형방지황탕으로부터 분리된 HTI의 항진균활성에 대한 연구 (In Vitro Antifungal Activity of HTI Isolated from Oriental Medicine, Hyungbangjihwang-tang)

  • 형방지황탕은 한의학에서 중요한 약처방으로 인간의 기력 회복에 약효가 탁월한 것으로 알려져 있다. 본 연구에서는 형방지황탕으로부터 항생 물질(HTI)을 분리하여 항진균활성 및 그 작용기작을 검토하였다. 그 결과 각종 병원성 진균에 대해 항진균 활성을 나타내었으며, 인간 적혈구에 대해서는 세포독성이 나타나지 않은 것으로 확인되었다. 또한 캔디다 알비칸스를 대상으로 스트레스 방어기작과 세포 이형성 유도에 HTI이 미치는 영향을 검토한 결과, 트리 할로즈의 증가와 이형성 유도에 영향을 미치는 것으로 나타나 이러한 현상이 항진균활성에 상관관계가 있음을 확인하였으며, 또한 진균의 세포주기에 미치는 영향도 항진균 활성의 요인이 됨을 밝혔다.

  • Antifungal effect of Ginkgo biloba leaves extracts conducted for Pityrosporum ovale. Antifungal effect verified by diffusion test, optical density test and colony counting test under various concentration. Extract of ginkgo biloba leaves performed with 40% ethanol and 60% water solution at $60^{\circ}C$ and major components analyzed by HPLC. The concentrated extract have bilobalide and ginkgolide A and ginkgolide B and their concentration were 153.0 mg/L, 8403.5 mg/L and 2723.0 mg/L respectively. Ginkgo biloba leaves extracts gave 99.1% of antifungal effect for Pityrosporum ovale examined by colony counting method.

Obacunone 황백성분의 Candida albicans에 대한 항진균효과 (Antifungal Effect of Obacunone on Candida albicans)

  • In the present study, we determined the antifungal effect of obacunone isolated from Phellodendri Cortex against Candida ablicans, a pathogenic fungus. The antifungal effect was analyzed by an in-vitro susceptibility test and in a murine model of disseminated candidiasis. Possible mechanism of the antifungal activity was also examined. Analyses of data resulting from the susceptibility test revealed that the compound inhibited C. albicans growth. At 25 ${\mu}g$ obacunone/ml, there was app. 45% reduction of CFUs (colony forming units) as compared to obacunone-untreated C. albicans yeast cells (P<0.01). In the murine model of disseminated candidiasis due to C. albicans, obacunone enhanced resistance of mice against disseminated candidiasis. During an entire period of 30-day observation, control animals all died within 14 days, whereas 60% of obacunone-treated mice survived (P<0.05). In addition, obacunone inhibited the hyphal production, a major virulence factor of C. albicans, from the blastoconidial form. Thus, obacunone appears to have antifungal activity for C. albicans infection. This may possibly be mediated by the blockage of hyphal production.

Microsporum canis에 대한 5가지 항진균제와 맥반석 가루의 항진균 효과 (Antifungal Activity of 5 Antifungal Agents and Granitic Powder on Microsporum canis)

  • The aim of this study was to determine tile inhibitory effect of granitic powder against Microsporum canis. Fourteen strains of M. canis isolated from dgs and cats with fungal dermatitis and two strains isolated from humans were used in this study. The in vitro antifungal activities of granitic powder and 5 commercialized antifungal agents (terbinafine, itraconazole, ketoconazole, griseofulvin and fluconazole) were compared. The antifungal effect was measured by the broth microdilution method and was expressed as the minimal inhibitory concentration (MIC). The MIC value of the granitic powder was ranged from 31.3 to 250mg/ml. Terbinafine showed the lowest MIC value among the 5 commercial antifungal agents $(0.0078-0.125{\mu}g/ml)$, while fluconazole showed the highest MIC values $(125-1,000{\mu}g/ml)$. The MIC range of itraconazole, griseofuvin and ketoconazole were $0.125-0.5{\mu}g/ml\;0.625-5{\mu}g/ml$ and $10-40{\mu}g/ml$ respectively. The Geometric mean(GM) MIC values of terbinafine and ketoconazole against M. canis isolated from human were $0.0078{\mu}g/ml\;and\;10{\mu}g/ml$, respectively, while the GM MIC values of these agents against M. canis isolated from animals were $0.063{\mu}g/ml\;and\;31.4{\mu}g/ml$, respectively. Other antifungal agents did not show any significant differences in antifungal activity against M. canis of animal or human origin. Although granitic powder was shown to have antifungal activity, it was much lower than that of the 5 commercialized antifungal agents.

Antifungal Activities of Isothiazoline/Cabamate based Organic Antifungal Agent Activated-Cement Mortars (AACM)

  • Antifungal agents are used to impart antibacterial or bactericidal properties to commodities and various articles used in industries and can be classified into two broad groups i.e organic and inorganic. Inorganic antifungal agents comprise of Ag, Zn, or Cu, etc. These elements tend to exhibit high level of antifungal activities, non-uniform dispersion in substrates, and have poor properties in expensive and cheap adhesiveness. In this study, the organic antifungal agent was used for the purpose of investigating the antifungal activity of antifungal agent activated-cement mortar (AACM) on the aspergilus niger of various fungus which can be easily discovered in the interiors and exteriors of buildings. In addition, an experiment on the basic physical properties of AACM such as compressive and flexural strength was carried out. The conclusion of this investigation revealed that a dosage increase of antifungal agent exhibits a high inhibitory effect on the aspergilus niger, and although there is a slight decrease in the strength of AACM, the strength of AACM was almost equal to that of inactivated cement mortar.

  • The purpose of this study was to evaluate the in vitro antifungal effect of several kinds of denture lining materials containing nystatin and silver-zeolite on Candida albicans. Three commercially available tissue conditioners (Soft-Liner, Coe-Comfort, Coe-Soft) and two direct denture relining materials (Tokuso Rebase, Durabase) were selected. In terms of the zone of inhibition and some basic physical properties, I could find the following results ; 1. Nystatin or silver-zeolite included in those relining materials had definite antifungal activities against the Candida albicans. 2. As times went on, both of the antifungal agents's activities decreased gradually. 3. Antifungal agents did not affect the relining materials' basic physical properties. 4 Direct chairside relining materials showed unfavorable dark discoloration with response to silver-zeolite.

In vitro Antifungal Activity of Limonene against Trichophyton rubrum

  • In this study, the antifungal activities of limonene against Trichophyton rubrum were evaluated via broth microdilution and vapor contact assays. In both assays, limonene was shown to exert a potent antifungal effect against T. rubrum. The volatile vapor of limonene at concentrations above $1{\mu}l$/800 ml air space strongly inhibited the growth of T. rubrum. The MIC value was 0.5% v/v in the broth microdilution assay. The antifungal activity of limonene against T. rubrum was characterized as a fungicidal effect.

Effect of method of synthesis on antifungal ability of ZnO nanoparticles: Chemical route vs green route

  • To compare the antifungal effect of two nanomaterials (NMs), nanoparticles of zinc oxide were synthesized by a chemical route and zinc oxide-based nanobiohybrids were obtained using green synthesis in an extract of garlic (Allium sativum). The techniques of X-Ray Diffraction (XRD), Infrared (IR) and Ultraviolet Visible (UV-Vis) absorption spectroscopies and Scanning (SEM) and Transmission Electron Microscopies (TEM) were used to determine the characteristics of the nanomaterials synthesized. The results showed that the samples obtained were of nanometric size (< 100 nm). To compare their antifungal capacity, their effect on Cercospora sp. was evaluated. Test results showed that both nanomaterials had an antifungal capacity. The nanobiohybrids (green route) gave an inhibition of fungal growth of ~72.4% while with the ZnO-NPs (chemical route), inhibition was ~87.1%. Microstructural studies using High Resolution Optical Microscopy (HROM) and ultra-structural analysis using TEM carried out on the treated strains demonstrated the effect of the nanofungicides on the vegetative and reproductive structures, as well as on their cell wall. To account for the antifungal effect presented by ZnO-NPs and ZnO nanobiohybrids on the fungi tested, effects reported in the literature related to the action of nanomaterials on biological entities were considered. Specifically, we discuss the electrical interaction of the ZnO-NPs with the cell membrane and the biomolecules (proteins) present in the fungi, taking into account the n-type nature of the ZnO semiconductor and the electrical behavior of the fungal cell membrane and that of the proteins that make up the protein crown.

의성개나리 과피(果皮) 성분(成分)의 항진균작용(抗眞菌作用)에 관한 연구(硏究)(I) (Antifungal Studies on Components from the Pericarp of Forsythia viridissima (I))

  • The antifungal studies on the triterpenoids from the pericarp of Forsythia viridissima LINDL. were conducted. The pericarp was obtained from the plant which was cultivated at Eusung, Kyungsang-bukkdo, Korea. Substances A and B were isolated by column fractionation and purified by recrystallization. Antifungal actions of substance A, acetyloleanolic acid, show significant inhibition effect against Trichophyton rubrum, T. mentagrophytes, T. tonsurans, Microsporum nanum, M. canis, and M. cookei. Substance B, viridissimic acid, shows negative effect against Candida albicans and shows lower effects against other test fungi.

Antifungal Effect of Chitosan as Ca2+ Channel Blocker

  • The aim of this study was to investigate antifungal activity of a range of different molecular weight (MW) chitosan against Penicillium italicum. Our results demonstrate that the antifungal activity was dependent both the MW and concentration of the chitosan. Among a series of chitosan derived from the hydrolysis of high MW chitosan, the fractions containing various sizes of chitosan ranging from 3 to 15 glucosamine units named as chitooligomers-F2 (CO-F2) was found to show the highest antifungal activity against P. italicum. Furthermore, the effect of CO-F2 toward this fungus was significantly reduced in the presence of $Ca^{2+}$, whereas its effect was recovered by ethylenediaminetetraacetic acid, suggesting that the CO-F2 acts via disruption of $Ca^{2+}$ gradient required for survival of the fungus. Our results suggest that CO-F2 may serve as potential compounds to develop alternatives to synthetic fungicides for the control of the postharvest diseases.
