Purification and Properties of Osteopontin from Bovine Milk (우유로부터 Osteopontin의 분리.정제 및 특성에 관한 연구)

  • The purpose of this study is to observe purification and properties of osteopontin(OPN) from bovine milk. The purification of osteopontin from bovine milk was performed by using ion-exchange and hydrophobic chromatography. SDS-PAGE analysis revealed that the protein migrated at Mw. 60,000. NH2-terminal sequence analysis of the first seven amio acids revealed the protein to be identical to that previously reported for bovine OPN. 35-wk-old chickens, including 3 Single Comb White Leghorn (SCWL), were used to produce egg yolk antibody(IgY) against OPNas a antigen. However, the anti-OPN antibody activities determined by ELISA. Immunological assy of OPN in milk was performed using radial immunodiffusion test based on the standard curve of pure OPN. The radial precipitation lines of four different milk samples indicated that the concentrations of OPN in the milk samples were within the range of 31.7 to 39.7${\mu}g$/ml. On inhibition with OPN on precipitation of calcium phosphate, OPN was slightly higher than casein phosphopeptide(CPP) and poly-glutamic acid.

  • The purpose of this study was to investigate to potective effects against porcine epidemic diarrhea virus (PEDV) infection in piglets by administration of the PEDV antiserum orally at 2 hrs, 24hrs and 36hrs after birth. six piglets administered the antiserum were experimentally infected with PEDV at five-day-old. Control group were four piglets infected with PEDV only. Serum antibody titers against PEDV were examined by serum neutralization (SN) test, dectection for PEDV or PEDV antigen from feces and small intestines was tested by reverse transcription-polymerase chain reaction (RT-PCR) and indirect immunoflurescence (IFA). The results obtained were as follows; 1. The piglets administered the PEDV antiserum showed higher antibody titers than those of control group and sustained during the experimental period. 2. The detection rate of PEDV in feces and small intestines by RT-PCR were 26.2% and 16.7% in PEDV antiserum treated group and 48.1 % and 75.0% in control group, respectively. 3. The detection rate of PEDV antigen in the small intestine by IFA were 0% in PEDV antiserum treated group and 50.0% in control group, respectively. It was concluded that oral administration of antiserum against PEDV to piglets was effective in preventing PEDV infection.

  • Purpose: This study was performed to develop a lateral flow strip sensor for the detection of pathogenic Escherichia coli O157:H7 in various samples. Also, feasibility of using an image analysis method to improve the interpretation of the strip sensor was evaluated. Methods: The lateral flow strip sensor has been fabricated based on nitrocellulose lateral-flow membrane. Colloidal gold and E. coli O157:H7 antibodies were used as a tag and a receptor, respectively. Manually spotted E. coli O157:H7 antibody and anti-mouse antibody on nitrocellulose membrane were used as test and control dots, respectively. Feasibility of the lateral flow strip sensor to detect E. coli O157:H7 were evaluated with serially diluted E. coli O157:H7 cells in PBS or food samples. Test results of the lateral flow strip sensor were measured with an image analysis method. Results: The intensity of the test dot started to increase with higher concentration of the cells were introduced. The sensitivities of the sensor were both $10^4$ CFU/mL Escherichia coli O157:H7 spiked in PBS and in chicken meat extract, respectively. Conclusions: The lateral flow strip sensor and image analysis method could detect E. coli O157:H7 in 20 min, which is significantly quicker than conventional plate counting method.

The prevalence of swine influenza viral antigens and serum antibodiesin Piglets in Jeju (제주지역 돼지에서 Influenza 바이러스 항원 및 혈중 항체 조사)

  • Ninety pigs under the age of 120-day-old requested at the diagnostic laboratory of animal diseases in Cheju National University were evaluated for the prevalence of tissue antigen and serum antibody to swine influenza virus (SIV). For histopathologic examination there was sampled at the consolidated area in cranioventral or dorsocaudal lobes of lungs. Lung tissues from all pigs were tested for the antigen of SIV type A by immunohistochemistry (IHC). Sera from 56 pigs were used for the antibody detection to SIV type A (subtype H1N1 and H3N2) by haemagglutinin inhibition test. Pneumonic lesions were observed in 72 cases (80%) of 90 pigs. Broncho-interstitial or interstitial pneumonia were more prevalent than suppurative or fibrinous bronchopneumonia. According to HI test, 46.4% of the tested sera showed seropositive. Positive sera were consisted with 5.3% for SIV H1N1, 28.6% for SIV H3N2, and 12.5% for both subtype to be tested, respectively. SIV antigens were detected in 51 cases(56.6%) of 90 pigs. Most SIV antigens were presented in the epithelium of the bronchi and bronchiole. Necrotizing bronchitis or bronchiolitis were observed in 28(31.1%) cases of all inspected pigs. These results suggested that SIV might be an important role to induce swine pneumonia in Jeju. Also IHC was very useful for the detection of SIV in the lung.

Indirect Particle Agglutination Antibody Testing for Early Diagnosis of Mycoplasma pneumoniae pneumonia in Children (소아에서의 마이코플라스마 폐렴의 진단을 위한 항체 검사에 관한 연구)

  • Objectives: Outbreaks of pneumonia caused by Mycoplasma pneumoniae (MP) occur every 3-4 years in Korea, most recently in 2011. The aim of our study was to determine the optimal time to perform indirect particle agglutination antibody assays to improve early diagnosis of MP pneumonia in children. Methods: A database of 206 pediatric patients treated for pneumonia at the Hanyang University Hospital from June to October 2011 was analyzed retrospectively for demographic characteristics and laboratory test results. Results: Among the 206 patients treated for pneumonia during the study period, there were 160 children (mean age, 5.44 years) diagnosed with MP pneumonia, who were studied further. The mean age of these MP pneumonia patients was 5.44 years. Antibody titers increased with increasing time between symptom onset and the collection of serum collection: MP titers were <1:640 for sera collected after 5.44 days and titers ${\geq}1:640$ for those collected after 8.58 days; P<0.001). Antibody titers were considered positive when they reached ${\geq}1:640$. In 42 MP pneumonia patients in whom there was a four-fold or greater increase in titer between successive serum samples, the optimal cut-off time-point for distinguishing between the initial and second titer groups was 7.5 days after the onset of symptoms (sensitivity, 90.5%; specificity, 92.9%). Conclusions: Negative MP antibody titers earlier than 8 days after the onset of symptoms in children with pneumonia may require repeating to confirm the diagnosis. This finding could optimize diagnosis and result in better therapeutic outcomes of MP pneumonia in children.

Production of a Specific Yolk Antibody against Enterotoxigenic E. coli F41 Fimbrial Antigen (장독성대장균 F41 섬모항원에 대한 특이난황항체 생산)

  • Enteric colibacillosis has economically become an important disease of young animals as a result of increasing intensification of farrowing management. The objective of this experiment is to isolate fimbrial antigen from enterotoxigenic E. coli F41, to develop specific polyclonal IgY which can effectively neutralize or reduce the proliferation of pathogens in feed or living animal system, and to apply IgY technologies to animal industry. The results obtained were as follows: The molecular weight of the purified F41 antigen was 29,500 dalton on sodium dodecyl sulfate-polyacrylamide gels. Fimbrial antigen was confirmed by the western blot method. It was observed that after immunization the level of serum antibody titer of laying hen was shown in two weeks and gradually increased. The antibody titer in egg yolk appeared two weeks after it was shown in serum antibody. The titers of egg yolk antibody were gradually increased to the maximum level of 320,000 (antigen 50${\mu}g$/$m\ell$), 450,000 (antigen 200${\mu}g$/$m\ell$), and 320,000 (antigen 600${\mu}g$/$m\ell$). According to the results of specificity test by ELISA, the anti-F41 antibodies from chicken serum and egg yolk reacted only with ETEC F41 antigen. There was no cross reaction with other ETEC strains (K88, K99, and 987P). In vitro condition, as a result of antigen binding ability of yolk antibodies, bacterial concentration was rapidly decreased to $10^5$ CFU/$m\ell$ from $10^9$ CFU/$m\ell$ when 2${\sim}$4 mg/$m\ell$ of freeze dried WSF (water soluble fraction) was added.

Influence of cadmium exposure on the immune response of olive flounder, Paralichthys olivaceus (Cadmium이 넙치(Paralichthys olivaceus)의 면역 반응에 미치는 영향)

  • Olive flounder, Paralichthys olivaceus known as an one of the major aquacultured species in Korea were exposed to cadmium(Cd) with different protocols and analyzed the effects of exposure on the immune response. Antibody levels in sera of the group exposed to Cd(20ppb) by immersion method from 2 weeks before immuniztion with formalinised Edwardsiella tarda(E. tarda) KFE entigen to the end of experiment reached to peak level faster than that of the non-exposed group. After this peaking time the levels decreased much at a faster rate compared to the non-exposed group. This tendency was also appeared in the numbers of specific antibody secreting cells(SASC) analyzed with the enzyme-linked immunospot (ELISPOT)-assay technique in the splenocytes of the experimental groups exposed to Cd with different ways. Interestingly, the group exposed to Cd for 2 weeks before immunization also showed increased numbers of SASC unlikely the antibody production and suggested a more critical influence of cadmium exposure in early stage of immune reaction. Artificial infection with live E. tarda KFE induces 100% mortality in the flounder exposed to cadmium throughout the experimental period from two weeks before the immunization. It may imply that some other factors related to specific immunity are involving in the defence system of flounder exposed to Cd. Taked together. Cd exposure may induce temporaily stimulatory or indhibitory effects on the immune reaction, but suppress the physiological systems for the resistant against the infective agents with other toxic effects.

Effects of Boosting Methods of H-Y Antigen on Production Rate and Titer of H-Y Antiserum and Studies on Sexing of Rabbit Embryos by H-Y Antiserum (H-Y 항원의 Boosting 방법이 H-Y 항혈청 생산율과 역가에 미치는 효과와 H-Y 항혈청에 의한 토끼 수정란의 성판별에 관한 연구)

  • The present study was carried out to sex effectively mouse and rabbit embryos using rat H-Y antiserum and to improve the rate of rat producing H-Y antiserum with H-Y antibody. The H-Y antiserum was prepared from inbred SD strain female rat immunized by intrasplenic injection and subsequent intraperitioneal booster of testis cell of syngenic male newborn rat. the titer of antiserum was identified by in vitro cytotoxicity test of mouse embryos. The rabbit embryos exposed to the H-Y antiserum was classified to the developed (H-Y negative) or delayed (H-Y positive) group. The H-Y negative rabbit embryos were transferred to recipients and sex of offspring was examined. 1. When mouse embryos were exposed to the rat H-Y antisera, the ratio of embryos developed vs delayed was various. The H-Y antisera where the ratio of embryos developed vs delayed showed the range of 40~60% were recognized as having titer of H-Y antibody. 2. When the subsequent intraperitioneal boosters were followed after priming of intrasplenic injection of H-Y antigen, the rate of rat producting the H-Y antiserum with H-U antibody was 13, 27, 70 and 73% in control, 1st B, 2nd B and 3rd B, respectively. The rate in 2nd B and 3rd B was significantly(P<0.05) higher than that in control and 1st B. 3. When the rabbit morulae were exposed to the rat H-Y antiserum with H-Y antibody, the ratio of morulae developed versus delayed was 42:58% and it was close to the natural sex ratio 50:50%. It was confirmed that the rat H-Y antiserum was cross reactive to the rabbit morulae. 4. When the H-Y negative rabbit embryos were transferred to the recipients, the pregnancy rate was 50% and 83% of the newborns were females. In conclusion, the rat H-Y antiserum with high titer of H-Y antibody was able to be obtained from the female rat immunized by the intrasplenic injection followed by the second intrapent oneal booster of testis cells at a week interval. When the rabbit embryos negative to the rat H-Y antiserum were transferred, 83% of the newborns were females.

  • The authors investigated the incidence of antithyroglobulin antibodies and antibodies and antimicrosomal antibodies measured by tanned red cell hemagglutination method in subjects suffering from various thyroid disorders. 1) In 15 normal patients, neither suffering from any thyroid diseases nor from any other autoimmune disorders, the anti thyroglobulin antibodies were all negative, but the antimicrosomal antibody was positive only in one patient (6.7%). 2) The antithyroglobulin antibodies were positive in 31.5% (34 patients) of 108 patients with various thyroid diseases, and the antimicrosomal antibodies were positive in 37.0% (40 patients). 3) of the 25 patients with Graves' diseases, 7 patients (28.0%) showed positive for the antithyroglobulin antibodies, and 9(36.0%) for the antimicrosomal antibodies. There was no definite differences in clinical and thyroid functions between the groups with positive and negative results. 4) Both antibodies were positive in 16(88.9%) and 17(94.4%) patients respectively among 18 patients with Hashimoto's thyroiditis, all of them were diagnosed histologically. 5) Three out of 35 patients with thyroid adenoma showed positive antibodies, and 3 of 16 patients with thyroid carcinoma revealed positive antibodies. 6) TRCH antibodies demonstrated negative results in 2 patients with subacute thyroiditis, but positive in one patient with idiopathic primary myxedema. 7) The number of patients with high titers ($>1:80^2$) was 16 for antithyroglobulin antibody and 62.5%(10 patients) of which was Hashimoto's thyroiditis. Thirteen(65.0) of 20 patients with high titers($>1:80^2$) for antimicrosomal antibody was Hashimoto's thyroiditis. TRCH test is a simple, sensitive method, and has high reliablity and reproducibility. The incidences and titers of antihyroglobulin antibody and antimicrosomal antibody are especially high in Hashimoto's thyroiditis.
