Abnormal changes in skin color induce significant cosmetic problems and affect quality of life. There are two groups of abnormal change in skin color; hyperpigmentation and hypopigmentation. Hyperpigmentation, darkening skin color by excessive pigmentation, is a major concern for Asian people with yellowe-brown skin. A variety of hypopigmenting agents have been used, but treating the hyperpigmented condition is still challenging and the results are often discouraging. Panax ginseng has been used traditionally in eastern Asia to treat various diseases, due to its immunomodulatory, neuroprotective, antioxidative, and antitumor activities. Recently, several reports have shown that extract, powder, or some constituents of ginseng could inhibit melanogenesis in vivo or in vitro. The underlying mechanisms of antimelanogenic properties in ginseng or its components include the direct inhibition of key enzymes of melanogenesis, inhibition of transcription factors or signaling pathways involved in melanogenesis, decreasing production of inducers of melanogenesis, and enhancing production of antimelanogenic factor. Although there still remain some controversial issues surrounding the antimelanogenic activity of ginseng, especially in its effect on production of proinflammatory cytokines and nitric oxide, these recent findings suggest that ginseng and its constituents might be potential candidates for novel skin whitening agents.

Cadmium (Cd) is well known as a spermatotoxic and gonadotoxic heavy metal ion. This study was performed to assess the possible protective effect of Enerbalance on Cd-induced spermiotoxicity and testicular damage. The control group received isotonic saline; Cd group received Cd (2 mg/kg BW per day) orally; extract-treated groups were orally administrated with Enerbalance (50 mg and 100 mg/kg BW per day) and Cd for 10 days. Morphological changes of testicular tissue, sperm characteristics, oxidative/antioxidative parameters from testis, and serum sexual hormone level were determined. Enerbalance was significantely increased sperm amount in cauda epididymis without changes of ratio of epididymis/body weight and testis/body weight. Cd caused a marked decrease in epididymal sperm concentration and chemotactic sperm motility, testicular superoxide dismutase (SOD), catalase (CAT), Enerbalance was significantly ameliorated loss of epididymal sperm concentration, sperm chemotactic motility, antioxidative parameters, and male hormone whereas decreased abnormal architecture by testis damage. Enerbalance was successfully attenuated these adverse effects of Cd and offers a dose-dependent protection. Our study demonstrated that Enerbalance could proffer a measure of protection against Cd-induced testicular damage and spermiotoxicity by possibly reducing oxidative stress and increasing the antioxidant defense mechanism in mice.
In the previous report, E-kong-san, which is usually used for recovering health in traditional medicine, has been shown to decrease cisplatin induced nephrotoxicity in vivo and in vitro. The significant reduction of E-kong-san on the cisplatin induced nephrotoxicity led us to investigate whether the effect of this water extract was a result of triggering antioxidation. In monkey kidney Vero cells, E-kong-san at $5{\sim}10\;mg/ml$ was able to attenuate 2mM cisplatin-stimulated cell death by 46.8% and 31.8%, respectively. E-kong-san showed strong free radical scavengering activities on 1,1-diphenyl-2-picrylhydrazil (DPPH) radical and xanthine/xanthine oxidase (XOD) generated superoxide anion radical $(O_2^{-.})$. We further studied the effects of E-kong-san on lipid peroxidation in rat liver microsomes induced by enzymatic and nonenzymatic methods. Moreover, E-kong-san exhibited significant inhibition on both ascorbic $acid/Fe^{2+}$ and $ADP/NADPH/Fe^{3+}$ induced lipid peroxidation in rat liver microsomes. Based on these results, we suggest that E-kong-san protects the cisplatin induced cytotoxicity by its antioxidative mechanism.
Objectives : This study was carried out to understand the immune responses of the Changchuldointanggami-bang(after referred to CDIT) on antioxidants, THP-1 cell and rheumatoid arthritis in Collagen-induced Arthritis(CIA) mice. Methods : The antioxidative effect of CDIT on DPPH free radical and SOD-like activity was measured. CDIT was administered to THP-1 cell, cytokine and mRNA associated with inflammation were measured. CDIT was orally administered to mice with arthritis by collagen II and then cytokine, PBMC in the serum were measured. Results : 1. The scavenging activity of CDIT on DPPH free radical was dose-dependent. 2. The effect of CDIT on SOD-like activity was dose-dependent. 3. The production of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ was decreased significantly in THP-1 cell. 4. The expression of $IL-1{\beta}$ mRNA, IL-6 mRNA, $TNF-{\alpha}$ mRNA were decreased significantly in THP-1 cell. 5. $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ were decreased significantly in the serum of CIA mice. 6. CD3+, CD4+, CD8+ were increased significantly, but CD3+/CD69+, CD3+/CD49b+, B220+/CD23+ were decreased significantly in PBMC of CIA mice. Conclusions : Taking all these observations into account, CDIT is considered to be effective in rheumatoid arthritis. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.
The 70% ethanol extract of Alpinia officinarum and its major flavonoid, galangin showed strong antioxidative effect on the lipid peroxidation of ethyl linolate with Fenton's reagent and free radical scavenging effect to DPPH radical generation. However, they did not reveal any pro-oxidant effect on bleomycin-Fe(III) dependent DNA degradation. They also showed the protective effect against $H_2O$$_2$, KO$_2$ or UV-induced cytotoxicity in mammalian cells. They also showed the suppressive effect of DNA damage induced by $H_2O$$_2$ or KO$_2$ with dose-dependent manner in single cell gel electrophoresis(SCGE) assay. On the other hand, they have an anticlastogenic effect against adriamycin-induced micronucleated reticulocyte in peripheral blood of mice. These results suggest that the mechanism of inhibition by 70% ethanol extract of Alpinia officinarum and galangin against reactive oxygen species (ROS)-induced genotoxicity or cytotoxicity is due, at least partly, to their antioxidative and free radical scavenging properties without pro-oxidant effect. All these results indicate that 70% ethanol extract of Alpinia officinarum and galangin may be useful for protection against ROS-induced cytotoxicity and DNA damage.

Effects of Sotosaja hwan on Blood Glucose, Hyperlipidemia, Polyol Pathway and Antioxidative Mechanism in ob/ob Mouse Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Sotosaja-hwan has been known to be effective for the antiaging and composed of four crude herbs. In male ob/ob mouse in severe obesity, hyperinsulinemia and hyperlipidemia, which are features of NIDDM, the hyperglycemic activites and mechanisms of Sotosaja-hwan were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 30 and 90 mg Sotosaja-hwan per 1 kg of body weight for 14 days. The effects of Sotosaja-hwan extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of superoxide anion radical $({\cdot}O_2)$, MDA+HAE, GSH/GSSG ratio, and also the enzyme activities involved in polyol pathway. Sotosaja-hwan lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Sotosaja-hwan treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Sotosaja-hwan-administered groups. Sotosaja-hwan inhibited the generation of ${\cdot}O_2$ in the kidney. Finally, MDA+HAE levels was increased and GSH/GSSG ratio was decreased in the ob/ob mice, whereas those were improved in the Sotosaja-hwan-administered groups. Sotosaja-hwan showed the antidiabetic and anti hyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging reactive oxygen species and reducing the MDA+HAE levels in the ob/ob mice.
In this study, the relationships between sensitivity to oxyfluorfen, absorption of the herbicide, protoporphyrin IX(Proto IX) accumulation and activities of antioxidative enzymes were examined to identify the tolerance mechanism against oxyfluorfen in various rice cultivars having different level of tolerance to this herbicide. Absorption of oxyfluorfen in tolerant rice cultivars was slower than in susceptible cultivars. Proto IX accumulation in various rice cultivars treated with oxyfluorfen was higher in susceptible cultivars than in tolerant ones. In susceptible cultivars especially, Proto IX accumlated rapidly during the herbicide treatment in the dark. Large amounts of Proto IX accumulation were considered to cause membrane lipid peroxidation in the light. However, among the tested rice cultivars, there was little relationship between their tolerance to oxyfluorfen and the activities of antioxidative enzymes. Therefore, it is assumed that differential susceptibility of rice cultivars to oxyfluorfen was due to difference in their capability to absorb the herbicide and to subsequently accumulate Proto IX.
To study whether the secondary toxic substances such as ethylene and reactive oxygen species(ROS) are induced by air pollutants in foliage plants, $SO_2$ was fumigated to Pachira aquatica, Spathiphyllum patinii, and Hedera helix. $SO_2$ was controlled to $1\;{\mu}L/L$ and then fiumigated to plants for 2 days(8 hrs/day). It resulted in visible injury in P. aquatica and H. helix while no symptom appeared in S. patinii. Photosynthetic rate and water use efficiency were most remarkably reduced in P. aquatica compared to other two species whereas least in S. patinii. Two days after $SO_2$ fumigation, ethylene evolution was quantified to 23.56, 10.43 and 4.79 nL/g/h in P. aquatica, H. helix and S patinii, respectively. On the other hand, antioxidative enzymes were clearly activated by $SO_2$ treatment in all tested plant species implying ROS production. In conclusion, we could suggest that ethylene and ROS have been intimately related to the defense mechanism against $SO_2$ and their induction degree increased with plant susceptibility to $SO_2$. Furthermore, it was found that S. patinii was tolerant and P. aquatica sensitive to $SO_2$ on the basis of antioxidative enzyme activity and ethylene evolution.

Chemical scavenging of reactive oxygen species (ROS) is considered a major mechanism of antioxidant effects, but preventing ROS generation can be more efficient in attenuating oxidative damage. In this study, the extracts of plants, Solanum lycopersicum, Ailanthus altissima, Equisetum arvense, and Oenothera biennis, were tested to determine whether their antioxidative effects are driven by the prevention of superoxide generation from mitochondria, a major ROS generator. While all the extracts efficiently attenuated the elevation of ROS levels in human fibroblasts and inflammation-induced mice, those of S. lycopersicum, A. altissima, and O. laciniata only suppressed mitochondrial ROS generation and reduced levels of lipofuscin and lipid peroxidation. Furthermore, the extracts of A. altissima and O. laciniata extended the lifespan of fruit flies. Our results suggest that plant extracts with anti-oxidative effects differ in their ability to prevent ROS generation, which may be associated with the attenuation of oxidative damage in cells and animal tissues.
G. japonicum is a perennial hem and the flowering plant has been used as a diuretic and an astringent in Japan and China. However, little information is available about the anti-inflammatory action of G. japonicum. Therefore, the objective of this study was to investigate the antiinflammatory action of fractions from G. japonicum methanol extract. Inhibition of NO production was observed when cells were cotreated with fractions of G. japonicum and lipopolysaccharide. We observed that ethyl acetate fraction of G. japonicum inhibited NO production by LPS-activated RAW 264.7 cells, and that the suppression induced by ethyl acetate fraction of G. japonicum was associated with antioxidant activity and direct NO clearance. In addition, only ethyl acetate fraction of G. japonicum inhibited stimulated $PGE_2,\;TNF-\alpha,\;IL-1\beta$ production, whereas water and methyl chloride fractions showed no such effects. The ethyl acetate fraction of G. japonicum methanol extract showed a remarkable scavenging activity on the 1,1-diphenyl-2 picrylhydrazyl radical. Based on the results, ethyl acetate fraction of G. japonicum may be useful source as natural antioxidants and antiinflammation. Therefore, the results obtained from this study provide an alternative protective mechanism of ethyl acetate fraction of G. japonicum and provide information on the potential use of ethyl acetate fraction of G. japonicum in chemoprevention or pathogenic conditions related to overproduction of NO and $PGE_2$. However, the mechanism of the inflammatory effect must be evaluated through various parameters for induction of NO production.
