A Study on the Pharmacetical Characteristics and Analysis of Green-tea Extract (녹차 추출물의 약리적 특성 및 분석)

  • From the result of pharmacetical characteristics and analysis of Green-tea extract, it could obtain some conclusions as follows. The extract experiment of Green-tea appeared about 10%-extraction ratio as semi-solid state, and after dried in freezing from Green-tea extract of semi-solid state, it obtained about 65% Green-tea extract as solid state. In the results on antimicrobial experiment of Green-tea extract, number of S-typhimurium and fungus in microbe decreased more and more according to the time passage. This phenomenon could show that Green-tea extract keeps antimicrobial effect. In the results on antioxidation experiment of Green-tea extract, DPPH scavenging activity of free radical showed that Green-tea extract appears more remarkable reduction ability than reference samples. This phenomenon means that antioxidation of Green-tea extract appears higher than Vitamin-C and BHA sample. In the results on intrument analysis, the fatty and aromatic components of aniline, acetaldehyde, acetic acid, trichloroethylene, caffeine etcs from Green-tea extract was detected with GC/MS analysis and inorganic components of Ca, Mg, Cu, Mo, Sb, Ti etcs from Green-tea extract was detected with ICP/OES analysis.

Effects of Ligustrum lucidum Fruits on Growth Performance, Antioxidation and Meat Quality in Arbor Acres Broilers

  • This study was conducted to evaluate the effects of Ligustrum lucidum (LL) on growth performance, antioxidation, and meat quality in broilers. 270 birds (1 d old) were allotted to 3 treatments with 6 replicates per treatment. The feeding program included a starter diet from d 1 to 21 of age and a grower diet from d 22 to 42 of age. The birds were given a basal corn-soybean meal diet supplemented with 0 (the control), 5 or 10 g of LL/kg, respectively. The results showed that in the starter, grower and overall phase, broilers fed with 5 or 10 g of LL/kg had better (p<0.01) average daily gain, but there were no differences in feed: gain between treatments. In the starter and overall phase, average daily feed intake of LL groups was greater (p<0.05) than that of the control, but this difference was not observed in the grower phase. Supplementation of LL significantly increased (p<0.05) superoxide dismutase activity and total antioxidant capability in serum of chickens. In breast muscle, birds fed 5 g of LL/kg had an increase (p<0.05) in superoxide dismutase activity. The LL supplementation significantly decreased (p<0.05) malondialdehyde contents. Adding 5 or 10 g of LL/kg to the diet significantly increased pH value and reduced drip loss of meat (p<0.05). The results of this study indicated that dietary LL could improve growth performance, increase pH value and reduce drip loss of meat by decreasing lipid peroxidation and by improving antioxidative status in broilers.

Antioxidative Properties of Amaranth Cauline Leaf and Suppressive Effect against CT-26 Cell Proliferation of the Sausage Containing the Leaf

  • The study investigated antioxidative properties and rectal cancer cell inhibition effect of amaranth (Amaranthus cruentus L.) cauline leaves (ACL) to produce the sausage with ACL powder (ACLP). Antioxidative effects of ACLP prepared with different stem lengths (10-45 cm) were evaluated through DPPH, ABTS, reducing power, total phenol, and total flavonoid. Inhibition effect on rectal cancer cells growth was also examined with CT-26 cell. To determine appropriate ACL amounts in sausage formula, response surface methodology was used. The sausages without ACL (control) and the sausage with ACL (ACLP sausage) were the subjected to the examinations of antioxidation, growth inhibition on CT-26, and physicochemical properties (pH and water content). ACLP made from the leaf with 15 cm length stem generally showed the highest antioxidative effect through results of DPPH, ABTS, reducing power, total phenol, and total flavonoid. ACLP also showed inhibition effect on the proliferation of CT-26, depending on concentration of ACLP. The surface response model showed that 4.87 g of ACLP was optimized amount for sausage production. Physicochemical properties between optimized ACLP and control sausages were not significantly different. Higher antioxidative effect of optimized ACLP sausage extract was observed (p<0.05) in antioxidation tests than control sausage extract except for DPPH. Cell viability of CT-26 cells were higher (p<0.05) in ACLP than in control sausage extracts. These results indicate that ACLP has functional effects on antioxidation activity and growth inhibition on CT-26 cell, and thus, it should be useful as a supplement in sausage, which may some effect as ACLP itself.

The Effects of Jehasuogamibang on Antioxidation Activity and Dietary Hyperlipidemia-induced Mice (제하수오가미방(製何首烏加味方)이 고지혈증 및 산화적 손상에 미치는 영향)

  • Objectives: This experiments were performed to determine the effects of the Jehasuogamibang (製何首烏加味方: JHGB) on antioxidationactivity and hyperlipidemia induced by a hypercholesterolemic diet in mice. Methods: After treatment with JHGB expert safety in cytotoxicity and toxicity of Human fibroblast cells and liver and kidney, effect on Reactive Oxygen Species, serum total cholesterol, LDL-cholesterol, triglyceride, glucose, HDL-cholesterol, lipid peroxid of liver tissue, significantly increased SOD and catalase. Results: 1. JHGB showed safety in cytotoxicity and toxicity of human fibroblast cells and liver and kidney. 2. JHGB showed significant inhibitory effect on reactive oxygen species. 3. JHGB significantly decreased serum total cholesterol, LDL-cholesterol, triglyceride, and glucose, and significantly increased serum HDL-cholesterol. 4. JHGB significantly decreased lipid peroxide of liver tissue and significantly increased SOD and catalase. Conclusions: These results suggest that Jehasuogamibang is effective in antioxidationactivity and dietary hyperlipidemia-induced mice.

The Effect of Samchoolgunbitanggamibang of the growth of rats (삼출건비탕가미방(蔘出健脾湯加味方)이 흰쥐의 성장에 미치는 영향)

  • Objectives : The purpose of this study is to find out the effect of antioxidation related to aging of Cheongeumyeonsudan which is written on Dongui-bogam experimentally. Method : 14 weeks aged SD albino rats were separated into uncontrolled group, controlled group and CGY group. As controlled and CGY groups were induced aging by subcutaneous injection of D-galactose, at the same time we administered the extract of Cheongeumyeonsudan to CGY group for 6 weeks. After then we drew blood from each group, and took measurements; the activity of SOD, GSH-px, catalase in erythrocytes, TBARS value, concentration of total lipid, tryglycende, total cholesterol, HDL-cholesterol in blood plasma. Results : The activities of SOD, GSH-px in erythrocytes were significantly increased in the CGY group compared with control group. The activity of catalase showed a tendency to increase, but it was nor remarkable. The concentration of total lipid, the values of TBARS and total cholesterol was significantly decreased in the CGY group compared with control group, and the concentration of plasma HDL-cholesterol was not remarkable. The concentration of tryglycende in plasma showed a tendency to decreased. Conclusions : it is suggested that Cheongeumyeonsudan decreased the activities of free radical, the concentration of lipid in plasma and generate enzyme which form lipid peroxide.

Differential antioxidation activities in two alfalfa cultivars under chilling stress

  • To understand the adaptability of alfalfa (Medicago sativa L.) to chilling stress, we analyzed the antioxidative mechanism during seed germination. The germination rates of six alfalfa cultivars were studied comparatively at $10^{\circ}C$. Xinmu No. 1 and Northstar were selected as chilling stress-tolerant and stress-sensitive cultivars for further characterization. After chilling treatment, Xinmu No. 1 showed higher seedling growth than Northstar. Xinmu No. 1 exhibited low levels of hydrogen peroxide and lipid peroxidation compared with Northstar. In addition, shoots in Xinmu No. 1 treated with chilling showed higher activities of the superoxide dismutase, ascorbate peroxidase (APX), and catalase than those of Northstar, whereas Xinmu No. 1 showed higher APX activity in roots that Northstar. These results indicated that high antioxidation activity in Xinmu No. 1 under chilling stress is well associated with tolerance to chilling condition during germination.

Amino Acid Analysis and Antioxidation Activity in Allium wakegi Araki (쪽파(Allium wakegi Araki)의 아미노산 분석과 항산화 활성)

  • The antioxidative activation of Allium wakegi Araki appeared differently according to parts, and the callus cultured with the addition of 2,4-D 1.0mg/L showed the activation of 280 $\mu\textrm{g}$ (RC$\sub$50/), and the bulbs and shoots in the natural condition showed 296 $\mu\textrm{g}$ (RC$\sub$50/), and 301 $\mu\textrm{g}$ (RC$\sub$50/). The total amino acid content was 59.779 mg% in the natural leaves and the leaves of bulbs, and was 73.725 mg% in the cultured callus(2,4-D 0.5mg/L). The total amino acid content of cultured callus was higher, but the content proportion of essential amino acid to the total amino acid in the natural bulbs was higher, which was 39.462 mg%.

Studies on the Mineral Component and Antioxidative Activity of Gastrodia elata Blum (천마의 무기성분 및 항산화 작용에 관한 연구)

  • The mineral components of the Gastrodia elata Blum were determined by inductively coupled plasma-atomic emission spectrophtometry. The mineral component contents of the Gastrodia elata Blum were greater in the order of Fe < Na < Mg < Ca < P < K. The antioxidative effects of the Gastrodia elata Blum extracts on refined perilla oil were investigated by the rancimat method, peroxide value and TBA value. The antioxidative effects of Gastrodia elata Blum added to the refined perilla oil was increased with dose-dependent fashion in concentration of Gastrodia elata Blum extracts. The peroxide value and TBA value for their antioxidation stability were also lower than that of control. Antioxidation stability was increased in a dose-dependent manner.

The Effect of Tocopherol and Ascorbate on the Enzyme Activity and Antioxidation in $CCl_4$ Induced Rats (Tocopherol과 Ascorbate 투여가 사염화탄소로 유도한 흰쥐의 효소활성 및 항산화적용에 미치는 영향)

  • The enzyme activities and antioxidative of tocopherol and ascorbate were investigated at the levels of liver, kidney homogenates and sera of SD-rats intoxicated with tetrachlorocarbon$CCl_4$. GOT and GPT activities of sera in the $CCl_4$ group were 3, 6 times increased compared to normal group. But they tended to decrease significantly in tocopherol and ascorbate administered group. As for BUN and total cholesterol they were the same. HDL-cholesterol in the $CCl_4$group was 42% decreased compared to normal group. HDL-cholesterol was about 26% increased in the tocopherol group and tocopherol ascorbate group compared to $CCl_4$ group. MDA and SOD activities in the liver and the kidney tissue homogenates were significantly increased in $CCl_4$ group compared to normal group. But they were decreased significantly in the tocopherol group and tocopherol-ascorbate group compared to $CCl_4$ group. In view of this study tocopherol and ascorbated were ascorbate were effective on the detoxication of liver and kidney injury.
