구기자 성분의 혈당강하작용 (Anti-diabetic Activity of Constituents of Lycii Fructus)

  • In the previous screening on antidiabetic effect of Lycii fructus by glucose transport method using $N_2$-STZ diabeted rat model, each extracts showed the potent antidiabetic activity. We obtained three compounds isolated from the water fraction, EtOAc fraction and n-BuOH fraction of Lycii fructus in the present work and their structures were identified as 1-carboxy-N,N,N-trimethylmethanaminium hydroxide inner salt, 2,4(1H,3H)-pyrimidinedione and 3,3',4',5,7-pentahydroxyflavone-3-rutinoside . Among the constituents separated from Lycii fructus, 2,4(IH,3H)-pyrimidinedione, 3,3',4',5,7-pentahydroxyflavone-3-rutinoside and ascorbic acid were shown a remarkable antidiabetic effect.

Medicinal plants used in the management of diabetes by traditional healers of Narok County, Kenya

  • The Maasai community from Kenya is highly esteemed for their strong adherence to traditional cultures and ethno medicine. This is attributed to their age-old traditional mechanisms of passing down knowledge to the younger generation. Adoption to new socio-economic lifestyle and urbanization has been associated with development of diabetes, which has been reported among some indigenous pastoral communities in Kenya. Documentation of traditional methods of treatment and management of diabetes by the Maasai has not yet been reported, yet it is noteworthy. Thirty traditional healers from Narok County were purposively selected and interviewed about traditional knowledge of antidiabetic medicinal plants, parts used, preparation dosage and administration. A total of 14 antidiabetic plant species distributed within 13 genera and 12 families were identified and documented as herbal medicine used in the management of diabetes. The most highly cited plant species was Dovyalis abyssinica (20%), the plant family Flacourtiaceae and Rhamnaceae (2 plant species each) recorded the highest number of plant species while the most frequently used plant part was the roots (46%). Literature review revealed that some of the cited plants have known phytochemicals with antidiabetic activity; the study recommends further scientific investigation to validate their efficacy and safety.

  • This study examined the antidiabetic and antihyperlipidemic activities of an ascidian (Halocynthia roretzi) in rats in vivo. Rats were fed on experimental diet including dried ascidian powder (200 mg/kg body weight) for 4 weeks, and then the triglyceride and total cholesterol levels in blood were analyzed. On the ascidian tunic powder diet, the triglyceride level decreased by more than 20.9% and the total cholesterol content decreased by more than 24.4%. In comparison, the triglyceride and total cholesterol level in the blood of rats fed ascidian meat powder decreased only slightly. Therefore, the ascidian tunic powder might be a healthy food with antidiabetic and antihyperlipidemic effects.

뽕잎분말을 첨가한 당면의 생리활성 평가 (A Study on the Physicochemical Activities of Dangmyon (starch vermicelli) Added with Mulberry Leaves Powder)

  • This study was performed to investigate the physicochemical activities such as mineral contents, antioxidantant activities, ${\partial}$-glucosidase activity, and angiotensin I-converting enzyme(ACE) inhibitory effect of Dangmyon (starch vermicelli) prepared with mulberry leaves powder. Mulberry leaves powder was added in ratio (w/w) of 0% (CON), 0.5% (MD1), 1.0% (MD2), 1.5% (MD3), and 2.0% (MD4), and then mineral contents, total polyphenol contents, antioxidant, antidiabetic, and antihypertensive activities of the Dangmyon were measured. The mineral contents were significantly increased by the addition of mulberry leaves powder (p<0.001). The total polyphenol contents and antioxidant activities of Dangmyon were significantly increased by the addition of mulberry leaves powder (p<0.05). In terms of antidiabetic and antihypertensive activities showed MD1~MD4 samples higher contents of ACE and ${\alpha}$-glucosidase compared to control (p<0.05). Our findings suggest that addition of mulberry leaves powder to Dangmyon may improve the effects of anti-oxidation, anti-DM, and anti-hypertension and provide health benefits of consumers.

Antidiabetic and Hypolipidemic Effects of Collybia confluens Mycelia Produced by Submerged Culture in Streptozotocin-Diabetic Rats

  • This investigation was undertaken to study the effects of oral administration (3 weeks) of Collybia confluens mycelial powder (CCMP) produced by a submerged culture on plasma glucose and other biochemical parameters in streptozotocin (STZ)-induced diabetic rats. Antidiabetic and hypolipidemic effects were proportionally increased with the increasing concentration of the CCMP for oral administration. The CCMP, at the dose of 400 mg/kg BW, substantially reduced the plasma glucose level by as much as 33.1 % as compared to the STZ-induced diabetic rats group. It also lowered the plasma total cholesterol, triglyceride, and low density lipoprotein (LDL) cholesterol by 22.9%, 19.9%, and 37.3%, respectively. The levels of total cholesterol and triglyceride in liver were reduced to the extent of by 13.5% and 18.8%, and the activity of alanine transaminase (ALT) and aspartate transaminase (AST) was decreased by 48.8% and 37.2%, respectively, under the influence of CCMP. The general components of CCMP were found to contain 26.18% carbohydrate, 3.67% crude ash, 4.02% crude fat, 22.55% crude protein, and 43.58% dietary fiber. The amino acid composition of the CCMP was also analyzed in detail.

  • Objecitivies : While Glechoma longituba (Nakai) Kupr(GL) has been known as a antiinflammatory, antidiabetic and antihypertension medicine in korea traditional medicine, there has been only a few researches about it in Korea. Thus, we examined antidiabetic effect of GL in diabetic rats induced by streptozotocin Methods : To verify the effects of GL on serum glucose, creatinine, BUN, total cholesterol and triglyceride through streptozotocin-induced diabetic rats, some rats were injected 1 time with 55mg/kg streptozotocin intraperitoneally to induce diabetes, and other rats were injected with buffer I.P. as control group. Results : GL extracts significantly reduced the amount of serum glucose, BUN, creatinine, and triglyceride level in diabetics rats induced by streptozotocin. Conclusions : According to above result, Glechoma longituba was expected to be applied to the treatment of diabetes mellitus.

홍국발효 대두의 항산화 및 항당뇨 활성 (Antioxidant and Anti-Diabetic Activities of Soybean Fermented with Monascus)

  • This study was performed to investigate the antioxidant and antidiabetic activities of soybean fermented with Monascus. Also, the changes in the content of isoflavones and Monacolin K were analyzed. It was observed that the glycoside forms of daidzin and genistin were converted to aglycones of daidzein and genistein within 6 days of fermentation. The product can be used as a health functional material that can increase bioavailability. Monacolin K production was found to increase significantly with the progression of fermentation with an increase to 0.04 mg/g and 0.44 mg/g on 6 and 12 days of fermentation, respectively. The DPPH radical scavenging activity of soybean fermented with Monascus was significantly increased compared to that of soybean. The protein expression of inflammation-related genes (TNF-α, IL-6, and COX-2) in the MIN cell was significantly increased in the presence of alloxan compared to the normal group, but a decrease was observed in the presence of soybean fermented with Monascus. In conclusion, soybean fermented with Monascus showed the highest antidiabetic and antioxidant effects. These results suggest that soybean fermented with Monascus has the potential to be used as a beneficial ingredient with antidiabetic and antioxidant effects.

Development of protein tyrosine phosphatase 1B (PTPIB) Inhibitors from marine sources and other natural products-Future of Antidiabetic Therapy : A Systematic Review

  • The incidence of both obesity and Type 2 Diabetes Mellitus( DM) is increasing proportionately so that causes of deaths from these has overtaken from that of malnourishment. Hence it has been recommended to treat the 2 in parallel considering the role of diabesity on health. Important causes of T2DM are insulin resistance (IR) and /or inadequate insulin secretion. Protein tyrosine phosphatase 1B(PTPIB) has a negative impact in insulin signaling pathways and hence plays crucial role inT2DM,since its overexpression might induce IR. Thus PTPIB is considered a therapeutic target for both obesity and T2DM, there has been a search for novel ,promising natural inhibitors. We conducted a pubmed search for articles related to PTPIB inhibitors from natural causes be it marine sources or other natural sources. Out of 988 articles we selected 100 articles for review. Thus various bioactive molecules isolated from marine organisms that can acts as PTPIB Inhibitors and thus possess antidiabetic activity both in vitro/ in vivo studies ,besides products from fruits like Chinese raspberry or curcumin used as routine spices are described with their chemical classes, structure-activity relationships and potency as assessed by IC 50 values are discussed. More work is required to make this a reality.

Simultaneous Determination of Anthraquinone, Flavonoids, and Phenolic Antidiabetic Compounds from Cassia auriculata Seeds by Validated UHPLC Based MS/MS Method

  • A systematic isolation and characterization study for Cassia auriculata (CA) seeds resulted in identifying antidiabetic compounds 1,3,8-trihydroxyanthraquinone and quercetin, quercetin-3-O-rutinoside, gallic acid, caffeic acid, ferulic acid, and ellagic acid. The ultra-high-performance liquid chromatography based triple quadrupole mass spectrometry methodology was developed and validated for simultaneous identification and confirmation of these compounds from CA seeds. Multiple reaction monitoring (MRM) based quantification method was developed with MRM optimizer software for MS1 and MS2 mass analysis. The method was optimized on precursor ions and product ions with the ion ratio of each compound. The calibration curves of seven bioactive analytes showed excellent linearity (r2 ≥ 0.99). The quantitation results found precise (RSD, < 10 %) with good recoveries (84.58 to 101.42%). The matrix effect and extraction recoveries were found within the range (91.66 to 102.11%) for the CA seeds. This is the first MS/MS-based methodology applied to quantifying seven antidiabetic compounds in CA seeds and its extract for quality control purposes.

추출용매 변화에 따른 바나바(Lagerstroemia speciosa Pers.) 잎 추출물의 항당뇨 효과 (Anti-diabetic Effects of Banaba Leaf Extracts (Lagerstroemia speciosa Pers.) through Solvents)

  • 본 연구는 바나바 잎 추출 용매에 따른 항당뇨 효과에 관한 것으로 세포 독성 실험을 통하여 인체에 대한 독성이 없음을 확인하였고 $\beta$-cell 인슐린 분비 실험을 통하여 항당뇨 효과를 알아보았다. 또한 당뇨 유발쥐를 이용한 인슐린 분비 실험, 체중변화 측정, 혈중 및 혈청의 glucose 농도, 총콜레스테롤 수치, HDL-cholesterol 농도, triglyceride 농도, free fatty acid 농도, langerhans islet에서의 인슐린 분비 의 확인을 통해 혈당조절 효과를 비교하였다. 물 혹은 에탄올을 단독으로 이용하여 추출한 추출물이나 70%, 90% 에탄올로 추출한 추출물 보다 열수로 먼저 추출한 후 에탄올로 다시 추출하여 혼합안 추출물이 항당뇨 효과가 더 뛰어남을 알 수 있었다. 이는 친수성(hydrophilic) 유효성분들과 소수성(hydrophobic) 유효성분들의 추출 유무나 함량에 의한 것이다. 특히, 많은 문헌에 언급된 코로소린산이라는 단일 성분(unique)에 의한 항당뇨 효과라기보다는 친수성 및 소수성 폴리페놀 및 항산화성분과 같은 유효성분들이 조합되어(mixture) 항당뇨 효과에 영향을 미치는 것으로 사료된다.
