Antifungal Activity of Lichen-forming Fungi against Colletotrichum acutatum on Hot Pepper

  • Antifungal activity of Korean and Chinese lichen-forming fungi (LFF) was evaluated against plant pathogenic fungus of Colletotrichum acutatum, causal agent of anthracnose on hot pepper. This is the first attempt to evaluate antifungal activity of LFF, instead of lichen thalli, against C. acutatum. Total 100 LFF were isolated from the lichens with discharged spore method or tissue culture method. Among the 100 isolates, 8 LFF showed more than 50% of inhibition rates of mycelial growth of the target pathogen. Especially, Lecanora argentata was highly effective in inhibition of mycelial growth of C. accutatum at the rate of 68%. Antifungal activity of other LFF was in the order of Cetrelia japonica (61.4%), Ramalina conduplicans (59.5%), Umbilicaria esculenta (59.5%), Ramalina litoralis (56.7%), Cetrelia braunsiana (56.5%), Nephromopsis pallescensn (56.1%), and Parmelia simplicior (53.8%). Among the tested LFF, 61 isolates of LFF exhibited moderate antifungal activity against the target pathogen at the inhibition rates from 30 to 50%. Antifungal activity of the LFF against C. acutatum was variable at the species level rather than genus level of LFF. This study suggests that LFF can be served as a promising bioresource to develop novel biofungicides.

향나무 심재 추출물로 염색된 직물의 방미성과 집먼지 진드기 기피효과 (Antifungal activity and house dust mite repellent effect of fabric dyed with Juniperus chinensis heartwood extracts)

  • This study was carried out to investigate the antifungal activity and house dust mite repellent effect of cotton and silk fabrics dyed with Juniperus chinensis heartwood extracts by hot-water and methanol. The chemical composition of Juniperus chinensis heartwood extracts by hot-water and methanol was analyzed by GC-MS. In the case of antifungal activity, dyed fabrics with hot water extract had no effect against chaetomium globosum fungus. But showing no growth in fabrics dyed with methanol extract, dyed fabrics with methanol extract showed very excellent antifungal activity. Silk fabric dyed with methanol extract showed excellent house dust mite repellent effect of 94.3%~96.0% against dermatophagodes farinae. The composition of Juniperus chinensis heartwood extracts from hot-water and methanol was different. There were no terpene and its analogue peaks in hot-water extract. However there were terpene and its analogue peaks in methanol extract. From these GC-MS results, terpene was identified in methanol extract of Juniperus chinensis heartwood. Efficient ingredient of antifungal activity and house dust mite repellent effect was assumed terpene.

Effects of Temperature and Culture Media Composition on Sporulation, Mycelial Growth, and Antifungal Activity of Isaria javanica pf185

  • The fungal isolate Isaria javanica pf185 has potential as a mycopesticide because it demonstrates insecticidal activity against the green peach aphid and antifungal activity against Colletotrichum gloeosporioides. For commercialization of this isolate, determination of the optimal and least expensive culture conditions is required; however, these data are not currently available. This study describes the conditions for optimal development of conidia and production of metabolites for the biocontrol of the fungal pathogen. The optimal culture conditions were examined using cultures on solid agar and liquid media. High growth temperature enhanced spore formation but reduced antifungal activity in both solid and liquid media. The highest spore yield was obtained in a medium containing glucose as a carbon source and yeast extract as a nitrogen source. Soybean powder and wheat bran were effective nitrogen sources that promoted spore production and antifungal activity of the isolate. These results revealed the basic, cost-effective growth media for commercial production of a biopesticide with insecticidal and antifungal properties for use in integrated pest management.

편백나무 건조 중 발생하는 휘발성 유기화합물의 항진균 및 항산화 활성 (Antifungal and Antioxidant Activities of Volatile Organic Compounds Generated During the Drying Process of Chamaecyparis obtuse)

  • 본 연구는 편백나무 건조 중 발생하는 휘발성 유기화합물로 구성된 응축수에 대해 항진균 및 항산화 활성을 평가하였다. 응축수는 hexane과 ethyl acetate로 각각 추출하였으며 추출액의 농도 100-500 mg/L에서 피부사상균에 대한 항진균 활성을 측정하였다. 추출액 농도가 증가할수록 항진균 효과도 증가하였으며 500 mg/L에서는 모든 피부사상균에 대해 높은 항진균 활성을 나타냈다. 추출용매에 따라서는 hexane으로 추출하였을 경우 높은 활성을 나타냈으며, 특히 Trichophyton rubrum에 대한 항진균 활성이 높았다. 항산화 활성을 측정한 결과 25 mg/L 이상의 농도에서는 90% 이상의 항산화 활성을 나타냈으며 추출용매에 따른 차이는 거의 없었다. 하지만 25 mg/L 이하의 농도에서는 hexane 추출물에서 높은 항산화 활성을 나타냈다. 각각의 추출물 구성성분을 분석한 결과 hexane 추출물의 주요 구성성분은 torreyol, alpha-cadinol, tau-cadinol이며, ethyl acetate 추출물의 주요 구성성분은 alpha-amorphene, alpha-cadinol, gamma-cadinene으로 나타났다.

Antifungal Activity and Inhibitory Modes of Volatile Vapours of Essential Oils

  • Antifungal activities of volatile vapours of essential oils were investigated. Volatile vapours of test essential oils except Cedar-wood and Pachouri showed inhibitory activity against test fungi. Volatile vapours of Birch essential oils exhibited fungistatic activity against dermatophytic filamentous fungi while others did fungicidal activity. Spores of dermatophytic filamentous fungi are more susceptible to volatile vapours of essential oils than mycelia.

Antifungal Activities of Bacillus thuringiensis Isolates on Barley and Cucumber Powdery Mildews

  • Fourteen Bacillus thuringiensis isolates having both insecticidal activity and in vitro antifungal activity were selected and tested for in vivo antifungal activity against tomato late blight, wheat leaf rust, tomato gray mold, and barley powdery mildew in growth chambers. All the isolates represented more than 70% disease control efficacy against at least one of four plant diseases. Specifically, 12 isolates exhibited strong control activity against barley powdery mildew. Under glasshouse conditions, four (50-02, 52-08, 52-16, and 52-18) of the isolates also displayed potent control efficacy against cucumber powdery mildew. To our knowledge, this is the first report of B. thuringiensis isolates that have disease control efficacy against powdery mildew of barley and cucumber as well as insecticidal activity.

유기질문화재 보존을 위한 천연 추출물의 항균·살충활성 연구 (Research on antifungal and insecticidal activity of Natural extracts for Protecting Organic Cultural heritages)

  • This study evaluated the activity of biocidal compounds isolated from natural products and confirmed applicable possibility as conservative agents for orgainic cultural heritages damaged by biological degradation. we used methanol extracts of 11 Korean trees and 17 medicinal plants as natural resources. Among these extracts, Phellodendron amurense was investigated on antifungal activity against Coniophora puteana, Lentinus lepideus, Tyromyces palustris and Aspergillus niger. Asarum sieboldii was investigated on insecticidal activity against adult of Lasioderma serricorne. First, the extract of Phellodendron amurense was partitioned into hexane, dichloromethan and ethylacetate fraction. Among them, the ethylacetate fraction had the most significant antifungal activity. In addition, Asarum sieboldii essential oil showed an effective antifungal and insecticidal activity. As a result, the extract of Phellodendron amurense and Asarum sieboldii essential oil could be useful for conservation of organic cultural heritage against biological deterioration by wood rot fungi and Lasioderma serricorne.

Influence of the Hydrophobic Amino Acids in the N- and C-Terminal Regions of Pleurocidin on Antifungal Activity

  • To investigate the influence of the N- or C-terminal regions of pleurocidin (Ple) peptide on antifungal activity, four analogs partially truncated in the N- or C-terminal regions were designed and synthesized. Circular dichroism (CD) spectroscopy demonstrated that all the analogs maintained an alpha-helical structure. The antifungal susceptibility testing also showed that the analogs exhibited antifungal activities against human fungal pathogens, without hemolytic effects against human erythrocytes. The result further indicated that the analogs had discrepant antifungal activities [Ple>Ple (1-22)>Ple (4-25)>Ple (1- 19)>Ple (7-25)] and that N-terminal deletion affected the activities much more than C-terminal deletion. Hydrophobicity [Ple>Ple (1-22)>Ple (4-25)>Ple (1-19)> Ple (7-25)] was thought to have been one of the consistent factors that influenced these activity patterns, rather than the other primary factors like the helicity [Ple>Ple (4-25) >Ple (1-22)>Ple (1-19)>Ple (7-25)] or the net charge [Ple=Ple (4-25)=Ple (7-25)>Ple (1-22)=Ple (1-19)] of the peptides. In conclusion, the hydrophobic amino acids in the N-terminal region of Ple is more crucial for antifungal activity than those in the C-terminal region.

  • To develop an environment-friendly fungicide for controlling tomato wilt diseases, antifungal active substance was isolated Rheum australe D. Don roots against Fusarium oxysporum f. sp. lycopersici, a pathogen of tomato wilt, in this study. Methanol extract obtained from Rheum australe roots was successively fractionated with hexane, chloroform, ethyl acetate, butanol and water. The ethyl acetate fraction, which showed the highest antifungal activity, was separated by column chromatography, and 60 subfractions were obtained. The 60 subfractions were anlayzed for antifungal activities by bioassay. The active compound was identified as 5-[(E)-2- (3-hydroxy-4-methoxyphenyl)ethenyl]benzene-1,3-diol (rhapontigenin) by NMR and GC-MS analysis. As a result of testing antifungal activity of rhapontigenin against Fusarium oxysporum, EC50 of rhapontigenin was showed strong antifungal activity at 7.48 mg/L. Therefore, this study showed that the Rheum australe roots extract can be a potential candidate which is a environment-friendly fungicide against Fusarium oxysporum.

Screening of Cyanobacteria (Blue-Green algae) from Rice Paddy Soil for Anti-fungal Activity against Plant Pathogenic Fungi

  • Soil cyanobacteria isolated from the rice paddy fields of 10 different locations across Korea were evaluated by agar plate diffusion test for antifungal activity. Aqueous, petroleum ether, and methanol extracts from one hundred and forty two cyanobacterial strains belonging to the 14 genera were examined for antifungal properties against seven phytopathogenic fungi causing diseases in hot pepper (Capsicum annuum L). Of total cyanobacteria, nine cyanobacteria (6.34%) exhibited antifungal effects. The nine cyanobacteria selected with positive antifungal activities were two species of Oscillatoria, two of Anabaena, three of Nostoc, one of Nodularia, and one of Calothrix. Alternaria alternata and Botrytis cinerea were inhibited by nine and eight species of cyanobacteria, respectively. Rhizopus stolonifer was suppressed by only methanol extract of Nostoc commune FK-103. In particular, Nostoc commune FK-103 and Oscillatoria tenuis FK-109 showed strong antifungal activities against Phytophthora capsici. Their antifungal activity at the late exponential growth phase is related to the growth temperature and not associated with the growth parameters such as cell biomass and $chlorophyll-{\alpha}$ concentration. The high inhibition levels of antibiotics were 22.5 and 31.8 mm for N. commune FK-103 and O. tenuis FK-109, respectively. The optimal temperature for antibiotic productivity was $35^{\circ}C$.
