Physicochemical components and antioxidant activity of Sparassis crispa mixture fermented by lactic acid bacteria (유산균 발효 꽃송이버섯 혼합물의 이화학적 성분과 항산화 활성)

  • The objective of this study was to investigate the nutritional composition and antioxidant activity of a mixture of rice bran and bodies of Sparassis crispa fermented with lactic acid bacteria (LAB). LAB-fermented S. crispa mixture had higher water, crude lipid and crude ash content than that of S. crispa. Insoluble dietary fiber contents of the dried powder of S. crispa and LAB-fermented S. crispa mixture were 46.13% and 33.46%, respectively. ${\beta}$-glucan was higher in dried S. crispa (38.03%) than in LAB-fermented S. crispa mixture (5.44%). Dried S. crispa contained mainly fructose and glucose instead of containing sucrose in LAB-fermented S. crispa mixture. No significant differences in the total polyphenol contents were found in between dried S. crispa and LAB-fermented S. crispa mixture. Total flavonoid content was significantly higher in LAB-fermented S. crispa mixture than in dried S. crispa. No significant differences were found in the DPPH radical scavenging activity and in the antioxidant index between dried S. crispa and LAB-fermented S. crispa mixture. Finally, ABTS radical scavenging activity of LAB-fermented S. crispa mixture was significantly higher than that of dried S. crispa. These results may provide the basic data for future studies for a better understanding of the biological activities of LAB-fermented S. crispa mixture.

Physicochemical Characteristics and Antioxidant Effects of Cooked Rice-Added Foxtail Millet according to Cooking Method (취반방법에 따른 조 첨가 밥의 이화학적 특성 및 항산화 효과)

  • This study was carried out to analyze the antioxidant components and activities of cooked mixed grain rice-added foxtail millet with different addition rates (5, 10, 15, and 20%) and cooking methods. The two foxtail millet varieties used in this study were non-waxy foxtail millet, 'Samdame', and waxy foxtail millet, 'Samdachal'. Cooked mixed grain rice-added foxtail millet was cooked by general and high pressure cooking methods with and without fermented alcohol. Except for breakdown viscosity, pasting characteristics of foxtail millet were reduced with increasing amounts of foxtail millet. Water-binding capacity and swelling power significantly decreased with increasing amounts of foxtail millet, whereas water solubility index significantly increased. Total polyphenol and flavonoid contents increased with increasing amounts of foxtail millet. DPPH and ABTS radical scavenging activities also increased with increasing amounts of foxtail millet. Moreover, foxtail millet cooked by general cooking method with fermented alcohol showed higher antioxidant effect compared to other cooking methods. In this study, antioxidant components and activities of cooked mixed grain rice-added foxtail millet with different addition rates and cooking methods can be used as basic data for manufacturing processed products.

The Effect of Eisenia bicyclis Extracts on Antioxidant Activity and Serum Lipid Level in Ovariectomized Rats (대황 추출물의 in vitro 항산화 활성 및 난소를 절제한 흰쥐의 혈중 지질함량에 미치는 영향)

  • Hormone replacement therapy (HRT) is an effective regimen that has been found to prevent these diseases in postmenopausal women. However, HRT is accompanied by an increased risk of unfavorable outcomes. This study was conducted to evaluate the effects of Eisenia Bicyclis extract on lipids in ovariectomized rats. Fifty 7-week-old female Sprague-Dawley rats were randomly assigned to four groups: sham-operated rats (SHAM), ovariectomized rats (OVX-CON), and ovariectomized rats that were treated with Eisenia bicyclis extracts. The extract-treated diets were fed to the rats for 6 weeks after operation. Antioxidant effects were measured by DPPH free radical scavenging activity. Antioxidant activity of the ethanol extract increased in a dose-dependent manner and was about 55.9% in a concentration of 100 ${\mu}g/ml$. We measured the total cholesterol content, triglyceride content, HDL-cholesterol content, LDL-cholesterol content, atherosclerotic index, cardiac risk factor in serum, and anti-platelet aggregation and blood rheology. The total cholesterol and triglyceride concentration in serum increased for the OVX-control group, but supplementation with the E. bicyclis extract caused these factors to decrease. Notably, the serum LDL-cholesterol concentration in the OVX-EB200 group was significantly lower than the OVX-CON group. In addition, the blood passage times in rats that received the E. bicyclis extract were more rapid than the times in the untreated group (OVX-CON). Microscopic evaluation revealed that whole blood passed more smoothly through the microchannels in rats in the E. bicyclis extract supplement groups. Our results clarified the effects of E. bicyclis extract on serum lipid content in ovariectomized rats, and consequently we expect positive effects from providing E. bicyclis extract to postmenopausal women with cardiovascular disease.

The Effects of Salinity Stress on the Antioxidant and Anti-inflammatory Activities of Crepidiastrum sonchifolium (염분 스트레스가 고들빼기의 항산화 및 항염증 활성에 미치는 영향)

  • This study was conducted to investigate the effect of salinity stress on the antioxidant and anti-inflammatory activities of Crepidiastrum sonchifolium. The plant was treated with NaCl at concentrations of 0, 50, 100, and 200mM for 6 weeks. After treatment, the whole plant was collected, and 70% ethanol extracts were prepared. The DPPH radical scavenging activity was highest in the order of NaCl treatment concentrations of 0, 100, and 50mM, while the 200mM treatment group showed the lowest radical scavenging. The total phenol and total flavonoid contents showed very similar results to the antioxidant activity depending on the NaCl concentration, confirming that the phenolic compounds of the plant can contribute to the antioxidant capacity against salinity stress. In addition, the investigation of the effect of NaCl-treated C. sonchifolium extract on the inhibition of NO production in the LPS-stimulated mouse macrophage cell line (Raw 264.7) revealed that NO production significantly decreased in the 1,000㎍/mL treatment group across all NaCl concentration groups. But, the high concentration (1,000㎍/mL) treatment of the 100mM and 200mM NaCl treatment groups was found to have a negative effect on cell survival. These results suggest that radical scavenging activity is highest in healthy plants and that they produce antioxidants to respond to NaCl salinity stress up to 100mM. However, a high NaCl concentration of 200mM has a negative effect on the physiological activity of the plants. Compared with the results of the previously reported growth index, it is thought that the growth and physiological activity of plants can be positively affected in an NaCl treatment environment of 50mM or less.

Physicochemical characteristics of onion with cold tolerance cultivated in Kangwon (강내한성 강원양파의 이화학적 특성)

  • This study was performed to provide the basic data for the prediction of the usefulness of onion with cold tolerance cultivated in Kangwon. The physicochemical properties and antioxidant activity of freeze-dried and hot-air-dried (40 and $60^{\circ}C$) onions were investigated. The moisture content of the raw onion was 90.55%. The crude protein and crude fat contents of the freeze-dried onions were slightly higher than those of the hot-air-dried onions ($40^{\circ}C$ and $60^{\circ}C$). As for the color values, the freeze-dried onion powder was highest in lightness (77.19), and the $60^{\circ}C$ hot air-dried onion was highest in redness (6.09) and yellowness (24.60). Moreover, the color difference (${\Delta}E$) between the freeze-dried and hot-air-dried ($40^{\circ}C$ and $60^{\circ}C$) onion powders was significant. The brown index was lower in the freeze-dried onion than in both hot-air-dried onions. The total phenol content and the 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging activity of both hot-air-dried onions were higher than those of the freeze-dried onion. These results indicate that the freeze-drying methods protected the physicochemical properties of the onion powder, whereas the hot-air-drying method enhanced the antioxidant activity and the total phenol content of the onion powder.

Effects of Eucommia ulmoides olivon Ethanol Extract on Lipid Metabolism and Antioxidant Enzyme Activities of Rats Fed High Fat Diet (고지방 식이를 투여한 흰쥐의 지질대사와 항산화 효소 활성에 미치는 두충 에탄올 추출물의 영향)

  • To investigate the effects of Ewomia ulmoides olivon extract on the serum and hepatic lipid status and anti-oxidant enzyme activities, male Sprague Dawley (SD) rats were given high fat diets and Eucomia ulmoides olivon ethanol extracts for 6 weeks. Rats were divided into (our groups consisting the control (C), Eucomia ulmoides oliuon extract fed group (CE), high fat diet group (CL) and the extract and high fat fed group (CLE). Eucomia ulmoides oliuon ethanol extract lowered total cholesterol, but increased HDL-cholesterol and HDL-cholesterol/total cholesterol of the serum compared to the control. On the other hand, the ratio of HDL-cholesterol/total cho-lcsterol (HTR) was increased by 55% (p<0.05). High fat diet significantly increased serum cholesterol contents, but decreased HDL-cholesterol level and HTR (p<0.05). Atherogenic index (AI) of CL group was increased by almost four times of the control (p<0.05). Triglycerides (TG) and phospholipids were also increased by high fat diet. Eucomia ulmoides oliuon ethanol extract decreased the levels of TG (p<0.05) and phospholipids (p<0.05). Although liver antioxidant engyme activities including glutathione sulfur transferase (GST), superoxide dismutase (SOD) and catalase were decreased by high fat diet, those were stimulated by the administration of Eucommia ulmoides olivon ethanol extract.

Antioxidant Effect of Edaravone on the Development of Preimplantation Porcine Embryos against Hydrogen Peroxide-Induced Oxidative Stress

  • Edaravone (Eda) is a potent scavenger of inhibiting free radicals including hydroxyl radicals ($H_2O_2$). Reactive oxygen species (ROS) such as $H_2O_2$ can alter most kinds of cellular molecules such as lipids, proteins and nucleic acids, cellular apoptosis. In addition, oxidative stress from over-production of ROS is involved in the defective embryo development of porcine. Previous study reported that Eda has protective effects against oxidative stress-like cellular damage. However, the effect of Eda on the preimplantation porcine embryos development under oxidative stress is unclear. Therefore, in this study, the effects of Eda on blastocyst development, expression levels of ROS, and apoptotic index were first investigated in preimplantation porcine embryos. After in vitro fertilization, porcine embryos were cultured for 6 days in PZM medium with Eda ($10{\mu}M$), $H_2O_2$ ($200{\mu}M$), and Eda+$H_2O_2$ treated group, respectively. Rate of blastocyst development was significantly increased (P<0.05) in the Eda treated group compared with only $H_2O_2$ treated group. And, we measured intracellular levels of ROS by DCF-DA staining methods and investigated numbers of apoptotic nuclei by TUNEL assay analysis is in porcine blastocyst, respectively. Both intracellular ROS levels and the numbers of apoptotic nucleic were significantly decreased (P<0.05) in porcine blastocysts cultured with Eda ($10{\mu}M$). More over, the total cell number of blastocysts were significantly increased (P<0.05) in the Eda-treated group compared with untreated group and the only $H_2O_2$ treated group. Based on the results, Eda was related to regulate as antioxidant-like function according to the reducing ROS levels during preimplantation periods. Also, Eda is beneficial for developmental competence and preimplantation quality of porcine embryos. Therefore, we concluded that Eda has protective effect to ROS derived apoptotic stress in preimplantation porcine embryos.

The Effect of Vitamin A Derivatives on the Activity of Drug-metabolizing Enzyme in Rat Liver (Vitamin A 유도체로 인한 간의 약물대사효소 변동)

  • It has been known that retinoids are intrinsically of critical importance for control of premalignant epithelial cell differentiation. In the absence of retinoids, normal cellular differentiation and growth does not occur in epithelia such as those of trachea and bronchi. Furthermore, it was also reported that retinoid deficiency enhanced susceptibility to chemical carcinogenesis in the respiratory system, in the bladder, and in the colon of the experimental animal. In 1974, Bollag examined the effects of synthetic retinoids in prevention of development of cancer and demonstrated synthetic retinoids to have more favorable therapeutic index than retinoic acid for causing regression of skin papilloma in mice. Therefore, it was assumed that this anticarcinogenic effect of vitamin A derivatives could be due to modification of the metabolism of the carcinogenic polycyclic hydrocarbon, which must first be activated to exert their effect. Hill and Shih reported that vitamin A compounds and analogs had inhibitory effect on drug metabolizing enzyme from liver and lung tissue of mouse and hamster. Lucy suggested that the chemoprevention effect of vitamin A derivatives is due to reaction with molecular oxygen, and it is possible that inhibition of hydroxybenzpyrene formation is a result of this property. On the other hand, butylated hydroxytoluene which is a potent antioxidant strongly inhibited the formation of mammary tumor induced by dimethylbenranthracene. Also, it was observed that this antioxidant inhibited cancer induction in rats by N-2-fluo-renylacetamide. The purpose of this experiment was to investigate the effect of vitamin A derivatives such as retinoic acid and retinoid on drug-metabolizing enzyme and to determine whether riboflavin tetrabutylate or vitamin E could prevent of modify any changes induced by vitamin A delivatives in the rats. The results obtained were as followings. 1) Body weight was significantly reduced by retinoic acid, but not by retinoid. 2) Retinoic acid markedly increased liver weight while retincid showed no effect on liver weight. Treatment of riboflavin tetrabutylate did not affect retinoic acid-induced change in both body weight and liver weight. 3) Both retinoic acid and retinoid remarkably decreased the activity of aminopyrine demethylase. Pretreatment of riboflavin tetrabutylate, however, prevented inhibitory effect of retinoic acid on the enzyme activity. 4) No significant effect of vitamin E on aminopyrine demethylase was observed in both groups treated with retinoic acid and retinoid.

Amelioration Effects of Irrigation-Aspiration on Renal Ischemia-Reperfusion Injury in Canine Model (개에서 신장의 허혈-재관류 손상에 대한관류-흡인의 감소효과)

  • Renal ischemia-reperfusion injury is great clinical important because viability of the transplanted organ depends on the tolerance of the graft to ischemia-reperfusion injury, an inevitable processing during surgery. The purpose of this study was to investigate the effects of irrigation-aspiration in ischemia-reperfusion injury model induced by cross-clamping of renal vessels. Blood samples were collected from these dogs for measurement of kidney function and antioxidant enzyme activity, and RI at the intrarenal artery was measured at different time intervals. And the kidneys were taken for histopathologic evaluation at day 14. Kidney function (Cr and BUN) showed a significant increasing in untreated group compared to treated group. Resistive index of intrarenal artery was no significant difference among the groups. Activity of antioxidant enzymes in plasma was significant decrease in untreated group compare to control group while in treated group was no significant difference compared to control group. In histopathologic finding, treated group was showed less damage than that of untreated group. This result suggests that the processing of irrigation-aspiration is useful to reducing ischemia-reperfusion injury.

Antioxidant effect of Omija (Schizandra Chinesis Baillon) Extracts (오미자 추출물의 항산화 효과)

  • Antioxidative activities of the extracts from Schizandra Chinesis Baillon (Omija) with various solvent were compared with some commercial antioxidants. AS (antioxidative index; induction period of oil containing extract/induction period of control oil) of Omija extracts from five kinds of solvents (MeOH, EtOH, BtOH, EA, PE) and other antioxidants were shown as following orders: 0.02% BHT > 0.05% EA > 0.01% MeOH > 0.05% EtOH > 0.1% EA > 0.05% PE > 0.05% BuOH >> 0.02% alpha-tocopherol. Antioxidative effects of 0.05% EA and 0.05% MeOH extracts during autoxidation (60${\pm}$ 2$^{\circ}C$) were higher than those (If the other extracts but were not greater than that of 0.02% BHT. However, Al of EA and MeOH, EtOH extracts during thermal oxidation (180${\pm}$2$^{\circ}C$) were greater than that of BHT. The antioxidant effect of alpha-tocopherol showed no apparent difference or a prooxidant effect as compared with result of control.
