Melatonin Enhances Hepatic Glutathione-peroxidase Activity in Sprague-Dawley Rats

  • Effects of melatonin on hepatic glutathione-peroxidase (GSH-Px) and glutathione-reductase (GSH-reductase) activities were studied in Sprague-Dawley (SD) rats administered i.p. (10 mg/kg body weight) with melatonin during 15 days. The activity of cytosolic GSH-reductase in the liver was not changed by melatonin. However, melatonin injection increased significantly the activity of liver cytosolic GSH-Px activity compared with those in saline-treated rats. At the same time, plasma GSH-Px was also increased significantly in melatonin-treated rats. Since GSH-Px, a major antioxidative enzyme, removes $H_2O_2$ and lipid peroxides which are formed during lipid peroxidation from cellular membrane, such elevation of heptatic GSH-Px activity may contribute to the improvement of antioxidative effects under oxidative damage in the liver.

  • Although the types of stress are various, environmental stresses generally increase the amounts of reactive oxygen species in plants. These reactive oxygen species stimulate stress-ethylene synthesis and accelerate senescence of plants. However, when stress-ethylene synthesis is suppressed through antioxidative enzymes and antioxidants, the resistance of plants against stress could be induced by limited production of ethylene.

Effects of Antioxidants on UV-B Susceptibility in Soybean (항산화 물질이 UV-B에 대한 콩의 감수성에 미치는 영향)

  • To determine whether the enhanced UV-B causes oxidative stress, and to test the relationship between plant growth response and biochemical defense response to UV-B, two soybean plants, Keunolkong, a highly UV-B susceptible cultivar, and Danyeubkong, a less UV-B susceptible cultivar, were subjected to the enhanced UV-B [daily dose : 0.06 (control) and 11.32 (enhanced UV-B) kJ $m^{-2}$ ; $UV-B_{BE}$] for 3 weeks. Contents of malondialdehyde and total carotenold were increased in Keunolkong compared with Danyeubkong by UV-B. In control plants, ascorbate level of Danyeubkong was 3 times higher than that of Keunolkong. The ratio of dehydroascorbate/ascorbate was highly increased in Keunolkong by UV-B . The activities of antioxidative enzyme such as superoxide dismutase, ascorbate peroxidase, monodehydroascorbate reductase and glutathione reductase were increased in both cultivars by UV-B. This results indicate that enhanced UV-B caused oxidative stress in both two cultivars, especially in Keunolkong. Susceptibility of two soybean cultivars to UV-B is closely related to the levels of antioxidants such as carotenoid and ascorbate.

  • Adriamycin is a major cancer chemotherapeutic agent against a me range of human neoplasms. However, its clinical application is limited since It has a variety of side effects, bone marrow suppression and cardiotoxity, and this toxicity appears by free radical. This study investigated the effect of Ampelopsis radix on the toxicity of adriamycin. The methanol fraction reduced slightly adriamycin induced lipid peroxidation and superoxide production at the dose of 50mg /kg. 1.p., respectively. During the adriamycin administration. Protein bound-SH, nonprotein bound-SH, and glutathione-5-transferase did not change, but methanol fraction treated group were markedly increase. These results indicated that Ampelopsis radix has a major influence on the thiol group and related enzyme activity on the antioxidative effects.

  • Oddi is a mulberry fruit that has anti-inflammatory and antioxidative effects. This study was carried out to investigate the quality characteristics of mulberry fruit wines manufactured by two different methods. Where they were processed according to the traditional Korean fruit wine methodology(A) or a modified(B). Sensory qualities, physiochemical characteristics, and biochemical activities of the Oddi wines were analyzed. From the results of the sensory evaluation, manufacturing method A showed higher acceptability. However, the two different methods was not significantly different overall. The method A wine had higher brightness(L)(77.65) redness(a), and yellowness(b). As for the antioxidant activities and ACE(angiotensin-converting enzyme) inhibitory abilities of the wines manufacturing method A had greater values.

Effects of Antioxidant Nutrient Supplementation on the Lipid Peroxidation and Antioxidative Enzyme Activities in Patients with Coronary Heart Disease

  • This study was carried out to evaluate whether antioxidant nutrient suppplementation with $\alpha$-tocopherol, vitamin C, $\beta$-carotene, and selenium reduces the lipid peroxide levels and increases the antioxidative enzyme activities in patients with coronary hart disease. Eighty nine patients participated in a randomized, double-blind, placebo-controlled trial. The antioxidant group (45 patients) was given daily doses of $\alpha$-tocopherol (400 IU), vitamin C (50 mg), $\beta$-carotene (15 mg), and selenium (50 $\mu\textrm{g}$) and forty four patients received a placebo. Thirty eight subjects (84.4%) of the antioxidant group and thirty nine subjects (88.6%) of the placebo group completed the three-month supplementation. Serum levels of tocopherol, vitamin C and $\beta$-carotene significantly increased in the antioxidant group compared with the baseline (p<0.05). Thiobarbituric acid-reactive substances(TBARS) decreased significantly (0.6 nmol MDA/mL) in the antioxidant group compared with that (0.09 nmol MDA/mL) in the placebo group (p=0.03). However, antioxidant supplementation did not affect the level of oxidized-LDL measured as autoantibodies against oxidized-LDL. The superoxide dimutase activity in red blood cells increased in the antioxidant group compared with the baseline (p<0.05). However, glutathione peroxidase activities did not change after supplementation in both groups, and catalase activity significantly decreased in the placebo group (p<0.05). These results suggest that antioxidant supplementation for 3 months with $\alpha$-tocopherol, vitamin C, $\beta$-carotene and selenium in patients with coronary heat disease may be partially protective against oxidative stress.

Physicochemical Property and Antioxidative Activity of Hot-Water Extracts from Enzyme Hydrolysate of Astragalus membranaceus (황기 효소분해물 열수추출액의 이화학적 특성 및 항산화 활성)

  • To enhance the yield and bioactivity of hot-water extract from herbal medicine, Astragalus membranaceus was hydrolyzed with carbohydrases, such as ClariSEB and Fungamyl. After hot-water extracts were prepared from each hydrolysate (HW-C/F), physicochemical property, antioxidant activity and sensory property were evaluated. The solid content ($^{\circ}Brix$) of HW-C/F was higher than hot-water extract from A. membranaceus no treated enzyme (control). Although pH of HW-C/F was lower than that of the control, the acidity was higher. Lightness of Hunter's color values was increased in HW-C/F whereas redness and yellowness were decreased. The contents of reducing sugar, flavonoid and polyphenol of HW-C/F were higher than the control but the content of ascorbic acid was not different from control. The inhibitory activity of HW-C/F against lipid peroxidation was slightly higher than control, but DPPH radical scavenging, ABTS reducing, metal chelating activities were significantly increased by HW-C/F. The sensory evaluation also revealed that the sensory panelists preferred HW-C/F to that of control. Therefore, hydrolysis by carbohydrases for preparation of hot-water extract from A. membranaceus is one of the good methods to improve antioxidative activity and sensory property of hot-water extract.

Effect of Inclusion of Chitosan-Oligosaccahariede in Drinking Water on the Blood Component Profile, Immunity and Antioxidative Enzyme in Broiler Chickens (키토산 올리고당의 음수내 급여가 육계의 혈액성상, 면역력 및 항산화 효소계에 미치는 영향)

  • The study was conducted to the effect of inclusion of chitosan-oligosaccharide in drinking water on the blood component profile, immunity and antioxidative enzyme of broiler chickens. A total of 28,000 broiler chickens (Arbor Acre) at 1 days of age were fed the commercial diet until 35 days of age, the treatment divided into two treatments, normal control group and chitosan-oligosaccharide in drinking water group. In concentration of glucose, treatment was significantly decreased than control (P<0.05), but not statistically different on the Triglyceride, Total cholesterol, HDL-cholesterol and LDL-cholesterol in the Blood component profile of broiler chicks. The concentration of glutamic oxalacetic transaminase (GOT) was statistically different but the concentration of glutamic pyruvate transaminase (GPT) was significantly decreased in treatment fed chitosan-oligosaccharide than control (P<0.05). Immunoglobullins in the blood, concentration of IgG was not significantly different among control and treatment but concentration of IgM was significantly increased in fed chitosan-oligosaccharide than control (P<0.05). Antioxidant and super oxide dismutase (SOD) was not different among control and treatment and concentration of catalase and glutathione peroxidase were significantly increased in fed chitosan-oligosaccharide than control (P<0.05).

Antioxidative activity, including Inhibitory activities of ACE, APN and $\alpha$-amylase, in Theaceae Plants Native to Jeju Island (제주도 자생 차나무과 식물의 ACE, APN, $\alpha$-amylase 저해 활성 및 항산화활성에 대한 연구)

  • Antioxidative activity, including inhibitory activities of angiotensin I converting enzyme(ACE), aminopeptidase N(APN) and $\alpha$-amylase, was investigated in the methanol extracts from Theaceae plants native to Jeju island, in order to select the plant species containing bioactive materials for functional food or medicines. ACE inhibitory activity was above 50% in Ternstroemia japonica(stem bark) and Cleyera japonica(leaf), and APN inhibitory activity was low to be positive only in C. japonica(leaf, stem bark) and T. japonica(stem bark). $\alpha$-Amylase inhibitory activity was above 30% in Camellia japonica(fruit), Eurya emarginata(stem), T. japonica(stem bark) and Thea sinensis(stem). The antioxidative activity, estimated by the DPPH radical scavenging capacity, was above 30% in C. japonica(stem bark), T. japonica(stem bark) and T. sinensis(leaf). Particularly, the antioxidative activity analyzed by dot-blot test was very high in C. japonica(stem bark) relatively to those of other plants, and remained high in the low concentration($1.25\;{\mu}g/m{\ell}$). From the TLC analysis of antioxidative compounds, EGC(Rf 0.26) was found to have high activity in stem bark of C. japonica and EGCG(Rf 0.09) was found to have high activity in stem bark of C. japonica, E. emarginata, and T. japonica. Five bands (Rf 0.54, 0.46,0.44, 0.16, 0.03) which were not identified as compared with catechins were detected as polyphenolic compounds on the TLC plates sprayed with the Folin-Ciocalteu solution or the Ferric chloride-alcohol solution. These results suggests that Theaceae plants except E. japonica could be potentially used as a resource of bioactive materials for functional foods or medicines and further research is reguired to identify the bioactive substances and determine the functions of them.

Effects of Salvia plebeia R. Br. on Antioxidative Enzyme Activities and Oxidative Damage in Rats Fed High-Fat and High-Cholesterol Diets (곰보배추섭취가 고지방과 고콜레스테롤 식이 랫드에서의 항산화 효소활성 및 산화적 손상에 미치는 영향)

  • The purpose of the present study was to investigate the effect of Salvia Plebia R. Br. (SP) powder on the antioxidative defense system and oxidative stress in rats which were fed a high fat high cholesterol diet. Accordingly, the rats were divided into four experimental groups which were composed of a high fat high cholesterol diet group (HF), HF diet with 5% SP powder supplemented group (PA), a HF diet with 10% SP powder supplemented group (PB), and a normal group (N). Consequently, the hepatic catalase activity of the HF group was decreased compared to the normal group (N), but it is recorded that of the PA and PB groups were significantly increased. With this in mind, the PA and PB groups resulted in the case of significantly increased activities of hepatic GSH-px and SOD. The hepatic superoxide radical and hydrogen peroxide contents of the PA and PB groups were significantly decreased, as compared to the HF group. The GOT and GPT activities of the PB group were also significantly decreased when thus compared to the HF group. Notably, the carbonyl values contents of the PA and PB groups were significantly reduced compared to the HF group. The hepatic TBARS values in the liver were significantly reduced as measured in the PA and PB groups. These results suggest that the SP powder may reduce the incidence of oxidative damage, by the activation of an antioxidative enzyme in rats fed with high fat high cholesterol diets.
