Effects of Methanolic Extract from Salvia miltiorrhiza Bunge on in vitro Antithrombotic and Antioxidative Activities (단삼 메탄올 추출물의 항혈전 및 항산화 효과)

  • Salvia miltiorrhiza Bunge is known to potentially prevent arteriosclerosis and hypertension, but its effects on platelet function are not clear. In this study, we evaluated the in vitro antithrombotic activities of the edible plant extract. Methanol extract of S. miltiorrhiza Bunge exhibited about 70% fibrinolytic activity compared to the plasmin control, and inhibited ADP- and collagen-induced platelet aggregation in a concentration-dependent manner with $IC_{50}$ values of 0.42 and 0.07 mg/mL, respectively. S. miltiorrhiza Bunge extract significantly prolonged the activated partial thromboplastin time (APTT) and prothrombin time (PT) compared with control. Moreover, 0.05 mg/mL S. miltiorrhiza Bunge extract contained 87.3% l,l-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. In conclusion, S. miltiorrhiza Bunge seemed to enhance antithrombotic activity due to its radical scavenging activity. Based on these data, further examination is required to determine the mechanism of platelet-dependent antithrombosis and the effect of polyphenols on platelet function.

Comparison of fibrinolytic activity from Korean indigenous insects (국내 토착 곤충의 항혈전 비교)

  • The fibrinolytic activity of aqueous extracts from Korean indigenous insects were studied. Fibrinolytic activity of aqueous extract from 5 insects (larva and adult parts of Meloimorpha japonica, Allomyrina dichotoma, Cetonia pilifera, Apis mellifera) showed 2.7-fold potent than that of plasmin used as a positive control. In addition, the fibrinolytic activity was observed through the degradation products by SDS-PAGE. The extracts efficiently hydrolyzed ${\alpha}-$, ${\beta}-$ and ${\gamma}$ chains of human fibrinogen. This study suggested that aqueous extracts from Korean indigenous insects have potential in developing a useful source of antithrombosis agent(s).

The Study on Antithrombosis and Inflammation according to The Broth Preparation Method of Gamijoukyungtang (가미조경탕(加味調經湯)의 전탕(煎湯) 방법에 따른 항혈전 및 염증에 관한 연구)

  • Purpose: In this study, we investigated the anti-thrombotic and antiinflammatory efficacy of "Gamijoukyungtang(GJKT)". Methods: We studied inhibitory effects of platelet aggregation, FXa activation, $TXB_2$ and $PGE_2$ biosynthesis and suppressive effects of GPIIb/IIIa activity and oxidative damage, pro-inflammatory cytokine reduction effects of 'GJKT(by press extractor)/GJKT-1(by pressless extractor)' in vitro. Also, we studied suppression of pulmonary embolism, AV shunt model in rats and shortening of Rat tail bleeding time in vivo. Results: GJKT/GJKT-1 extract showed inhibitory effects on GPIIb/IIIaactivities and platelet aggregation induced by ADP, epinephrine, collagen and arachidonic acid. They suppressed biosynthesis of $PGE_2$ but GJKT-1 only supressed biosynthesis of $TXB_2$. In FXa assay, they inhibited activation of FXa. they suppressed pulmonary embolism triggered by collagen and epinephrine. In AV shunt model, they decreased the weights of AV shunt thrombus. they inhibited pro-inflammatory cytokines and decreased oxidative damages caused by DPPH. Conclusion: We confirmed the anti-thrombosis, and ant-inflammatory efficacy of 'GJKT(by press extractor)/GJKT-1(by pressless extractor)'.

Antithrombotic Activity of Sunghyangjunggisan

  • As apart of our continuing search for antistroke agents from the herbal medicinal resources, we examined in vitro, ex vivo and in vivo the possibility of Sunghyangjunggisan and its ingradients as a novel antithrombotic agent. In vitro ADP- and collagen-induced rat platelet aggregations were potently inhibited by Arisaematis Rhizoma, Cinnamomi Cortex and Zingiberis Rhizoma in a dose-dependent manner, but not by Sunghyangjunggisan. However, Sunghyangjunggisan significantly inhibited ex vivo rat platelet aggregation. Arisaematis Rhizoma, Atractylodis Rhizoma Alba, and Pinelliae Rhizoma also significantly inhibited ex vivo rat platelet aggregation. Sunghyangjunggisan, Alpiniae Fructus and Zingiberis Rhizoma showed significant protection from death due to pulmonary thrombosis in mice. Therefore, Sunghyangjunggisan can express the antithrombotic action, when it is orally administered.

The effect of Propolis on Endotoxin-induced thrombosis (Endotoxin에 의한 혈전증에 미치는 Propolis의 효과)

  • Propolis, a natural resinous compound collected from honey bees, contains many biochemical constituents and has been used for traditional medicines as early as 300 B .C. Recently, it has been reported to possess many biological activities such as antibacterial, antiviral, fungicidal, local anaesthetic, immunostimulating, antiinflammatory and free radical scavenging properties. To investigate activities of chrysin, one of propolis effective compounds for blood coagulation system was injected endotoxin (4000 EU/kg, i.v.) in rats at 1 hr after administered chrysin (20 mg/kg, p.o.). This study was resulted that chrysin has antiplatelet aggregation activity in vitro, delay of blood clotting time and prothrombin time, and reduction of fibrinogen and FDP in vivo. Chrysin has increased SOD activity, GSH content and GST activity, and decreased MDA content in liver. The result suggests that the antithrombosis effect of chrysin is suppressive activity for a blood coagulation system and antioxidative activity.

Antithrombotic Activities of Chungpesagantang, Sunghyangjunggisan and Yangkyuksanwhatang

  • The possibility of Chungpesagantang, Sunghyangjunggisan and Yangkyuksanwhatang, which have been recommended for stroke patients with constipation in traditional Korean clinics, was evaluated as a novel antithrombotic agent. Chungpesagantang potently inhibited ADP- and collagen-induced rat platelet aggregation in vitro, but Sunghyangjunggisan and Yangkyuksanwhatang did not inhibit it. However, all tested herbal formulae inhibited in vivo ADP- and collagen-induced rat platelet aggregation and extended the bleeding time of mice tails. All these herbal formulae did not affect certain plasma clotting times such as APTT, PT and TT. Among these herbal formulae, Chungpesagantang showed significant protection against death due to pulmonary thrombosis in mice. Based on these findings, these herbal formulae should prevent thrombosis and cardiovascular diseases such as myocardial infraction stroke, and arteriosclerosis.

  • Ophiopogonin D (OPD) is a steroidal glycoside derived from Ophiopogon japonicus, a traditional Chinese medicine with diverse biological activities, including antithrombosis, anti-inflammation, and antitussive effects. To investigate the cellular effects and mechanisms of OPD on oral squamous cell carcinoma, cell viability was explored, and the effects of OPD on cell cycle regulators, apoptotic marker proteins, and key proteins involved in metastasis and signaling pathways were examined by MTT assay and Western blotting in YD38 cells. OPD strongly inhibited cell proliferation and induced caspase-dependent apoptosis of YD38 cells by suppressing the cell cycle and activating caspase-3 and poly ADP ribose polymerase. Additionally, OPD suppressed the expression of vital proteins regulating metastasis and proliferation within the integrin/matrix metalloproteinases/FAK and AKT/PI3K/mTor pathways. Thus, OPD can be a potential treatment candidate for gingival cancer.

Effect of Alkaline Ionized Water on Stabilization of Antioxidation, Antithrombosis and Antibacterial Activities (항산화, 항혈전 및 항세균 활성의 안정화에 미치는 알칼리 이온수의 영향)

  • In this study, the stable maintenance of bioactivity in alkaline ionized water (AIW) and antibacterial effects of AIW were evaluated to confirm benefits of AIW. As controls, purified water (PW) and tap drinking water (DW) were used. The pH and ORP (oxidation-reduction potential) of AIW, PW and DW used were 9.5 and 120 mV, 7.2 and 144 mV, and 7.3 and 564 mV, respectively. High level of minerals was observed in DW (DW>AIW>PW of mineral contents). Concentrations of $Ca^{++}$ and $Na^+$ in DW were 14.5, and 8.4 mg/l, respectively, while no $Ca^{++}$, $Mg^{++}$, $K^+$, and $Na^+$ were detected in PW. Evaluation of antioxidant activities for AIW, PW and DW showed that the waters did not act as antioxidants. However, the DPPH (1,1-diphenyl-2-picryl hydrazyl) or superoxide radical scavenging activities or reducing power of vitamin C were stably maintained in AIW and PW, though not in DW, against heat treatment ($60^{\circ}C$) or vigorous shaking (120 rpm) at $37^{\circ}C$. Similarly, after aspirin treatment at $60^{\circ}C$ for 1 hr, the antithrombosis activity in PW and AIW was 62.6% and 55.3%, while that of DW was 52.1%. Furthermore, cell growth analysis and viable cell count of Escherichia coli H7:O157 in PW, AIW and DW showed that AIW and DW, not DW, have antibacterial activities. Our results suggest that the state of water, for example pH, ORP and mineral contents of water, should be considered in medicine or food industries, and that AIW has high potential for utilization in various fields.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Dioscorea batatas Decne (마(Dioscorea batatas Decne)의 항균, 항산화 및 항혈전 활성 평가)

  • Yam (Dioscorea batatas Decne) has been recognized as healthy food due to its various biological activities, such as anti-obesity, anti-constipation, anti-proliferation, anti-mutagenic activity, and decrease of blood glucose and cholesterol level. In this study, the methanol extract and its organic solvent fractions were prepared from dried Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activity were evaluated, respectively. The 66.7% of methanol extract was fractioned into water residue, and most of total-polyphenol/total-flavonoids were found in ethylacetate fraction. This ethylacetate fraction at $500\;{\mu}g/disc$ concentration showed strong antibacterial activity against gram-negative and gram-positive bacteria, except Staphylococcus aureus. Antifungal activity was not observed in methanol extract and its fractions. But, strong antithrombin activity was found in methanol extract. At 4.8 mg/mL concentration, thrombin time was 99.5 sec, which is coincides with 30% activity of aspirin, a commercial available antithrombosis agent. The ethylacetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $80.5\;{\mu}g/mL$), compared to vitamin C ($IC_{50}$ of $15.2\;{\mu}g/mL$). Also, the ethylacetate fraction showed strong SOD-like activity and reducing power, which are coincide with 43% of vitamin C and 82.7% of butylated hydroxytoluene activity, respectively.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Aerial Bulbils of Dioscorea batatas Decne (영여자의 항균, 항혈전 및 항산화 활성 평가)

  • The annual productions of yam and its aerial bulbils are estimated to 5,000 and 2,500 ton, respectively. But the majority of bulbils had been discarded without specific use. In this study, methanol extract and its organic solvent fractions were prepared from bulbils of Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activities were evaluated, respectively. The methanol extract contained 58.98% of water-soluble materials as like yam's extract. But the bulbils's extract contained 12-folds of total polyphenol and 3.4-folds of total flavonoids compared than yam's extract, respectively. For antimicrobial activity the hexane and ethyl acetate fraction showed strong antibacterial activity at $500{\mu}g/disc$ concentration against gram-negative and gram-positive bacteria. Antifungal activity was not observed in any fractions. Strong antithrombin activity was found in the hexane fractions. At 4.8 mg/mL concentration thrombin time (TT) was over 300 sec, which is 4-folds extended than the TT of yam. In a while, the ethyl acetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $38.1{\mu}g/mL$), SOD-like activity and reducing power, which are comparable to vitamin C or BHT. Our results suggest that the bulbils of yam as yam tuber have useful bio-activities, such as antibacterial, antithrombosis, and antioxidant activity.
