Selection of Optimum Expression System for Production of Kringle Fragment of Human Apolipoprotein(a) in Saccharomyces cerevisiae

  • Recombinant Saccharomyces cerevisiae expression systems were developed to pro­duce a novel human anti-angiogenic protein called LK8, an 86 amino-acid kringle fragment pro­tein with three disulfide linkages. Galactose-inducible LK8 expression plasmid was constructed, and LK8 production levels by four S. cerevisiae strains were compared in order to select an op­timal host strain. S. cerevisiae 2805 was the most efficient among the strains tested. Elevating the LK8 gene copy number through multiple integration using 8-sequences as target sites re­sulted in more than a two-fold increase in the LK8 production level compared with the plasmid­based expression system. The maximum LK8 protein concentration of 25 mg/L was obtained from batch cultivation of the yeast transformant that harbors 16 copies of the LK8 gene. In con­clusion, the strain integrated with the multiple LK8 gene secreted the protein with relatively high yield, although, the increased LK8 gene dosage over 11 copies did not lead to further en­hancement in batch cultivations.

  • Very low density lipoprotein (VLDL) of commercial broiler (CB) and crossbred village chicken (AK) was purified using Fast Protein Liquid Chromatography (FPLC). The fraction collected was then confirmed as VLDL using 4% polyacrylamide gel electrophoresis and transmission electron microscopy (TEM). The particle size of VLDL is 46.8${\pm}$8.6 nm. The VLDL fraction was then subfractionated and the apolipoprotein (apo) profile was studied by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDSPAGE). The CB and AK have almost similar types of apo in both subfractions 1 and 2. The AK showed the presence of apoAI, AIV, D and E whereas the CB had apo AIV, D, E and H. The apo AIV and apo E were present in both subfractions of AK and CB.

Sulfasalazine attenuates tamoxifen-induced toxicity in human retinal pigment epithelial cells

  • Tamoxifen, a nonsteroidal estrogen receptor (ER) antagonist, is used routinely as a chemotherapeutic agent for ER-positive breast cancer. However, it is also causes side effects, including retinotoxicity. The retinal pigment epithelium (RPE) has been recognized as the primary target of tamoxifen-induced retinotoxicity. The RPE plays an essential physiological role in the normal functioning of the retina. Nonetheless, potential therapeutic agents to prevent tamoxifen-induced retinotoxicity in breast cancer patients have not been investigated. Here, we evaluated the action mechanisms of sulfasalazine against tamoxifen-induced RPE cell death. Tamoxifen induced reactive oxygen species (ROS)-mediated autophagic cell death and caspase-1-mediated pyroptosis in RPE cells. However, sulfasalazine reduced tamoxifen-induced total ROS and ROS-mediated autophagic RPE cell death. Also, mRNA levels of tamoxifen-induced pyroptosis-related genes, IL-1β, NLRP3, and procaspase-1, also decreased in the presence of sulfasalazine in RPE cells. Additionally, the mRNA levels of tamoxifen-induced AMD-related genes, such as complement factor I (CFI), complement factor H (CFH), apolipoprotein E (APOE), apolipoprotein J (APOJ), toll-like receptor 2 (TLR2) and toll-like receptor 4 (TLR4), were downregulated in RPE cells. Together, these data provide novel insight into the therapeutic effects of sulfasalazine against tamoxifen-induced RPE cell death.

Relationships of Potential Atherogenic Indices to Anthropometric Measurements, Dietary Intake and Dietary Behavior in Korean Obese Children

  • Epidemiological studies have indicated that a relationship between overweight and cardiovascular disease exists. To assess the relationship of unfavorable changes in serum lipid concentrations, apolipoprotein B, lipoprotein(a), blood sugar and blood pressure to anthropometric variables, nutrient intakes and dietary behavior,64 obese children were chosen as subjects and 24-hour dietary intake, dietary behavior as well as blood pressure, apolipoprotein B, lipoprotein(a), three kinds of cholesterol concentrations were measured in November 1976. More than half(57.8%) of the children had serum cholesterol concentrations over 175mg/d1, and 26.6% of them had over 200mg/d1 and this group could be classified as a high risk group. Abdominal obesity as measured with waist circumference and waist hip ratio was negatively co..elated to high density lipop개tein(HDL) cholesterol. Dietary behavior rather than nutrient intakes appeared to have associations with unfavorable lipid profiles. Total dietary behavior scores and fish and bean product consumption had strong correlations with potentially atherogenic lipoprotein concentrations. This study suggests that fir the early prevention of cardiovascular disease, waist hip ratio together with obese rate should be included in selective programs to identify risk group of children in Korea. furthermore, interventions in cardiovascular disease risk groups in obese children should emphasize maintenance of ideal body weight through reducing body fat and adopting desirable dietary behaviors such as increasing intake of fish or bean products. Triglyceride-Lowering Effects of Two Probiotics, Lactobacillus plantarum KY1032 and Lactobacillus curvatus HY7601, in a Rat Model of High-Fat Diet-Induced Hypertriglyceridemia

  • The triglyceride-lowering effect of probiotics Lactobacillus plantarum KY1032 and Lactobacillus curvatus HY7601 were investigated. Male SD Wistar rats were randomly divided into three groups and fed high-fat diet (HFD), HFD and probiotics (5 × 109 CFU/day of L. plantarum KY1032 and 5 × 109 CFU/day of L. curvatus HY7601), or normal diet for 6 weeks. Probiotic treatment significantly lowered the elevated plasma triglyceride and increased plasma free fatty acid, glycerol, and plasma apolipoprotein A-V (ApoA-V) levels. The probiotic-treated group showed elevated hepatic mRNA expression of PPARα, bile acid receptor (FXR), and ApoA-V. These results demonstrate that L. plantarum KY1032 and L. curvatus HY7601 lower triglycerides in hypertriglyceridemic rats by upregulating ApoA-V, PPARα, and FXR.

Comparative Analysis of Serum Proteomes of Moyamoya Disease and Normal Controls

  • Objective : The etiology and pathogenesis of moyamoya disease remain unclear. Furthermore, the definitive diagnostic protein-biomarkers for moyamoya disease are still unknown. The present study analyzed serum proteomes from normal controls and moyamoya patients to identify novel serological biomarkers for diagnosing moyamoya disease. Methods : We compared the two-dimensional electrophoresis patterns of sera from moyamoya disease patients and normal controls and identified the differentially-expressed spots by matrix-assisted laser desorption/ionization-time-of flight mass spectrometry and electrospray ionization quadruple time-of-flight mass spectrometry. Results : We found and analyzed 22 differently-expressed proteomes. Two proteins were up-regulated. Twenty proteins were down-regulated. Complement C1 inhibitor protein and apolipoprotein C-III showed predominantly changed expressions (complement C1 inhibitor protein averaged a 7.23-fold expression in moyamoya patients as compared to controls, while apolipoprotein C-III averaged a 0.066-fold expression). Conclusion : Although our study had a small sample size, our proteomic data provide serologic clue proteins for understanding moyamoya disease.

Curcumin modulates the apolipoprotein B mRNA editing by coordinating the expression of cytidine deamination to uridine editosome components in primary mouse hepatocytes

  • Curcumin, an active ingredient of Curcuma longa L., can reduce the concentration of low-density lipoproteins in plasma, in different ways. We had first reported that curcumin exhibits hypocholesterolemic properties by improving the apolipoprotein B (apoB) mRNA editing in primary rat hepatocytes. However, the role of curcumin in the regulation of apoB mRNA editing is not clear. Thus, we investigated the effect of curcumin on the expression of multiple editing components of apoB mRNA cytidine deamination to uridine (C-to-U) editosome. Our results demonstrated that treatment with $50{\mu}M$ curcumin markedly increased the amount of edited apoB mRNA in primary mouse hepatocytes from 5.13%-8.05% to 27.63%-35.61%, and significantly elevated the levels of the core components apoB editing catalytic polypeptide-1 (APOBEC-1), apobec-1 complementation factor (ACF), and RNA-binding-motif-protein-47 (RBM47), as well as suppressed the level of the inhibitory component glycine-arginine-tyrosine-rich RNA binding protein. Moreover, the increased apoB RNA editing by $50{\mu}M$ curcumin was significantly reduced by siRNA-mediated APOBEC-1, ACF, and RBM47 knockdown. These findings suggest that curcumin modulates apoB mRNA editing by coordinating the multiple editing components of the edito-some in primary hepatocytes. Our data provided evidence for curcumin to be used therapeutically to prevent atherosclerosis.

야생 돌복숭아(Prunus persica Batsch var. davidiana Max.) 추출액이 선천성 고혈압 흰쥐의 지질성분 및 혈압 저하에 미치는 영향 (Effects of the Feral Peach (Prunus persica Batsch var. davidiana Max) Extract on the Lipid Compositions and Blood Pressure Level in Spontaneously Hypertensive Rats)

  • 야생 돌복숭아(Prunus persica Batsch var. davidiana Max.)의 생리활성 추출물질이 선천성 고혈압 흰쥐(SHR)의 지질대사 이상 및 고혈압 예방과 개선효과 등에 생리생화학적 효능이 있을 것으로 추정되어 Wistar 계 수컷 SHR을 사용하여, 기본식이와 물만을 급여한 대조군인 Control군과 SHR에 돌복숭아 5.0g% 추출액을 급여한 군(5g% Ex.군) 및 SHR에 돌복숭아 10.0g% 추출액을 섭취시킨 10g% Ex.군을 33일간 실험 사육하여 혈청 지질성분 및 혈압 저하 효과를 생리적 측면에서 검토하기 위하여 본 실험을 수행하였다. 혈청 중의 총 콜레스테롤 농도, 중성지방, LDL-콜레스테롤, 유리 콜레스테롤 및 동맥경화지수 등에서 돌복숭아 생리활성물질 5.0g%, 10.0g% 추출액을 섭취시킴으로서 유의적인 농도 등의 감소 효과를 보였으며, HDL-콜레스테롤 농도 및 총 콜레스테롤에 대한 HDL-콜레스테롤 비는 상승되는 것으로 나타났다. 한편, 간장 및 뇌 중의 총 콜레스테롤, 중성지방 농도는 돌복숭아 5.0g%, 10.0g% 추출액을 섭취시킨 군에서 감소되는 것으로 나타났다. 반면, 혈청 apolipoprotein(Apo) A-I, Apo A-II 농도 등은 돌복숭아 5.0g%, 10.0g% 추출액을 섭취시킨 군에서 유의적으로 증가되는 것으로 나타났다. 또한 Apo C-II, Apo C-III, Apo E 및 Apo A-I에 대한 Apo B의 농도 비는 감소되었으며, 혈압의 변화는 SHR에 돌복숭아 5.0g% 및 10.0g% 추출액을 섭취시킴으로서 혈압이 저하됨을 관찰할 수가 있었다. 돌복숭아 5.0g% 농도(5g% Ex.군)와 10.0g% 농도(10g% Ex.군)의 추출액 섭취에 따른 두 군간에 각종 지질성분과 apolipoprotein 분획 농도 및 혈압 변화의 비교는 유의적인 차이는 없었다 따라서 야생 돌복숭아 중의 생리활성물질 추출액 섭취가 선천성 고혈압 흰쥐에 대한 혈청 지질개선 및 심장순환기계 질환, 고혈압의 예방과 치료개선에 효과가 있을 것으로 생각된다.

녹두 첨가로 인한 탈지대두 Grits(Defatted Soybean Grits) 발효물의 in vitro 상에서의 콜레스테롤 개선능 상승효과 (Cholesterol Improvement Synergistic Effects of Fermented Soybean Grits Caused by Added with Mung Bean in vitro)

  • 본 연구에서는 Bacillus subtilis NUC1균주로 발효한 탈지대두 grits 발효물인 FD와 탈지대두 grits에 녹두를 2.5, 5, 10%로 각각 첨가하여 발효한 FDM을 각각 제조하여 in vitro 상에서의 콜레스테롤 개선능을 검색하였다. FD와 FDM군의 콜레스테롤 흡착능을 살펴본 결과, 모든 군에서 70% 이상의 흡착능을 보였다. 특히 2.5% 녹두를 첨가하여 발효시킨 FDM군(2.5% FDM)은 90%의 가장 높은 흡착능을 보였다. 2.5% FDM군은 42%의 HMG-CoA reductase 저해 활성을 보였고, 또한 HepG2 세포를 이용하여 측정한 세포 내의 콜레스테롤 함량과 apolipoprotein AI, CIII의 개선효과에서도 가장 우수한 개선효과를 보였다. 따라서 2.5% 녹두를 첨가하여 발효시킨 FDM군은 고콜레스테롤 예방에 도움을 줄 것으로 생각된다.

Kisspeptin-10 Enhanced Egg Production in Quails Associated with the Increase of Triglyceride Synthesis in Liver

  • Our previous results showed that kisspeptin-10 (Kp-10) injections via intraperitoneal (i.p.) once daily for three weeks notably promoted the egg laying rate in quails. In order to investigate the mechanism behind the effects of Kp-10 on enhancing the egg laying rate in birds, this study focused on the alternations of lipids synthesis in liver after Kp-10 injections. 75 female quails (22 d of age) were allocated to three groups randomly, and subjected to 0 (control, Con), 10 nmol (low dosage, L) and 100 nmol (high dosage, H) Kp-10 injections via i.p. once daily for three weeks, respectively. At d 52, quails were sacrificed and sampled for further analyses. Serum $E_2$ concentration was increased by Kp-10 injections, and reached statistical significance in H group. Serum triglyceride (TG) concentrations were increased by 46.7% in L group and 36.8% in H group, respectively, but did not reach statistical significance, and TG contents in liver were significantly elevated by Kp-10 injections in a dose-dependent manner. Serum total cholesterol (Tch) concentrations significantly decreased in H group, while in H group the hepatic Tch content was markedly increased. The level of non-esterified fatty acid (NEFA), apolipoprotein A1 and B (apoA1 and apoB) were not altered by Kp-10 injections. The genes expression of sterol regulatory element binding protein-1 (SREBP-1), fatty acid synthetase (FAS), apolipoprotein VLDL-II (apoVLDL-II), cholesterol $7{\alpha}$-hydroxylase (CYP7A1) and vitellogenin II (VTG-II) were significantly up-regulated by high but not low dosage of Kp-10 injection compared to the control group. However, the expression of SREBP-2, acetyl-CoA carboxylase ($ACC_{\alpha}$), malic enzyme (ME), stearoyl-CoA (${\Delta}9$) desaturase 1 (SCD1), apolipoprotein A1 (apoA1), fatty acid binding protein 2 (FABP2), 3-hydroxyl-3-methyl glutaryl-coenzyme A reductases (HMGCR), estrogen receptor ${\alpha}$, ${\beta}$($ER{\alpha}$ and ${\beta}$) mRNA were not affected by Kp-10 treatment. In line with hepatic mRNA abundance, hepatic SREBP1 protein content was significantly higher in H group. Although the mRNA expression was not altered, the content of $ER{\alpha}$ protein in liver was also significantly increased in H group. However, SREBP-2 protein content in liver was not changed by Kp-10 treatment. In conclusion, exogenous Kp-10 consecutive injections during juvenile stage significantly advanced the tempo of egg laying in quails, which was associated with the significant elevation in hepatic lipids synthesis and transport.
