Effect of Fermentation on the Antioxidant Activity of Rice Bran by Monascus pilosus KCCM60084

  • In this study, we optimized fermentation conditions for the solid state fermentation of rice bran with Monascus pilosus KCCM60084, and the antioxidant activities were investigated. Optimal fermentation conditions were determined by the production of Monacolin K, a functional secondary metabolites with cholesterol lowering activity. The highest Monacolin K production were 2.88 mg/g observed on day 10 with 45% moisture content in the substrate when inoculated with 5% inoculum (w/w). Reducing power, iron chelating activity and $ABTS^+$ radical scavenging activity were significantly enhanced after fermentation by 60, 80, and 38% respectively. Furthermore, the content of total flavonoid were found to be increased by 4.58 fold. Based on these results, Monascus-fermented rice bran showed strong possibility to be used as a natural antioxidant agent due to its enhanced antioxidant activity.

  • The antioxidant in the oriental drug, NP-S, traditionally used as a therapeutic agent for Hepatitis B was identified. The NP-S was divided into two fractions at molecular weight cut-off of 1000 by ultrafiltration and the fraction having molecular weight less than 1000 was named as NP-S(l)10. Both NP-S and NP-S(l)10 had antioxidant activity. An antioxidant that was extracted by chloroform from water-insoluble fraction in lyophilized NP-S(l)10 had a little smaller activity than NP-S(l)10 and identified as cyclooctasulfur(S8) by mass spectrum.

  • Flavonoids are phenolic compounds widely distributed in wide variety of edible plants including leafy vegetables, fruits, beverages. Quercetin is one of bioflavonoid compounds and has anti-tumor effect by suppressing tumor growth in vitro and in vivo, including multiple biological effects by antioxidant and effective anti-inflammatory agent. The present study investigated whether quercetin can enhance antioxidant enzyme activities (glutathione peroxidase: GPX, superoxide dismutase : SOD, catalase: CAT) and intracellular reactive oxygen intermediate (ROI) levels in the presence of taurine on B16F10 murine melanoma cells. From this result, the antioxidant enzyme activities of quercetin in the presence of taurine was enhanced. In addition, the same treatments decreased intracellular reactive oxygen intermediate levels on B16F10 murine melanoma cells. Taken together, these results demonstrate that the antioxidant effect of quercetin can enhance in the presence of taurine and it might play an important role in anti-tumor effect.

Antioxidant and Liver-protective Effects of Caesalpinia sappan (소목 추출물의 항산화 및 간보호작용)

  • The heartwood of Caeralpinia sappan L. (Leguminosae) has been used to activate blood flow, remove blood stasis, reduce swelling and relieve pain in Korean folk medicine. In this study. the antioxidant effects of this crude drug and its hepatoprotective activity on CCl$_4$-induced liver injury in rats were evaluated. The levels of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) reduced by CCl$_4$treatment, were recovered by this crude drug. It is suggested that Caesalpinia sappan L. have antioxidant effect. The increased levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) by CCl$_4$were also recovered by treatment with this crude drug. These results can be attributed to the agent's antioxidant and membrane - stabilizing actions.

A Study on the Antioxidative and Depigmentation Activities of the Ethanol Extract of Saururus Herba (삼백초(三白草) 에탄올추출물의 항산화능 및 피부 미백작용 연구)

  • Objectives : The purpose of this study is to investigate the etahnol extract of Saururus Herba and their dermal boiactivity properties related to cosmeceuticals such as depigmentation and radical scavenging effect. Methods : Using B16/F10 Melanin cells, we measured depigmentation effect of the etahnol extract of Saururus Herba. Antioxidant activities of the etahnol extract of Saururus Herba were determined by DPPH radical scavenging activity. Results : The etahnol extract of Saururus Herba showed considerable radical scavenging activity($SC_{50}$ : 25 ${\mu}g$/ml). Antioxidant activity is in a dose dependant manner. The etahnol extract of Saururus Herba down-regulated melanin synthesis effectively. B16 cells incubated with the etahnol extract of Saururus Herba showed reduces melanin formation and tyrosinase activity. The etahnol extract of Saururus Herba alone markedly suppressed melanin content and tyrosinase activity. Conclusions : These results suggest that the ethanol extract of Saururus Herba is a potent depigmentaion agent and it may be a candidate for antioxidant agent.

  • The rhizome of Cibotium barometz has been used for variety of bone disease as a traditional medicine. In the present study, we examined the antioxidant and anti-inflammatory activities of 85% methanol extract of C. barometz. C. barometz exhibited potent scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide radical and nitric oxide radical. In IFN-$\gamma$/LPS-stimulated mouse peritoneal macrophage model, C. barometz suppressed nitric oxide production and IL-6 secretion dose-dependently. Moreover, C. barometz showed decreased iNOS and COX-2 expression without notable cytotoxicity. These results suggest that C. barometz may be an useful agent as an antioxidant and anti-inflammatory agent.

Effects of the Grapevine Shoot Extract on Free Radical Scavenging Activity and Inhibition of Pro-inflammatory mediator Production in RAW264.7 Macrophages (포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과)

  • Free radical scavengers or quenching agents for reactive oxygen species (ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract (GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory mediator production. Utilizing 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and xanthine oxidase (XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the $IC_{50}$/ values of 34.5 and 155 $\mu\textrm{g}$/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E$_2$ (PGE$_2$) and nitric oxide (NO) production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells with the $IC_{50}$/ value of 6.4 and 14.5 $\mu\textrm{g}$/ml, respectively. This result suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

Higher Extraction of Phytochemical Compounds from Tartary Buckwheat Seeds by the Application of Surfactant Formulation

  • The aim of this study was to determine the suitability of surfactant to extract higher phenolic compound, flavonoid and antioxidant activity from Tartary buckwheat and evaluate the potentiality of surfactant as a screening agent for breeding purpose. Primarily, we employed two types of surfactant (Hydrophilic: Tween 20 and Lipophilic: Span 80) to select the suitable surfactant agent for the extraction of optimum bioactive compounds. Between two surfactants, Tween 20 showed highest efficiency at 4 mM concentration to extract total phenolic content (TP), total flavonoid (TF) and antioxidant activity (AA). Tween 20 at 4 mM concentration was fixed for further analysis along with hot water ($90^{\circ}C$) treatment as a control. In our findings, highest TP (118 mg/g), TF (38 mg/g) and AA (76%) was achieved in KW21 and KW22 among the fifteen accessions of Tartary buckwheat. In other way, TP, TF and AA was 200%, 120% and 110% higher in surfactant formulation compared with control treatment, respectively.

Effect of Artemisia anomala S. Moore on Antioxidant Activity and Melanogenesis (유기노의 항산화 활성 및 멜라닌 생성 억제 효과)

  • In mammalian melanocytes, melanin synthesis is controlled by tyrosinase, the key enzyme in the pigment synthesis. In this study, to develop a new whitening agent, we have investigated the antioxidant and the inhibitory effect of Artemisia anomala extract on tyrosinase activity and melanigenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of butanol fraction from A. anomala was higher than that of arbutin ($97.5{\pm}0.5%$ at the concentration of 2 mg/ml). The butanol fraction was shown scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radicals in a dose dependent manner. The highest inhibitory activity of melanogenesis was also butanol fraction ($25.0{\pm}3%$ at the concentration of $200\;{\mu}g/ml$). From these results, we suggest that the A. anomala extract might be used to be a potential agent for skin whitening.
