토복령의 항산화작용 및 항균 효과에 대한 연구 (Studies on the Antioxidation and Antimicrobial Effect of Smilacis Glabrae Rhizoma)

  • Objectives : This study was performed to investigate the effects of Smilacis GlabraeRhizoma on antioxidation and antimicrobial activity. Methods : In this study, we investigated the effects of peroxide radicals on hydrogen donating activity and linoleic acid, and the MDA contents on the hepatic lipids of rats, via methanol extractions and subfractions of Smilacis Glabrae Rhizoma. Results : 1. Hydrogen donating activity was very great for the radical scavenging effects, depending on the additional concentration at the fraction level of chloroform and ethyl acetate. 2. The peroxide radicals in linoleic acid were lower depending on the additional concentration, at the fraction level of ethyl acetate, than the controls. We concluded that both had significant anti-oxidation effects. 3. MDA contents of the hepatic lipid had also inhibition effects on lipid radicals, in proportion to the concentration of n-hexane, chloroform, and ethyl acetate fraction level. 4. After extracting Smilads Glabrae Rhizoma with 80% methanol, we experimented with the extracts the antibiosis each concentration, for 5 bacilli, Bacilus subtilis, staphylococcus aureus, Escherichia coli, Salmonela typhimurium, and Alcaligenes faecalis. While the effects showed differentiations by concentration, they had usually the significant inhibition effect for the multiplication at 37.5~75ug/ml. To identify the effective constituents, we identified the antibiosis of the fractions assaying cyclically hexane, chloroform, ethyl acetate, and butanol. The result showed that antimicrobial activation of both Gram-positive and Gram-negative bacillus except for E. coli was measured highest at the fraction level of BuOH and water. Conclusions : This result suggest that the extractions of Smilacis Glabrae Rhizoma, at ethyl acetate fraction, had significant anti-oxidation effects and at BuOH and water fraction had relatively strong antimicrobial activity against Bacilus subtilis, staphylococcus aureus, Salmonela typhimurium, and Alcaligenes faecalis.

감초 추출물의 약리적 특성 및 분석 (A Study on the Pharmacetical Characteristics & Analysis of Glycyrrhizin Extract)

  • From experiment results on pharmacetical characteristics and analysis of Glycyrrhizin extract, some conclusions are obtained as follows. From results on extract experiment of Glycyrrhizin, it appeared about 8%-extraction ratio as semi-solid state, and after dried in freezing from Glycyrrhizin extract of semi-solid state, it obtained about 70%-Glycyrrhizin extract as solid state of yellow gold color. From results on antimicrobial experiment of Glycyrrhizin extract, number of S-typhimurium and Fungus in microbe decreased more and more according to time passage. This phenomenon shows that Glycyrrhizin extract keeps antimicrobial effect. From results on antioxidation experiment of Glycyrrhizin extract, DPPH scavenging activity of free radical shows that Glycyrrhizin extract appears more remarkable reduction ability than reference samples. This phenomenon means that antioxidation of Glycyrrhizin extract appears higher than Vitamin-C and BHA. From results on instrument analysis, the fatty and aromatic components of 2-pentanone, cyclohexasiloxane, tetrasiloxane, benzoquinoline-2-carboxylic acid etcs from Glycyrrhizin extract was detected with GC/MS and inorganic components of Ca, Mg, Ti, Zn, Fe etcs from Glycyrrhizin extract was detected with ICP/OES.

흑마늘의 항산화, 항균 및 항혈전 활성 (Antioxidation, Antimicrobial and Antithrombosis Activities of Aged Black Garlic (Allium sativum L.))

  • 흑마늘을 이용한 식의약품 기능성 소재 개발 연구의 일환으로, 생마늘과 흑마늘 추출물을 조제하여 항산화, 항균, 항혈전 활성을 비교하였으며, 이후 흑마늘 추출물의 순차적 용매 분획물을 조제하여 활성분획을 확인하고자 하였다. 흑마늘의 열수 추출효율은 40.6%로 생마늘의 4배의 효율을 나타내었으며, 고온 숙성과정을 통해 생마늘보다 총 폴리페놀은 4배, 총 플라보노이드 함량은 1.56배, 총 당 및 환원당은 각각 3.36배 및 6.75배 증가되었다. 항산화 활성 평가 결과, 흑마늘 추출물은 DPPH 음이온에 대한 소거능은 생마늘 추출물보다 미약하였으나, ABTS 양이온 소거능 및 nitrite 소거능은 생마늘 추출물보다 우수하였다. 특히 흑마늘 추출물의 ethylacetate 분획은 매우 강력한 항산화 활성을 나타내었다. 흑마늘 추출물은 생마늘 추출물에 비해 항균 및 항혈전 활성을 상실한 반면, 흑마늘 추출물의 ethylacetate 분획은 Staphylococcus aureus 및 Bacillus subtilis에 대한 항세균 활성과 프로트롬빈, 혈액응고인자 저해, 인간 혈소판 응집저해를 나타내어 강력한 항혈전 활성을 확인하였다. 흑마늘 열수 추출물 및 분획물은 적혈구 용혈활성은 나타나지 않았다. 본 연구결과는 흑마늘 추출물의 ethylacetate 분획이 향후 항혈전 활성을 위한 기능성 식의약품 소재로 개발 가능함을 제시하고 있다.

오미자 물추출물이 나박김치의 저장성에 미치는 영향 (Effects of Water Extract from Omija (Schizandra chinensis Baillon) on Nabak Kimchi Preservation)

  • 나박김치의 저장성 향상을 위한 천연보존제로서 이용 가능성을 검토하기 위해 오미자 물추출물을 제조하여 이들 시료의 농도를 0.5, 1.0, 1.5, 2.0%로 사용하여 항산화력 및 전자공여능을 측정하고 나박김치 발효에 관여하는 젖산균에 대해 항균활성을 조사하였다. 항산호력은 TBA값을 측정하였는데, 오미자 물추출물의 농도가 높을수록 항산화 효과가 뚜렷이 나타났고, 시간이 경과할수록 그 효과는 현저하였다. 전자공여능 또한 오미자 물추출물의 농도가 높을수록 높은 전자공여능을 나타냈고, 1.0% 이상이 높은 전자공여능을 보였다. 나박김치에서 잦산균을 분리한 결과 117균주를 1차 분리하였고, 이 가운데 성장이 우수하고 산 생성력이 강한 4균주를 2차 선별하여 Lactobacillus brevis, Lactococcus faecalis, Leuconostoc mesenteroides, Lactobacillus plantarum을 확인하였다. 이들의 항균활성을 측정한 결과 Lactobacillus brevis를 제외한 세 균주 모두 오미자 물추출물 첨가에 의한 뚜렷한 성장 억제현상을 나타내었다. 따라서 나막김치 담금시에 항산화성과 항균활성이 있는 오미자 물추출물을 이용할 수 있는 가능성이 높을 것으로 생각되었다.

현토고본환이 노화유발 백서의 항산화능에 미치는 영향 (Effect of Hyuntogobon-hwan on Antioxidation Activity in Induced Aging Rats)

  • Objectives : This experiment was done to evaluate the effects of Hyuntogobon-hwan(HTG) on antioxidant capability and lipidic concentration in blood, both of which are presumed to be related to aging. Methods : 12 week-old SD rats were divided into controlled group, uncontrolled group and HTG group. As controlled and HTG groups were given subcutaneous injection of D-galactose(50mg/kg/rat), at the same time HTG group was administered extract of Hyuntogobon-hwan(270mg/200g). HTG injections continued for 6 weeks. After initial injections, blood was drawn from each group and the following were measured: the activity of SOD, GSH-px, catalase in erythrocytes, TSARS value, concentration of total lipid, tryglyceride in blood plasma. Results : The activities of SOD and GSH-px in erythrocytes increased significantly in HTG group compared with controlled group. The activity of catalase seemed to increase slightly, but it was barely noticeable. The concentration of total lipid in plasma decreased significantly in HTG group compared with controlled group. The value of TBARS in plasma seemed to decrease slightly, but it was barely noticeable. Conclusions : According to the above results, Hyuntogobon-hwan has an influence on aging by virtue of activation of antioxidative enzyme systems in erythrocytes and concentrations of lipid in blood plasma.

Antioxidant and Anti-hyperglycemic Activity of Polysaccharide Isolated from Dendrobium chrysotoxum Lindl

  • Although polysaccharide is believed to play an important role in the medicinal effect of Dendrobium chrysotoxum Lindl (DCL), its role as an antioxidant and in anti-hyperglycemic induction was not reported. In this study, polysaccharide with molecular weight of approximately 150 kDa, herein named DCLP, was isolated from the stem of DCL, and its antioxidative, hypoglycemic and immune stimulating effects were evaluated using various in vitro and in vivo assay systems. DCLP inhibited hydroxyl radicals ($^{\cdot}$OH)-mediated deoxyribose degradation by scavenging hydroxyl radicals directly as well as by chelating iron ions. DCLP also showed dose-dependent scavenging activity on superoxide anions ($O_2^{{\cdot}-}$) and offered significant protection (p < 0.001) against glucose oxidase-mediated cytotoxicity in Jurkat cells. DCLP had immune stimulating effects, as evidenced by the DCLP-mediated increases in the level of DNA synthesis, viability, and cytokine secretion in mouse lymphocytes. Moreover, oral administration of DCLP produced a significant reduction in blood glucose level in alloxan-induced diabetic mice. These findings suggest that DCLP has a potential utility in treating patients who require enhanced antioxidation, immune function and/or hypoglycemic activity.

Rooibos Tea(Aspalathus linearis)에탄올 추출물의 항산화 효과 및 식품에 대한 응용 (Antioxidative Effects of Ethanol Extract Obtained from Rooibos Tea(Aspalathus linearis) and It's Application of Food)

  • This study was designed the antioxidative effect of the ethanol extract obtained Rooibos tea (Aspalathus linearis) on various kinds of oil, and examined the synergistic effect of Rooibos tea extract by addition of citric acid to the antioxidation activity and also investigate to antioxidation effect of the extract in food production and storage peroids. The antioxidative activity of 0.2mg of Rooibos tea extract was showed similar to same doses of $\alpha$-tocopherol, BHA and BHT in linolieic acid-ethanol system. The antioxidative effect of the mixture with 0.1% Rooibos tea extract on lard was more effective than that of the mixture with same doses of $\alpha$-tocopherol, BHA and BHT. The antioxidative effect of Rooibos tea extract was showed slightly effects on lard or soybean oil. Antioxidative effects of Rooiboe tea extract in addition of citric acid as synergist showed more effective in linoleic acid-ethanol system, but did not showed in the other oils. In the application of Rooibos tea extract to food prodctuction and storage period, the antioxidative effect was more effective in biscult, preparation added lard mixed with 0.1% Rooibos tea extract.

수산건제품의 갈변에 관한 연구 붕장어육 및 유의 산화, 갈변 물질의 성상 (Studies in Browning Reaction in Dried Fish Lipid Oxidative Browning in Dried Conger eel and Properties of Browning Products)

  • This studies was carried out in order to investigate the browning reaction of lipid originated compound with nitrogenous compound in dried conger eel. The major fatty acids were $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{20:5}\;and\;C_{22:6}$. The nonpolar lipid contained the highest percentage of $C_{16:0}$, while the polar lipid contained the highest percentage of $C_{22:6}$. The browning reaction there was a rapidly developed with the beginning of the decline in carbonyl value and remarkable decrease in polyunsaturated fatty acids such as $C_{20:5},\;C_{22:5},\;C_{22:6}$ compared with the other fatty acid, in the water soluble fraction of the browning product obtained from tile fish was detected some antioxidation activity but in the lipid soluble fraction which covers most of the browning reactions in the fish meat antioxidation activity was not detected. In the test of conger eel oil, the phosphatidylcholine was largest in quantity and browning products provided in this experiment showed very low reducing activity.

아연-고함유 효모 Saccharomyces cerevisiae FF-10 세포액의 항산화효과 (Antioxidative Activity of Zinc-Enriched Saccharomyces cerevisiae FF-10 in In vitro Model Systems)

  • 아연 고함유 효모 S. cerevisiae FF-10의 항산화능을 검토하기 위하여 DPPH 전자 공여능, linoleic acid을 이용한 ferric thiocyanate법과 TBA법에 의한 과산화지질 생성 정도 및 흰쥐 간 조직 생체막을 이용한 TBARS법에 의한 과산화지질 생성 정도를 측정하였다. 본 실험은 효모 생육배지인 YM 기본배지와 아연 생산량을 증대시키는 YM 최적배지에서 각각 배양된 S. cerevisiae FF-10의 세포 파쇄액의 항산화 활성을 비교하였다. DPPH 전자 공여능은 양성 대조구로 사용한 BHT에서 가장 높았고, YM 기본배지 보다는 YM 최적배지에서 배양된 FF-10 세포 파쇄액에서 항산화 활성이 높게 나타났다. 간 조직 생체막 과산화지질 생성 정도는 BHT > 최적 생산배지 > 기본배지 순으로 저해되었다. Linoleic acid를 이용한 과산화지질 생성정도는 음성 대조구에서 반응 1일째부터 급격히 증가한 후 반응종료일까지 계속 그 수준이 유지되었고, 양성 대조구인 BHT 처리구에서는 과산화지질 생성이 억제되어 높은 항산화활성이 확인되었으며, YM 기본배지 보다는 YM 최적배지에서 높은 과산화지질 생성 저해활성을 보였다. 이상의 결과에서 in vitro 항산화 실험계인 DPPH radical scavenging activity, 간 조직 생체막과 linolic acid 지방산을 이용한 ferric thiocyanate and TBARS 측정에서 항산화 활성은 양성 대조구인 BHT 보다는 낮았으나 최적배지에서 배양된 아연 고함유 효모 S. cerevisiae FF-10 균주의 세포 파쇄액에서 모두 높게 나타나 in vivo 항산화 실험계에서도 확인이 필요한 것으로 사료되어 진다.
