한국산 옻나무로부터 추출.분리한 생리활성 물질들의 항산화 효과 및 세포독성 (Antioxidative and Cytotoxicity Activities of Compounds Isolated from Korean Rhus verniciflua S.)

  • 한국산 옻나무의 메탄올추출물과 유기용매 분획의 항산화 효과를 조사하였다. 항산화 효과는 수소공여 억제능, 과산화지질 형성 억제능, xanthine oxidase저해 활성과 아질산염 소거능으로 측정한 결과, ethyl acetate 분획이 가장 뛰어난 효과를 나타내었다. 이 분획의 생리활성 물질을 분리하기 위하여, rotatory locular counter current chromatography(RLCCC), Sephadex LH-20 column chromatograpy와 HPLC등의 방법을 사용하여, 1,2,3-trihydroxybenzene, methyl 3,4,5-trihydroxybenzoat와 3,4,5-trihydroxybenzoic acid를 분리하였다. 이중 methyl 3,4,5-trihydroxybenzoate는 butylated hydroxyanisole(BHA)이나 butylated hydroxytoluene(BHT)보다도 강한 항산화 효과를 나타내었다. 그러나 세포독성 실험에서는 이들 물질들이 암세포주에 대하여 활성이 높지 않았다. 이상의 결과로 methyl 3,4,5-trihydroxybenzoate은 뛰어난 항산화 효과를 지닌 식품첨가물로서의 이용 가능성을 제시할 수 있다.

Optimization of Alcalase for Krill Byproduct Hydrolysis and Antioxidative Activities by Response Surface Methodology

  • Krill byproduct was hydrolyzed with Alcalase 2.4L to produce functional ingredients for high antioxidative activities against 1,1-dimethyl-2-picryl-hydrazyl (DPPH) radical and Fe. The objective of this study was to investigate the optimum condition for degree of hydrolysis and antioxidative activity of enzymatic hydrolysate produced with the commercial Alcalase using response surface methodology (RSM) with a central composite rotatable design (CCRD). The ranges of independent variables were pH 7.6~10.4 for initial pH and $50.9{\sim}79.1^{\circ}C$ for hydrolysis temperature and their dependent variables were degree of hydrolysis, Brix, amount of phenolic compounds, DPPH-scavenging activity and Fe-chelating activity. RSM with CCRD was well designed to investigate the optimum condition for functional ingredients with high antioxidative activities using Alcalase 2.4L because of their high $R^2$ values of the range of 0.93~0.99 except the $R^2$ value of 0.50 for the amount of total phenolic compounds. The optimum hydrolysis conditions were pH 9.5 and $62^{\circ}C$ for degree of hydrolysis (DH) and pH 9.1 and $64^{\circ}C$ for DPPH-scavenging activity by response surface methodology. The yield of DH and DPPH-scavenging activity were $14.1{\pm}0.5%$ and $10.5{\pm}0.2%$, respectively. It is advantageous to determine the optimum hydrolysis conditions of krill and its by-products for the creation of different kinds of food products, as well as to increase the usage of marine protein sources.

갓김치의 발효과정 중 Chlorophylls 및 Carotenoids의 변화와 동획분의 항산화성 (Changes in Chlorophylls and Carotenoids of Mustard Leaf Kimchi during Fermentation and Their Antioxidative Activities on the Lipid Oxidation)

  • 갓김치의 발효과정 중 chlorophylls, chlorophyll de-rivatives 및 carotenoids의 함량 변화와 갓김치에서 얻어진 이의 조획분의 항산화성을 살펴보았다. 갓김치 발효기간에 따른 chlorophylls 및 carotenoids의 함량은 발효 7일 동안 chlorophylls가 pheophytins로 급격히 전환되었으며 chlorophylls와 함께 chlorophyllides는 높은 농도는 아니지 만 15일 까지는 존재하다가 그 이후 모두 pheophytins와 pheophorbides로 변했다. 갓김치 발효기간 동안 total chlorophylls에는 큰 변화없이 거의 일정하게 유지 되었으나 carotenoids는 발효 말기인 25일째에 초기 함량의 43.7%로 감소되었다. 갓김치에서 얻어진 chlorophylls와 carotenoids 조분획이 지방질의 자동산화에 미치는 영향을 조사하였을 때 $\alpha$-tocophe-rol 보다 높은 항산화성을 나타내었으며 발효기간에 따른 갓김치의 항산화성을 비교하였을 때 유의적으로 차 이는 없었다. 갓김치의 chlorophylls와 carotenoids 분획이 지방산의 산화에 대한 저해작용을 나타내는 동안 chlorophylls와 caratenoids의 감소율을 비교했을 때 총 chlorophylls가 총 caratenoids 보다 안정성이 높았다.

  • Phytochemical examination of Barks of Ulmus macrocarpa isolated two flavanone, three flavanonol, three flavan 3-ol and one procyanidin compounds. We also determinated the antioxidative activity of these compounds by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals. Three flavan 3-ol (catechin, epicatechin and catechin-7-O-$\beta$-O-xylopyranoside) and procyanidin B1 showed significant antioxidative activity. (omitted)

약용버섯의 생리활성 분석 (Analysis of Biological Activities of Medicinal Mushrooms)

  • This study was carried out to observe the antioxidative activity, fibrinolytic activity, nitrite savenging ability and adenosine content of three medicinal mushrooms. Among water and ethanol extract of each mushroom, water extract of Inonotus obliquus (Czech) showed the highest antioxidative activity $(57.1{\mu}g/ml)$, nitrite scavenging ability (52.04%), fibrinolytic activity (86.8%) and adenosin content $(94.3{\mu}g/g)$, whereas nitrite scavenging ability of ethanol extract of Phellinus linteus showed higher than that of water extract. Apart from the above statements, water was effective than ethanol as extracting solvent in general. These results suggest that Inonotus obliquus (Czech) showed higher activities such as antioxidative activity, nitrite scavenging ability, fibrinolytic activity and adenosin content and water was effective as solvent except for nitrite scavenging ability.

Antioxidative Activity of Urushiol Derivatives from the Sap of Lacquer Tree (Rhus vernicifera Stokes)

  • The authors isolated four olefinic catechols, commonly referred to as urushiol, from the sap of Korean lacquer tree(Rhus vernicifera STOKES) with stronger antioxidative activities than $\alpha-tocopherol$. The hexane extract with a free radical scavenging activity was purified by silica and ODS gel column chromatography. The active compounds were identified by MS and $^1H-NMR$ as 3-[8'(Z),11'(Z),14'-pentadecatrienyl]catechol, 3-[8'(Z),11'(Z)-pentadecadienyl]catechol, 3-[8'(Z)-pentadecenyl] catechol, and 3-pentadecylcatechol. All of these compounds showed strong free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, in which 3-pentadecylacatechol exhibited the highest activity ($IC_{50}$: $1.2{\mu}g/ml$). They also showed a significant inhibitory activity on lipid peroxidation ($IC_{50}$: 2.1 - 3.5 ${\mu}g/ml$). The antioxidative activity of 3-pentadecylcatechol on DPPH radical and lipid peroxidation is approximately two times greater than that of $\alpha$-tocopherol. The results suggest that the urushiol derivatices may contribute to the preservative characteristics effective against oxidative stress and could be a good source for industrial applications including a coating material.

Studies on the Biological Activity of Astragalus membranaceus Extracts

  • Physiological activities of hot water extract and solvent fractions isolated from Astragalus membranaceus were examined and the antioxidative, fibrinolytic, thrombin inhibitory and a-glucosidase inhibitory activity were measured. The hot water extract of Astragalus membranaceus was fractionated into hexane, chloroform, ethyl acetate, butanol and water fractions, and each of these fractions was individually assayed. The antioxidative activities of ethyl acetate and chloroform fractions were 89.96% and 87.36%, respectively. Using the fibrin plate method, only the hot water extract showed a plasmin activity of 0.41 units/ml. The thrombin inhibitory activity of the ethyl acetate fraction was the highest with a value of 82.73%. The hot water extract displayed a-glucosidase inhibitory activity of 64.91%. In conclusion, the hot water extract and the ethyl acetate fraction can be used as materials for the development of biofunctional foods to prevent cardiovascular diseases.

  • Changes of chemical and oxidative/antioxidative characteristics chlorophylls(CHLs) and their derivatives in the model system of mustard leaf kimchi(MLK) were investigated. During fermentation of MLK(at 15oC, for 25days, 2.3$\pm$ 0.1% salt content) pH and total acidity were decreased/increased from 5.6 and 0.4%(initial day) to 3.6 and 1.07%(final day) resceptively. Activities of lipoxygenase and peroxidase were decreased gradually, however, these of chlorophyllase was increased in the first 10 days of fermentation. CHLs of MLK in the initial stage of fermentation were degraded rapidly and all CHLs and chlorophyllides were converted to pheophytins and pheophorbides in the final stage. Deg radation effects of CHLs(a & b) and their derivatives(pheophytins a & b) fractionated from MLK and carotene on the autoxidation and lipoxygenase oxidation of linoleic acid were observed, and also stronger antioxidative activities of CHLs and pheophytins were shown in their autoxidation/enzymtic oxidation of linoleic acid.

  • Four diarylheptanoids oregonin, hirsutanonol, 1,7-bis- (3,4-dihydroxyphenyl)-heptane-5-O-$\beta$-D-xylopyanoside and 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-0-$\beta$-D-glucopyranoside isolated from Alnus hirsuta were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$and/or Fe$^{2+}$ to rat liver homogenate. All these diarylheptanoids showed significant inhibitory activities against the DPPH radical and demonstrated strong activities in preventing the lipid peroxidation induced by $H_2O$$_2$.>.

함초 첨가식이가 당뇨유발 흰쥐의 항산화효소 활성에 미치는 영향 (Effects of Salicornia Herbacea L. Supplementation on Antioxidative Enzyme Activities in Streptozotocin-Induced Diabetic Rats)

  • This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on hepatic antioxidative enzyme activities in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The activities of Xanthine oxidase (XOD), glutathione- S-transferase (GST), glutathione peroxidase (GPX), glutathione reductase (GR), superoxide dismutase (SOD) and CAT (CAT) were measured in the homogenates of liver. The activity of CAT was lower in the supplementary group with glasswort compare to the STZcontrol group but it was not significantly different. The activity of SOD was not significant in all of experimental groups. The activity of GR was significantly increased in the normal supplementary group with glasswort, and GPX activity was significantly increased in STZ-GW 10% group compare to the STZ-control group. The activity of XOD was significantly decreased in the all of supplementary groups with glasswort. The activity of GST was significantly increased in the N-GW 20% group and it was significantly decreased in the STZ-GW 20% group. These results show that the supplementation of glasswort may have favorable influence on antioxidative status in diabetic rats and it may be useful for the diabetic complications as functional food.
