus L. (saffron) and its active compound, crocin, on the withdrawal behavior induced after repeated administration of ethanol, in two regimens of prophylactic (administration of drugs concomitant with the induction of dependence) and treatment (administration of drugs during the period of ethanol withdrawal) in mice which received ethanol. Methods: Ethanol dependence was induced by oral administration of 10% v/v ethanol (2 g/kg) for 7 days. The aqueous extracts of saffron (40, 80 and 160) and crocin (10, 20 and 40 mg/kg) were administered to mice in two regimens of prophylactic (along with ethanol) and treatment (during withdrawal period). Diazepam (1 mg/kg) was used as a positive control. Six hours after discontinuation of the ethanol, seizure was evaluated by the sub-convulsive dose of pentyleneltetrazole (PTZ) (30 mg/kg). The open field test and Rota rod test were used for evaluation of locomotor activity and motor incoordination, respectively. Results: Both extracts and crocin increased the number of crossed lined in the open field test. PTZ kindling seizure was inhibited in animals received extract (80 and 160 mg/kg) in both regimens. Motor incoordination was only improved following administration of crocin. Conclusion: The aqueous extract of saffron and crocin can be considered as safe agents and reliable alternative to diazepam in management of ethanol withdrawal syndrome.

Antioxidant and Anti-inflammatory Activity of Grateloupia filicina extract (지누아리(Grateloupia filicina) 추출물의 항산화 및 항염능)

  • In this study, the antioxidant ability, anti-inflammatory ability and whitening effect of the extracted Grateloupia filicina under optimal conditions were confirmed. The useful substances of Grateloupia filicina were extracted with various ethanol aqueous solutions to measure DPPH, ABTS, and nitrite radical elimination capabilities, and the polyphenol and flavonoid contents were analyzed. When extracted with a 20% ethanol aqueous solution, the radical elimination ability was the best, and the content of polyphenol and flavonoids was also the highest. From this, it was confirmed that the 20% ethanol aqueous solution efficiently extracted the antioxidant and anti-inflammatory substances of Grateloupia filicina. Meanwhile, the cytotoxicity and anti-inflammatory properties of the Grateloupia filicina extract were confirmed using RAW 264.7 cell lines. Grateloupia filicina extract did not show toxicity at a concentration of 200 ㎍/mL, and showed NO production inhibitory capacity of 50% or more at this concentration. As a result, it was confirmed that the Grateloupia filicina extract can be used as a functional cosmetic raw material having antioxidant and anti-inflammatory properties.

Effect of the Gardenia jsaminodes and Glycine max on Hepatotoxicity of D-Galactosamine in Rats (D-Galactosamine으로 유발된 흰쥐의 간손상에 대한 치자와 두시 추출액이 미치는 영향)

  • Aim : To investigate the hepatotective effect of Gardenia jsaminodes and Glycine max aqueous extract against D-galactosamine (d-GalN, 300mg/kg body weight) was administered to the male Sprague Dawley (SD) rats. Materials and Methods : The study was carried out on male SD rats (age matched, weight $250{\pm}10g$). Experimental groups divided four: Normal group (Nor) was administered saline, Control (Con) group was administered saline after d-GalN treatment. Experimental group (Exp) was administered Gardenia jsaminodes (200 mg/kg; Ga group), Glycine max (700 mg/kg; Gl group), and Gardenia jsaminodes+Glycine max (200 mg/kg+700 mg/kg, GG group) during 14 days(n=5). Results : d-GalN administration induced hepatotoxicity in rats which was manifested by increased levels of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase but decreased total cholesterol and triglyceride. Treatment with Gardenia jsaminodes extract significantly protected the liver in d-GalN administered rats. Conclusion : Gardenia jsaminodes aqueous extract and Gardenia jsaminodes+Glycine max extract possesses hepatoprotective potential, thus validating its use in alleviating toxic effects of d-GalN.

Allelopathic effects of red pepper (Capsicum annuum L.) and coriander (Coriandrum sativum L.) on early seedling growth of wheat (Triticum aestivum L.)

  • A pot experiment was conducted to assess the effects of red pepper (Capsicum annuum) and coriander (Coriandrum sativum) on seedling growth of wheat (Triticum aestivum). The aqueous extracts treatment of red pepper and coriander showed a significant (p < 0.05) reduction in root, shoot and seedling length, number of leaves and seedling dry weight of wheat (T. aestivum) as compared to control. The inhibitory different effect on growth of wheat (T. aestivum) was directly proportional to the increasing concentration (1, 2, 3, 4 and 5%) of aqueous extracts of red pepper and coriander as compared to control treatment (0%). The root, shoot, seedling length and number of leaves of T. aestivum significantly p < 0.05 decreased at 5% concentration of red pepper as compared to control. The root, shoot and seedling growth of T. aestivum was also significantly reduced at 1, 2, 3, 4 and 5% concentration of coriander as compared to control. The root, shoot and leaves dry weight of T. aestivum at 5% coriander extract treatment concentration decreased as compared to control. The tolerance in seedlings of T. aestivum to red pepper and coriander extract treatment was dose dependent as compared to control. The seedlings of T. aestivum showed low percentage of tolerance to pepper extract treatment than coriander extract treatment.

Inhibitory effect of medicinal plant extracts on xanthine oxidase activity (약용식물 추출물의 xanthine oxidase 활성 저해 효과)

  • This study was performed to certify the inhibitory effect of aqueous extracts from sixty-seven medicinal plants on the activity of xanthine oxidase in vitro. Among the sixty-seven medicinal plants, Scutellaria baicalensis Georgi, Citrus aurantium L., Polygonum multiflorum Thunb., Pueraria thunbergiana (Sieb. et Zucc.) Benth., Citrus unshiu Marcor., Rubus coreanus Miquel, Camellia sinensis L., and Poncirus trifoliata Raf. were regarded as effective anti-gout sources. The active substances of P. multiflorum root extract were very stable at pH 2.0 and high temperatures. Xanthine oxidase activity was proportionally inhibited when concentrations of P. multiflorum extract increased. The aqueous extract from P. multiflorum root at a concentration of 2.0 mg/0.1 mL inhibited xanthine oxidase by 73.8%.

Anti-tumor activities of Naesosungma-tang (내소승마탕(內消升麻湯)의 항암효과(抗癌效果) 연구(硏究))

  • This study was carried out to evaluate antioxidative, cytotoxic and chemosensitizing effects for anti-tumor activities of Naesosungma-tang (NST). The results were as follows ; 1. The oxidative effects were measured by DPPH radical scavenging activity. NST water extract was showed more effective than ethanol extract and also various solvent fractions from NST showed effective in the following order : butanol fraction > ethyl acetate fraction > hexane fraction > aqueous fraction. 2. NST water extract was showed cytotoxic effects on the human cancer cells Calu-6 cell and SNU-601 cell, but not on MCF-7 cell. 3. The viability of cells was measured in 4 kinds of solvent fractions from water extract of NST by MTT assay. The results were as followings : 1) On the NIH3T3 cell, $IC_{50}$ value was $200{\sim}300\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 2) On the Caco-2 cell, effective only in the butanol fraction. 3) On the Calu-6 cell, most effective in hexane fraction($IC_{50}:80\;{\mu}g/m{\ell}$) and effective in ethyl acetate and butanol fraction($IC_{50}:100{\sim}120\;{\mu}g/m{\ell}$). 4) On the MCF-7 cell, $IC_{50}$ value was similar in three fractions except aqueous fraction. 5) On the SNU-601 cell, $IC_{50}$ value was within $100\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 6) On the HCT-1l6 cell, $IC_{50}$ value was $150{\sim}200\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 7) On the AML-2/WT cell, not effective in four kinds of solvent fractions. 4. In combined effects of NST and vincristine on AML-2/D100 cell, water and ethanol extracts of NST and hexane fraction of water extracts of NST more effectively inhibited proliferation of AML-2/D100 cell. These results Suggest that NST has antioxidative and cytotoxic effects against Caco-2, Calu-6, MCF-7, SNU-601, HCT116 and NIH3T3 cell especially Calu-6 and SNU-601 cell, and also chemosensitizing effects against AML-2/D100 cell in combined vincristine.

In vitro antimicrobial activity of aqueous and ethanol extracts of Euphorbia hirta

  • Euphorbia hirta, locally called 'ara tanah' or 'susun nabi' in Malaysia is a small annual herb common to the tropical countries and belongs to the same family as the tic and tapioca. E. hirta has had a long history of usage in the treatment of various ailments. In this current study, in vitro sensitivity test of crude aqueous and ethanol extracts of leaves and barks of E. hirta was carried out against bacteria (Escherichia coli, Salmonella enteritidis, Staphylocccus aureus and Bacillus subtilis) and fungi (Microsporum canis, Aspergillus fumigatus, Candida albicans and Candida tropicalis) using the discs diffusion method. The extract-impregnated discs (20, 40 and $80\;{\mu}g/{\mu}l$), the E. hirta extracts inhibited the growth of all the bacteria tested. The growth of C. albicans was inhibited in a concentration dependent manner by the aqueous leaves and barks extracts. C. tropicalis was found to be sensitive to the aqueous leaves extracts. The results were compared to antibacterial drugs of chloramphenicol, ampicilin, penicillin G, and enrofloxacine; and to antifungal drug of ketoconazole, itraconazole and miconazole. In this current study, it can be concluded that this plant has antimicrobial activity that is as potent as the standard antimicrobial drugs against certain microorganisms.

Antiinflammatory Principle of Bupleurum longiradiatum Roots

  • A methanol extract of the roots of bupleurum longiradiatum (Umbelliferar) showed antiinflammatory activity in rats. Hexane, chloroform, butanol, and aqueous fractions from the methanol extract were tested with the result that only the hexane fraction exhibited positive activity. Fractionation of the hexane extract resulted in isolation of arborinone as an active principle.
