N-(2치환스티릴) 카르복사미드류의 합성 (Synthesis of N-(disubstituted styryl) Carboxamides)

  • For the synthesis of tuberin derivatives, N-(disubstituted styryl) carboxamides, the series of cinnamic acids were transformed through chlorides, azides to isocyanates. And then isocyanates were reduced separately by Dibal and Grignard reagent. As a result of antimicrobial susceptibility test, N-(3,4-dichlorostyryl) formamide and N-(3,4-dichlorostyrl) acetamide showed comparatively large activity against some bacteria that is, MIC was respectively 50 ppm, $6.25{\sim}50\;ppm$. MIC of other derivatives was similiar to that of tuberin, about 100.

항균성 Monascus 균주의 Screening 및 영양원과 배양조건이 항균활성에 미치는 영향 (Screening of Monascus Strains for Antimicrobial Activity and Effect of Change of Nutrients and Incubation Conditions on Antimicrobial Activity)

  • 흥국으로부터 분리된 Monascus 균주로부터 항균활성이 우수한 2균주, 분리주 No.116및 No.481을 선정하여 영양원 및 배양조건의 변화에 따른 항균활성에 대한 검토한 결과 분리주에 따라 차이를 보였으며, 분리주 No.116의 경우 탄소원으로서는 sucrose 8%, 질소원으로 $(NH_4)_2SO_40.8%,$ 인산염으로 $KH_2PO_40.5%$ 그리고 금속성분으로 $MgSO_40.5%$ 첨가시 뚜렷이 향상된 결과를 보였으며, 분리주 No.481의 경우에는 각각 sucrose 8%, $(NH_4)_2SO_41.6~2%,$ $K_2HPO_40.1%,$ $MgSO_40.5%$ 그리고 $FeSO_40.5%$ 첨가시 항균활이 크게 향상된 결과를 보였다. 또한 두 균주 모두 배양 초기의 pH는 5.3, 배양온도는 $32.5^{\circ}C에서$ 가장 우수한 항균물질을 생성할 수 있었다. 상기 배양 최적조건에서 분리주 No.116과 No.481 모두Bacillus subtilis, Staphyloccus aureus, Listeria monocytogenes 및 Enterococcus faecium에 대해 51시간 이상 뚜렷한 생육저해를 나타냈다.

Genotyping, Phage Typing, and Antimicrobial Resistance of Salmonella Typhimurium Isolated from Pigs, Cattle, and Humans

  • Salmonella enterica serovar Typhimurium (ST) is one of the most common serovars isolated from humans and animals. It has been suggested that ST infections in Koreans are largely due to the consumption of contaminated pork and beef. To investigate the genotypes, phage types, and antimicrobial resistance patterns for ST isolates of different origins, a total of 70 ST strains, including 19 isolates from humans, 44 isolates from pigs, and 6 isolates from cattle, were analyzed using pulsedfield gel electrophoresis (PFGE), phage typing, and antimicrobial susceptibility tests. Forty-three distinct PFGE patterns were generated from 70 ST isolates, which were grouped into 14 PFGE groups (from A to N) at the level of 75% similarity. The most prevalent group was the A (A1-A17 subtypes) group, encompassing 54.5% (38/70) of ST isolates. ST isolates from pigs and cattle mostly belong to groups A and L, whereas ST isolates from humans mostly belong to groups F and C. Antimicrobial susceptibility tests using 11 antimicrobial agents showed that resistance to tetracycline (TE) (81.4%) was highly prevalent, followed by streptomycin (S) (64.3%) and nalidixic acid (NA) (31.4%) resistance. A total of seventeen antimicrobial resistance patterns were observed. Only 8.6% of isolates, including a reference strain, were susceptible to all antimicrobial agents tested. The most prevalent resistance pattern was TE-S (37.1%), which was seen in 66.6% of bovine, 40.8% of swine and 21.1% of human isolates. Three ST isolates from humans (15.9%) showed resistance to 7-8 antimicrobials. The most predominant phage type (PT) was U302 (64.3%), followed by DT170 (10.0%). PFGE types did not coincide with antimicrobial resistance patterns and phage types; therefore, the combination of those types allowed for further differentiation between tested ST isolates.

근관세정제와 상호작용시 Hydroxyethylidene bisphosphonate (HEBP)의 조직용해능력, 항균효과에 대한 연구 (Interaction of Hydroxyethylidene bisphosphonate (HEBP) with other endodontic irrigants on tissue dissolving capacity and antimicrobial effect)

  • 목적: 근관 세정제로서 Hydroxyethylidene bisphosphonate (HEBP)가 다른 근관 세정제인 Sodium hypochlorite (NaOCl), Etylenediaminetetraacetic acid (EDTA) 상호 작용 시 조직용해 능력, 항균효과와 온도에 따른 조직 용해 능력의 변화에 대해 알아보았다. 연구 재료 및 방법: 소의 근육으로 제작된 동일한 단면적($2{\times}2{\times}2mm$)을 가진 80개의 시편을 8개의 그룹(n = 10)으로 임의로 나누었다. 초기 무게 측정 후 각각의 제조된 근관 세정제에 넣어 5분, 10분, 15분 후 무게를 측정하여 무게변화율을 구하였다. 항균 효과 조사를 위해 Enterococcus faecalis를 접종한 agar diffusion test를 시행하였다. 결과: 조직 용해 능력은 NaOCl group (G4, 5)이 가장 뛰어났으며, NaOCl과 HEBP의 혼합물(G7, 8)이 다음으로 나타났다. 또한 가열된($40^{\circ}C$) NaOCl과 NaOCl/HEBP의 혼합물의 경우 실온($25^{\circ}C$)보다 우수한 조직용해 능력이 나타났다. 항균 효과의 경우 EDTA > EDTA + 1% NaOCl > $1%\;NaOCl{\geq}1%\;NaOCl$ + HEBP 순으로 나타났다. 결론: HEBP는 NaOCl 의 항균 효과를 방해하지 않으며, 조직 용해 능력에도 기존의 킬레이팅 제재에 비해 적은 영향을 미친다. 가열된 NaOCl, NaOCl과 HEBP의 혼합재제는 실온과 비교 시에 효율적으로 조직을 용해할 수 있다.

청열약(淸熱藥)의 질내(膣內) 미생물(微生物)에 대(對) 항균효과(抗菌效果) (Antibiotic Effect of Heat-Clearing Medicinals on Vaginal Microorganisms)

  • Purpose : This study was conducted to investigate the in vitro inhibitory effects of heat-clearing medicinal on common bacterias in gynecology. Methods : The heat-clearing medicinals ( Trichosanthis Radix, Sophorae Fructus, Phragmitis Rhizoma, Buddleiae Flos, Bambusae Folium, Anemarrhenae Rhizoma, Celosiae Semen, Gardeniae Fructus, Prunellae Spica, Sophorae Radix, Dictamni Radicis Cortex, Coptidis Rhizoma, Gentianae Scabrae Radix, Scutellariae Radix, Phellodendri Cortex) were used in this study. Staphylococcus aureus, MRSA, Candida albicans and Gardnerella vaginalis were used for vaginal pathogenic microorganisms. Streptococcus spp., Escherichia coli HB101, Lactobacillus gasseri were used for normal vaginal florae. We evaluated antibiotic effect by the optical density and the colony test. Results : The optical density and colony test showed that Celosiae Semen, Prunellae Spica, Scutellariae Radix and Phellodendri Cortex of herbs among heat-clearing medicinal had antimircobial effect. Celosiae Semen and Prunellae Spica had antimicrobial susceptibility and selective toxicity in MRSA. Scutellariae Radix and Phellodendri Cortex had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis. Conclusion : According to the above results, we could suggest that Celosiae Semen, Prunellae Spica, Scutellariae Radix and Phellodendri Cortex among heat-clearing medicinal be available to antimicrobial agent of vaginal pathogenic microbial species in vitro.

와송 에틸아세테이트 분획물의 항균효능 평가에 관한 연구 (A Study on the Evaluation of Antimicrobial Effect of Orostachys Japonicus A. Berger Ethyl Acetate Fraction)

  • 본 연구에서 와송 에틸아세테이트 분획물을 항균활성 및 화장품 소재로 활용하기 위해 연구를 진행하였다. 와송 에틸아세테이트 분획물의 항균활성을 측정한 결과 Paper disc method에서 S. aureus, S. epidermidis, P. aeruginosa 균의 생육을 저해하였으며, 특히 S. aureus에 대한 생육저해환은 0.5 g/mL농도에서 18.35 ± 1.5 mm로 대조군인 methyl paraben(16.83 ± 1.0 mm)보다 우수한 항균활성을 나타내었다. MIC 측정을 통한 최소저해농도를 평가에서는 Candida. A를 제외한 나머지 S. aureus, S. epidermidis, E. coli P. aeruginosa 균주에서 17.5 mg/mL의 최소저해농도를 보였으며 화장품 에멀션 제형에 적용하여 Challenge test를 통해 방부 효능을 평가한 결과, S. aureus에 대하여 와송 에틸아세테이트 분획물을 0.3% 첨가한 에멀션에서 7일 후 100.0%균의 생장을 사멸시켜 화장품에 적용 시 우수한 방부효능을 나타내었다.

쑥 첨가가 빵과 떡의 저장성 향상에 미치는 영향 (Effect of Mugwort on the Extention of Shelf-Life of Bread and Rice Cake)

  • The purpose of this study is to investigate the antimicrobial effect of mugwort (Artemisia asiatica Nakai) on the rice cake and bread preservation, and to identify their antimicrobial compounds. The mugwort extracts showed complete inhibition on the growth of Bacillus subtilis and Staphylococcus aureus at 250 $\mu\textrm{g}$/ml level. Antimicrobial activi쇼 of mugwort extract were stronger than that of commercial antimicrobial agent. Five % of sodium propionate solution showed complete inhibition on the growth of B. subtilis, E. coli and S. aureus, but L. plantarum was inhibited 50.87% at the same concentration. When various amounts of freeze-dried mugwort powder were added in sulgis (steamed rice cake), 3% ssooksulgi (mugwort powder added sulgi) had quite lower level of total bacterial count (5.5$\times$$10^/5 CFU/g) compared with the control group (1.4$\times$$10^/7 CFU/g) at ambient temp. (30$\pm$1$^{\circ}C$) after 72 hr. Three % addition of mugwort showed 2 days extention of shelf-life of rice cake. The sensory qualities of ssooksulgi has no significant difference in moistness, consistency, cohesiveness, afterswallowing and overall quality compared with control group. Ssooksulgi with 3% of mugwort powder had the best overall quality in sensory test. The methanol extract of 500 $\mu\textrm{g}$/ml of mugwort could lead the successful retardation of the growth of putrefactive microorganism during the incubation of rice cake at 37$^{\circ}C$ for 24 hr. On the other hand, coumarin (Sigma) had 54% inhibitory effect at 500 $\mu\textrm{g}$/ml level, and (E,E)-2,4-decadienal completely inhibited the growth of putrefactive microorganism of whitesulgi at 100 $\mu\textrm{g}$/ml level during the incubation at 37$^{\circ}C$ for 48 hr.48 hr.

Antimicrobial Activity of a Bacteriocin Produced by Enterococcus faecalis KT11 against Some Pathogens and Antibiotic-Resistant Bacteria

  • In this study, the antimicrobial activity of a bacteriocin produced by Enterococcus faecalis KT11, isolated from traditional Kargı Tulum cheese, was determined, and bacteriocin KT11 was partially characterized. The results showed that bacteriocin KT11 was antagonistically effective against various Gram-positive and Gram-negative test bacteria, including vancomycin- and/or methicillin-resistant bacteria. The activity of bacteriocin KT11 was completely abolished after treatment with proteolytic enzymes (proteinase K, ${\alpha}$-chymotrypsin, protease and trypsin), which demonstrates the proteinaceous nature of this bacteriocin. Additionally, bacteriocin KT11 remained stable at pH values ranging from 2 to 11 and after autoclaving at $121^{\circ}C$ for 30 min. In addition, the activity of bacteriocin KT11 was stable after treatment with several surfactants (EDTA, SDS, Triton X-100, Tween 80 and urea) and organic solvents (chloroform, propanol, methanol, ethyl alcohol, acetone, hexane and ethyl ether). Cell-free supernatant of E. faecalis KT11 was subjected to ammonium sulfate precipitation and then desalted by using a 3.5-kDa cut-off dialysis membrane. The bacteriocin activity was determined to be 711 AU/mL in the dialysate. After tricine-SDS-PAGE analysis, one peptide band, which had a molecular weight of ~3.5 kDa, exhibited antimicrobial activity. Because the bacteriocin KT11, isolated from E. faecalis KT11, exhibits a broad antimicrobial spectrum, heat stability and stability over a wide pH range, this bacteriocin can be used as a potential bio-preservative in foods. Additionally, bacteriocin KT11 alone or in combination with conventional antibiotics may provide a therapeutic option for the treatment of multidrug-resistant clinical pathogens after further in vivo studies.

우슬 추출물의 Clostridium difficile에 대한 항균 효과 (Antimicrobial Effect of Achyranthes japonica Nakai Extracts against Clostridium difficile)

  • 본 연구에서는 대장염 유발균인 C, difficile에 대한 항균활성이 우수한 천연항균물질을 검색하기 위하여 40여 가지 생약재 에탄올 추출물에 대하여 C. difficile에 대한 생육저해활성을 측정하였다. 결과, 우슬, 희첨, 황백은 1,000 ppm농도에서 90% 이상의 높은 항균 활성을 나타내었고, 우슬 에틸아세테이트분획물(AJNEA)은 가장 높은 활성을 나타내었으며 이의 최소저해농도(MIC)는 $625{\mu}g/mL$였다. 또한 기능성 소재로의 활용가능성을 확인하기 위하여 AJNEA의 유산균 생육에 미치는 영향을 조사한 결과, B. longum, L. helveticus와 S. thermophilus를 제외한 나머지 균주들에 대해서 생장 억제 효과를 나타내지 않았다. 이들 결과로부터 우슬 추출물은 C. difficile 관련 질환에 대하여 우수한 항균제제로 이용될 수 있을 것으로 확인되었다.

왕우럭(Tresus keenae)에서 분리된 Bacillus species의 고분자 유기물질 분해능력과 항균활성 (Degradation capability of macromolecular organic matters and antimicrobial activities of Bacillus species isolated from surf clam (Tresus keenae))

  • The production of enzymes that help digestion, assimilation of essential nutrients, and prevent pathogenic bacteria are important for probiotics used in aquaculture. The objective of this study was to investigate enzyme activities for macromolecular organic matters and antimicrobial properties of the selected potential probiotics isolated from gut of surf clam (Tresus keenae) against well-known shellfish-pathogenic bacteria. Among 65 isolates from guts of 60 surf clams, seven Bacillus strains with outstanding degradation capability of macromolecule organic matter were selected as potential probiotics as follows: TKI01 (B. vietnamensis), TKI02, TKI26 (B. thuringiensis), TKI14, TKI32, TKI42 (B. amyloliquefaciens), and TKI18 (B. stratosphericus). After in vitro antimicrobial activity test was performed against five shellfish-pathogenic bacteria including Listonella anguillarum, Vibrio parahaemolyticus, V. splendidus, V. harveyi, V. tubiashii, PCR assay was performed to detect bacteriocin-producing strain. PCR results revealed that the five Bacillus strains possessed diverse bacteriocin genes including ericinA, coagulin, surfactin, iturin, bacyllomicin, fengycin, bacylisin, subtilin, and lantibiotics. In the present study, the selected seven Bacillus strains showed different enzyme activities according to types of macromolecule organic matters. And their antimicrobial activities varied based on the species of pathogenic bacteria. In addition, at least five Bacillus strains had genetic potential to produce several natural lipopeptide antibiotics that may help biological control of surf clam aquaculture. Therefore, mixed use of probiotics might show co-operative effect and increase the efficiency of probiotics rather than separate use. To the best of our knowledge, it is the first report on antimicrobial properties of Bacillus species isolated from surf clam.
