Ehrlichia canis Infection in a Dog (개 엘리키아 감염 1 증례)

  • 3-year-old female Pitbull terrier dog that had been moved to Republic of Korea was diagnosed with Ehrlichia canis infection. Abnormal findings on a complete blood count (CBC) and serum chemistry profile were moderate anemia, mild thrombocytopenia, hyperproteinemia and hyperglobulinemia. Serologic screening test by a commercial ELISA kit for Ehrlichia canis was positive, and serum antibody titer was markedly high (> 1 : 10240). The morula of Ehrlichia organisms was not detected in buffy coat blood smears. Polymerase chain reaction (PCR) was done using the peripheral blood and the result was negative. Based on the serologic test results and the clinical signs, the dog was diagnosed as ehrlichiosis. The dog responded well to doxycycline and was uneventfully recovered.

Two adolescent patients with coexistent Graves' disease and Moyamoya disease in Korea

  • Moyamoya disease is a cerebrovascular condition that results in the narrowing of the vessels of the circle of Willis and collateral vessel formation at the base of the brain. Although relationships between Graves' disease and cerebrovascular accidents in Moyamoya disease are obscure, the coexistence of the two diseases is noteworthy. Moyamoya disease has been rarely reported in adolescent patients with thyrotoxicosis. Recently, we encountered two adolescent Korean patients with Moyamoya disease associated with Graves' disease who presented with episodic right-sided hemiparesis and syncope. These two girls who had Graves' disease had no history of other diseases or head trauma. A thyroid function test revealed a euthyroid state and a high thyroid-stimulating hormone (TSH) receptor antibody titer at that time. The patients were diagnosed with Moyamoya disease based on brain magnetic resonance angiography and cerebral four-vessel angiography. The patients underwent cranial revascularization by encephalo-duroarterio-synangiosis as soon as a diagnosis was made, which resulted in successful symptom resolution. They fared well and had no additional neurological symptoms as of their last follow-up visits. Here, we report these two cases of confirmed Moyamoya disease complicated by Graves' disease with a review of the literature, and discuss the possible association between the two diseases. To our knowledge, this is the first report in South Korea on Moyamoya disease associated with Graves' disease in adolescents with a euthyroid.

Effects of the Butanol Extact of Head of Panax Ginseng on Type II Collagen-induced Arthritis in DBA/1J Mice

  • In order to evaluate the improvement effects of head of Panax ginseng on chronic arthritis, we have investigated the activity of butanol fraction (BuOH fraction) in vitro and in vivo system. BuOH fraction showed significant inhibition on the elastase activity. Anti-arthritic activity of BuOH fraction was also examined on type II collagen-induced arthritis in DBA/1J mice. Mice were immunized with injection of type II collagen emulsified in Freund's complete adjuvant, followed by a booster injection 21 days later. BuOH fraction(BHPG) was administered at an oral dose of 500mg/kg for 2 weeks from the 1st day boost. The hind paw edema was significantly decreased in the group of treatment with BuOH fraction compared to control. In collagen-induced DBA/1J mice, BuOH fraction did not affected the collagen antibody titer but significantly inhibited the tumor necrosis factoralpha(TNF-${\alpha}$) activity. These results were confirmed with histological evaluation of joint tissues. This study may raise the possibility that the usage of BuOH fraction of head of Panax ginseng as alternative medicine for the relief and prevention of rheumatoid arthritis symptoms.

Development of inactivated Akabane and bovine ephemeral fever vaccine for cattle

  • Akabane and bovine ephemeral fever (BEF) viruses cause vector-borne diseases. In this study, inactivated Akabane virus (AKAV)+Bovine ephemeral fever virus (BEFV) vaccines with or without recombinant vibrio flagellin (revibFlaB) protein were expressed in a baculovirus expression system to measure their safety and immunogenicity. Blood was collected from mice, guinea pigs, sows, and cattle that had been inoculated with the vaccine twice. Inactivated AKAV+BEFV vaccine induced high virus neutralizing antibody (VNA) titer against AKAV and BEFV in mice and guinea pigs. VNA titers against AKAV were higher in mice and guinea pigs immunized with the inactivated AKAV+BEFV vaccine than in animals inoculated with vaccine containing revibFlaB protein. Inactivated AKAV+BEFV vaccine elicited slightly higher VNA titers against AKAV and BEFV than the live AKAV and live BEFV vaccines in mice and guinea pigs. In addition, the inactivated AKAV+BEFV vaccine was safe, and induced high VNA titers, ranging from 1 : 64 to 1 : 512, against both AKAV and BEFV in sows and cattle. Moreover, there were no side effects observed in any treated animals. These results indicate that the inactivated AKAV+BEFV vaccine could be used in cattle with high immunogenicity and good safety.

  • Lyme disease, or Lyme borreliosis. is an infection caused by spirohete Borrellia burgdorferi. This disease was recognized in Lyme, Conneticut U.S.A. in 1975. The onset of the disease is usually heralded by the appearance of a pathognomic skin lesion, known as erythema chronicum migrans, and accompanied by flue like or meningitis like symptoms. Unless treatment is initiated early, the disease usually disseminated, often resulting carduac, neurologic, or joint manifestations. All stages of the disease are usually curable by appropriate antibiotic therapy, and can prevent severe late cardiac, neurologic, and joint complications. Lyme disease is typically defined by clinical evidence supported by serologic test. The diagnosis require serologic confirmantion of erythema chronicum migrans, occurring in patient in nonendemic countries. Determination of antibody titer against B. burgdorferi by enzyme linked immunosorbent assay(ELISA) currently the most practical diagnostic test. Currently Lyme disease occurs in U.S.A. Europe, and Australia. It has recently recognized in China, Japan, and Soviet Union also. In United States, Lyme disease is most common vector borne infection. There is no reported case of patients with this disease in Korea. But the vector of this disease, -Ixodes ticks- had been identified in Korea. And Korea is geographically closely related to China and Japan where Lyme disease is already reported. We expect first case of Lyme disease could he reported in near future. We review the clinical manifestations and diagnostic method of Lyme disease.

Early Growth Response-1 Plays a Non-redundant Role in the Differentiation of B Cells into Plasma Cells

  • Early growth response (Egr)-1 is a $Cys_2-His_2-type$ zincfinger transcription factor. It has been shown to induce survival and proliferation of immature and mature B cells, respectively, but its role in the differentiation of B cells into plasma cells remains unclear. To examine the effects of Egr-1 deficiency on the activation of B cells, naive B cells from $Egr1^{-/-}$mice and their wild-type (WT) littermates were activated to proliferate and differentiate, and then assayed by FACS. Proportions of cells undergoing proliferation and apoptosis did not differ between $Egr1^{-/-}$ and WT mice. However, $Egr1^{-/-}$ B cells gave rise to fewer plasma cells than WT B cells. Consistently, $Egr1^{-/-}$ mice produced significantly lower titer of antigen-specific IgG than their WT littermates upon immunization. Our results demonstrate that Egr-1 participates in the differentiation program of B cells into plasma cells, while it is dispensable for the proliferation and survival of mature B cells.

Megaoesophagus Seconday to Myasthenia Gravis in Three Dogs (중증근무력증에 의한 개의 거대식도증 3례)

  • Three dogs (An 8 years-old intact female Poodle, a 7 years-old intact male Schunauzer, and an 8 yearsold Golden Retriever) were presented due to acute vomiting, dyspnea, and generalized weakness. Megaesophagus was confirmed through radiographic examination in all 3 dogs. Relative oesophageal diameter (ROD) was measured and results of ROD measurements showed the possibility of megaesophagus secondary to myasthenia gravis in three dogs. Thus we performed anticholinesterase test as screening test for myasthenia gravis. In all three dogs, esophageal diameter was reduced after neostigmine methylsulfate administration. For definite diagnosis of acquired myasthenia gravis, serum acetylcholine receptor antibody titer was measured, but definite diagnosis was confirmed only in one case. However, based on history, radiographic findings, anticholinesterase test, ROD measurement, other two cases were still suspected as megaesophagus secondary to myasthenia gravis. Treatment with pyridostigmine bromide was initiated in all dogs, and improvement of esophageal diameter was shown in all dogs. One dog was successfully managed for 15 months after initial treatment and, is still alive, but other two dogs were died shortly after initial treatment, because of severe aspiration pneumonia.

엔비로의 첨가.급여가 계사내 유해가스와 육계의 생산성에 미치는 영향

  • To investigate the influence of feeding Enviro on noxious gas of environmental controlled poultry house and performance of broiler chicks. Diets contained 21.5, 19.00 CP and 3,100, 3,200 kcal/kg ME, respectively for starting and finishing period. Enviro were supplemented with 0, 0.2, 0.4, 0.6 and 0, 0.1, 0.2, 0.3% in Expt 1and 2, respectively. CO$_2$and ammonia gas were periodically detected for finishing period in Expt 1. Weight gain, feed intake and feed conversion(FC) were measured for five weeks in Expt. 2. ND antibody titer were also measured after primary and booster vaccination at two and four weeks old. In Expt 1, chicks fed 0.2% Enviro showed 32.8ppm and 1719 ppm of ammonia gas, CO$_2$in poultry house and significantly decrease compared to that of control(P<0.05). Weight gain of chicks fed 0.1 and 0.2% Enviro was significantly increased for starting period(P<0.05) but was not statistically different for finishing period. Feed intake was no significantly different of all treatments. Chicks showed lower FC in 0.3% Enviro treatment for starting period(P<0.05) than others, but was not for finishing period. ND AB titet of chicks fed Enviro was prone to be higher than control, but was not significantly different. The results of these experiments indicated that 0.1% Enviro supplement would be maximize the performance of broiler chicks.

Discrimination of Hantaviruses from the Tissues of Infected Hamsters to 5 Different Serotype Hantaviruses by Nested RT-PCR using Hantavirus Serotype Specific Primers (한타바이러스 혈청형 특이 Primer를 이용한 Nested RT-PCR 방법으로 5가지 혈청형 한타바이러스에 감염된 햄스터 조직에서 바이러스 검출)

  • We developed a sensitive, nested reverse transcription-polymerase chain reaction (RT-PCR) to detect Hantaan, Seoul, Belgrade, Puumala and Sin Nombre viruses in animal tissues. Total RNA was extracted from blood, lung or kidney samples of experimentally-infected hamsters by using the guanidine isothiocyanate buffer-acid phenol-chloroform method. Genus-reactive outer primers were derived from the consensus region of the G1 gene sequences of several hantaviruses. Serotype-specific primers were selected within the region amplified by the outer primers. To examine the sensitivity and specificity of the test, we diluted known quantities of Hantaan, Seoul, Belgrade, Puumala and Sin Nombre viruses in human or hamster immune sera before performing the nested RT-PCR. We could detect as little as 1 pfu of virus, even in the presence of high-titer neutralizing antibodies, and the serotype-specific primers amplified only homologous serotype viruses. RT-PCR with these primers demonstrated virus in the blood of experimentally-infected hamsters as early as four days to as late as 30 days after infection. A comparison of a standard immunofluorescent antibody screening test (IFAT) to nested RT-PCR with RNA extracted from lung or kidney tissues of the hamsters, demonstrated that RT-PCR to be more sensitive for identifying viruses in these tissues.

Survey on antibody against bovine herpesvirus 1 (BoHV-1) in cattle in Korea

  • This study was performed in Korea to get serological information for bovine herpesvirus 1 (BoHV-1), most commonly found in cattle. Antibodies against BoHV-1 were examined by targeting infectious bovine rhinotracheitis (IBR) in unvaccinated and vaccinated cattle, using viral neutralization (VN) test. In 2013, among 261 sera collected from IBR-unvaccinated herds, 7 sera (2.7%) were found seropositive and their VN titers were ranging from 1:4 to 1:32. Among 315 sera collected from IBR-vaccinated herds in large capacity farms, 303 sera (96.2%) were found to be seropositive for BoHV-1 and their VN titers were in the range of 1:4 to 1:2048. It was found that the IBR-vaccinated herds had higher levels of VN titer than IBR-unvaccinated herds. The results indicated that it may be due to heavy vaccination in vaccinated herds and no or a little infection in unvaccinated herds. At the end of the study it was concluded that although the seropositivity in IBR-unvaccinated herds was low, the monitoring of IBR should be continuously practiced to control and prevent the disease because of exportation of living cattle causing its nationwide outbreaks.
