Hepatoprotective Effect of Green Tea (Camellia sinensis) Extract against Tamoxifen-induced Liver Injury in Rats

  • Tamoxifen citrate (TAM), is widely used for treatment of breast cancer. It showed a degree of hepatic carcinogenesis. The purpose of this study was to elucidate the antioxidant capacity of green tea (Camellia sinensis) extract (GTE) against TAM-induced liver injury. A model of liver injury in female rats was done by intraperitoneal injection of TAM in a dose of $45\;mg\;Kg^{-1}\;day^{-1}$, i.p. for 7 successive days. GTE in the concentration of 1.5%, was orally administered 4 days prior and 14 days after TAM-intoxication as a sole source of drinking water. The antioxidant flavonoid; epicatechin (a component of green tea) was not detectable in liver and blood of rats in either normal control or TAM-intoxicated group, however, TAM intoxication resulted in a significant decrease of its level in liver homogenate of tamoxifen-intoxicated rats. The model of TAM-intoxication elicited significant declines in the antioxidant enzymes (glutathione-S-transferase,glutathione peroxidase, superoxide dismutase and catalase) and reduced glutathione concomitant with significant elevations transaminase) levels. The oral administration of 1.5% GTE to TAM-intoxicated rats, produced significant increments in the antioxidant enzymes and reduced glutathione concomitant with significant decrements in TBARS and liver transaminases levels. The data obtained from this study speculated that 1.5% GTE has the capacity to scavenge free radical and can protect against oxidative stress induced by TAM intoxication. Supplementation of GTE could be useful in alleviating tamoxifen-induced liver injury in rats.

Evaluation of the Antioxidant Activities of Natural Components of Artemisia iwayomogi

  • The antioxidant activities of 29 components isolated from the aerial parts of Artemisia iwayomogi were evaluated in vitro and in cell culture. Among the tested compounds, 2, 6, 8, 10, 13, and 14 exhibited the greatest peroxyl radical-scavenging activities in the oxygen radical absorbance capacity (ORAC) assay, and 2, 10, and 14 also showed significant reducing capacities. However, all compounds showed weak metal chelating activities. Their cellular antioxidant activities were evaluated in HepG2 cells. At $10{\mu}M$, compounds 6, 8, and 14 exhibited stronger protection against 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress than compounds 2, 10, and 13. Moreover, Compounds 2 and 8 were more effective in protecting against $Cu^{2+}$-induced oxidative stress than compounds 6, 10, 13, and 14 at $10{\mu}M$. These results suggest that the phenolic compounds in A. iwayomogi have the potential to be developed as natural antioxidants for the treatment of oxidative stress-related diseases.

Anti-osteoporotic and Antioxidant Activities by Rhizomes of Kaempferia parviflora Wall. ex Baker

  • In this report, we investigated the antioxidant (peroxyl radical-scavenging and reducing capacities) and anti-osteoporotic activities of extracts and isolated constituents (1 - 16) from the rhizomes of Kaempferia parviflora Wall. ex Baker on pre-osteoclastic RAW 264.7 cells. Compound 5 exhibited significant peroxyl radical-scavenging capacity, with TE value of $8.47{\pm}0.52{\mu}M$, while compound 13 showed significant reducing capacity, with CUPRAC value of $5.66{\pm}0.26{\mu}M$, at $10.0{\mu}M$. In addition, flavonoid compounds 2, 4, 6, 8, 10, 12, and terpene compound 15 showed significant inhibition of tartrate-resistant acid phosphatase (TRAP) in NF-${\kappa}B$ ligand-induced osteoclastic RAW 264.7 cells, with values ranging from $16.97{\pm}1.02$ to $64.67{\pm}2.76%$. These results indicated that K. parviflora could be excellent sources for the antioxidant and anti-osteoporotic traditional medicinal plants.

구연산 및 효소 불활성화 처리가 오디술의 C3G 안정성과 항산화능에 미치는 영향 (The effect of citric acid and enzyme inactivation treatment on C3G stability and antioxidant capacity of mulberry fruit alcoholic drink)

  • 안토시아닌 색소는 일반적으로 pH에 따라서 색이 변하는 매우 불안정한 색소이며, 유기용매인 ether, ethyl acetate, acetone에는 녹지 않고 물과 알콜에 잘 녹는 수용성 색소로 알려져 있다. 또한 금속이온과 chelate compound를 형성하여 특유의 색을 띄며, sucrose, glucose, fructose, xylose, fomiate, luvuliniate, 5-hydroxymethylfurfural 및 furfural 등에 의해 분해가 촉진되고, 산소가 없을 때는 안정하나 산소가 존재하면 갈변하는 성질을 갖고 있다. 오디를 첨가하여 술을 제조하는 경우에도 여러 가지 인자에 의해 C3G 색소의 함량 변화 및 갈변 현상이 발생한다. 이를 해결하고 오디의 이용을 촉진하기 위해 C3G 색소를 안정화시킬 수 있는 가공 방법을 개발하고자 하였다. 그 결과를 요약하면 다음과 같다. 1. 냉동된 오디는 전자렌지를 이용하여 3분간 blanching 하는 것이 항산화능을 안정화시키고 색소의 안정성에 관여하는 효소를 불활성화 시킴으로써 저장기간에 따른 C3G 색소의 파괴를 억제시켰다. Sucrose 2.5% 및 citric acid 0.1%를 첨가하는 것이 C3G 함량이 높았다. 2. 냉동건조 오디를 사용할 경우 오디색소 함량과 항산화능이 처리구 중 가장 높았으나 동결건조 비용이 부담되므로 경제적으로 바람직하지 않다. 3. 보관용기(투명유리병, 갈색유리병, 알루미늄 호일, 녹색유리병, 반투명유리병)보다 효소 불활성화 처리 및 구연산의 첨가가 항산화색소 C3G의 안정성에 영향을 미치는 것으로 판단된다.

TOSC 법을 이용한 당알코올의 항산화 활성 평가 (Evaluation of Antioxidant Activity of Sugar Alcohols Using TOSC (Total Oxy-radical Scavenging Capacity) Assay)

  • Although animal and epidemiological studies have suggested oxidative stress as an etiological factor in pathogenesis including cancer, inflammation, sepsis, fibrosis, cardiovascularlneurodegenerative diseases and aging-related disorders, conflicting results have been obtained in clinical trial with antioxidants. The reason for this discrepancy remains unknown but may be due, in part, to the lack of a validated assay system for evaluating antioxidant capacity. The antioxidant activity of a series of sugar alcohols against peroxyl radicals, hydroxyl radicals and peroxynitrites was determined by the total oxy-radical scavenging capacity (TOSC) assay and cell-based assay using H4IIE cells. Specific TOSC values calculated from the slope of the linear regression for erythritol, xylitol, sorbitol or mannitol against peroxyl radicals was $2.1{\pm}0.2,\;3.7{\pm}0.3,\;9.1{\pm}0.3$ or $8.7{\pm}1.1$ TOSC/mM, respectively. Specific TOSC values for erythritol, xylitol, sorbitol or mannitol against peroxynitrite was $1.9{\pm}0.3,\;3.9{\pm}0.4,\;7.8{\pm}0.7$ or $7.7{\pm}0.5$ TOSC/mM, respectively. These results suggest that oxy-radical scavenging capacity is dependent on the number of aliphatic hydroxyl group in sugar alcohols of monosaccharide. Tert-butylhydroperoxide (t-BHP)-induced cell toxicity determined by MTT assay was marginally attenuated by 10 mM erythritol, but completely inhibited by 10 mM xylitol, 2 mM sorbitol or 0.75 mM maltitol, a disaccharide alcohol. Oxidative stress markers, such as glutathione (GSH) and malondial-dehyde (MDA) levels, were measured in t-BHP-treated cells using HPLC equipped with a fluorescence detector and a reverse phase column. Erythritol did not change the levels of GSH and MDA in H411E cells treated with t-BHP. The t-BHP-induced changes in cellular GSH and MDA levels were ameliorated by 10 mM xylitol and completely blocked by 10 mM sorbitol and maltitol. These results indicate that sugar alcohols protect cells against oxidative stress via scavenging oxy-radical and suggest that TOSC assay in conjunction with cell-based assay is a valid method for evaluating antioxidant capacity of natural and synthetic chemicals.

Effects of dietary marigold extract supplementation on growth performance, pigmentation, antioxidant capacity and meat quality in broiler chickens

  • Objective: This experiment was conducted to investigate the effects of dietary supplementation with marigold extract on growth performance, pigmentation, antioxidant capacity and meat quality in broiler chickens. Methods: A total of 320 one-day-old Arbor Acres chickens were randomly divided into 5 groups with 8 replicates of 8 chickens each. The chickens of control group were fed with basal diet and other experimental groups were fed with basal diet supplemented with 0.075%, 0.15%, 0.30%, and 0.60% marigold extract respectively (the corresponding concentrations of lutein were 15, 30, 60, and 120 mg/kg). Results: The results showed that marigold extract supplementation increased the yellowness values of shank, beak, skin and muscle and the redness ($a^*$) value of thigh muscle (linear, p<0.01). Marigold extract supplementation significantly increased the total antioxidant capacity, and the activities of superoxide dismutase in liver and thigh muscle (linear, p<0.01) and significantly decreased the malondialdehyde contents of liver and thigh muscle (linear, p<0.01). Marigold extract supplementation significantly decreased the drip loss and shear force of thigh muscles (linear, p<0.01). There was no significant effect on growth performance with marigold extract supplementation. Conclusion: In conclusion, dietary supplementation of marigold extract significantly increased the yellowness values of carcass, antioxidant capacity and meat quality in broiler chickens.

Agricultural Characters, Phenolic and Nutritional Contents, and Antioxidant Activities of Pigeon Pea (Cajanus cajan) Germplasms Cultivated in the Republic of Korea

  • Pigeon pea (Cajanus cajan) is an important legume species that produces seeds that are rich in phenolic compounds and dietary nutrients. The aim of the present study was to evaluate the agricultural characters, phenolic and nutritional contents, and antioxidant capacities of seeds from 82 pigeon pea germplasms cultivated in the Republic of Korea. The accessions exhibited compact (24.39%), semi-spreading (74.39%), or spreading (1.22%) growth habits and determinate (89.02%), indeterminate (8.54%), or semi-determinate (2.44%) flowering patterns. Days to 75% maturity ranged from 30 to 72 d, and yield per plant ranged from 6.00 to 148.60 g. Meanwhile, total phenolic, crude protein, crude fiber, and dietary fiber contents ranged from 16.42 ± 0.62 to 29.67 ± 0.43 mg gallic acid equivalent per g of dried extract, from 16.76 ± 6.74% to 22.61 ± 0.05%, from 4.70 ± 0.24 to 8.63 ± 0.02%, and from 12.98 ± 0.71 to 33.19 ± 1.50%, respectively. In addition, DPPH radical-scavenging capacity ranged from 1.61 ± 0.10 to 16.04 ± 2.30 mg ascorbic acid equivalent per g of dried extract, and Trolox equivalent antioxidant capacity ranged from 3.03 ± 0.86 to 42.24 ± 0.72 mg Trolox equivalent per g of dried extract. Phenolic content was correlated with both DPPH radical-scavenging capacity (r = 0.63) and Trolox equivalent antioxidant capacity (r = 0.29). Nine accessions (IT170290, IT170291, IT170270, IT170276, IT170379, IT170386, IT170388, IT170418, and IT170340) exhibited early maturity, compact and erect growth habits, and above average antioxidant activities and phenolic and protein contents. In addition, accessions IT170290 and IT170291 were especially promising pigeon pea germplasms to grow, owing to various favorable characteristics (e.g., high yield and dietary fiber content). Hence, these accessions could be useful cultivars to the Republic of Korea if considered in future agricultural systems.

계통별 유색보리의 항산화효과 (Antioxidant Capacity of Colored Barley Extracts by Varieties)

  • 본 연구에서는 기능성이 강화된 유색보리 계통을 선발하고 유색보리 계통 육성 및 활용에 기초자료로 사용하기 위해 유색보리 40계통과 대조군인 춘추쌀보리, 겉보리를 $0.1\%$ TFA를 용매로 사용하여 3회 반복 추출 후 감압 농축 후 anthocyanin 함량, 색도 및 항산화효과를 측정하였다. 유색보리의 anthocyanin 색소함량과 색도를 측정한 결과 계통별로 큰 차이를 보였고, 색의 짙음과 옅음으로 그룹 I ,그룹 II, 그룹 III을 나눌 수 있었다. 계통별로 항산화효과에서도 상당한 차이가 있었으나, 제놀함량이 높았던 유색보리 계통이 전자 공여능, SOD-like 활성, lecithin 산화 저해활성능, hydro-gen radical 소거능이 높음을 알 수 있었으며, 여러가지 항산화실험에서 항산화능이 높게 측정되었던 No. 25, 27, 34, 35, 38이 anthocyanin을 함량이 높은 그룹 I에 포함되어 있어 anthocyanin을 함량과 항산화효과는 정의 상관관계로 나타났다. 따라서, 다른 유색 함유 작물처럼 색깔이 진한 유색보리 품종이 개발되어 다양한 생리활성이 입증된다면 새로운 기능성 식품소재로 활용가능성이 높을 것으로 사료된다.

Comet Assay를 이용한 케일, 명일엽, 당근, 돌미나리 녹즙의 Chinese Hamster Lung 세포 DNA 손상 보호 효과 (Protective Effect of Yellow-Green Vegetable Juices on DNA Damage in Chinese Hamster Lung Cell Using Comet Assay)

  • The present study was attempted to investigate the antioxidant capacity of popular yellow-green vegetable juices (kale, Angelica keishei, carrot, small water dropwort) and to investigate the effect of vegetable juices on protecting oxidative damage to DNA in cultured Chinese hamster lung (CHL) cells. Antioxidant capacity was analyzed by TRAP assay (Total radical-trapping antioxidant potential). Cellular DNA dmamage was measured by SCGE (single-cell gel electrophoresis, also known as comet assay. Cells incubated in medium with PBS (negative control) or with various concentration of the freeze dried green juices (25, 50, 100, 250 $\mu\textrm{g}$/$m\ell$) resuspended in PBS were treated with $H_2O_2$ (200 ${\mu}{\textrm}{m}$) as an oxidative stimulus for 5 min at 4$^{\circ}C$. The physiological function of each vegetable juice on oxidative DNA damage was analyzed and expressed as tail moment (tail length X percentage migrated DNA in tail) . Kale juice had the highest TRAP value suggesting that kale has the highest antioxidant capacity followed by Angelica keishei, small water dropwort and carrot. Cells treated with $H_2O_2$ had extensive DNA damage compared with cells treated with PBS or pre-treated with vegetable juice extracts. All green juices inhibited $H_2O_2$-induced DNA damage with kale being the most effective juice among the tested juices. These results indicate that green juice supplementation to CHL cells followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species. Lysophosphatidylethanolamine (LPE) Improves Fruit Size, Color, Quality and Phytochemical Contents of Sweet Cherry c.v. '0900 Ziraat'

  • Lysophosphatidylethanolamine (LPE) affects the quality of flowers, fruits, and other horticultural products. Studies have provided evidence that LPE can accelerate ripening of fruits and prolong shelf-life at the same time. In this study, the influence of LPE on anthocyanin accumulation and phytochemical characteristics of sweet cherry was investigated. LPE ($10mg{\cdot}L^{-1}$) was applied to a commercial sweet cherry c.v. '0900 Ziraat' orchard two and four weeks before harvest for two treatment years (2011 and 2012). Preharvest applications of LPE resulted in significant improvement in both pomological and phytochemical attributes at harvest. LPE treatment led to a 17% increase in fruit weight and a 6% increase in soluble solid content when averaged over two experimental years. Fruit phytochemical content and antioxidant capacity were increased significantly. The average total phenolic content of LPE-treated fruits for the two years was $703{\mu}g$ gallic acid equivalent (GAE)/g fresh weight (g FW) compared to $569{\mu}g$ GAE/g FW in the untreated control. Fruits treated with LPE had a 27% and 16% more anthocyanin than the control fruits in 2011 and 2012. Antioxidant capacity of fruits, as measured by TEAC (Trolox equivalent antioxidant capacity) assay, was 12.5 and $11.4{\mu}mol$ TE/g FW in LPE-treated and untreated control fruits, respectively, when averaged over two experimental years. Our results suggest that preharvest application of LPE may have the potential to increase anthocyanin accumulation, improve fruit quality and enhance phytochemical characteristics of sweet cherries.
