한국산 고등균류의 항보체활성 효과에 관한 연구 (Studies on the Anticomplementary Activity of Korean Higher Fungi)

  • 국내에서 자생하는 61주의 고등균류로부터 면역증강효과를 검색하기 위해 이들로부터 성분을 분리하여 이들의 항보체활성을 측정한 결과, krestin의 보체활성도인 20.6%보다 우수한 보체활성 효과를 나타낸 것은 11주 였으며, 이 중에는 Ganoderma lucidum 5주, Lentinus edodes 3주, Cordyceps sp. 3주 및 Agaricus campestris 1주이다.

보리(Hordeum vulgare)추출물의 항보체 및 면역증강 효과 (Anticomplementary Activity and Immune-Stimulating Effect of the Extracts from Barley (Hordeum Vulgare))

  • By using several solvents, barley extracts containing the anticomplementary activities in classical pathway were prepared (250 $\mu\textrm{g}$/ml): methanol (83.1%), ethanol (71.9%), water extract (25.4%), M-1 (250 $\mu\textrm{g}$/ml), and the soluble part of methanol extract which showed the highest activity (83.4%) and the yield. Anticomplementary activity of methanol extract as well as protease digestion in classical pathway showed 82.4% and 78.4% in the concentration of 250 $\mu\textrm{g}$/ml, respectively. It was found that protein was not involved in anticomplementary activity in the classical pathway and the methanol extract made an impact on classical pathway, but not on alternative pathway. For the immune-stimulating effect, the T cell proliferation effect of the protease digestion displayed little effect irrespective of the dose. In addition, the T cell proliferation effect of methanol extract showed 13-fold higher proliferation effect compared with positive control. It was revealed that the substance containing protein serves as an important factor for the immune proliferation. Therefore, the anticomplementary activity ${\beta}$-glucan in classical pathway and alternative pathway displayed the lowest activity, showing 2.2%, 22.3% respectively. However, the immune-stimulating effect of ${\beta}$-glucan showed the T cell stimulating effect 13 times higher than positive control.

  • The anticomplementary activity of stilbenes from medicinal plants in Korea was investigated in vitro. 3,5-Dihydroxy-4'-methoxystilbene (3) was most potent with $IC_{50}$ value of $1.5{\times}10^{-4}M$ followed by rhapontigenin (4), oxyresverastrol (2), 2,3,4',5-tetrahydroxystilbene-2-O-beta-glucoside (9), rhaponticin (8), resverastrol (1), and piceid (7). The activity was found to be increased by a methylation on a hydroxy group of C-4' of 1, but decreased by further methylation on hydroxy groups of C-3 and C-5 and glucosylation on any hydroxy group of 1. Addition of hydroxy group on C-2' of 1 or C-3' of 3 was little affected on the anticomplementary activity but the activity was increased by O-glucosylation on C-2 of 1.

  • A very high content (at least 0.23%) of ergosterol peroxide was isolated from Naematoloma fasciculare Karst. Not only ergosterol peroxide but also ergosterol showed very strong anticomplementary activity on the classical pathway, the $IC_{50}$ values being $5.0 {\mu}M$ and $1.0 {\mu}M$, respectively. The $ ^{1}H $and $^{13}C$ NMR data of ergosterol peroxide were revised and completely assigned by DEPT, $^{1}H-^{1}H$ COSY, HMQC and HMBC correlations.

  • Gel filtration과 ion exchange chromatography, reversed-phase 및 ion exchange FPLC를 이용하여 고려인삼 뿌리에서 subunit 분자량 약 25 kDa의 주요단백질(이하 GMP)을 분리하였다. PAS 염색을 통하여 GMP는 carbohydrate moiety를 갖는 당단백질일 가능성을 보여주었고, 이는 glycosidase 처리후 protein band shift 실험으로 확인되었다. GMP는 native polyacrylamide 전기영동 및 gel permeation FPLC를 통해 native form은 약 63 kDa의 분자량을 갖는 dimer로 판단된다. GMP의 생리활성 측정결과, inflammation mediator의 기능탐색에 쓰이는 anticomplementary activity를 보여주었다.

  • Antitumor polysaccharides were known to activate complement system and to increase specific serum proteins in mouse, and researcher reported that antitumor activity of polyasccharides might be correlated with their biological properties such as activation of complement system and increase of specific protein $L_{A}$, $L_{B}$ and $L_{C}$ within the mouse serum. In case of several Ganoderma lucidum, there was no correlation between their antitumor activities and their bioloical properties, but the antitumor activities against sarcoma 180 of the alkali extracted crude polysaccharide fractions of the Ganoderma lucidum IY 009. AS, T, AI and M were correlated with their bioloical properties such as anticomplementary activity and intensity of mouse serum protein $L_{A}$, $L_{B}$ and $L_{C}$.

  • The quality of an intravenous immunoglobulin preparation (IVIG) is reflected by the degree of nonspecific activation of complements, the so-called anticomplementary activity (ACA). ACA of aggregates in IVIG was investigated using method by the European Pharmacopoeia and Clq-coated microtiter enzyme-linked immunosorbent assay (ELISA). Both the EP method and the ELISA method showed a dose response curve with the amount of complements bound increasing with the percentage content of aggregates in immunoglobulin standard. The correlation between the two tests was good (r=0.96, r=0.99). However, the correlation was not found when the ACA (EP method) of IVIG product was compared with its aggregate percentage. These results emphasize that the method of aggregate formation affects ACA and that estimation of the percentage distribution of aggregates by HPLC may not reflect ACA. In analysing WIG product for Clq binding activity test with the ELISA, the result by using Protein A-HRP correlated with aggregate percentage (r=0.84). But the correlation decreased (r=0.48) when the result used Protein A-AP(having poorer sensitivity than HRP) was compared with aggregate percentage. As a result, some variation between the two methods, due to differences in assay principles, is to be expected. However, ELISA technique has the advantage in that it is easier to perform, more precise and less subject to reagent variability, and is the more suitable screening method than HPLC analysis.

  • Anticomplementary activity of hederagenin and related saponins isolated from Dipsacus asper was investigated in vitro. HN saponin F (3) was most potent with $IC_{50}$ value of$ 3.7{\times}10^{-5} M$ followed by 3-O-${\beta}-D-glucopyranosyl-(1{\rightarrow} 3)-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}2)-{\beta}-L-arabinopyranosyl$ hederagenin $28-O-{\beta}-D-glucopyranosyl-(1{\rightarrow}6)-beta$-D-glucopyrano side (8), $3-O-{\beta}-L-arabinopyranosyl$ hederagenin $28-O-{\beta}-D-glucopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (5), dipsacus saponin A (4), and hederagenin (1) on the classical pathway (CP) of complement system, while the saponins 3-5 did not show the inhibition of hemolysis and rather increase the hemolysis on the alternative pathway (AP). However, all of C-3 monodesmosides [prosapogenin CP (2), dipsacus saponin B (6), and dipsacus saponin C (7)] evoked hemolysis directly on the erythrocytes.

Ganoderma lucidum IY009 균사체로부터 추출된 ganoderan의 항암 및 항보체 활성에 미치는 탄소원의 영향 (The effects of carbon sources on antitumor and anticomplementary activities of Ganoderan extracted from the mycelium of Ganoderma lucidum IY009)

  • 영지 균사체로부터 종양 면역 활성이 우수한 다당류를 얻기 위하여 여러 탄소원을 이용하여 균사체를 배양하였다. 배앙된 균사체로부터 ganoderan을 추출하고 이들의 조성과 항암 및 항보체 활성능을 알아보았다. 여러 탄소원은 영지 균사체에 의해 기질로 이용될 수 있으나 ${\beta}-glucan$성 다당류인 ganoderan의 생성에는 상당한 차이를 보였다. 균사체 생육이 우수한 탄소원에서 배양된 균사체의 경우, 비수용성(WI) 다당류의 생성이 증가하였고, 생육이 저하된 탄소원에서는 수용성(WS) ganoderan이 상대적으로 증가하였으며, 포도당을 탄소원으로 사용할 경우 약 $5.2{\sim}16%$가 알칼리 가용성 ganoderan으로 도입되었다. 탄소원에 따라 수획된 ganoderan은 포도당을 주 구성당으로 이루어진 ${\beta}-glucan$성 다당류이며 만노오스, 과당 등이 포함되어 있었으나, 비수용성 ganoderan의 경우 수용성보다 포도당의 함량이 약 $20{\sim}40%$ 더 많이 포함되었으며 약 16개의 아미노산으로 구성된 소량의 단백질이 결합되어 있었다. WI은 glucose가 주 구성당이었으나 WS는 여러 단당들이 상대적으로 많이 포함되어 있는 heteroglucan의 성격을 보였다. 탄소원에 따라 배양된 균사체의 알칼리 추출된 ganoderan 가운데 항암 및 항 보체 활성이 가장 우수한 것은 갈락토오스를 탄소원으로 이용한 ganoderan이었으며 항보체 활성(ITCH 50,%)치가 35.7%였고, 특히 균사체의 생장이 낮은 탄소원에서 수획한 ganoderan이 항보체 활성이 높게 나타났다. Sarcoma 180이 피하이식된 마우스에 20 mg/kg씩 복강 투여하여 항암 활성을 검토한 결과, 항보체 활성실험에서와 마찬가지로 갈락토오스를 탄소원으로 이용한 ganoderan-GAL이 83.6%의 암 저지율을 보여 항암 활성이 가장 높았으며, 이는 약 5%의 단백질이 결합된 ${\beta}-glucan$성 다당류로 55%의 포도당, 13%의 만노오스, 12%의 과당 및 소량의 galactose, xylose, ribose등으로 구성된 분자량 30 kD의 다당류이다.

In Vitro Anticomplementary Activity of Phenylpropanoids from Agastache rugosa

  • In searching for anticomplementary compounds, three phenylpropanoids were isolated from the roots of Agastache rugosa and identified as rosmarinic acid (RA), rosmarinic acid methyl ester (RAM) and caffeic acid methyl ester (CAM) by NMR analyses. RA and RAM exhibited strong inhibitory activity on both the classical pathway (CP) and the alternative pathway (AP) of the complement system, in vitro, but CAM did far less than RA and RAM. $RAM-M1{\sim}-M5$, the methylated derivatives from the RAM, showed that the inhibitory activity was decreased in inverse proportion to the number of methylated groups and $RAM-M 2{\sim}-M4$, the isomers of two methylated hydroxyl groups, exhibited different inhibition activity.
