SEROLOGICAL RELATIONSHIPS BETWEEN POTATO VIRUS Y STRAINS (감자바이러스 Y 계통간의 혈청학적 관계)

  • Two PVY strains (PVY-VB and PVY-VN) isolated from tobacco in Korea were compared for their serological relationship with other 8 strains which were obtained from tobacco or potato in different countries. One of these strains, PVY-Argentina showed the spur reaction to PVY-VB and PVY-VN antisera in SDS-agar gel double diffusion plates. The two Korean PVY strains were closely related to other strains except for one, PVY-Argentina when antigen-antibody reciprocal absorption tests were conducted. It is suggested that the strain, PVY-Argentina, is a new serotype containing a specific antigenic site different from other 9 strains tested.

  • A competitive enzyme-linked immunosorbent assay (ELISA) was developed for the quantitative detection of organophosphorus insecticide cyanophos. An analogue (hapten) of cyanophos was synthesized and was coupled to BSA to produce polyclonal antibodi es from rabbits. The antisera were screened against another hapten coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigen-coated ELISA was developed, which showed an I50 of 310 ng/mL with the detection limit of 20 ng/mL. The antibodies showed negligible cross-reactivities with other organophosphorus pesticides except for parathion-methyl, which makes the assay suitable for the selective detection of cyanophos.

  • Species of scotochromogenic mycobacteria of Group II isolated from sputa of patients with pulmonary tuberculosis and tuberculosis-like diseases from 1979 to 1984 were identified by simple biochemical tests using. nitrate reduction, Tween-80 hydrolysis, arylsulfatase and urease test, and serotypes of the isolates belonging to M. scrofulaceum were differentiated by bacterial agglutination test. Of 39 strains. tested, 11(28.2%) proved to be M. scrofulaceum, 15(38.5%) M. flavescens and 13(33.3%) M. gordonae. But none of the isolates belonged to M. szulgai and M. xenopi known as major pathogens of mycobacteria of Group II. Of 11 strains of the isolates identified as M. scrofulaceum 3 strains(27.3%) each belonged to serotype 41 and 42, and 4 strains(36.4%) belonged to serotype 43, but one strain was not typable because of its inagglutinability by any one of the type specific sera. In addition, the sensitivity and specificity of rabbit immune sera against type strains of serotype 41, 42 and 43 of M. scrofulaceum were analysed by bacterial agglutination test. In the sensitivity of microplate test with 11 isolates of M. scrofulaceum, a comparative tandem test using 2 units and one unit of absorbed antisera against three serotypes appeared to be superior to a conventional microplate test using one unit of type specific antisera.

  • This study was performed in order to identify changes of the plasma membrane proteins in rat submandibular gland tumors induced by 7,12-dimethylbenz[a]anthracene [DMBA] and X-irradiation. Two kinds of tumor associated membrane proteins (protein A and B) were isolated with 3 M KCl extraction from rat submandibular gland tumors induced by DMBA and X-irradiation. To identify their antigenicities, immunoelectrophoresis and double immunodiffusion was carried out with various proteins extracted from liver, heart, skin and pancreas of adult rats and from embryonic liver, heart and skin. The rabbit antisera against the protein A did not cross-react with any of the proteins extracted from the above mentioned tissues, suggesting that protein A might be tumor specific antigen. However, the rabbit antisera against protein B was precipitated with proteins extracted from the liver of adult and embryonic rats. Polyacrylamide gel electrophoresis of these two proteins (A and B) showed that protein A was a dimer with molecular weights of 69,000 and 35,000 dalton, whereas protein B was a monomer with molecular weight of 50,000 dalton.

  • Pasteurella multocida is one of the important animal pathogen causing widespread infections in various domestic animals. In swine, it causes severe respiratory diseases such as atrophic rhinitis and pneumonic pasteurellosis. To develop the efficient subunit vaccine against swine atrophic rhinitis, we investigated protective antibodies and humoral immunity of outer membrane protein H (OmpH) which is one of the major outer membrane proteins in P. multocida. Outer membrane fraction of P. multocida was immunologically detectable using antisera from both mice groups vaccinated by formalin-killed whole cells and by commercial vaccine. The expression vector for production of recombinant OmpH was constructed and the recombinant OmpH was expressed and purified from E. coli. Recombinant OmpH showed high antigenic and immunogenic properties in mice vaccination and ELISA with antisera.

Isolation and Characterization of Bacillus thuringiensis Toxic to Spodoptera Species in Kora (거세미나방속 해충에 독성을 가지는 Bacillus thuringiensis 군주의 분리 및 특성)

  • To isolate Bacillus thuringiensis toxic to Spodoptera species, we collected soil samples in Korea. In these samples, we characterized 7 B. thuringiensis isolates toxic to spodoptera exigua or S. litura from soil, granary and sericultural farm samples. The 7 isolates were named B. thuringiensis STB-1, STB-2, STB-3, STB-4, STB-5, STB-6 and STB-7, respectively. The bioassay of these isolates against S. exigua and S. litura showed highly insecticidal activity. The serotypes of them were determined by agglutination tests using 33 antisera ; STB-1 an STB-2 are identical to B. thuringiensis subsp. kurastaki, and STB-3, STB-4 and STB-5 are identical to subsp. kenyae. STB-6 and STB-7 did not react with 33 antisera. STB-1 and STB-3 which have different gene types from B. thuringiensis subsp. kurastaki and subsp. kenyae are identified new isolates. STB-6 and STB-7 which show no agglutination in serological tests havd cryIA(a), cryIA(b), cryIC, and cryII genes are also identified new isolates. Molecular weights of parasporal inclusions of all isolates were determined approximately 130 kDa by SDS-polyacrylamide gel elctrophoresis.

Immunohistochemical studies on somatotropes and mammotropes in hypophysis of Korean native goat (한국재래산양 뇌하수체의 성장자극세포 및 젖샘자극세포에 관한 면역조직화학적 연구)

  • The localizations and morphological characteristics of immunoreactive cells for prolactin(PRL) and growth hormone(GH) antisera were studied with double immunohistochemistry in the adenohypophysis of the Korean native goat. PRL immunoreactive cells(mammotropes) and GH immunoreactive cells(somatotropes) were present in the pars distalis and pars tuberalis, but not in the pars intermedia. A few cells were stained with both PRL and GH antisera (somatomammotropes). The possessional percentages of mammotropes, somatotropes and somatomammotropes were 37.0%, 32.6%, 1.0% in females and 35.6%, 32.6%, 1.1% in males, respectively. Mammotropes were oval or round in shape, and $10{\sim}20{\mu}m{\times}17{\sim}25{\mu}m$ in size. These cells were distributed throughout the pars distalis, but were more abundant on the dorsal part adjacent to the hypophyseal cavity and along the lateral and ventral peripheral regions. Somatotropes were elliptical, triangular or polygonal in shape, and $8{\sim}17{\mu}m{\times}17{\sim}18{\mu}m$ in size. The distribution pattern of somatotropes was similar to that of mammotropes. Some somatomammotropes were intercalated between clusters of mammotropes while the others were dispersed independently among the secretory cells.

Adhesion of Oral Pathogens to Human Submandibular-Sublingual Salivary Proteins (악하선-설하선 혼합타액내 주요 단백질에 대한 구강내 주요 균주의 부착)

  • The present study was performed to investigate the binding between salivary proteins(low-molecular-weight mucin;MG2, amylase, proline-rich proteins;PRPs) and oral pathogens(Streptococcus gordonii, Actinomyces viscosus, Staphylococcus aureus) by using solid-phase assay. In the case of transferring proteins to Immobilon-P, S. gordonii binds to MG2. A. viscosus binds to MG2, amylase, and PRPs, and S. aureus binds to MG2 and amylase. On nitrocellulose membrane, S, gordonii and A. viscosus bind to MG2, amylase, and PRPs. S. aureus binds to MG2 and PRPs. However, rabbit anti-A. viscosus antisera and rabbit anti-S. aureus antisera showed cross reactivity to PRPs adsorbed to only nitrocellulose membrane in negative control experiments, which were done without bacterial overlay. The results were different according to the membrane used as solid-phase, which reflected the assay-sensitive nature of binding experiment. PRPs and amylase are known to be components of tooth enamel pellicle. In addition, there was experimental evidence that PRPs and MG2 may covalently bind to oral mucosal epithelium. Considering above facts, the results of the present study can provide information on the interactions between salivary proteins and oral bacteria on tooth and oral mucosal surfaces.

IMMUNE REACTION OF SPECIFIC SERUM ANTIBODIES TO PORPHYROMONAS ENDODONTALIS ANTIGEN IN PATIENTS WITH PERIAPICAL LESION (치근단 병소가 있는 환자에서 Porphyromonas endodontalis 항원에 대한 혈청 특이 항체의 면역 반응 연구)

  • Porphyromonas endodontalis is a black-pigmented anaerobic Gram-negative rod which is associated with endodontal infections and this microorganism possesses a potential for pathogenicity. The purpose of this study was to compare the membrane components of Porphyromonas endodontalis and Porphyromonas gingivalis and to study the immune reaction patterns of Porphyromonas endodontalis with patients with periapical lesion. Porphyromonas endodontalis (ATCC 35406), Porphyromonas gingivals serotypea (381), serotype b(W50), serotype c(A7A1-28) were cultured in anaerobic condition. Rabbit antisera were prepared by intravenous injection of formalized whole cells and human sera were obtained from patients and dental students. Indirect immunofluorescence method was used to study on the cross reaction between Porphyromonas endodontalis and Porphyromonas gingivalis serotype a, b, c antigen. Total membrane protein profiles of Porphyromonas endodontalis antigen were studied by sodium dodecyl sulfate polyacrylamide gel electrophoresis and the reactivity of antigenic components of Porphyromonas endodontalis against sera of patients and rabbit anti-Porphyromonas endodontalis antisera were assessed by Immunoblotting method. The following results were obtained : 1. Antigens of Porphyromonas endodontalis has multiple antigenic components, and both patients with periapical lesion and normal healthy individual showed immune response to this. 2. Patients group and healthy individual group showed a diversity of immune reaction pattern but they showed immune response against 43kd protein. 3. Patients with periapical lesion showed more diverse immune response than healthy individual and in some patients, much more bands appeared to lower molecular weight protein. 4. According to indirect immunofluorescence and Immunoblotting study, Porphyromonas endodontalis did not share common antigen with Porphyromonas gingivalis serotype a, b, c.

Studies on Sexing of Mouse Embryos with Rat H-Y Antisera (흰쥐 H-Y 항혈청을 이용한 생쥐배의 성감별에 관한 연구)

  • These expriments were carried out to investigate existence of H-Y antibody in the rat serum immunized against H-Y antigen from rat spleen cells and effect of H-Y antiserum on development of mouse male embryos. The results obtained were summerized as follows : 1. When mouse embryos were cultured for 48∼72 hrs in the Ham's F10 containing 16% of FBS(fetal bovine serum) or RNS(rat normal serum), percentages of embryos developed from 2, 4, 8 and 16-cell embryo to morulae were 20, 27, 94 and 100%, respectively, in FBS and 8, 7, 94 and 100%, respectively, in RNS. Eight to 16-cell embryos showed no difference in development rate between FBS adn RNS. 2. When 8∼16-cell mouse embryos were cultured for 24∼48 hrs in the Ham's F10 containing FBS, RNS+GPC(guinea pig complement) and RAS(rat antiserum)+GPC, proportions of embryos developed to the expanded blastocyst stage were 100, 82.4 and 52.1∼53.6%(ave.52.9), respectively, so that it was suggested that rat antiserum suppressed development of male embryos. 3. When 8∼16-cell mouse embryos were cultured for 24∼48 hrs in the Ham's F10 containing FBS, RNS, RNS+GPC and RAS+GPC, proportions of embryos developed to the expanded blastocyst stage were 94.5, 90.9, 82.3 and 47%, respectively, and the embryos developed in the medium containing RAS+GPC seemed to be female. These results indicated that the antisera prepared through immunized against H-Y antigen from rat spleen cell, possessed H-Y antibody which supressed development of male embryos.
