Functional Properties of Egg Shell Membrane Hydrolysate as a Food Material (난각막 분해물의 식품 소재로서 기능적 특성)

  • The functional properties of egg shell membrane hydrolysate by Bacillus licheniformis(EESMH) and NaOH-ethanol(AESMH) as a food material were investigated.. The yield of egg shell membrane hydrolysate was about 15% by Bacillus licheniformis, whereas that was 70% by NaOH-ethanol. Histidine content was higher in EESMH (18.69%) than in AESMH (2.56%). Both EESMH and AESMH showed high protein solubility (>95%). Emulsi-fying activity and stability of EESMH were higher than those of AESMH. foaming capacity and stability of AESMH were 2 times higher than those of EESMH in the pH ranges from 2 to 12. The AESMH had antioxidative activity whereas EESMH had not. Therefore, both AESMH and EESMH can be used for industrial food materials from the results of functional properties.

Effects of the Antioxidative Components to Ginsenoside in the Liver of 40-week-old Mice (40주령의 생쥐 간에 미치는 ginsenoside의 항산화효과)

  • To study on antioxidant effects of saponin fractions, we investigated effects in the liver of 40-week-old mice to which were pretreated with 5 mg/kg per body weight of saponins for 5 days. The ability of saponins to protect against oxidative damage to the mouse liver was examined by determining the level malondialdehyde (MDA),hydrogen peroxide and the activity of superoxide dismutase (SOD) and catalase (CAT). The only panaxadiol (PD)among the ginseng saponin fractions significantly increased the hepatic SOD activities (p<0.01), Whereas PD, panaxatriol (PT), ginsenoside Rd (G-Rd) (p<0.01) and ginsenoside Re (G-Re) (p<0.05) significantly decreased the contents of hydrogen peroxide. It was only G-Rd that significantly increased CAT activities (p<0.05). The level of MDA was significantly decreased by G-Rd and PD. In conclusion, PD and G-Rd among the saponin fractions were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation.

Study on the Protective Effect of Corni Fructus against Free Radical Mediated Liver Damage (산수유의 유리자유기에 의한 간손상 보호효과)

  • Carbon tetrachloride ($CCl_4$)-induced liver injury depends on a toxic agent that has to be metabolized by the liver NAPDH-cytochrome P450 enzyme system to a highly reactive intermediate. Alternations in the activity of cytochrome P450 enzymes affect the susceptibility to hepatic injury from $CCl_4$. In this study, we evaluated the potential protective activity of the traditional Korean medicinal herb, Corni fructus (CF), against an experimental model of hepatotoxicity induced by $CCl_4$. The CF exhibited a hepatoprotective activity against $CCl_4-induced$ liver damage in Sprague-Dawley (SD) rats, as measured by GOT, GPT, ALP and histological observation. The CF also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. In addition, the expression of CYP2E1, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, was significantly decreased in the liver of CF treated SD rats. But $CCl_4$ and CF has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of CF possibly related to antioxidative effects and regulation of CYP2E1 expression.

Luteolin and luteolin-7-O-glucoside protect against acute liver injury through regulation of inflammatory mediators and antioxidative enzymes in GalN/LPS-induced hepatitic ICR mice

  • BACKGROUND/OBJECTIVES: Anti-inflammatory and antioxidative activities of luteolin and luteolin-7-O-glucoside were compared in galactosamine (GalN)/lipopolysaccharide (LPS)-induced hepatitic ICR mice. MATERIALS/METHODS: Male ICR mice (6 weeks old) were divided into 4 groups: normal control, GalN/LPS, luteolin, and luteolin-7-O-glucoside groups. The latter two groups were administered luteolin or luteolin-7-O-glucoside (50 mg/kg BW) daily by gavage for 3 weeks after which hepatitis was induced by intraperitoneal injection of GalN and LPS (1 g/kg BW and $10{\mu}g/kg\;BW$, respectively). RESULTS: GalN/LPS produced acute hepatic injury by a sharp increase in serum AST, ALT, and $TNF-{\alpha}$ levels, increases that were ameliorated in the experimental groups. In addition, markedly increased expressions of cyclooxygenase (COX)-2 and its transcription factors, nuclear factor $(NF)-{\kappa}B$ and activator protein (AP)-1, were also significantly attenuated in the experimental groups. Compared to luteolin-7-O-glucoside, luteolin more potently ameliorated the levels of inflammatory mediators. Phase II enzymes levels and NF-E2 p45-related factor (Nrf)-2 activation that were decreased by GalN/LPS were increased by luteolin and luteolin-7-O-glucoside administration. In addition, compared to luteolin, luteolin-7-O-glucoside acted as a more potent inducer of changes in phase II enzymes. Liver histopathology results were consistent with the mediator and enzyme results. CONCLUSION: Luteolin and luteolin-7-O-glucoside protect against GalN/LPS-induced hepatotoxicity through the regulation of inflammatory mediators and phase II enzymes.

Evaluation on antioxidative and antiaging effect of immature fruits from Citrus unshiu Markovich (미숙감귤의 항산화 및 항노화 활성에 대한 평가)

  • Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.

Protective effects of mealworm (Tenebrio molitor) extract on N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced cellular toxicity in SH-SY5Y neuroblastoma cells (SH-SY5Y 인간 신경모세포종 세포에서 MPTP 유발 세포 독성에 대한 거저리(Tenebrio molitor) 추출물의 보호효과)

  • Purpose: Edible insect extracts have been used as an alternative source for medicinal supplements due to their significant antioxidative and anti-inflammatory activity. Recent studies have reported that anti-microbial peptides from insects have neuroprotective effects on dopamine toxins. The purpose of this study was to investigate the protective functions of mealworm (Tenebrio molitor) extract (MWE) on N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced cellular toxicity in SH-SY5Y neuroblastoma cells. Methods: Cellular toxicity induced by the MPTP toxin and the impact of MWE on cell survival were analyzed using MTT assays. DAPI staining was performed to observe apoptotic phenomena caused by MPTP. Changes in caspase-3 activity and protein expression were observed using enzyme activity assays and western blot assays, respectively. Results: MWE exerted significant antioxidant activity, which was measured by both DPPH and ABTS radical assays, with a dose-dependent relationship. Furthermore, MWE resulted in cellular proliferation in SHSY5Y cells in a dose-dependent manner. Furthermore, MWE pretreatment significantly inhibited MPTP-induced cytotoxicity, with a dose-dependent relationship. The morphological characteristics of apoptosis and increased reactive oxygen species induced by MPTP were also significantly reduced by MWE pretreatment. Conclusion: MWE treatment significantly attenuated MPTP-induced changes in the levels of proteins associated with apoptosis, such as caspase-3 and PARP. These findings suggest that MWE exerts neuroprotective effects on human neuroblastoma SH-SY5Y cells subject to MPTP-induced dopaminergic neurodegeneration.

Inhibitory Effect against Angiotensin Converting Enzyme and Antioxidant Activity of Panax ginseng C. A. Meyer Extracts (인삼 추출물의 Angiotensin Converting Enzyme 저해 효과와 항산화 활성)

  • The study was performed for elucidating angiotensin converting enzyme (ACE) inhibitory activity and comparing antioxidative activity of Panax ginseng extracts prepared at different conditions. Total phenolic content, inhibitory activity on ACE and antioxidative effects were tested on 10 ethanolic extracts and correlation coefficient between total phenolic content and physiological activity was calculated. Yield and total phenolic content of 50% ethanolic extract prepared at $85^{\circ}C$ exhibited the highest value as 42.52% and 0.82%, respectively. Among the fractions obtained from 50% ethanolic extract prepared at room temperature, water fraction showed the highest value in yield as 72.08% and ethyl acetate fraction did in total phenolic content as 6.59%. In the test on ACE inhibitory activity, 50% ethanolic extract obtained at room temperature indicated the strongest effect of 93.8% which was higher than 85.2% of commercialized ACE inhibitor and solvent fractions showed potent inhibitory activity in order of hexane fraction, diethyl ether fraction, ethyl acetate fraction, butanol fraction and water fraction at concentration of $4000{\mu}g/ml$. 50% Ethanolic extract prepared at $85^{\circ}C$ had the most potent inhibition effect on human LDL oxidation as 78.2% at $200{\mu}g/ml$ and the other extracts also did above 60%. Diethyl ether fraction and ethyl acetate fraction showed strong inhibition activity $(34.38%{\sim}78.13%)$ on LDL oxidation at concentration of $10{\sim}200\;{\mu}g/ml$. From the statistical analysis via SAS program, correlation coefficient between total phenolic content and ACE inhibitory effect was 0.6353 at P<0.05. Conclusively, this report showed that the most efficient extraction condition for elevating inhibitory activity on ACE and LDL oxidation, phenolic content and yield from Panax ginseng was 50% ethanol extraction at room temperature or high temperature condition. And Panax ginseng would be used for preventing hypertension or atheroscrelosis for man via inhibitory action on ACE and LDL oxidation.

Isolation and Characterization of Lactic Acid Bacteria with Angiotensin-Converting Enzyme Inhibitory and Antioxidative Activities (안지오텐신 전환효소 저해 활성 및 항산화 활성을 가진 젖산균의 분리 및 특성)

  • In this study, we isolated and characterized plant-associated lactic acid bacteria which are able to produce angiotensin-converting enzyme (ACE) inhibitory and antioxidative activities. Five lactic acid bacteria were isolated from plants (grape and leek), a plant-associated fermentative product (Kimchi) and Korean traditional alcohol (Dongdongju). Strains K-1 and K-21 from Kimchi, strain L-5 from leek, strain G-3 from grape, and strain D-3 from Dongdongju were identified as Pediococcus pentosaceus, Lactobacillus plantarum, Weissella cibaria, L. plantarum, and L. brevis, respectively, by 16S rRNA gene analysis. ACE inhibitory activities of isolated strains ranged from 44.3 to 71.9% in the MRS broth. G-3, L-5 and K-1 strains especially showed high ACE inhibitory activities (59.8-98.69%) in the MRS broth containing skim milk. DPPH radical scavenging activities of the strains were in the range of 42.5-82.7%. All strains showed varying levels of resistance in artificial gastric fluid (pH 2.5), retaining viability ranging from 42.2 to 88.1% after 3 hr of incubation. All strains showed high resistance to 0.3% oxgall after 24 hr of incubation; survival rates were in the range of 55.4-112.8%. Isolated strains were found to be antagonistic to some pathogens including Pseudomonas aeruginosa.

Physiological Damages and Biochemical Alleviation to Ozone Toxicity in Five Species of genus Acer

  • We investigated physiological damages and biochemical alleviation of five species of genus Acer under ozone fumigation in order to assess their tolerant ability against ozone toxicity. At the end of 150 ppb $O_3$ fumigation, photosynthetic characteristics were measured, and chlorophyll contents, malondialdehyde (MDA) and antioxidative enzyme activities were analyzed in the leaves of five maple trees (Acer buergerianum, A. ginnala, A. mono, A. palmatum, and A. palmatum var. sanguineum). The reduction of chlorophyll (chl) a in ozone-exposed plants was 16.8% (A. buergerianum) to 26.7% (A. ginnala) of control plants. For the content of chi b, A. ginnala and A. palmatum var. sanguineum represented the high reduction of 26.3% and 23.6%, respectively. The highest reduction on the chi a:b ratio was observed in the leaves of A. palmatum. The reduction of net photosynthesis in five species varied from 2.4% to 37.6%. Among five species, A. ginnala showed remarkable reduction (37.6%) for net photosynthesis in comparison with control. Carboxylation efficiency differed significantly (P < 0.05) among species and between control and ozone treatment. The reduction of carboxylation efficiency was the highest in the leaves of A. ginnala (44.7%). A. palmatum var. sanguineum showed the highest increase (41.7%) for MDA content. The highest increase of superoxide dismutase (SOD) activity represented in A. palmatum (26.1%) and the increase of ascorbate peroxidase (APX) activity ranged from 16.5% (A. ginnala) to 49.1% (A. palmatum var. sanguineum). A. mono showed the highest increase (376.6%) of glutathione reductase (GR) activity under ozone fumigation and A. buergerianum also represented high increase (42.3%) of GR activity. Catalse (CAT) activity increased in the leaves of A. ginnala, A. palmatun and A. palmatum var. sanguineum under ozone exposure, whereas A. buergerianum and A. mono decreased in comparison with control plants. In conclusion, physiological markers such as chlorophyll content and photosynthesis that responded sensitively to $O_3$ in maple trees were considered as the very important indicators in order to evaluate the tolerance against $O_3$ stress, and parameters were closely related with each other. Among anti oxidative enzymes, SOD and APX might be contributed to alleviate to $O_3$ toxicity through the increase of activity in all maple trees. Therefore, these compounds can be used as a biochemical maker to assess the stress tolerance to $O_3$.

Comparison of Biological Activities of Epimedium koreanum Nakai Produced in Korea and China (국내산과 중국산 삼지구엽초의 생리활성 비교)

  • This study was carried out to compare the biological activities of Epjmedium koreanum Nakai grown wild in Korea and China. The antioxidative effect of E. koreanum Nakai-extracts grown wild in Gangwondo was 78%, which was higher than that in China as 71%. The inhibition ratio of growing cancer cells was estimated as 89, 91 and 86% for human liver, lung and breast cancer cell lines, respectively in adding 0.1 g/l of ethanol extracts of E. koreanum grown in Korea. The hepatoprotective effect and Angiotensin Converting Enzyme (ACE) inhibiting effect of Korean one were also observed as 124 and 87%, respectively, which were also higher than those produced in China. There was not much difference between Korean and Chinese one in inhibiting ${\alpha}-glucosidase$ activities in all ranges of supplement. It was proved that most effective extraction solvent was mixed type as water and ethanol (1:1, v/v) to have higher biological activities from both Korean and Chinese ones.
