Optimization of Immunosasay Procedures for the measurement of Progesterone (Progesterone 측정을 위한 면역분석법의 최적조건에 관한 연구)

  • This research was carried out to investigate the optimal conditions ssociated with the RIA procedures such as a bridging phenomena, prozone effects and a new separation methods etc. The results obtained were summarized as follows: 1. The lgG fractions of donkey anti-rabbit IgG sera were purified by Protein-A-Sepharose affinity column, which indicates that Protein-A an affinity for IgG class of donkey antiserum. 2. In coating the IgG fraction on polystryene tubes, incubation conditions made no differences between 2 hr at room temperature and overnight at 4$^{\circ}C$. 3. There were no significant differences between 1st antibody-coated tube and 2nd antibody-coated tube as a separation method when compared in terms of reproducbility. A better reproducibility may be expected if the titers of 1st antibody for the progesterone to be assayed and of corresponding 2nd antibody are reasonably high. 4. The titers of anti-progesterone antibody for 3H-progesterone and progesterone-11HS-125I were 1:300 and 1:700 in liquid-phase, and 1:100 and 1:300 in solid-phse for the separation methods. 5. A bridging phenomena in which a standard curve is long and shallow were observed when progesterone-11HS-125I was used for the tracer, but not in 3H-progesterone. 6. A prozone effect in a solid-phase system, especially 1st antibody-coated tube method was observed which the degree of inhibition was significantly different although zero bindings look the same. In this case, the titration curve should be made both in the absence and in the presence of a, pp.opriate amount of competiter, standard, respectively.

  • To identify immune response of leukocytes in peripheral blood of cattle vaccinated with an attenuated live Akabane virus vaccine, leukocytes were reacted with monoclonal antibodies which are specific to bovine lymphocyte surface antigens and assayed by the flow cytometry. Serum neutralizing (SN) test was used to measure antibody titers after vaccination, SN antibody was appeared to 7 days post-vaccination (PV) and 2-8 antibody titers were observed in 14 days PV. Proportion of $CD8^-$ MHC $class II^+$ expressing cells were rapidly increased at 3 days PV. $CD8^+$ MHC $class II^-$ cells were increased at 7 days PV. $CD4^+CD8^-,\;WC^+CD4^-,\;CD4^+CD8^+,\;WC1^-CD4^+, \;WC1^-CD8^+$, and $CD4^-CD8^+$ cells were highly increased at 3, 3, 7, 7, 14, 14 days PV, respectively.

  • Objective: The effectiveness of Staphylococcus protein A (SPA) in improving the penetration ability of sperm and reducing antisperm antibody (ASA) titers in immunologically infertile males was evaluated. Methods: Seminal fluid samples were obtained from 15 infertile men, and ASA titers were assessed with the latex agglutination test. Identification of immunoglobulin (Ig) classes and characterization of the antigens involved in the immune response were performed using indirect immunofluorescence. Local ASAs typically present as a mixture of IgG and IgA classes. The capillary tube penetration method was used to assess the capability of spermatozoa to penetrate the cervical mucus (CM). Results: ASAs associated with the neck region of sperm showed a significantly lower migration distance in the CM of infertile females than ASAs associated with the head or tail segments. ASA-positive seminal fluid exhibited significant increases in the mean migration distance (2.6 ± 1.4 cm vs. 1.54 ± 1.1 cm, respectively; p< 0.001) and sperm concentration (174 ± 121.0 × 103/mL vs. 101 ± 93.7 × 103/mL, respectively; p= 0.033) after treatment with SPA compared to pre-treated samples. A significant reduction (p< 0.01) in the recorded ASA titer was detected. Conclusion: These results indicate that SPA can be used as a sorting regimen for insemination programs. However, further studies are warranted to assess its influence on pregnancy rate.

Studies on the effects of immunization against Babesia gibsoni antigen and Theileria sergenti as a non-specific antigen in dog (Babesia gibsoni 항원접종과 Theileria sergenti를 비특이 항원으로 접종한 개의 면역효과에 관한 연구)

  • To examine the effects of vaccination against Babesia gibsoni(B gibsoni) infection in dogs, 15 normal mixed-breed dogs(5 months to 1 year old) were divided into 3 groups with 5 dogs in each group. One of them was selected as control group(group A) and other were selected as experimental groups(group B and C). The group B was vaccinated with antigen which were mixed 0.2% of formalin treated B gibsoni and sonicated one. The group C was inoculated Theileria sergenti as a non-specific antigen. The results obtained in this experiment were summarized as follows; 1. After first vaccination, antibody titers of group B and C were increased 5 times(1:200) than those of control group(1 : 40). The antibody titers of group C were increased more than that of group B after second vaccination. When challenged with the living protozoa(Babesia gibsoni), the antibody titers of C group were elevated higher than that of B group and maintained steadly. Those were not exceeded over 1 : 5,000 for 4 weeks in all 3 groups. 2. After challenge, the peak time of the parasitemia appeared nearly on the 15th day(12~18 days) in all groups. During this period, the rate of parasitized erythrocytes in control group was $55.0{\pm}5.4$‰. But that of group B and C were $41.3{\pm}38.8$‰ and $15.2{\pm}16.3$‰, respectively. 3. After challenge with B gibsoni, all of the values of PCV, Hb, RBC and total leukocytes counts were decreased in both of the experimental and the control. 4. In all groups, there were increased lymphocytes and monocytes after challenge with the protozoa.

Influence of Antithyroid Antibodies in Euthyroid Women on IVF-ET Outcome (정상 갑상선기능을 가진 여성에서 항갑상선항체가 체외수정시술결과에 미치는 영향)

  • The present study was designed to investigate if antithyroid antibodies (ATA) could affect the pregnancy outcome in euthyroid women undergoing in vitro fertilization and embryo transfer (IVF-ET). From October 1995 to September 1996, 28 euthyroid women with ATA who underwent IVF-ET were studied. Fifty-one euthyroid women without ATA who underwent IVF-ET served as control. Thyroid peroxidase antibody (TPOA) and thyroglobulin antibody (TGA) were assayed using radio ligand assay kits as ATA. All patients included in study and control groups had only tubal factor in infertility. Long protocol of gonadotropin-releasing hormone agonist (GnRH-a) was used for controlled ovarian hyperstimulation (COH) in all patients. There were no significant differences between study and control groups in patient characteristics such as age, infertility duration and hormonal profile. There were also no significant differences between two groups with respect to the clinical response to COH and IVF results such as number of retrieved oocytes, fertilization rate, number of embryos frozen and number of embryos transfered. There were no correlations between ATA (TPOA and TGA) titers and fertilization rate. The clinical pregnancy rate per cycle seemed to be lower in the study group than in the control group (26.3% vs 39.3%), but the difference was not statistically significant. The biochemical pregnancy rate per cycle and miscarriage rate were significantly higher in the study group at 18.4% (7/38) and 40.0% (4/10) compared with 5.6% (5/89) and 11.4% (4/35) in the control group. In the study group, both TPOA and TGA titers were significantly higher in the biochemical pregnancy group than in the clinical pregnancy group or non-pregnancy group. In 10 women with ATA who achieved pregnancy following IVF-ET, both TPOA and TGA titers were significantly higher in the miscarriage group than in the ongoing or delivery group. In conclusion, euthyroid women with ATA appear to represent a less favorable subset within other tubal factor patients when treated with IVF-ET.

  • In this study, effect of levamisole, selenium and tocopherol on the lymphocyte blastogenesis and antibody production in Korean native goat were evaluated in vitro and in vivo. Lymphocyte blastogenesis of goat blood increased significantly (p<0.01 and p<0.05) when the cells were treated in vitro with levamisole at the concentration of $50{\sim}500{\mu}g/ml$, with selenium at the concentration of $0.062{\sim}1.0{\mu}g$ and with tocopherol nt the concentration on $12.5{\mu}g$. Increased lymphocyte blastogenesis was detected from 2 to 24 hours after oral administration of levamisole (2.5mg/kg of body weight). After 7 days, increased mitogenic response of lymphocytes was not detected. Meanwhile increased blastogenesis of lymphocyte from goats given the selenium-tocopherol mixture (selenium $100{\mu}g$-tocopherol 200IU/head/day) was detected from 10 days after feeding, and the tendency continued throughout the entire experimental period. When immune responses of goats against PPD were subjected to test by ELISA, the mean IgG titers of levamisolc group (1 : 1,800) and selenium-tocopherol group (1 : 960) were higher than that of control group (1 : 600) at 2 weeks after lst inoculation. At 3 weeks after lst inoculation and 1 week after 2nd inoculation, the significant (p<0.05) differences in IgG titers were detected among the three groups. The mean IgG titers of levamisole group, selenium-tocopherol group and control at that time were 1 : 20,480, 1 : 5,120 and 1 : 2,640, respectively. The IgG production of levamisole group was significantly (p<0.01) higher than that of control group.

In the western blot, the lane A revealed specific two bands on the regions of 54kd and 100kd, respectively. 2. After the first vaccination, the antibody titers of group B and C were higher 5 times(1 : 200) than those of control group(1:40). After the second vaccination, the antibody titers of group B and C have not changed. When challenged with the protozoa(Babesia gibsoni), the antibody titers(1 : 5,000) were elevated in all groups. But these were not exceeded over 1 : 5,000 for 4 weeks. 3. After challenge, the peak time of increased numbers of the protozoa was the 15th day (12-18 days) in all groups. During these days, the rate of parasitized erythrocytes in control group was 55.0${\pm}$5.4%. But those of group B and group C were 26.0${\pm}$6.4%, and 15.6${\pm}$7.8%, respectively. 4. After challenge, all of the values of PCV, Hb, RBC were shown to decrease in all of the control and experimental groups. 5. The total leukocytes counts are shown a tendency of reduction in all groups after challenge. 6. In all groups, there were increase in lymphocytes and monocytes after challenge.

A STUDY ON THE CHANGES OF THE ROOT SURFACE PLAQUE FLORA AND GINGIVAL CREVICULAR FLUID ANTIBODY TITERS AFTER ANTIMICROBIAL VARNISH TREATMENT (치근면에 항균 varnish 처치 전후의 치태 세균 및 치은열구액내 항체수준 변화에 관한 연구)

  • In the prevention of root surface caries, antimicrobial therapy for the control of subgingival and supragingival plaque is seriously considered as a long term suppression of pathogenic microflora. Recently, varnishes containing antimicrobial agents have been developed to control the supragingival microflora. The purpose of this study was to determine the antimicrobial effects of 20% chlorhexidine varnish and 2.6% silane fluoride varnish with sealant. In clinical experiments, 12 subjects were selected from the periodontally treated patient and divided into 3 groups. After a dental prophylaxis, the subjects were treated with single application of placebo varnish (group I), 20% chlorhexidine varnish (group II), and 2.6% silane fluoride varnish (group III). Root surface plaque samples were taken before (baseline) and one, two, four, and 8 weeks after the treatments. Microbiological examinations of root surface plaque were performed with culture study and indirect immunoflorescence (I.I.F.) study, and immunological examination of gingival crevicular fluid antibody titers was performed with ELISA study. The results were as follows: 1. Pathogenic microflora on the root surface including S. mutans, S. sanguis, S. mitis, A. naeslundii, A. viscosus were 24 - 37% on I.I.F. study. 2. S. mutans, S. sanguis, S. mitis, A. naeslundii, A. viscosus of the root surface plaque was significantly reduced from 1 week to 8 weeks after antimicrobial varnish treatment, but showed generally increasing tendency in control group. 3. Gingival crevicular fluid antibody titers were significantly reduced from 1 or 2 weeks to 4 weeks after antimicrobial varnish treatment.

Development of ELISA for detection of canine brucellosis (Canine brucellosis 검출을 위한 ELISA 진단법 확립)

  • This study was carried out to develope enzyme-linked immunosorbent assay (ELISA) for detection of canine brucellosis in dogs experimentally inoculated with Brucella abortus 1119-3 and B. canis RM666. Groups A, B and C of dogs (each group consisting of three dogs) were orally inoculated with approximately $5{\times}10^9$ colony-forming units of B. abortus and B. canis, and with sterile pyrogen-free PBS, respectively. The animals were monitored at regular intervals upto the 12th week post inoculation (PI) by standard tube agglutination test (STAT), plate agglutination test (PAT), Rose Bengal test (RBT), 2-mercaptoethanol rapid slide agglutination test (2ME-RSAT) and ELISA. The induced antibody titers in group A dogs were detected from the first week PI to the eighth week PI in STAT, PAT and RBT using the inactivated whole cells of B. abortus 1119-3 as antigens, while no sera in groups B and C dogs reacted with the antigens. In 2ME-RSAT using whole cells of B. canis M-strain as antigens, the induced antibody titers in group B dogs were observed at the second week PI and persisted for the 12th week PI, while sera of groups A and C dogs did not react with the whole cells. In ELISA using cytoplasmic fractions antigen of B. abortus 1119-3, the mean optical density of antibodies in groups A and B was detected from the first and second weeks PI, respectively, and persisted for 12th week PI, while sera of group C did not cross-react with the fractions antigen. However, in ELISA using the hot saline extracts of B. canis M- as an antigen, the induced antibody titers in only group B dogs were detected from second week PI and persisted for until the end of this study. These results indicate that the ELISA using B. abortus 1119-3 cytoplasmic fractions as antigens can be a good candidate for detection of brucellosis by B. abortus as well as B. canis in dogs.
