Virulence genes and antimicrobial resistance of pathogenic Escherichia coli isolated from diarrheic weaned piglets in Korea

  • For efficient prevention and treatment of enteric colibacillosis, understanding about latest virulence factors and antimicrobial resistance of Escherichia coli is essentially needed. The aim of this study was to survey antimicrobial resistance and determine the prevalence of fimbriae and enterotoxin genes among 118 pathogenic E. coli isolates obtained from Korean pigs with diarrhea between 2016 and 2017. The genes for the toxins and adhesins were amplified by polymerase chain reaction (PCR). The susceptibility of the E. coli isolates to antimicrobials were tested using the standard Kirby-Bauer disk diffusion method. The most prevalent fimbrial antigen was F18 (40.7%), followed by F4 (16.9%), and the most prevalent combinations of toxin genes were Stx2e (21.2%), STb:EAST-1 (19.5%), and STa:STb (16.9%), respectively. Among the pathotypes, enterotoxigenic E. coli (ETEC) was the most predominant (67.8%), followed by Shiga-toxin producing E. coli (STEC, 23.7%). We confirmed high resistance rates to chloramphenicol (88.1%), tetracycline (86.4%), streptomycin (86.4%), and ampicillin (86.4%). And the majorities of isolates (90.7%) showed multi-drug resistance which means having resistance to 3 or more subclasses of antimicrobials. Results of this study can be a source of valuable data for investigating the epidemiology of and control measures for enteric colibacillosis in Korean piggeries.

  • E coli strains isolated from pigs were investigated with respect to antimicrobial resistances and prevalence of R-plasmids. Also determined were properties of R-plasmids by plasmid conjugation, curing and southern hybridization using gene probes. All of 400 E coli strains were resistant to CL and SU, and 0.3% to 96.8% of the strains were resistant to most antimicrobials such as TC, PG, AM, SM, CP, GM, EM, NM, etc, while all strains were sensitive to AK. All strains were also multiply resistant to three to twelve antimicrobials. The resistances to PG, SM, TC, AM, CP, SU and ST were transferable and supposed to be mediated by R-plasmids which were opportunistic for transposition into chromosome. Plasmids bigger in size than chromosomal DNA were considered as R-plasmids and most plasmids in small size (<4Kb) proved as cryptic plasmids or nonconjugative R-plasmids. In a strain(No 99), AM resistant property was determined from both chromosomal DNA and R-plasmid DNA which is bigger in size than chromosome.

  • Species and antimicrobial susceptibility of nonfermenting gram-negative bacilli(NFB) isolated from clinical materials at Yonsei Medical Center during the period of September 1980 to August 1981 were analyzed and the following results were obtained. 1. NFB were isolated from 17.1% of sputum, 14.8% of pus, 5.0% of urine, 3.3% of throat and 1.4% of blood specimens. 2. Among the isolates 57.6% were Pseudomonas aeruginosa and 32.7% were Acinetobacter calcoaceticus. P. maltophilia and P. cepacia were 3.2% and 2.8% respectively. Other bacteria were rarely isolated. 3. The monthly isolation rates were high during the June to November period(8.8-12.0%), and low during the December to May period(4.2-8.4%). 4. Many of the isolates showed resistance to various antimicrobials. Although there were variations depending on the bacterial species, amikacin, colistin, gentamicin, tobramycin and co-trimoxazole showed more antibacterial activities than other antimicrobials.

  • The emergence of antibiotic-resistant pathogens is a Serious clinical problem in the treatment of infectious diseases that increase mortality, morbidity, hospitalization length, and the cost of healthcare. In particular, $Streptococcus$ $pneumoniae$ is a major etiologic pathogen of pneumonia, sinusitis, otitis media, and meningitis. As the definition of penicillin resistance to $S.$ $pneumoniae$ was recently changed, macrolide-resistant $S.$ $pneumoniae$ is a major resistant pathogen in the community. Infections caused by antibiotic-resistant strains are associated with incorrect use of antibiotics and critical clinical outcomes. For the appropriate use of antibiotics to treat infections, physicians always should have up-to-date information on the current epidemiologic status of antibiotic resistance for common pathogens and their susceptibility to antimicrobials. Appropriate selection of antimicrobials, strict control of infection, vaccination, and development of a feasible national policy of infection control are important strategies for the control of antimicrobial resistance. This review article focuses on the current status of antibiotic-resistant $S.$ $pneumoniae$ in community-acquired pneumonia in Korea.

Antimicrobial Resistance and Virulence Genes Presence in Escherichia coli Strains Isolated from Gomso Bay, Korea

  • In total, 131 Escherichia coli isolates from surface seawater of the Gomso Bay, of Korea, were analyzed for their susceptibility to 22 different antimicrobials and for genes associated with antimicrobial resistance and virulence. According to the disk diffusion susceptibility test, the resistance to tetracycline was most prevalent (33.6%), followed by that to ampicillin (22.1%), ticarcillin (22.1%), and trimethoprim (16.8%). More than 46.6% of the isolates were resistant to at least one antimicrobial, and 22.9% were resistant to three or more classes of antimicrobials; these were consequently defined as multidrug resistant. We further found that 29 ampicillin-resistant isolates possessed genes encoding TEM-type (93.1%) and SHV-type (6.9%) ${\beta}$-lactamases. Among the 44 tetracycline-resistant isolates, tetA and tetC were found in 35 (79.5%) and 19 (43.2%), respectively, whereas tetB was detected in only three isolates (6.8%). With regard to virulence genes, merely 0.8% (n = 1) and 2.3% (n = 3) of the isolates were positive for the enteroaggregative E. coli-associated plasmid (pCVD432) gene and the enteropathogenic E. coli-specific attaching and effacing (eae) gene, respectively. Overall, these results not only provide novel insight into the necessity for seawater sanitation in Gomso Bay, but they help reduce the risk of contamination of antimicrobial-resistant bacteria.

개와 고양이에서 분리한 methicillin 내성 및 감수성 Staphylococcus pseudintermedius (Methicillin-resistant or susceptible Staphylococcus pseudintermedius isolates from dogs and cats)

  • Staphylococcus pseudintermedius is an important opportunistic pathogen of dog and cats. Since 2006 there has been a significant emergence of methicillin-resistant S. pseudintermedius (MRSP) mainly due to clonal spread. The aim of this study was to investigated the prevalence of antibiotic resistance and presence of mecA and femA gene in 91 S. pseudintermedius isolates isolated from dogs and cats associated with various clinic infections. Methicillin resistance was confirmed by oxacillin disc diffusion method. MRSP isolate was detected 19 isolates (20.9%). MRSP and methicillin-resistant S. pseudintermedius (MSSP) isolates were highly resistant to penicillin, kanamycin, tetracycline, erythromycin, trimethoprim-sulfamethoxazole, clindamycin, ciprofloxacin, enrofloxacin and choloramphenicol (100~47.3% and 90.3~33.3%, respectively). About 90% of MRSP isolates were multi-drug resistance (resistance to at least five or more antimicrobials), and MSSP isolates was ca 74%. Among the 91 isolates, mecA gene was detected in 25 isolates (27.5%, 19 in MRSP isolates and 6 in MSSP isolates), but none carried the femA gene. Our results indicated MRSA isolates show a strong resistance to antimicrobials commonly used in veterinary medicine. A continuous surveillance and monitoring should be called for to prevent the contamination and spread of MRSP in dogs and cats.

In vitro에서 Edwardsiella tarda와 Streptococcus iniae에 대한 항균제와 천연 유래 항균물질의 병용효과 (In vitro combination effects of natural substances and antimicrobials against Edwardsiella tarda and Streptococcus iniae)

  • The purpose of this study is to explore the possibility of essential oils and flavonoids improving the performance of antibiotics, proving a higher efficacy in the presence of natural substances against Edwardsiella tarda and Streptococcus iniae. Increased efficacy was observed in several cases: both essential oils and flavonoids increased antibacterial efficacy of oxytetracycline and flumequine against E. tarda by 2-fold; essential oils and flavonoids increased antibacterial efficacy of nalidixic acid against E. tarda by 4-fold; flavonoids increased antibacterial efficacy of josamycin against S. iniae by 4-fold. The results obtained in this study suggest that essential oils and flavonoids might be especially useful in increasing the antimicrobial activity of nalidixic acid and josamycin against E. tarda and S. iniae in fish. Natural compounds examined in this study could also be useful to help decrease the overuse of antibiotics in fish.

Antimicrobial Susceptibility of Coagulase-Negative Staphylococci Isolated from Bovine Mastitis Between 2003 and 2008 in Korea

  • A total of 1,444 coagulase-negative staphylococci (CNS) isolates from bovine mastitic milk samples collected during 2003-2008 in Korea were identified to the species level. Of 14 species identified, S. simulans, S. haemolyticus, and S. sciuri accounted for over 60% of the isolates. All the CNS isolates were tested for susceptibility to eight antimicrobials commonly used in dairy cattle. With a few exceptions, similar resistance patterns were observed among the CNS species: penicillin and ampicillin showed the lowest activity, whereas amikacin, cephalothin, and gentamicin were highly effective. About 39% (557/1,444) of the CNS isolates were pan-susceptible, whereas 12% (175/1,444) showed resistance to four or more antimicrobials tested.

전북지역 소 설사유래 병원성대장균 감염실태 조사 (Prevalence of enterovirulent Escherichia coli from diarrhea of cattles in Jeonbuk, Korea)

  • Enterovirulent Escherichia coli are among the most important causes of diarrhea in cattles. Between January and December, 2017, a total of 150 stool specimens from cattles were investigated for enterohaemorrhagic E. coli (EHEC), enteropathogenic E. coli (EPEC), enteroaggregative E. coli (EAEC), enterotoxigenic E. coli (ETEC) and enteroinvasive E. coli (EIEC) using real-time PCR. 131 E. coli were isolated from feces. The most frequently isolated pathotype in feces was EHEC (37 isolates). EPEC, ETEC and EAEC were detected in feces with 14, 7 and 3 respectively. EIEC was not detected. Antimicrobial resistance test was performed by agar disc diffusion method with 14 antimicrobials. Enterovirulent E. coli isolates showed the highest antimicrobial resistance to ampicillin 61.3%, followed by tetracycline 54.5% and streptomycin 45.5%. They had low resistance to amikacin 11.4%. Of 44 isolates, 37 (84.1%) were resistant to more than 2 antimicrobials. futher study a highest antimicrobial susceptibility to trimethoprim/sulfamethoxazole 50.0% and florofenicol 47.7%.

시판 젓갈에서 분리한 Bacillus cereus의 독소 유전자 및 항균제 내성 분석 (Profiles of Toxin Genes and Antimicrobial Resistance of Bacillus cereus Strains Isolated from Commercial Jeotgal)

  • Twenty-three Bacillus cereus strain isolated from commercial jeotgal were investigated for 11 toxin genes and susceptibility to 25 different antimicrobials. The hemolytic enterotoxins hblA, hblC, and hblD were detected in 13.0%, and non-hemolytic enterotoxins nheA, nheB, and nheC were detected in 26.1%, 100%, and 100% of the isolates, respectively. The positive rates of cytK, entFM, becT, hlyII, and ces were 73.9%, 60.9%, 26.1%, 8.7%, and 0.0%, respectively. According to the disk diffusion susceptibility test, all of the strains studied were resistant to cefuroxime, followed by cefoxitin (78.3%), oxacillin (78.3%), ampicillin (69.6%), penicillin G (69.6%), and amoxicillin (65.2%). However, all the strains were susceptible to 11 other antimicrobials, including amikacin, chloramphenicol, and ciprofloxacin. The average minimum inhibitory concentrations of amoxicillin, ampicillin, and cefuroxime against B. cereus were 462.9, 235.0, and 135.0 ㎍/mL, respectively. These results highlight the need for sanitizing commercial jeotgal, and provide evidence to help reduce the risk of jeotgal contamination by antimicrobial-resistant bacteria.
