The Effect of Opuntia ficus-indica var. saboten Fruit on Gastric Lesion and Ulcer in Rats

  • The effects of the dried fruit powder of Opuntia ficus-indica var. saboten (OF-f) were investigated on gastric lesion and ulcer models in rats. It showed significant inhibition in HCl ethanol and HCl aspirin induced gastric lesion at a dose of 600 mg/kg, p.o. OF-f also showed significant inhibition in indomethacin induced gastric lesion at the doses of 200 and 600 mg/kg, p.o. However, it did not affect aspirin and Shay ulcer in rats. It also did not affect the gastric juice secretion, acid output and pH. These data suggest that OF-f possesses pronounced inhibitory action on gastric lesion of rats.

괴화산(槐花散)이 항소화성궤양(抗消化性潰瘍) 및 혈액(血液) 응고작용(凝固作用)에 미치는 실험적(實驗的) 연구(硏究) (A Study of Goiwhasan's Antigastric ulcer and Blood Hemostasis)

  • The purpose of this research was to investigate the efficacy of Goiwhasan extract powder on the gastric injuries, antiulcer, gastrointestinal tract and blood hemostasis. Animals were used through this studies mice and rats. All animals were divided into 3 groups, contol group(no treatment), sample Ⅰ group(375mg/kg administration), sample Ⅱ group(750mg/kg administration). The gastric injuries and ulcer have been made by using pyloric ligation, indomethacin, HCI-ethanol, acetic acid and then The histological observation was followed. In the gastrointestinal tract, gastric juice secretion, gastric acidity, pepsin output, blood gastrin and secretin level, transport potentials in the small and large intestine were checked. And studies on blood hemostasis were performed on normal hemostatic activities and plasma prothrombin time, plasma recalcification time, plasma fibrinogen levels in the hypoprothrombinemic mice induced by warfarin. The results were as follows: 1. The antigastric ulcer effects on the pyloric ligation, indomethacin, HCl-ethanol, acetic acid induced gastric injuries were shown in Sample Ⅱ group(p<0.05). 2. Through the morphologic examination on the acetic acid induced ulcer, Sample Ⅰ group showed mild regeneration of epithelium and slight decrease of periulcer edema then that of Control group, while Sample Ⅱ group showed more retraction of round ulcer site, remarkable loss of swelling and edema then that of Control group, and revealing the regenerated epithelium in the surrounding ulcer site. Thus it was noted that both Sample groups have antigastriculcer effects on the experimentally induced gastric ulcer. 3. The inhibitory effects on gastric juice were noted in both Sample Ⅰ group(p<0.05) and Sample Ⅱ group(p<0.01). However, only Sample Ⅱ group showed the inhibitory effects on total acidity and pepsin output(p<0.05). 4. The significant inhibition of blood gastrin level showed at 30 min.(P<0.05) and 90 min.(P<0.05) after starting medication in only Sample Ⅱ group, but significance of blood secretin level in both groups was not recognized. 5. Any significant changes in barium sulfate transport in the small intestine of mice was not recognized in both groups, but the significantly inhibitory effect in large intestine was recognized in both Sample Ⅰ group(p<0.05) and Sample Ⅱ group(p<0.001). 6. In hemostatic effect on both normal mice and hypoprothrombinemic mice induced by warfarin, the significantly shortening effect on coagulation time was seen in only Sample Ⅱ group(p<0.01). 7. On plasma prothrombin time in hypoprothrombinemic rat induced by warfarin, Sample Ⅱ group have shortened the prothrombin time significantly(p<0.001). 8. On plasma reclcification time in hypoprothrombinemic rat induced by warfarin, the recalcification time have been shortened significantly in both Sample Ⅰ group(p<0.05) and Sample Ⅱ group(p<0.01). 9. On plasma fibrinogen levels in hypoprothrombinemic rat induced by warfarin, the fibrinogen contents in Sample Ⅱ have been decreased significantly(p<0.01). Overall the above results suggest that Goiwhasan has an therapeutic efficacy on antigastric ulcer and blood hemostasis. Further studies would be needed on the interaction of its herbal medicine and its mechanism in the future.

지유가 흰쥐의 급성위염 및 소화성 궤양에 미치는 영향 (The Effects of Ethylacetate Fraction of Sanguisorba officinalis L. on Experimentally-induced Acute Gastritis and Peptic Ulcers in Rats)

  • 본 실험에서 흰쥐에 실험적으로 급성위염, 위궤양 및 십이지장궤양을 유발하고 지유 에틸아세테이트분획을 투여하여 항위염 및 항궤양에 미치는 영향을 검토하였던 바 다음과 같은 결과를 얻었다. 지유 에탄올 추출물을 n-헥산, 클로르포룸, 에틸아세테이트 및 n-부타놀과 물로 계통분획하여 농축하고 in vitro에서 항산화력을 측정한 결과 에틸아세테이트분획이 가장 우수하였다. 지유 에틸아세테이트분획 (SOL & SOH)의 HCI-EtOH급성위염에 대한 억제율은 각각 $27.7\%$, $31.7\%$로 대조약물로 사용된 cimetidine(CMT, $47.2\%$)의 억제효과에는 미치지 못했으나 CON에 비해 우수하였다. Indomethacin유발 위궤양에서, SOL & SOH의 위궤양억제율은 각각 $48.28\%$, $51.08\%$로 대조약물로 사용된 CMT($58.26\%$)의 억제효과에는 미치지 못했으나 CON에 비하며 유의 성있는 억제효과를 나타냈다. Shay 위궤양에서, 지유 에틸아세테이트분획을 십이지장에서 위내로 투여하였을 때 궤양 억제율은 SOL이 $42.86\%$로 CON에 비하여 억제되었으나 유의성 있는 효과는 아니었으며, 용량을 증량하여 투여한 SOH는 $47.71\%$로 대조약물인 omeprazol(OMP)의 저해효과($62.00\%$)에 미치지는 못했으나 CON에 비해 유의적민 효과를 나타내었다. 위액량 및 총위산분비량에서, 지유 에틸아세데이트분획 투여로 CON에 비하여 $32.11\%$$47.69\%$, $24.28\%$$34.69\%$의 감소로 유의성 있는 효과를 나타냈으며 OMP($55.69\%$, $41.62\%$)에 근접하게 감소되었다. pepsin 활성은 지유 에틸아세테이트분획 투여로 CON에 비해 각각 $19.55\%$, $\%21.45$가 저하되었으나 유의성있는 변화는 아니었다. 수침 $\cdot$ 구속 스트레스에 의해 유발된 궤양의 경우, SOL는 궤양억제율이 $9.23\%$ 로 CON와 유사하게 출혈을 동반한 깊은 궤양이 관찰되었으나 용량을 증량하여 투여한 SOH는 $48.45\%$로 CON에 비해 궤양의 깊이도 얕아졌고 출혈을 동반한 궤양도 감소되어 유의적인 억제율을 나타내었다. Cysteamine 유발 십이지장궤양에서, CMT는 유발된 십이지장 궤양을 CON에 비해 $54\%$ 정도 억제하여 우수한 항궤양효과를 나타냈고 시료투여군인 SOL 및 SOH는 CMT보다 우수하지는 않았으나 약 $35\∼42\%$ 정도의 궤양억제율을 나타냈다. 이상의 실험결과에서 지유의 항위염 및 항궤양효과는 항산화물질에 의한 결과로 판단되어지며, 앞으로 위염 및 위궤양 개선을 위한 건강 기능성 식품으로 개발이 기대된다

  • The effects of the dried stem powder of Opuntia ficus-indica var. saboten (OF-s) were investigated on gastric lesion and ulcer models in rats. It showed significant inhibition in HCl·ethanolinduced gastric lesion at the doses of 200 and 600 mg/kg p.o. and in HCl·aspirin-induced gastric lesion at 600 mg/kg p.o. OF-s also showed significant inhibition in indomethacin-induced gastric lesion at the doses of 200 and 600 mg/kg, p.o. However, it did not affect both the aspirin-induced and Shay ulcers in rats. It also did not affect gastric juice secretion, acid output and pH. These data indicate that OF-s only possesses pronounced inhibitory action on gastric lesion without antiulcer activity in rats.

오리나무 열수 추출물이 실험적으로 유발된 흰쥐의 위염 및 위궤양에 미치는 영향 (Effect of Hot Water Extract of Alnus japonica Steud on the Experimentally-induced Acute Gastritis and Peptic Ulcers in Rats)

  • Alnus japonica Steud (A. japonica) have long been used in the traditional medicine for gastric disorder, hepatitis and fatty liver in Korea. Antiulcer effects of A. japonica hot water extract (AJ ext) were evaluated by in vitro antibacterial activity against H. pylori, by the inhibitory action against the in vitro gastric $H^+/K^+$-ATPase and using rat models of gastric mucosal damage and gastric ulcer induced by HCl-ethanol, indomethacin, and restraint and water-immersion stress. For the determination of antibacterial activity of AJ ext against H. pylori, the activity of urease which released from H. pylori was measured in culture. AJ ext showed weak antibacterial activity against H. pylori with the growth inhibitions of 37% and 61% by adding final concentrations of 500 and $1000{\mu}g/ml$ culture, respectively at 24 h. To observe the inhibitory activity of AJ ext against the $H^+/K^+$-ATPase in hog gastric membrane vesicle, $IC_{50}$ value of AJ ext was $806.3{\mu}g/ml$. Pretreatment of AJ ext (200, 500 mg/kg, p.o.) prevented in a dose-dependent manner the acute gastritis in HCl-ethanol model and the formation of gastric ulcer in indomethacin model and restraint and water-immersion stress model. These results suggest that the AJ ext can be used for prevention and treatment of gastric mucosal damage and ulcers induced by various stress.

화담청화탕이 acetic acid로 유발된 흰쥐 위궤양 점막의 내분비세포와 점액의 조직화학적 변화 (The Changes of Mucin and Endocrine cells of Mucosa of Acetic Acid-Induced Gastric Ulcer after Administration of Whadamcheongwha-tang Extract in Rat)

  • We examined the effects of Whadamcheongwha-tang(WDCWT) extract on the acetic-acid induced antigastric ulcer in rats. These experiments investigated the numerical changes of gastrin and histamine secreting cells of the gastric mucosa by immunohistochemical method, and the changes of mucin of gastric mucosa by PAS-AB stain methods after the oral administration of WDCWT extract(1.0ml/day) and omeprazole(0.2mg/day) for 1, 3 and 6 weeks. The result are as follows; 1. When WDCWT extract was administrated for 1, 3, 6 weeks, in result, gastrin secreting cells in gastric mucosa were increased compared to the control group. 2. When WDCWT extract was administrated for 1, 3, 6 weeks, in result, the density of immunoreactive gastrin cells was increased compare to the control group. 3. When WDCWT extract was administrated for 1, 3, 6 weeks, in results, the changs of mucosal thickness stained by PAS/PAS-AB was increased compared to the control group. 4. When WDCWT extract was administrated for 1, 3, 6 weeks, in results, the density of PAS stain was decreased compare to control group, but density of AB stain was increased compare to control group. The results suggest that WDCWT extract inhibits a gastric acid secretion in rat gastric mucosa, and is useful in the treatment of the hyperacidity and gastric ulcer.

  • Although the 'Insamyangwee-tang' has been widely used in clinical purposes in the oriental medicine, its clinical efficacy is only documentated for the cases of gastritis, gastric ulcer and enteritis, but the experimental study on these has not been undertaken. So, to investigate the clinical efficacy of 'Insamyangwee-tang' comparing with animal experiments, This study was carried out. The results showed that relaxing action was shown on the isolated ileum in mice and that strong antagonistic actions were seen on $BaCl_2$, acetylcholine and histamine induced contraction of the ileum in mice, rats, rabbits and guinea-pigs that the relaxing effect of the intestinal smooth muscle was recognized. Inhibitory effects on transport rate in the small intestine of mice. Strong antagonistic actions were seen on acetylcholine induced contraction of duodenum in rats and remarkably inhibiting actions were seen of duodenum in rats. Inhibitory action on the secretion of gastric juice and pepsin, anti-ulceration effect was recognized.

위염 및 위궤양에 미치는 Propolis 추출물의 영향 (Effects of Propolis Extract on Antigastritic and Antiulcer)

  • 벌집으로부터 채취한 수지상의 물질인 propolis는 다양한 생화학적 성분을 함유하며 기원전 300년부터 사용되어 온 전통약물이다. 최근 항균, 항 바이러스, 항진균, 국소마취, 면역 활성, 항염, 항산화 작용등의 생물학적 활성이 보고된 바 있다. 본 연구에서는 위염과 위궤양에 미치는 propolis추출물과 분획물의 약조학적 효과를 평가하고자 에탄올 추출물을 hexane, toluene, ethyl acetate로 분획하여 항위염 및 항위궤양 활성을 조사하였다. Propolis 에탄을 추출물은 염산·에탄을 위손상, Shay의 위액 분비 실험에서 용량 의존적인 보호효과를 나타내었고 hexnae과 toluene 분획물은 염산·에탄을 위손상, aspirin에 의한 위손상, Shay의 위액 분비 실험에서 위손상과 위액분비를 감소시켰다. propolis 추출물과 분획물의 위염 및 위궤양에 대한 보호효과는 H+/K+ATPase activity의 억제에 의한 위액분비의 감소에 의한 것으로 생각된다.
