Antitussive Effect of Fritillaria unibracteata Hsiao et K. C. Hsia

  • This experiment was conducted to clarify the antitussive effect of Fritillaria unibracteata Hsiao et K. C. Hsia. The antitussive effect in ethanol (75, 50, 30%) extracts from F. unibracteata was relatively high. The ethanol extracts of 20, 40 mg/kg concentrations were especially good antitussive effect. The ethanol extracts of F. unibracteata showed lower antitussive activities of 20~30% than sodium chloride injection used as a blank space control.

Antitussive, Expectorant, and Anti-inflammatory Effects of Mahwangyounpae-tang, a Polyherbal Formula in ICR Mice (ICR 마우스를 이용한 마황윤폐탕(麻黃潤肺湯)의 진해, 거담, 및 항염 효과 평가)

  • Objectives: The antitussive, expectorant, and anti-inflammatory effects of Mahwangyounpae-tang (MHYPT) aqueous extracts were observed in appropriate animal models of various respiratory disorders. Methods: MHYPT aqueous extracts were orally administered once a day for 11 days at dose levels of 400, 200, and 100 mg/kg. The effect of MHYPT was determined by comparing its antitussive effect with theobromine (TB), its expectorant effect with ambroxol (AM), and its anti-inflammatory effect with dexamethasone (DEXA). Results: MHYPT aqueous extracts (400 mg/kg) showed favorable antitussive effects comparable to those of TB (50 mg/kg) in the NH4OH-exposure coughing mouse model and expectorant effects comparable to those of AM (250 mg/kg) in the phenol red-secretion mouse model, but MHYPT (400 mg/kg) showed less anti-inflammatory activity compared to DEXA (1 mg/kg) in the xylene-induced acute inflammatory mouse ear model under the experimental conditions used. Conclusion: MHYPT aqueous extracts administered at dosage levels of 400, 200, and 100 mg/kg induced dose-dependent and favorable antitussive, expectorant, and anti-inflammatory activities that occurred by simultaneous modulation of the activity of mast cells and respiratory mucous production under the experimental conditions used in this study.

Experimental Study on Anti-inflammatory, Antitussive, and Expectoration Effects of Friltillariae Thunbergii Bulbus (절패모(浙貝母)의 항염 및 진해거담 효과에 대한 실험연구)

  • Objective: This study aimed to evaluate anti-inflammatory and antitussive expectoration effects of Friltillariae Thunbergii Bulbus (FTB) in a mouse model. Materials and Methods: To evaluate the anti-inflammatory effects of the FTB, we conducted in vitro experiments using RAW264.7 cells. An MTT assay and enzyme-linked immunosorbent assay (ELISA) were carried out to examine the anti-inflammatory effects of FTB. The expectorant effect on phenol red secretion, the antitussive effect on cough induced by ammonia solution, and leukocyte increased inhibition effects in acute airway inflammation in the animal model were confirmed. Results: FTB did not show cytotoxicity in the experimental group at 10, 30, 50, 100, 300, or 500 ㎍/ml and significantly inhibited the increase of NO, TNF-α and IL-6 in the experimental groups at 30, 50, 100, 300, and 500 ㎍/ml concentrations. In sputum, cough, and acute airway inflammation animal models, FTB significantly increased phenol red secretion in the 400 mg/kg administration group. FTB significantly reduced the number of coughs and significantly increased cough delay time in both 200 and 400 mg/kg dose groups. FTB decreased the white blood cell count in BALF (bronchoalveolar lavage fluid) in the 400 mg/kg administration group. Conclusion: Our study revealed that FTB elicits antitussive and expectorant effects by inhibiting inflammatory cytokines, increasing sputum secretion, suppressing cough, and reducing inflammatory cells. We concluded that FTB is a highly promising agent for respiratory tract infection with therapeutic opportunities.

  • According to the study of literature on RADIX STEMONAE about its efficacy, pharmacological action, and clinical adaptive disease, the results are as follows; 1. About the efficacy of RADIX STEMONAE, it is known as moistening the lungs to arrest cough, and intestinal parasites from ancient to now, and dispelling phlegm is also known. 2. The clinical adaptation of RADIX STEMONAE is chronic bronchitis, pertussis, pulmonary tuberculosis in respiratory disease, and ascaricide for Ascaris, Enterobius vermicularis or eczema, pruritus, destroy louse for endermic liniment. 3. The pharmacological action of RADIX STEMONAE are antitussive, tuberculostatic, anthelmintic, antibacterial, antivirus, antifungal. 4. The antitussive mechanism of RADIX STEMONAE is central inhibitor for cough center of medulla oblongata, and the mechanisms of bronchial smooth muscle relax, and expectorant is also known.

Prophetic Medicine-Nigella Sativa (Black Cumin Seeds) - Potential Herb for COVID-19?

  • Coronavirus disease-19 (COVID-19) is caused by the severe acute respiratory syndrome coronavirus 2 (SARS CoV-2). Currently, the management of patients with COVID-19 depends mainly on repurposed drugs which include chloroquine, hydroxychloroquine, lopinavir/ritonavir, ribavirin, remdesivir, favipiravir, umifenovir, interferon-α, interferon-β and others. In this review, the potential of Nigella sativa (black cumin seeds) to treat the patients with COVID-19 analyzed, as it has shown to possess antiviral, antioxidant, anti-inflammatory, anticoagulant, immunomodulatory, bronchodilatory, antihistaminic, antitussive, antipyretic and analgesic activities. Medline/PubMed Central/PubMed, Google Scholar, Science Direct, Directory of open access journals (DOAJ) and reference lists were searched to identify articles associated with antiviral and other properties of N.sativa related to the signs and symptoms of COVID-19. Various randomized controlled trials, pilot studies, case reports and in vitro and in vivo studies confirmed that N.sativa has antiviral, antioxidant, anti-inflammatory, immunomodulatory, bronchodilatory, antihistaminic, antitussive activities related to causative oraganism and signs and symptoms of COVID-19. N. sativa could be used as an adjuvant therapy along with repurposed conventional drugs to manage the patients with COVID-19.

  • A description of simple, isocratic and precise reversed phase HPLC methods is given for simultaneous quantification of pholcodine and ephedrine hydrochloride together with either carbinoxamine maleate or terfenadine in antitussive-antihistaminic oral pharmaceutical formulations. Separations were carried out on X-Terra and symmetry shield C18 column $(250\;{\times}\;4.6\;mm,\;5\;{\mu}m)$. The used isocratic elution systems were either $0.02\;M\;KH_2PO_4-acetonitrile$ in the ratio of 75 : 25 and pH adjusted to 7.70 with orthophosphoric acid or sodium hydroxide, for syrup (method A), or 0.02 octanesulphonic acid sodium salt solution-acetonitrile-acetic acid in the ratio of 75 : 25 : 0.5 for suspension (method B). The elution of both mixtures was achieved with a flow rate of 1 ml/min. Detection was carried out by UV absorbance at wavelengths of 220 and 250 nm for syrup and suspension, respectively. The quantification of the components in synthetic mixtures and actual syrup and suspension were calculated using the internal standard technique with metoclopramide HCl and codeine phosphate as internal standards (IS), respectively. The methods, for both mixtures, were validated and met all the requirements for the quality control analysis recommended by FDA and ICH.

Effect of Benincasa hispida Extract on Mucus Secretion in Rat Tracheobronchial Lumen (동과추출물이 흰쥐의 기관·기관지 내 점액분비에 미치는 영향)

  • Mucus in the respiratory system is a factor to cause not only respiratory diseases but also chronic diseases. In this regard, this study is to investigate whether Benincasa hispida extracts (BHE) would stimulate secreting mucus in trachea and tracheobronchial lumen of rats. As a result, effect of stimulating the mucus secretion into trachea and trachebronchial lumen of rats is significantly increased in accordance with the administration concentration of Benincasa hispida seed extracts (BHSE). The secretion amounts of phenol red (PR) and sodium fluoreseceine (NaFI) are significantly increased by 25% and 34%, respectively, when 70% EtOH extracts of Benincasae pericarpium (BPE) are administrated to rats at the concentration of 200 mg/kg. In addition, the secretion amounts of PR and NaFI are significantly increased when BuOH fraction and water fraction are intraperitoneally administrated to rats at the concentration of 200 mg/kg. It seems the result implies the physiologically active substance that stimulates the secretion of mucus is contained in Benincasa hispida. Thus, Benincasa hispida is considered to be effective for Korean medicine treatment as a medicinal plant having effect of antitussive and apophlegmatic, and it also can be useful in developing functional food and beverage.

  • In order to investigate experimentally the clinical effects of Chungsimjeongchun-Tang that have been widely used for cardiac asthma, dyspnea and coughs secondary to the appoplexy and cardiac diseases, the water extract of Chungsimjeongchun-Tang was conducted to the effects on the anti-histamine action and respiratory system in mice, guinea-pigs, rabbits and cats. The following results of Chungsimieongchun-Tang were obtained. Spontaneous motility of the isolated ileum of mice and rabbits was strongly suppressed and contractions of the isolated ileum of mice and guinea-pigs induced by acetylcholine chloride, barium chloride and histamine were remarkably inhibited. Anti-histamine action was noted. Antitussive action on the mechanically irritative cough in cats was recognized. Expectorant action on the leakage of dye in the respiratory tract of rabbits was also shown.

  • Prunus serrulata var. spontanea(Rosaceae) is a large sized tree widely distributed throughout Korea. The red fruits are edible and are used in traditional folk medicine against heart failure from beriberi. dropsy. mastitis, an emmenagogue, and toothache. Also. the bark of P. serrulata var. spontanea has been used for detoxification. and relaxation and as an antitussive in traditional Korea medicine. (omitted)
