Effect of Deer Antler Extract on Muscle Differentiation and 5-Aminoimidazole-4-Carboxamide Ribonucleoside (AICAR)-Induced Muscle Atrophy in C2C12 Cells

  • The effect of deer antler extract on muscle differentiation and muscle atrophy were evaluated to minimize muscle loss following aging. Various deer antler extracts (HWE, hot water extract of deer antler; FE, HWE of fermented deer antler; ET, enzyme-assisted extract of deer antler; UE, extract prepared by ultrasonication of deer antler) were evaluated for their effect on muscle differentiation and inhibition of 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR)-induced muscle atrophy in C2C12 cells. Morphological changes according to the effect of antler extracts on muscle differentiation were confirmed by Jenner-Giemsa staining. In addition, the expression levels of genes related to muscle differentiation and atrophy were confirmed through qRT-PCR. In the presence of antler extracts, the length and thickness of myotubes and myogenin differentiation 1 (MyoD1) and myogenic factor 5 (Myf5) gene expression were increased compared to those in the control group (CON). Gene expression of AMP-activated protein kinase (AMPK), MyoD1, and myogenin, along with the muscle atrophy factors muscle RING finger-1 (MuRF-1) and forkhead box O3a (FoxO3a) upon addition of deer antler extracts to muscle-atrophied C2C12 cells was determined by qRT-PCR after treatment with AICAR. The expression of MuRF-1 and FoxO3a decreased in the groups treated with antler extracts compared to that in the group treated with AICAR alone. In addition, gene expression of MyoD1 and myogenin in the muscle atrophy cell model was significantly increased compared that into the CON. Therefore, our findings indicate that antler extract can increase the expression of MyoD1, Myf5 and myogenin, inhibit muscle atrophy, and promote muscle differentiation.

The Effect of Fermented Antler Extract in Prevention of Osteoporosis or Reduced Physical Activity in Females during Menopause

  • We was confirmed that the antler extract increases the expression of marker genes expressed in the process of bone formation, and that the effect on the increase in the expression of the gene is further increased by fermentation of the antler extract. In addition, the mouse model in which menopausal was induced by ovary extraction significantly reduced the movement distance and exercise time of mice compared to the control group. But the decrease was somewhat alleviated by the administration of the antler extract, and completely restored when the fermented antler extract was administered. In the menopause-induced mice, the body weight ratio of heart, liver, and spleen weights increased compared to the control group, but the antler extract and the antler ferment extract restored the body weight ratio of various organ weights to the level of the control group in the menopause-induced mice. Consequently, this has led to mitigating changes in the metabolism affected by menopause.

  • The aim of this study was to estimate the factors that can be used as a predictive indicator of antler productivity in elk. Body weight and antler data from 115 stags raised at the Animal Genetic Resources Research Center in Korea were used in this study. The function for the average antler weight was derived by age, which was $y=-3.4693+5.3747x-0.8x^2+0.0424x^3$, and $R^2$ was 0.991. The analysis of the relationship between body weight at birth and the 1st and 2nd year and the antler weight after the 2nd year showed a correlation between them. There was a correlation between the antler weight of 1 and 2 year old stags and the weight of 2 year old stags as well; especially, both antler weights for the 1st and 2nd year were highly correlated with the antler weight of the 4th year. However, there was no relationship between the antler main beam length for the 1st and 2nd year and the antler weight after the 3rd year. Therefore, it is expected that a large amount of antler will be obtained in the future if proper management is carried out for young deer from birth to 2 years old. Additionally, the results of this study are expected to be useful for the development of a deer breeding program and the deer industry.

가미생화탕(加味生化湯) 가(加) 녹용(鹿茸) 처방(處方)으로 산모(産母)의 빈혈(貧血)이 개선(改善)된 치험 3례 (A Clinical Study on Three Cases of Anemia in Postpartum Women Treated by Gamisaengwha-tang with Deer Antler)

  • Objectives: The purpose of this study is to report the effect of Gamsaengwha -tang with Deer antler on three postpartum women with anemia. Methods: The three postpartum women with severe anemia among postpartum women who admitted our hospital were treated by oriental treatments such as herbal medicine(Gamisaengwha-tang with Deer antler). Results: After treatment, the measurements of hemoglobin was increased and Anemia was improved. Conclusion: After delivery, many postpartum women have tendency to become anemic. Saengwha-tang and Deer antler became known to be good for anemia in oriental medicine. So the three postpartum women with severe anemia among postpartum women who admitted our hospital were treated by Gamisaengwha-tang with Deer antler. After treatment, the measurements of hemoglobin were increased comparative quickly, and anemia was improved. This case report shows that the Gamisaengwha-tang with Deer antler is effect for treatng anemia. Anemia will improve not only treating by Saengwha-tang except Deer antler but also treating by iron supplement, so we need more comparative study and observation and need more practical use of herbal medicine with Deer antler for anemia and disease of postpartum women.

사육 엘크의 녹용혈과 체녹혈의 혈액성분 (Biological Contents of Velvet Antler and Femoral Venous Blood in Farmed Elk(Cervus canadensis))

  • This study was performed to investigate the blood contents of velvet antler and femoral venous blood in farmed elk (Cervus canadensis). Blood samples were collected from 12 young adult elk (3.5-4.5 years old) with a mixture of fentanyl-azaperone-xylazine (Fentazine$^{(R)}$). Values for hematological and clinicochemical components of the clinical healthy elk were determined. The results were grouped for comparison according to velvet antler and femoral venous blood. Lower mean platelet counts and fibrinogen values were found in the velvet antler than femoral venous blood (p<0.01). White blood cell count of velvet antler was significantly higher than that of femoral venous blood 0<0.05). The clinicochemical parameters (total protein, albumin, globulin, alanine aminotransferase, aspartate aminotransferasei blood urea nitrogens calciums magnesium and phosphonls) obtained in this study far farmed elk are consistent with the values between velvet antler and femoral venous blood.

분말녹용의 약물활성 연구(I) (Pharmacological Studies on Powdered Whole Part of Unossified Antler)

  • Systematic pharmacological studies on powdered whole part of unossified antler obtained from Cervus nippon Temminck var. mantchuricus have been carried out in mice and rats. Powdered antler, with a single oral administration, showed a very weak acute and subacute toxicity; its MLD being >5g/kg orally in mice. On daily oral treatments of the antler for 14 days, it did not cause any significant differences in body weight gain, various organ weights and serum transaminase activities compared to those of the control rats. Powdered antler, with a single oral administration, showed a remarkable analgesic activity as evaluated by writhing syndrome and tail-pressure test, a weak CNS depressant activity as well as a weak immunopotentiating action as evaluated by carbon clearance test in mice. Powdered antler, with repeated treatments, showed a moderate antifatigue effect against immobilized stress and showed significant increases in both adrenal weight and its ascorbic acid content in rats, suggesting that the mode of antifatigue effect of antler is related to adrenal and its components.

Effect of Antler Development Stage on the Chemical Composition of Velvet Antler in Elk (Cervus elaphus canadensis)

  • This study was conducted to provide the basic information to allow improved scientific assessment of velvet antler's quality by investigating the change of chemical composition during different antler growth stages in elk (Cervus elaphus canadensis). Twenty four antlers were harvested from elk stags (aged 4-5 years) on 65 days (VA65), 80 days (VA80) and 95 days (VA95) after button casting, and the chemical composition of each antler was determined in five sections (top, upper, middle, base, and bottom). Crude protein and ether extract content was the highest in the top section, whereas ash content was the highest in the bottom section in all groups (p<0.05). Glycosaminoglycan (GAG) content was higher in the VA65 group than in the VA95 group in the upper section of antler (p<0.05). The collagen content was higher in the VA65 group compared to the VA95 group in the middle and bottom sections (p<0.05), and increased downward from the top to the bottom section. The proportions of certain amino acids, including aspartic acid, glutamic acid and isoleucine were higher (p<0.05), whereas proline and glycine were lower in the top section of antler compared to all other sections (p<0.05). The proportion of linoleic acid, 11,14,17-eicosatrienoic acid, total ${\omega}$-3 and ${\omega}$-6 fatty acids and polyunsaturated fatty acids (PUFAs) for all sections in the VA65 group was higher than in the VA95 group (p<0.05). These results suggested that the quality of velvet antler is strongly influenced by antler development stage.

골다공증 유발 동물모델에서 녹용 추출물의 경구 투여 효과: 체계적 문헌고찰 및 메타분석 (The Effects of Oral Administration of Deer Antler Extracts on an Osteoporosis-induced Animal Model: A Systematic Review and Meta-analysis)

  • Objectives This study aimed to assess the effects of oral administration of deer antler extracts on an osteoporosis-induced animal model. We analyzed the results of using deer antler single extracts on animal models with osteoporosis through a systematic review and meta-analysis. Methods We included osteoporosis studies in animal experiments that administrated deer antler extracts orally. We searched the following 13 databases without a language restriction: PubMed, EMBASE, Cochrane Library, Cumulative Index to Nursing and Allied Health Literature (CINAHL), China National Knowledge Infrastructure (CNKI), Wanfang, Korean Medical Database (KMbase), National Digital Science Library (NDSL), Korean Traditional Knowledge (Koreantk), Oriental Medicine Advanced Searching Integrated System (OASIS), Research Information Sharing Service (RISS), Korea Institute of Science and Technology Information (KISTI), and Koreanstudies Information Service System (KISS). We used Systematic Review Centre for Laboratory Animal Experimentation's risk of bias tool for assessing the methodological quality of the included studies. Results A total of 299 potentially relevant studies were searched and 11 were included for a systematic review. Nine studies used a single deer antler extract. A study compared the effects of single extracts of deer antler and antler glue, while another study compared the effects of three single extracts of deer antler, old antler, and antler glue. For evaluating the intervention effect, bone mineral density (BMD) was measured as the primary outcome, while the histomorphometric indicators of the bone and serum alkaline phosphatase and osteocalcin levels were used as the secondary outcome variables. On conducting a meta-analysis of studies on single deer antler extract, BMD was observed to be significantly increased compared to that in control group (standardized mean difference [SMD]=2.11; 95% confidence interval [CI]=1.58~2.65; Z=7.75; p<0.00001; I2=56%). As a result of meta-analysis, according to the concentration of deer antler, the group with high concentration showed statistically significantly higher BMD than the group with low concentration (SMD=1.28; 95% CI=0.74~1.82; Z=4.63; p<0.00001; I2=9%). Conclusions The research shows that the deer antler extracts have significant anti-osteoporotic effects on the osteoporosis-induced animal model. However the studies included in this research had a high methodological risk of bias. This indicates the requirement of considerable attention in the interpretation of the study results.

녹용수침액(鹿茸水浸液)이 Mouse의 Stress 방어작용에 미치는 영향(影響) (Effect of the Antler (Deer Horn) Water Extract on the Stress Resistance in Mice)

  • Antler, deer horn, occupies a particular place in oriental folk medicine as so called tonic remedy. In order to study on the effects of Antler water extract on stress resistance in mice, quantitative response was measured for the change of spontaneous activity by chemical stress drugs in the control group or the Antler water extract pretreated group. Spontaneous activity in mice was measured by counting the number of interruptions of light. The results of experiment were summerized as follows; I) In case of administrating Antler water extract 10mg/kg, 20mg/kg, 50mg/kg, and 100mg/kg, no significant change was observed in spontaneous activity in comparison with the control group. II) In case of administrating Antler water extract for 5 days or 10 days, no significant change was observed in spontaneous activity by chemical stress drugs, caffein and chloropromazine, in comparison with the control group.

Hyphal Growth Inhibition by Deer Antler Extract Mimics the Effect of Chitin Synthase Deletion in Candida albicans

  • Chitin synthase null-mutants propagate in yeast form in RPMI medium with suppression of hyphal growth. This hyphal suppression is also observed in the wild type culture grown in RPMI medium supplemented with deer antler extract. To identify the possible target of deer antler extract, the enzymatic activities of chitin synthases were examined. The enzymatic activities of three chitin synthases, CAChsl, CAChs2, and CAChs3, were found to be differentially inhibited by deer antler extract. Of them, CAChsl, was the most sensitive to the extract. These results indicate that deer antler extract causes hyphal suppression, which resembles the effects of chitin synthase deletion, probably through direct inhibition of chitin synthases.
