HPLC 및 신속검출방법을 이용한 우유내 Sulfamethazine의 분석에 관한 연구 (Determination of Sulfamethazine Using High Performance Liquid Chromatography and Several Screening Methods)

  • Sulfonamides, a therapeutically important group of antimicrobial drugs, are widely used to treat and prevent the acute systemic and skin infections in dairy cattle. They also pose an economic hazard through inhibition of growth of dairy starter cultures. This study was carried out to compare four screening methods for detection of sulfamethazine in milk and determine the positive milk sample by HPLC method. Sulfamethazine was used to spike at five levels of sulfamethazine. The Lac-Tek test and CharmII test were also consistent better than TTCII test and Delvo SP test in sulfamethazine detection. Analysis probabilities of obtaining a positive response with TTCII test and Delvo SP test assay at 50 ppb sulfamethazine level in milk samples were only 14%, 42% each. Whereas using the Lac-Tek test and CharmII test would have resulted in 100% identification of the five levels. Determination of sufamethazine using the HPLC method in the spiked milk were 10.64, 19.30, 30.76,38.83 and 50.23 ppb, respectively.

  • 항균제내성(抗菌劑耐性)인 Salmonella typhi를 공시(供試)하여 chloramphenicol(Cm), tetracycline(Tc), ampicillin(Ap), kanamycin(Km) 및 rifampicin(Rif)의 항균작용(抗菌作用)을 한천평판희석법(寒天平板稀釋法)(평판법(平板法))과 broth 희석법(稀釋法)(broth법(法))으로 비교(比較)하였다. 공시균(供試菌)의 최소발육저지농도(最少發育沮止濃度)(MIC)를 보면 Cm, Tc 및 Rif에 있어서는 평판법(平板法)과 broth법(法) 사이에 유의(留意)할만한 차이(差異)는 없었으나 Km에 있어서는 broth법(法)의 MIC가 평판법(平板法)보다 높은 것이 약(約) 반수(半數)있었고 Ap에 있어서도 소수(少數) 있었다. Cm과 Rif는 대부분(大部分)의 공시균(供試菌)에 대(對)하여 정균적(靜菌的)으로 작용(作用)하였으나 Tc, Ap 및 Km은 살균적(殺菌的)으로 작용(作用)하였고 그 작용(作用)은 균주(菌株)에 따라 차이(差異)가 있었다.

  • The cosmetic and toiletries are necessary health care & household for common life. However we need antiseptic which is effecting harmlessly to the human body. There are propolis, Lavender, Lemon, essential oil in the natural antiseptic materials. This work proceeded design Natural-antiseptic system with three materials as above-mentioned. Natural-antiseptic system was accomplished with propolis (2%), Lavender essential oil (0.3%), Lemon essential oil (0.3%) safety out of Polysorbate 20 (0.5%), Polysorbate 80 (0.5%), PEG (60) hydrogenated castor oil (0.45%), ethanol (5%). The antimicrobial test was experimented on E. coli and Bacillus stearothermophilus. In this antimicrobial test, we found that the effect of antisepsis against E. coli and Bacillus stearothermophilus with propolis 0.3%, Lavender essential oil 0.045% and Lemon essential oil 0.045% was improved. Therefore could expect Natural-antiseptic system product for moisturizing skin toner for face, nourishing essence and wet tissue for clean other things.

Identification of the Bacteria Isolated from Oral Cavities in Korea

  • The aim of this study was to identify bacteria isolated from the oral cavities and to determine their antimicrobial susceptibility against eight antibiotics. The bacterial strains were obtained from the Korean Collection for Oral Microbiology (KCOM). The bacteria were identified by comparing 16S rDNA sequences at the species level. The data showed that 77 bacterial strains were predominantly identified as streptococci (49.4%) and staphylococci (14.3%). Minimum inhibitory concentrations (MIC) were determined using a broth dilution assay to test the sensitivity of the bacterial strains. The MIC values of the oral bacterial strains against antibiotics were different. Streptococci were sensitive to clindamycin, cefuroxime axetil, and vancomycin, and they were resistant to tetracycline. Staphylococci also were sensitive to clindamycin, cefuroxime axetil, and vancomycin, and they were resistant to penicillin antibiotics. Gramnegative bacterial strains were sensitive to tetracycline and were resistant to clindamycin. These results suggest that the antimicrobial susceptibility test is necessary in deciding the prescription for antibiotics, to prevent the misuse or abuse of antibiotics.

강원 동해안 지역 돼지 폐렴에서 분리한 puteurella multocida 에 대한 조사 (A Survey of Puteurella Multocida Isolated from Pigs Affected with Pneumonia in Eastern Kangwon)

  • The present study was conducted to investigate the incidence of pasteurella multocida(p. multocida) infection and some properties of the isolated organisms from the swine herds in Eastern Kangwon during the periods from March 1993 to November 1993. The results obtained were summerized as follows: 1. The lungs of 180 slaughtered pigs were sampled and p. multocida was isolated from 38 pigs (21.1%) and cultured positive. 2. The majority of biochemical md cultural properties of the p. multocida isolates were identical to those of the standard strains. 3. We investigated the capsular serogroup and drug susceptibility of 38 Isolates of p. multocida from pigs with pneumonic lesions 4. p. multocida isolateds were typed for capsular serogroupes A by hyaluronidase inhibition of capsule and D by acriflavin auto agglutination. Most isolates(55.3%) were type A, 15.8% were type D, and the remaining 28.9% were untypable 5. In antimicrobial susceptibility test these isolates of p. multocida were susceptible in order of colistin(94.7%), ampicillin(94.7%), cepalothin(92.1%). gentamicin(92.1%), amikacin(89.5%), but the majority of them were resistant in order of neomycin(26.3%), teracycline(23.7%), streptomycin( 15.8%)

레이온계 여름용 직물에서 매염제 종류에 따른 오배자 염색의 색채발현 및 기능성에 관한 연구 (Coloring Effect and Functionality of Rayon Based Cool Fabrics Treated by Various Mordant and Gallnut Extract)

  • Gallnuts are known to exert various pharmaceutical properties that include anti-inflammatory, antimicrobial, antioxidant, and detoxifying effects. Gallnut extract is thought to be a particularly safe antimicrobial agent for textile application due to its natural origins. Hence, textiles were dyed with gallnut to develop multi-functional clothing material with no harmful effects. In this study, three kinds of cool touch woven fabrics, each comprised of 100% viscose rayon filament, viscose rayon filament/high absorption & quick dry (polyester) filament, and viscose rayon $filament/tencel^{(R)}$ spun yarn, were prepared and used as substrates for gallnut dyeing to especially develop functional cool touch textiles for summer clothing material. On the other hand, gallnut dyeing was conducted with mordanting with iron, lime or alum, which induced different colors on the dyed textiles. Dyed textiles were then investigated by SEM, FTIR, antibacterial test, and antioxidant test. Subsequently, gallnut dyed textiles indicated excellent antibacterial ability and deodorization activity regardless of mordant species. However, only gallnut dyed textiles mordanted with iron and alum showed significant antioxidant ability. In addition, the dyeing processes impair the cool touch feeling of the textiles by changing the micro structure of the textile surface.

Cytotoxicity and antimicrobial effects of the methanolic extract of Sophora flavescens Ait. (IV)

  • This study was carried out to evaluate cytotoxicity of the methanol extract from Sophora flavescens Ait. against L1210 (lymphocytic leukemia) and $P388D_1$ (lymphoid neoplasma) Cells in vitro. We have determined cytotoxicity by the MTT (3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H- tetrazolium bromide) assay. The order of cytotoxicity of Sophora flavescens Ait. extracts against L1210 and $P388D_1$ cells in vitro is as follows: Fr. 4 > Fr. 3 > Fr. 5 > Fr. 2 > Fr. 1. These results suggest that the fraction 4 of the methanol extracts from Sophora flavescens Ait. may be a valuable choice for the development of antitumor agents. In order to develop an antimicrobial agent, dried Sophora flavescens Ait. was extracted with hot methanol, and then antimicrobial activity (MIC test) was investigated. In this study, the fraction 3 of the methanol extracts from the roots of S. flavescens showed strong growth inhibition activity against gram-positive and gram-negative bacteria (MIC, $3.125\;{\mu}g/ml$) such as S. mutans, S. epidermidis and P. putida. These results indicate that fractions 3 and 4 inhibit tumor cells and bacteria.

Antimicribial and Antioxidant Activities of Ethanol Extracts of Medicinal Plants

  • The objective of this study was to determine the radical scavenging activity, total phenolic content, antimicrobial activity, minimum inhibitory concentration (MIC) of ethanol extracts of 32 medical plant species that have been commonly used in medicinal plants. Total phenolic index of T. chebula exhibited the highest value (498.01㎎/g), followed by R. coreanus miquel (400.33㎎/g), Sanguisorba officinalis (368.25㎎/g), P. thumbergiana (259.74㎎/g) and Eugenia aromaticum (229.38㎎/g). Radical scavenging activity for the DPPH radical was highest in T. chebula (40.91%, p<0.01), followed by C. sappan (36.50%), S. officinalis (32.92%), R. coreanus miquel (26.54%) and P. thumbergiana (24.50%). The extracts from T. chebula, R. coreanus muquel, C. sappan, E. aromaticum, S. officinalis and C. japonica possessed outstanding antimicrobial activity against Escherichia coli, Salmonella Typhimurium, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis and Lactobacillus plantarum. MIC was determined on those extracts that showed high efficacy against the test organisms. The most potent MIC values were seen for T. chebula extract against P. aeruginosa, S. aurusa, E. coli, B. subtilis, L. plantarum and S. Typhimurium at 7.8, 7.8, 15.6, 7.8, 125 and 31.2㎍/mL, respectivley. Furthermore, the total phenolic content and radical scavenging activity were very closely correlated for all samples (r=0.78). The coefficient correlations between total phenolic index and antimicrobial activity were 0.91 (E. coli), 0.91 (B. subtillis), 0.79 (P. aeruginosa), 0.79 (S. Typhimurium) and 0.70 (L. plantarum).

Simplified the Screening and In Vitro Appraisal of Antioxidant, Cytotoxic, Thrombolytic, Antimicrobial and Membrane Stabilizing Activities of Lablab Purpures at a Time

  • To simplify the different biological investigation of the methanolic extract and solvent-solvent partitioning of Lablab purpures (L. purpures) bark. In-vitro anti-oxidant study was determined using total DPPH radical scavenging assay. In vitro antimicrobial study was measured by observing zone of inhibition. The cytotoxic activity was studied using brine shrimp lethality bioassay and thrombolytic activity by clot disruption method. The antioxidant potential was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Folin-Ciocalteau reagents using butylated hydroxytolune (BHT) and ascorbic acid as standards. The Aqueous soluble fraction revealed the highest free radical scavenging activity ($IC_{50}=48.76{\mu}g/mL$). The antimicrobial screening of the bark of L. purpures exhibited mild to moderate activity in test microorganisms. The CSF showed the maximum relative percentage inhibition against Salmonella parathyphi (34.2%) for bacteria and C. albicans (28.8%) for fungi whereas, lowest relative percentage inhibition against Sarcina lutea (22.0%) for bacteria and Aspergillus niger (24.4%) for fungi. In the brine shrimp lethality bioassay, The $LC_{50}$ values of Carbon tetrachloride and N-Hexane soluble fraction were found $92.18{\mu}g/mL$, and $68.95{\mu}g/mL$ respectively while the $LC_{50}$ values of standard Vincristine sulphate was $1.37{\mu}g/mL$. The methanolic extract and its organic soluble fractions of Lablab purpureus at concentration 2.0 mg/mL, significantly protected the lysis of erythrocyte membrane induced by hypotonic solution and heat as compared to the standard, acetyl salicylic acid (0.10 mg/mL). The MSF and AQSF produced 61.48 % and 53.75% inhibition of hemolysis of RBC caused by hypotonic solution respectively, whereas acetyl salicylic acid (0.10 mg/mL) showed 76.42%. Ethanol extract of L. purpures and all of its different partitions exhibited moderate thrombolytic activity of 37.25%-2.40%. Very good preliminary screening and simplified experiments were able to show the different biological activity of methanolic extract and its soluble fractions of L. purpures at a time.

Antimicrobial and Synergistic Effects of Silver Nanoparticles Synthesized Using Soil Fungi of High Altitudes of Eastern Himalaya

  • Fifty three fungi isolated from soils of different microhabitats of eastern Himalayan range (3,400-3,600 msl) were screened for mycosynthesis of silver nanaoparticles (AgNPs) and their efficacy as antimicrobials were assessed in combination with commonly used antibiotics. Three isolates $Aspergillus$ $terreus$ SP5, $Paecilomyces$ $lilacinus$ SF1 and $Fusarium$ sp. MP5 identified based on morphological and 18S rRNA gene sequences were found to synthesize AgNPs. These nanoparticles were characterized by visual observation followed by UV-visible spectrophotometric analysis. The AgNPs synthesized by $Aspergillus$ $terreus$ SP5, $Paecilomyces$ $lilacinus$ SF1 and $Fusarium$ sp. MP5 showed absorbance maxima at 412, 419, and 421 nm respectively in the visible region. Transmission electron microscopy micrograph showed formation of spherical AgNPs of 5-50 nm size. The antimicrobial activity of the mycosynthesized nanoparticles were investigated alone and in combination with commonly used antibiotics for analysis of growth inhibition zone against test organisms, namely, $Staphylococcus$ $aureus$ MTCC96, $Streptococcus$ $pyogenes$ MTCC1925, $Salmonella$ $enterica$ MTCC735 and $Enterococcus$ $faecalis$ MTCC2729. The mycosynthesized nanoparticles showed potent antibacterial activity and interestingly their syngergistic effect with erythromycin, methicillin, chloramphenicol and ciprofloxacin was significantly higher as compared to inhibitions by AgNPs alone. The present study indicates that silver nanoparticles synthesized using soil borne indigenous fungus of high altitudes show considerable antimicrobial activity, deserving further investigation for potential applications.
