Effects of β-Glucan Supplementation on Lymphocyte Proliferation, Macrophage Chemotaxis and Specific Immune Responses in Broilers

  • Immunomodulatory feed additives might offer alternatives to antimicrobial growth promoters in poultry production. This experiment was carried out to test the effect of $\beta$-glucan supplementation on the growth performance and immune response in broilers. Total of 160 day-old broilers were randomly assigned to 4 treatment groups fed corn-soybean diets containing 0, 0.012, 0.025 or 0.05% of $\beta$-glucan supplement in a 6 week feeding experiment. Growth performance, antibody titer against New Castle vaccine, lymphocyte blastogensis, and peritoneal macrophage chemotaxis activity of broilers were evaluated. Results showed that there were no significant differences in weight gain and feed efficiency among the treatments, and no differences in antibody titer was observed. Supplementation of $\beta$-glucan did not elevate the lymphocyte blastogensis among treatments, following stimulation with different mitogens. However, supplementation with 0.025 and 0.05% $\beta$-glucan enhanced the macrophage chemotaxis activity of broilers. These results suggest that $\beta$-glucan may enhance some cell-mediated immune responses of chickens by modulate macrophages ability.

만손주혈흡충 충란항원 연속주입에 의한 면역학적 반응 (Immunological Responses by Soluble Egg Antigen of Schistosomu munsoni in Mice)

  • 만손주혈흠충 충란의 soluble eff antigen (SEA)을 mini-pump를 이용하여 마우스, 복강내에 지속적으로 주입시키면서 말초혈액내의 호산구수 변동, delayed hypersensitivity인 ear swelling test, 간조직병변, 효소 표식면역법을 이용한 혈청내 항체가를 경시적으로 관찰한 결과 다음 성적을 얻었다. 1. 말초혈액 호산구수는 SEA투여후 2~3주후에 최고에 달하였고 그후 서서히 감소하였다. 2. Ear swelling test 실시에서 SEA투여 2주후에 종창이 최고에 달하고 그후 서서히 감소하였다. 3. 간조직병변은 SEA투여후 4주에 육아종형성을 관찰하였고 그후 조직손상이 복귀하였다. 4. 현청에서의 항체가는 SEA투여후 4주후에 양성대조군보다 높았으며 시간이 경과함에 따라 항채가가 증가하였다.

CFC-101(녹농균 백신)의 능동 및 수동면역 효과 (Active and Passive Protective Effect of CFC-101 (Pseudomonas Vaccine) in Mice)

  • The treatment of pseudomonal infection is a perplexed problem because of its modest susceptibility to most of the major antibiotics. A novel Pseudomonas vaccine(CFC-101) was prepared from the outer membrane protein fractions of several Pseudomonas strains. In this study, we examined CFC-101's effectiveness in both active and passive immunization models. CFC-101 in mice at 0.2 mg/kg, i.p., given three times at two-day intervals, completely prevented the death caused by Pseudomonas aeruginosa. Antibody titer, in accordance with the protective effect in this active immunization, was elevated to its peak level following three consecutive administrations of CFC-101. Thereafter, antibody titer stayed at a constantly high level. Each outer membrane protein fraction from the four CFC-101 producers, exhibited good cross-protective effects in mouse infection models against different Fisher types of P. aeruginosa. In the passive immunization model, 21~336 $\mu\textrm{g}$/kg of anti-rabbit IgG to CFC-101, when mice being infected with a challenge strain, prevented the Pseudomonhas-induced death up to 60%. Therefore, the preventive effect of CFC-101 was verified in both the active and passive immunization models.

한국 소아에서 Haemophilus influenzae type b (Hib) 기초 예방 접종 후 항체 지속과 추가 접종에 대한 반응 (Antibody persistence after Haemophilus influenzae type b (Hib) primary vaccination and response to boosters in Korean children)

  • 목 적 : 우리나라에서 Hib 백신을 기본 예방접종에 포함시키는 것을 고려할 때 우리나라의 Hib 질환의 역학, 우리나라 소아의 Hib 백신에 대한 면역 반응에 대한 연구가 있어야 한다. 본 연구는 우리나라 소아에서 면역원성에 근거한 추가접종의 필요성 여부를 확인하고자 하였다. 방 법 : 2006년 6월부터 2006년 12월까지 이화여자대학교 동대문 병원, 강남 차병원, 관동대 명지병원에 내원하여 검사를 위해 혈액 채취의 기회가 있었던 12-23개월 사이의 소아 144명을 대상으로 하였다. 분리된 혈청으로 항 PRP 항체가를 효소면역법으로 측정하였고, 혈청 살균 능력 측정을 시행하였다. 결 과 : 추가접종까지 완료한 군의 항 PRP 항체의 기하평균은 기초접종을 완료한 군이나 접종하지 않은 군의 기하평균보다 높았다(P<0.05). 항 PRP 항체가가 $1.0{\mu}g/mL$ 이상인 비율은 추가접종까지 완료한 군은 96.5%로 다른 세 군에 비해, 기초접종을 완료한 군은 68.6%로 접종하지 않은 군의 30.0%에 비해 양성률이 높았다(P<0.05). 평균 살균 역가는 추가접종까지 완료한 군이 11,205로 기초접종을 완료한 군의 3,946보다 높았다(P<0.05). 항 PRP 항체가와 살균 역가는 양의 상관관계에 있었다(R=0.60). 결 론 : 우리나라 소아는 Hib 백신의 기초 접종 후 12-23개월의 나이에 많은 경우에서 방어 농도 이상의 항체가를 여전히 가지고 있었다. 추가접종에 대한 면역 기억 반응도 좋아 추가 접종 받은 소아의 항체가는 받지 않은 소아에 비해 의미 있게 높았다. 우리나라에 Hib 백신의 도입과 접종 횟수를 결정하기 위해서 지속적인 연구가 필요하다.

가토화·계태화 우역 생백신의 유효기간 설정을 위한 안정성 및 효능 평가 (Evaluation of stability and potency of live attenuated rinderpest vaccine of lapinized-avianized tissue culture strain origin for the establishment of expiration period)

  • In this study, we evaluated the stability and potency of live attenuated rinderpest vaccines of lapinized-avianized tissue culture strain origin, which had been produced annually from 2005 to 2008. When immune responses to the vaccines were evaluated using two Holstein calves weighing 100~150 kg, neutralizing antibody titer of 1 : 16 was induced at 21 days post vaccination. When calves were also inoculated with vaccines lots that had been stored for 39 months at ${4^{\circ}C}$, same level of antibody titer was observed. Using the virus titer test, we found that all batches of the vaccine that had been kept for 3, 10, 15, 22, 27, 34, 39, and 45 months showed no significant loss of titers, and fulfilled the requirement necessary ($\geq$ 3 $logTCID_50$) to be used as the national rinderpest vaccine reserve in Korea. In this study, we demonstrated that stability and potency of the rinderpest vaccines were maintained over three years when kept at ${4^{\circ}C}$ storage. This indicates that it maybe feasible to extend the expiration period of this vaccine from one year to three years.

오골계의 뉴캣슬바이러스 면역원성 조사 (Studies on the immunization against Newcastle disease virus in Ogol chicken)

  • This study was conducted to determine whether the vaccination programs for the control of Newcastle disease(ND) would affect the immune status of Ogol chicken in Yeonsan, Chungnam province. The results obstained are summerized as followes ; 1. Maternal antibody titers level of Ogol chickes was 2 $log_2$. 2. The efficacy of the Newcastle spray and drinking water vaccine was evaluated by the antibody responses and the protection rates(over 90% ) after challenge with a virulent Newcastle disease vims at 14 days. 3. Spray vaccine caused on visible side reaction like respiratory symptoms, but it is declined of the 4~5 days.

돼지의 유행성폐렴 원인균(Mycoplasma hyopneumoniae)에 대한 항체가 분포도 조사 (Studies on Enzyme-Linked Immunosorbent Assay(ELISA) for Detection of antibody to Mycoplasma hyopneumoniae)

  • Mycoplasmal pneumonia of swine(MPS) cause by Mycoplasma hyopneumoniae has been recognized as a serious impediment to swine production due to chronic respiratory disorder which result in the weight loss and decreased feed conversion. The disease causes a great economic losses in pig industry by characterizing with high morbidity, low mortality, growth retardation and low feed efficiency. The present study was conducted to investigate the titers of antibody against M hyopneumoniae from the regional and seasonal groups of the slaughtered pigs by enzyme-linked immunosorbent assay(ELISA). The result have shown that the average seropositive rate of M hyopneumoniae infection was 84.6% . The regional seropositive rate in Korea showed 87.4% in Kyonggj, 83.4n in Kangwon, 89.2% in Chungnam and 77.6% in Chungbuk area, respectively. Also the seasonal seropositive rate was appeared as 78.6% in spring,90.1% in summer, 76.9% in autumn and 83.8% in winter, respectively.

국내 야생따쥐(Crocidura lasiura) 의 Hantavirus 항체 보유실태 및 원인체 분리 (Hantavirus infection and isolation from wild shrews(Crocidura lasiura) in Korea)

  • Eleven shrews were caught from three areas of Korea. All of them were confirmed in the same species, Crocidura lasiura. All of sera from wild shrews were examined by indirect immunofluorescent test against Hantaan-related virus. The antibody to Hantaan-related virus was detected by 2 out of 11 shrews. Just 2 of 7 shrews from BG area were sero-positive for Hantaan-related virus antigen and none from other. All of sero-positive for Hantaan-related virus antigen belonged to male with antibody titer of 1:40 to 1:80. Two Hantaan-related viruses were isolated in vivo and in vitro.

Acquired myasthenia gravis in a domestic shorthair cat with cranial mediastinal mass

  • A 7-year-old castrated male domestic short-hair cat presented with anorexia, constipation, depression, and voice alteration. Physical and neurological examinations revealed hyperthermia ($40.5^{\circ}C$), ventroflexion of the neck, reduced responses to external stimuli, generalized muscle weakness, and exercise intolerance. Thoracic radiographs revealed the presence of a cranial mediastinal mass. The history, clinical signs, and other examination results were compatible with acquired myasthenia gravis (MG). Acetylcholine receptor (AChR) antibody titers were determined to confirm MG and the serum AChR antibody concentration was 1.24 nmol/L (reference interval, < 0.3 nmol/L). This is the first diagnosis of acquired MG in a cat in Korea.

충청북도 소 백혈병 항체 조사 연구 (Survey on Antibodies Against Bovine Leukemia Virus in Chung Buk Province)

  • A serological survey was carried out for the detection of antibody of Bovine Leukemia Virus (BLV) in nothern parts of Chung Buk area. The results were summarized as followed. 1. The overall positive rate was revealed as high as 15% with 48 positive cases out of 319 heads examined. 2. According to age, cattle of 4 to 7 ages showed relatively higher positive rate of 15% than other ages. 3. Seasonal differences of positive rate were not recognized. 4. BLV antibody titer of scales of cattles that from 5 to 15 heads farm were the highest. 5. With the result of blood test that of BLV positive cattle, the number of WBC was slightly Increased, but other records were normal.
