  • The synthetic study of 3',5-cyclic phosphates of 2'-substituted 2',3'-secouridines and 2',3'-secoribavirins toward development of new antiviral agents is described. These cyclic phosphates were synthesized from their respective 4-nitrophenyl 3',5'-cyclic phosphate triesters. These triesters were prepared from the corresponding 2'-azido and 2'-bromo 2',3'-seconucleosides.

COVID-19 Antiviral and Treatment Candidates: Current Status

  • The coronavirus disease 2019 (COVID-19) pandemic caused by the severe acute respiratory syndrome coronavirus 2 has severely impacted global health and economy. There is currently no effective approved treatment for COVID-19; although vaccines have been granted emergency use authorization in several countries, they are currently only administered to high-risk individuals, thereby leaving a gap in virus control measures. The scientific and clinical communities and drug manufacturers have collaborated to speed up the discovery of potential therapies for COVID-19 by taking advantage of currently approved drugs as well as investigatory agents in clinical trials. In this review, we stratified some of these candidates based on their potential targets in the progression of COVID-19 and discuss some of the results of ongoing clinical evaluations.

In vitro Antiviral Activities of Korean Marine Algae Extracts against Fish Pathogenic Infectious Hematopoietic Necrosis Virus and Infectious Pancreatic Necrosis Virus

  • To investigate the antiviral activity of marine algae against fish pathogenic viruses, which are often the causes of viral disease in aquaculture, the 80% methanolic extracts of 21 species collected from the coast of Korea were screened for their in vitro antiviral activities on infectious hematopoietic necrosis virus (IHNV) and infectious pancreatic necrosis virus (IPNV), using a flounder spleen (FSP) cell-line. Among them, Monostroma nitidum (10 ${\mu}g/mL$) exhibited the strongest inactivation on IHNV, showing a 2 log reduced virus titre as compared to the control in the determination of direct virucidal activity. In addition, Polysiphonia morrowii (100 ${\mu}g/mL$) remarkably reduced the virus titres of treated cells by 2-2.5 log, for both IHNV and IPNV, in the determination of cellular protective activity, implying the existence of substances that may modulate innate host defense mechanisms against viral infections. These results reveal that some marine algae could be promising candidates as sources of antiviral agents or as health-promoting feeds for aquaculture.

Synthesis of Certain 6-(Arylthio)uracils and Related Derivatives as Potential Antiviral Agents

  • New series of 6-(arylthio)uracils, 6-(4-substituted-1-piperazinyl)uracils, 2,4,5-trioxo-1H,3H-benzothiopyrano[2,3-d]pyrimidine and 5-aryl-2,4-dioxo-1H,3H-pyrimido[5,4-f]benzo[1,4]thiazepines have been prepared and screened for their in vitro activity against herpes simplex-1 virus (HSV-1) and human immunodeficiency virus-1 (HIV-1). The in vitro cytotoxic activity was also evaluated. The results of biological testing revealed that compound 5b showed marginal activity against HSV-1, while compounds 5b and 5f exhibited marginal activity against HIV-1. The rest of the tested compounds were found devoid of antiviral activity against both HSV-1 and HIV-1.

항바이러스제 Transdermal Patch의 제조와 평가 (Evaluation of Transdermal Patch for Antiviral Agent)

  • Transdermal patches and polymer membrane were prepared and evaluated for their ability to antiviral agent in vitro. The membrane perpared with styrene and HEMA by 0.5 and 10% of styrene composition. And the transdermal patches were fabricated with this membrane and silastic silicone sheeting. The antiviral agents used were ACV, BVDU and FEAU. The higher HEMA content membranes exhibited relatively high release rate of each drug. Permeation was enhanced by increasing of drug concentration. The parameters of each drug in diffusion experiment with styrene-HEMA membrane were investigated.

  • In this study; a series of 2',4'-doubly branched carbocyclic nucleosides (8,9,10) were synthesized from simple acyclic ketone derivative as starting material. The installation of the 4'-quaternary carbon needed was carried out using a 〔3,3〕-sigmatropic rearrangement. In addition, the introduction of a methyl group in the 2'-position was accomplished by Grig-nard reaction. Bis-vinyl was successfully cyclized using a Grubbs' catalyst II. The natural bases (adenine, cytosine, uracil) were efficiently coupled with the use of a Pd(0) catalyst. Although all the synthesized compounds were assayed against several viruses, only cytosine analogue 9 showed weak antiviral viral activity (EC$_{50}$=45.4 $\mu$M) against CoxB3 virus.s.

  • Eight new D-1.3-dioxolanyl and L-1,3-oxathiolanyl nucleosides containing 5-phenyl- selenyl pyrimidine bases which are expected to have antiviral activity were synthesized. Condensation of D-1,3-dioxolane acetate and L-1,3-oxathiolane acetate with 5-phenylselenyl pyrimidines gave anomeric mixtures of their nucleosides which were separated by silicagel column chromatography.

  • Based on (-)-neplanocin A with the 5'-hydroxyl substituted with fluoro, azido, or amino group, the corresponding xylo- and arabino derivatives were synthesized from D-ribose using the Mit-sunobu reaction as a key step. None of the final nucleosides did show either significant antiviral activities or cytotoxicities.
