각종 변이원에 대한 쇠비름 추출물의 돌연변이 억제 효과 (Inhibitory Effects of Poturaca oleracea L. Extract on the Mutagenicity of Various Mutagen)

  • This study was performed to determine the effects of antimutagenicity of Porturaca oleracea L. in Korea. In Ames test, the ethanol extract of Poturaca oleracea L. inhibited mutagenic activity of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), , 4-nitroquinoline-1-oxide(4NQO), benzo($\alpha$)pyrene (B($\alpha$)P) and 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indole (Trp-P-1) in Salmonella typhimurium TA98 and TA100. But hot-water extract Poturaca oleracea L. only Inhibited mutagenic activity of MNNG in Salmonella typhimurium TA100, On 4NQO, the ethanol extract 100-1,600$\mu\textrm{g}$/plate of Porturaca oleracea L. showed a slight inhibitory effect of 13-48%, 4-47% in TA98 and TA100, respectively, but on MNNG, it showed higher inhibitory effect of 6-86% in TA100, And the treatment of 1,600$\mu\textrm{g}$/plate of ethanol extract of Porturacea L. had strong antimutagenicity with 74-87% inhibition against TA98 and TA100 induced by B(a)P and with 85-93% inhibition against TA98 and TA100 induced by Trp-P-1. The ethanol extract was fractionated with ether. chloroform, ethylacetate, butanol and water. Among them, most of the fraction except water fraction showed strong antimutagenicity effects against mutation induced by 4-NQO, MNNG, B(a)P and Trp-P-1. Chloroform fraction had strong antimutagenicity with 91% inhibition against TA100 induced by MNNG, diethyl etherfraction had strong antimutagenicity with 92%, 98% inhibition against TA98 and TA100 induced by 4NQO, Chloroform fraction had strong antimutagenicity with 97% inhibition against TA100 induced by B (a)P and diethyl etherfraction had strong antimutagenicity with 98% inhibition against both strain Induced by Trp-P-1, respectively.

  • 마(산마, 재배마)의 생리활성 물질의 기능성을 밝힐 목적으로 마의 갈변물질, 점질물 및 식이섬유를 분리 정제하여 항변이원성을 검색하여 다음과 같은 결과를 얻었다. 1. 마의 갈변효소를 cathol, hydroquinion, resorcinol, pyrogallol 등에 반응시켜 얻은 갈변물질의 항변이원성은 sodium azide에 대하여 기질에 따른 차이가 없었으나, 2-aminofloureno에 대해서는 모든 기질에서 유의적으로 높았다. (P<0.01) 2. 마의 점질물과 식이섬유($\alpha$-cellulose, hemicellulose, pectin) 의 sodium azide와 2-ami-noflourene에 대한 항변이원성은 $\alpha$-cellulose와 hemicellulose에서 유의적으로 높았으며(p<0.01), $\alpha$-cellulose와 흡착 반응시킨 2-amino-flourene에 대한 항변이원성은 5시간 반응시 가장 높았고 반응시간이 경과함에 따라 감소하였다.

보리(Hordeum vulgare) 추출물의 항변이원성 (Studies on the Antimutagenicity of extract from Barley (Hordeum vulgare))

  • This study was performed to determine the effects of antimutagenicity from Barley (Hordeum vulgare). In Salmonella typhimurium reversion assay (In vitro test), the extract of Barley (Hordeum vulgare) inhibited mutagenic activity of 4-NQO and Trp-p-1 with 59 mix. in Salmonella typhimurium TA98 and TA100. In Micronucleus test (In vivo test), the methanol extract of Barley (Hordeum vulgare) inhibited micronucleus formation in bone marrow by cyclophosphamide. The $\beta$-glucan of Barley (Hordeum vulgare) showed inhibitory effects of 59-77% in mutagenic activity of 4-NQO by Salmonella typhimurium TA100. The mutagenicity of Trp-p-1 with S9 mix. by Salmonella typhimurium TA98 showed inhibitory effects of 24-56%. The methanl extract (M) was fractionated with ether (MI), ethylacetate (M2), buthanol (M3) and water (M4). The Antimutagenicity of Trp-p-1 with 59 mix. by Salmonella typhimurium TA98 in Barley fraction showed the following: methanol extract (99.58%)>ether fraction (98.05%)>buthanol fraction (56.90%)>water fraction (56.72%)>ethyl acetate fraction (28.72%). Among them, ether fraction in TA 98 showed strong antimutagenicity effects (85.56%, 98.05%) against mutation induced by 4-NQO and Trp-p-1. As concentration of the methanol extract increased (1.25~5 g/kg/10 cc), micronucleus formation in bone marrow by chemical mutagen (CP) showed inhibitory effects of 50% (p< 0.05).

노루궁뎅이 버섯 열수 추출물의 이화학적 특성 및 항산화성, 항돌연변이성, cytotoxicity 분석 (Physicochemical Characteristics and Antioxidant activity, Antimutagenicity, and Cytotoxicity of Hot-water Extract of Hericium erinaceus)

  • The physicochemical characteristics and biological activities, including antioxidant activity, antimutagenicity, and cytotoxicity of hot-water extract of fruiting body of Hericium erinaceus, were investigated in this study. Hot-water extract of fruiting body of Hericium erinaceus contained carbohydrate (7.86%), protein (10.91%), and ${\beta}$-glucan (3.62%). Water solubility of hot-water extract was 42.58%. Antioxidant activities of the extract were evaluated by ABTS assay and FRAP assay. The $IC_{50}$ value was 312.21 ${\mu}g/mL$ in ABTS assay. Antimutagenic activity of the extract was evaluated by Ames test. Antimutagenicity of hot-water extract (5 mg/mL) on Salmonella Typhimurium TA100 mutagenated by sodium azide (0.15 ${\mu}g/mg$) was 69.2%. Cytotoxicity of hot-water extract was also evaluated by MTT and SRB assay. The cytotoxicity was highest (83.95%) on Hep3B treated with 2,000 ${\mu}g/mL$ of hot-water extract in SRB assay. Therefore, it is suggested that hot-water extract of fruiting body of Hericium erinaceus has high antioxidant activity, antimutagenicity, and cytotoxicity.

Purification and Evaluation of Rice Bran Hydrolysates with Antimutagenicity

  • A 3% suspension of heat-stabilized defatted rice bran was treated with papain, followed by inactivating the enzyme by heat, and centrifuged. The supernatant was subjected to ultrafiltration, and fractions with various molecular sizes, F1 (>30 kDa), F2 (10-30 kDa), F3 (5-10 kDa), F4 (3-5 kDa), and F5 (3 kDa<), were freeze-dried, and evaluated for antimutagenicity by Ames test using Salmonella typhimurium TA 100 against phenazine methosulfate. The F3 fraction containing highest antimutagenicity from ultrafiltration was separated into 6 fractions by DEAE-Sephadex A-25 ion-exchange column chromatography (F3-1-F3-6). Each fractions having protein contents were pooled, dialyzed, freeze dried, and evaluated for antimutagenicity. Among the six fractions, the F3-1, F3-2, and F3-6 fractions showed antimutagenicity, which were 80.2, 53.4, and 58.6% at concentration of $100\;{\mu}g/plate$, respectively. These F3-1, F3-2, and F3-6 fractions were subjected to Sephadex G-50 gel filtration column chromatography for further purification. Among the purified fractions, the F3-1-1, F3-2-2, and F3-6-1 fractions showed antimutagenicity of 84.5, 58.6, and 69.8% at concentration of $100\;{\mu}g/plate$, respectively. It is thought that these peptides can find application for nutraceutical and pharmaceutical products.

Bile and Acid Tolerance of Lactic Acid Bacteria Isolated from Dadih and Their Antimutagenicity against Mutagenic Heated Tauco

  • Antimutagenicity of milk cultured with lactic acid bacteria isolated from dadih on the mutagenicity of heated salty and sweet tauco was examined using streptomycin dependent (SD) 510 strain of Salmonella typhimurium TA 98 as a tester culture. Cultured milk samples exhibited widely antimutagenic activity against mutagenic heated salty and sweet tauco. Lc. lactis subsp. lactis R-22, Lc. lactis subsp. casei R-35, Lc. lactis subsp. casei R-52 and E. faecalis subsp. liquefaciens R-55 exhibited no inhibitory effect on the mutagenic heated salty tauco. Mutagenicity of heated sweet tauco was inhibited by cultured milks stronger than that of heated salty tauco. Milk cultured with Lc. lactis subsp. cremoris R-48, Leuc. mesentroides R-51 and Lc. lactis subsp. casei R-68 showed high inhibition against the mutagenicity of both heated salty and sweet taucos. Antimutagenic activity of the cultured milks against mutagenic heated tauco was attributed to the bacterial cells. Among the three strains which showed high antimutagenicity, only Leuc. mesentroides R-51 was tolerant to both acid and bile; so this strain can be used as probiotic in preventing the occurrence of mutagenesis caused by mutagenic heated food like tauco.

대두 사포닌이 Aflatoxin B1으로 유도된 세포돌연변이에 미치는 영향에 관한 연구 (Effect of Soybean Saponins on Aflatoxin B1-induced Mutagenicity)

  • Free radicals formed during the metabolism of environmental chemicals are known to induce mutagenicity, while different types of antioxidants suppress this event. The purpose of this study was to determine the antioxidative and antimutagenic effects of soybean saponins, and to examine the relationship between these two effects for the elucidation of mechanisms involved in the anticarcinogenicity of soybean saponins. Also, antioxidative and antimutagenic effects of soybean saponins were compared with those of kinown antioxidants. For the measurement of antioxidative capacity, soybean saponins, L-ascorbic acid, $\alpha$-tocophoerol, and BHT at concentrations between 005 and 1.0mg/ml were tested for their ability to donate hydrogens and to reduce the formation of thiobarbituric substances(TBARS). Antimutagenic activity was examined using the Ames salmonella test system at concentrations of 600, 900 or 1200ug/ml. Study results showed soybean saponins and all of the other antioxidants tested possessed dose-dependent antioxidative activities. The ability of hydrogen-donation to DPPH was in the order of L-ascorbic acid>$\alpha$-tocopherol=>BHT>soybean saponins. TBARS formation was also inhibited by these compounds, in the order of BHT>$\alpha$-tocopherol=L-ascorbic acid>soybean saponins. Soybean saponins and other antioxidants also showed antimutagenicity in a dose-dependent manner. Especially, soybean saponins and BHT were excellent antioxidants compounds, inhibiting near 80% of the mutagenic effects at a concentration of 1200ug/ml. The correlation coefficients between antioxidative capacity and antimutagenicity for each compund was statistically significant at p<0.05. These results indicate that soybean saponins possess antioxidative and antimutagenic capacities. Also, antimutagenicity of saponins and other antioxidats is partly due to their antioxidative activities.

한방 소갈약 추출물의 혼합물에 의한 항돌연변이 효과 (Antimutagenic Effect of the Extract Complex of Korean Anti-thirst Herb Medicines)

  • The three crude drugs of the Kalopanax pictus (Araliaceae) roots (K), Pueraria thunbergiana (Leguminosae) flowers (P), and the Rhus verniciflua (Anacardiaceae) heartwood(R) used for anti-thirst drugs in Oriental herb medicine were extracted with MeOH, respectively, and the successive fractionation of the extract gave EtOAc extract. Certain amount ratios of the three extracts were also prepared to compare the antimutagenicity in Ames test. In N-methyl-N(-nitro-N-nitrosoguanidine (MNNG; 0.4 (g/plate)-induced test, the activities of complex mixture were observed between the highest antimutagenic activity of K extract and the lowest P extract. In aflatoxin (AFB$_1$)-induced test, the EtOAc complex (K : P : R=l : 1 : 3) labeled E-113 decreased the revertants of Salmonella typhimurium TA100 by 95%, which activity were highest among other extracts or complexes mixture used. Fractionation of organic solvent mostly increased the antimutagenicity. These trends were also observed in the antimutagenicity test of the mixture of each active component of kalopanaxsaponin A, tectorigenin and sulfuretin. These results supported that many kinds of anti-thirst herb medicine in the prescription could effectively prevent cancer disease.

Melanoidin의 항산화성 및 항돌연변이원성 (Antioxidative and Antimutagenic Characteristics of Melanoidin Related Products)

  • Melanoidins, as brown colored polymers, are formed through the diversified reaction systems of Maillard type and other reactions. Melanoidins are important components in relation to food quality and also are known to have antioxidative, mutagenic and antimutagenic activities. Since these aspects have been extensively reviwed elsewhere, only the recent studies regarding to their antioxidative and antimutagenic activities are discussed in this review. Even though their mechanisms are not clearly identified, melanoidins or specific fractions isolated from their mixtures have shown varied antioxidative activities depending on the reaction systems and reaction conditions. Those activities presumely are derived from the complex functional properties of hydrogen / electron donors and metal chelating power, which are originated from their reductone structure and others. It is considered that pyrolysate and other mutagens are formed by the given conditions in some cases during browning reaction, whereas melanoidins and their fractions have antimutagenic effects on chemical and other mutagens. There are positive correlationship among the color intensity, antioxidative activity and antimutagenicity of melanoidins or their fractions. These suggest that the antimutagenicity of melanoidins could be attributed to their antioxidative properties, however, it might also be due to other factors, because the relevant responses for antimutagenicity are very complicate and not clear. Accordingly, further studies are required to determine the actual acitivities and mechanisms involved in antioxidation and (anti)mutagenicity of melanoidins by reaction systems / conditions and by the isolated fractions. And also, additional studies are needed to evaluate the applications of melanoidins and their relevant effects to food and human health.

In vivo Antimutagenicity of Dadih Probiotic Bacteria towards Trp-P1

  • In vitro acid- and bile-tolerant lactic acid bacteria isolated and identified from Indonesian traditional fermented milk dadih might be considered as potential probiotic strains after further characterization with animal models, especially for their therapeutic properties. Five dadih lactic bacteria isolates each had moderate survival rate for 2 h at pH 2.0, as well as bile tolerance. The aim of this research was to identify candidate probiotic lactic bacteria among indigenous dadih lactic isolates originated from Bukit Tinggi, West Sumatra, especially their in vivo antimutagenic property. Milk cultured with Enterococcus faecium IS-27526 significantly lowered fecal mutagenicity of rats as compared to the control group, skim milk, and milk cultured with L. plantarum IS-20506. These results suggest that Enterococcus faecium IS-27526 may serve as a potential probiotic strain with its antimutagenicity.
